This study investigated the high-efficient process for bioethanol from barley by various condition. First, higher concentrations of ethanol could be produced without loss of yield by using reducing water consumption. This is because it could prevent to increase viscosity despite reducing water consumption. Second, the ethanol yield could be improved by using reducing particle size of biomass (increase of enzyme reactive surface). Third, The addition of protease could have a considerable effect on yield of fermentation, which provides nutrients to the yeast. This results showed that bioethanol production would provide efficient ethanol production and lower production costs.
알바비스커스 잎으로부터 세포외 지질을 대량 생산하는 효모를 분리하고 동정한 결과 Rhodotorula graminis SW 214로 확인하였으며, $25^{\circ}C$에서 8일간 진탕배양시 Rhodotorula graminis SW 214 세포의 지질 생산량은 7.31g/l였다. 배지내의 질소원과 탄소원의 농도는 세포의 지질 생산에 큰 영향을 주었는데 배지내의 질소원이 효모 성장에 거의 이용되고 탄소원이 아직 남아 있을때 지질 생산이 활발히 이루어졌다. 배지내에 질소원 농도가 높은 동안에는 지질 생산이 더디게 이루어졌다. Rhodotorula graminis SW 214 세포의 지질 생산량의 최적조건으로는 gluaminis SW 214 세포의 지질 생산량의 최적조건으로는 glucose 8%, yeast extract 2.5g/l, $KH_{2}PO_{4}\;1g/l,\;MgSO_{4}{\cdot}7H_{2}O\;0.2g/l$, pH6에서 8일간 배양할 때임을 알 수 있었고 효모건조 균체량은 8.05g/l, 지방 생산량은 8.89g/l였다. Rhodotorula graminis SW 214 세포의 지질의 TLC에 의한 분별 정량 결과는 triglyceride 75.16%, phospholipid 10.77%, free fatty acid 5.88, esterified sterol 4.94%, free sterol 3.25%였다. Rhodotorula graminis SW 214 세포의 지방의 주된 지방산 조성은 3-hydroxypentadecanate 37.90%, 3-hydroxypentadecanate 20.80%, trans-9-octadecanate 11.53%, cis-9-headecanate 10.93%, 15-methylhexadecanate 9.82%, 18-methylnonadecanate 4.63%, octadecanate 2.64%, 3-hydroxytridecanate 1.75%였다.
The objective of this study was to optimize industrial-grade media for improving the biomass production of Weissella cibaria JW15 (JW15) using a statistical approach. Eleven variables comprising three carbon sources (glucose, fructose, and sucrose), three nitrogen sources (protease peptone, yeast extract, and soy peptone), and five mineral sources (K2HPO4, potassium citrate, ⳑ-cysteine phosphate, MgSO4, and MnSO4) were screened by using the Plackett-Burman design. Consequently, glucose, sucrose, and soy peptone were used as significant variables in response surface methodology (RSM). The composition of the optimal medium (OM) was 22.35 g/l glucose, 15.57 g/l sucrose, and 10.05 g/l soy peptone, 2.0 g/l K2HPO4, 5.0 g/l sodium acetate, 0.1 g/l MgSO4·7H2O, 0.05 g/l MnSO4·H2O, and 1.0 g/l Tween 80. The OM significantly improved the biomass production of JW15 over an established commercial medium (MRS). After fermenting OM, the dry cell weight of JW15 was 4.89 g/l, which was comparable to the predicted value (4.77 g/l), and 1.67 times higher than that of the MRS medium (3.02 g/l). Correspondingly, JW15 showed a rapid and increased production of lactic and acetic acid in the OM. To perform a scale-up validation, batch fermentation was executed in a 5-l bioreactor at 37℃ with or without a pH control at 6.0 ± 0.1. The biomass production of JW15 significantly improved (1.98 times higher) under the pH control, and the cost of OM was reduced by two-thirds compared to that in the MRS medium. In conclusion, OM may be utilized for mass producing JW15 for industrial use.
This work was aimed at utilizing rice bran as a substrate for $\beta$-carotene production by Rhodotorula glutinis DM 28 under optimized conditions of solid-state fermentation. The biomass and $\beta$-carotene content of Rhodotorula glutinis DM 28 grown on rice bran as a sole substrate under solid-state fermentation were 54 g/kg rice bran and 1.65 mg/kg rice bran, respectively. Its biomass and $\beta$-carotene content, however, could be improved by 60% and 30%, respectively, using the Central Composite Design for the optimization of its cultivation conditions. The optimized conditions obtained were a pH of 5, a moisture content of 70% (w/w), and a carbon-to-nitrogen ratio of 4. Under these conditions, rice bran containing R. glutinis DM 28 had nutritional values of $\beta$-carotene, protein, and fat higher than those of rice bran alone. Yeast-grown rice bran could be suitable, therefore, to use as a $\beta$-carotene-enriched supplement in animal feeds.
Among various pretreatment processes for bioethanol production, extrusion pretreatment, one of cheap and simple process was investigated to efficiently produce fermentable sugars from micro alga, Chlorella sp. The biomass was pretreated in a single screw extruder at five different barrel temperatures of 45, 50, 55, 60 and $65^{\circ}C$, respectively with five screw rotation speed of 10, 50, 100, 150 and 200 rpm. The pretreated biomass was reacted with two different hydrolyzing enzymes of cellulase and amyloglucosidase since the biomass contained different types of carbohydrates, compared to cellulose of agricultural by-products such wheat and corn stovers, etc. In general, higher glucose conversion yield was obtained as 13.24 (%, w/w) at $55^{\circ}C$ of barrel temperature and 100 rpm of screw speed conditions. In treating 5 FPU/glucan of cellulase and 150 Unit/mL of amyloglucosidase, ca. 64% of cellulose and 40% of polysaccharides in the micro alga were converted into glucose, which was higher yields than those from other reported data without applying an extrusion process. 84% of the fermentable sugars obtained from the hyrolyzing processes were fermented into ethanol in considering 50% of theoretical maximum fermentation yield of the yeast. These results implied that high speed extrusion could be suitable as a pretreatment process for the production of bioethanol from Chlorella sp.
Lipase (triacylglycerol lipase, EC 3.1.1.3) is an enzyme capable of hydrolyzing triacylglycerol, to produce fatty acids and glycerol and reverse the reaction of triacylglycerol synthesis from fatty acids and glycerol through transesterification. Applications of lipase are quite widespread in the industrial sector, including in the detergent, paper, dairy, and food industries, as well as for biodiesel synthesis. Lipases by yeasts have attracted industrial attention because of their fast production times and high stability. In a previous study, a lipase-producing yeast isolate was identified as Zygosaccharomyces mellis SG1.2 and had a productivity of 24.56 U/mg of biomass. This productivity value has the potential to be a new source of lipase, besides Yarrowia lypolitica which has been known as a lipase producer with a productivity of 0.758 U/mg. Lipase production by Z. mellis SG1.2 needs to be increased by optimizing the production medium. The aims of this study were to determine the significant component of the medium for lipase production and methods to increase lipase production using the optimum medium. The two methods used for the statistical optimization of production medium were Taguchi and RSM (Response Surface Methodology). The data obtained were analyzed using Minitab 18 and SPSS 23 software. The most significant factors which affected lipase productivity were olive oil and peptones. The optimum medium composition consisted of 1.02% olive oil, 2.19% peptone, 0.05% MgSO4·7H2O, 0.05% KCl, and 0.2% K2HPO4. The optimum medium was able to increase the lipase productivity of Z. mellis SG1.2 to 1.8-fold times the productivity before optimization.
The distribution of methanol-assimilating yeasts on three different sources (elm bark, soil and fresh-water mud) and the growth conditions of a new strain of Candidaboidinii (SIO) wereexamines. From 150 samples, 91 methanol yeasts were isolated through enrichment culture ; they were identified as 77 strains of Candida boidinii including four new strains, 5 isolates of Torulopsis pinus, 3 strains of Hansenula polymorpha and one sstrain of Pichia pastoris respectively. The comparison of these yeasts with three sources indicated that decaying bark of elm tree other two, and that Gandida boidinii was most frequently distributed in all three sources. Four new strains of Candida boidinii were freshly isolated and their taxonomical properties were discussed. Of them, SIO strain was selected and characterized for its growth on methanol. This yeast could grow well on less than 1%(v/v) methanol. However, its growth was inhibited at 10% methanol. The cell yield was 3.1g (dry weight) per 1000ml of mineral mediurr, containing 1%(v/v) methanol as well as 01.% yeast extract as additive. The concentration of 0.1% yeast extract appears to be effective for the biomass production. Optimum conditions for growth on methanol was found to be : $28^{\circ}C,\;NH_4^+$ as nitrogen sources, thiamine as vitamin, and pH 4.5 to 6.0. The cell composition was as follows : crude protein and nucleic acids were 54% and 7% respectively. The amino acids were also described.

Yeasts that are present in marine environments have evolved to survive hostile environments that are characterized by high exogenous salt content, high concentrations of inhibitory compounds, and low soluble carbon and nitrogen levels. Therefore, yeasts isolated from marine environments could have interesting characteristics for industrial applications. However, the application of marine yeast in research or industry is currently very limited owing to the lack of a suitable isolation method. Current methods for isolation suffer from fungal interference and/or low number of yeast isolates. In this paper, an efficient and non-laborious isolation method has been developed and successfully isolated large numbers of yeasts without bacterial or fungal growth. The new method includes a three-cycle enrichment step followed by an isolation step and a confirmation step. Using this method, 116 marine yeast strains were isolated from 14 marine samples collected in the UK, Egypt, and the USA. These strains were further evaluated for the utilization of fermentable sugars (glucose, xylose, mannitol, and galactose) using a phenotypic microarray assay. Seventeen strains with higher sugar utilization capacity than the reference terrestrial yeast Saccharomyces cerevisiae NCYC 2592 were selected for identification by sequencing of the ITS and D1/D2 domains. These strains belonged to six species: S. cerevisiae, Candida tropicalis, Candida viswanathii, Wickerhamomyces anomalus, Candida glabrata, and Pichia kudriavzevii. The ability of these strains for improved sugar utilization using seawater-based media was confirmed and, therefore, they could potentially be utilized in fermentations using marine biomass in seawater media, particularly for the production of bioethanol and other biochemical products.
백합나무를 원료로 바이오 에탄올을 생산하기 위하여 알칼리 가수분해 처리 후 잔재물을 상업용 혼합 셀룰라아제(Celluclast 1.5L과 Novozym 342)를 사용하여 효소당화 후, 발효하여 바이오 에탄올을 생산하였다. 알칼리 가수분해 후 51.1%의 목재 성분이 회수 되었으며, 이중 셀룰로오스가 82.2%, xylan이 17.6%와 리그닌 2.0%의 조성을 보였다. 백합나무의 알칼리 가수분해과정에서 셀룰로오스 96.9%, xylan 38.0%, 리그닌 5.7%가 잔류하였다. 알칼리 가수분해 잔류물을 상업용 혼합 셀룰라아제에 의한 효소 당화결과, 셀룰로오스의 glucose 전환율은 87.0%였으며 xylan의 xylose로의 전환율은 87.2%였다. 분해된 단당류를 발효효모를 사용하여 바이오 에탄올을 생산하였는데 Saccharomycess cerevisiae 균주는 대부분의 glucose를 발효에 사용하였고, 0.4-1.4%의 소량의 glucose만을 잔류 시킨데 대하여, xylose의 경우는 92.1-99.5%가 잔류하여 이 균주는 발효과정에서 xylose를 거의 사용하지 않았다. 24시간 발효에서 에탄올의 농도는 57.2 g/L수준이었지만 발효 균주에 의한 에탄올 소비로 인하여 48시간 및 72시간 발효에서 에탄올 농도가 각각 56.2 g/L와 54.3 g/L로 점차 감소하였다.
장수상황버섯(Phellinus baumii)의 균사체 성장 및 세포외 다당체(exopolysaccharides) 생산을 위한 액체배양의 최적 배지 및 배양조건에 관한 실험을 수행한 결과, 최적배양 온도와 초기 pH는 각각 3$0^{\circ}C$와 5.0으로 결정되었다. 탄소원의 경우, cellobiose와 maltose가 균사체 성장에는 양호하였으나 저조한 세포외 다당체 생산을 보였으며, 반면에 fructose 와 mannitol 은 상대적으로 높은 수율의 세포외 다당체 생산과 균사체 성장 또한 양호하였다. 최적 질소원으로는 yeast extract가 균사체 성장뿐만 아니라 세포외 다당체 생산에 효과적인 것으로 결정되었다. 최적 배지의 조성은 fructose 20 g/L, yeast extract 20 g/L 및 $CaCl_2$ 0.55 g/L이었으며, 최적 배양조건 하에서 5-L 교반 발효조를 운전한 결과, 최대 균사체 성장(17.43 g/L)과 최대 세포외 다당체 생산(3.6 g/L)을 얻을 수 있었다. 회수된 세포외 다당체는 glycoprotein이었으며 그 조성은 amino acid분석 결과 주로 arginine (14.1%)과 glycin (12.0%)이었으며, cabohydrate의 경우는 mannose (48.7%)와 arabinose (38.4%)이 주성분이었다.
