  • In order to investigate the pathogenic genes and genetic relationships of Y. pseudotuberculosis, we isolated 9 strains of Y. pseudotuberculosis from about 380 spring water sites in Seoul and carried out antibiotic susceptibility test, biological test and molecular typing. All isolated strains were distributed throughout the northeast area in Seoul (Mt. Bookhan, Mt. Soorak, Mt. Boolam and etc...).Antibiotic susceptibility test revealed that all the strains were susceptible to chloramphenicol, gentamicin, neomycin and amoxicillin/clavulanic acid, but were resistant to novobiocin and vancomycin. • In order to investigate the pathogenicity and development of differential identification technique in the Yersinia species and other entericbacteria, we isolated 5 strains of Y.pseudotuberculosis from spring water sites in Seoul. The biochemical characteristics of isolated strains revealed that indole, VP($25^{\circ}C$, $37^{\circ}C$), $H_2S$, phenylalanine, lysine, arginine, ornithine, gas from glucose, lactose, sucrose, sorbitol, oxidase and motility($37^{\circ}C$) were all negative and urease, glucose, mannitol, salicin, catalase and motility($25^{\circ}C$) were all positive. To detect the causative agent of pseudotuberculosis(Y.pseudotuberculosis), we carried out a study using a PCR with inv primers complementary to the pathogenic region and found that all strains were positive, this revealed that strains from spring waters were pathogenic. Also 16S rDNA for total 5 strains of Y. pseudotuberculosis were amplified and a stretch of approximately 1,450 nucleotides were sequenced and analyzed. The 16S rDNA nucleotide sequence homologies among Yersinia species ranged 97.5% to 100% and between Y.pseudotuberculosis and other entericbacteria they ranged 93.0% to 95.1%. The Phylogenetic tree generated from the sequence analysis of the 16S rDNA gene showed 3 coherent clusters that could be separated into Y.pseudotuberculsis strains, some Yersinia species strains and other entericbacteria strains.

  • Kikuchi disease(subacute necrotizing lymphadenitis), first reported by Kikuchi and Fujimoto in 1972, is a benign self-limiting illness characterized by fever, neutropenia and cervical lymphadenopathy and develops predominantly in young women, especially in Asia. The cause of Kikuchi disease is unknown, but postinfectious(virus or bacteria) hyperimmune reaction has been suggested. Few pediatric cases have been reported. We experienced a case of Kikuchi disease associated with Yersinia pseudotuberculosis infection in 12-year-old girl. After she was admitted with cervical lymphadenopathy and headache, followed by sustained fever with leukopenia. All symptoms and signs did not seem to be responded to antimicobial treatment. Open biopsy of cervical lymph node was performed and showed findings consistent with subacute necrotizing lymphadenitis. Serologic studies were all negative except for Y. pseudotuberculosis. Fever subsided and lymphadenopathy improved after administration of oral steroid for 1 week.

  • Yersinia pseudotuberculosis, a gram-negative coccobacillus, is a member of the enterobacteriaceae family, and the illness has fever, abdominal pain, diarrhea, mesenteric lymphadenitis, terminal ileitis, arthropathy, erythema nodosum. Specially in children, it is known as an important cause of acute renal failure due to acute tubulointerstitial nephritis. It has been recognized in a worldwide distribution. For the first time, the cases of acute renal failure caused by this organism were reported in Japan in 1980's. In Korea, the first case accompanying with ARF was reported in 1988. Since that, many cases have been reported in Seoul area, but no case in other places. So we report 2 cases of Y.P. infection accompanying with ARF in Pusan area, for the first time in other place than Seoul area.

  • Caseous lymphadenitis (CLA) is a chronic and contagious disease of sheep and goats caused by Corynebacterium (C.) pseudotuberculosis. A four-year-old female Saanen dairy goat was submitted to the Animal Disease Diagnostic Center at National Veterinary Research and Quarantine Service. The clinical signs of the goat were emaciation, abortion and quadriplegia. The multifocal nodules of lymph nodes were encapsulated and filled with whitish caseous contents on the cut surface. Histopathologically, lymph nodes displayed suppurative and necrotizing granulomas. Caseous necrosis was diffusely observed in the center of the lymph nodes. Gram positive bacilli were shown in the lesions. C. pseudotuberculosis was isolated and confirmed by the biochemical tests and PCR assay. Based on clinical signs, histopathological examination and bacterial isolation, we diagnosed this case as CLA. To our knowledge, this is the first report of CLA in a Saanen dairy goat in Korea.

  • This study was performed to investigate the distribution of Yersiniae and correlation between Yersiniae and indicator organism by time and area in spring water located in northern part of Seoul. Samples collected from 46 spring waters located in four mountains(Dobong, Bukhan, Surak, Bulam) were inspected to detect Yersiniae and indicator organisms. And also there were examined bioserological characteristics and resistance of ahtibiotics of the isolated Yersiniae. The result were as follows. 1. The isolation rate of Yersiniae was 22% in February and 20% in April. The isolated species were 6 strains of Y. enterocolitica, 6 strains of Y. aldova, 4 strains of Y. intermedia and 43 strains of Y. pseudotuberculosis. The serotype of Y. pseudotuberculosis isolated from was all O:5 and biotype of Y. enterocolitica isolated from was all O:3. 2. The Geometric mean of standard plate count, coliform, and psychrotrophilic bacteria were 3.4 CFU/ml, 1.2 MPN/100 ml and 33.0 CFU/ml in February and 3.1 CFU/ml, 1.5 MPN/100 ml and 20.5 CFU/ml in April respectively. There was no significant difference by time and area but the indicator organisms were correlated significantly with each other (p<0.05). 3. Because detection of Yersiniae was not statistically associated with indicator organism, Yersiniae can be detected in the spring water approved microbiologically (p<0.05). 4. The Yersiniae isolated were resistant to Ampicillin, Colistin, Carbenicillin and Coilstin. All isolaed Y. enterocolitica were resistant to Ampicillin (100%). In the case of Y. pseudotuberculosis, only 1 of 3 isolated was resistant to Colistin but susceptible to other antibiotics.

  • It was perfomed to investigate for Yersinia species from 2,841 spring waters in Seoul, from 1994 to 1998. Of them, Yersinia spp. were isolated 86 isolated (3.3%). Of 86, sixty two isolates(71.1%) were Yesinia enterocolitica, followed by Y. aldovae (11 strains), Y. pseudotuberculosis (5 strains), Y. frederiksenii (3 strains), unclassified Yersinia spp. (5 strains). Yersinia spp. were highest isolated from Nowon-Gu (22 samples asd Bukhan Mountain Park isolated(18 samples). We tested 1.186 samples for SPC and colifrom from 1996 to 1998. of these tests, the positive rate of coliform was 23.6%, SPC, 9.1%, and either coliform of SPC positive 27.1%. The positive rates of coliform and SPC were decreased 26.7%, 12.7% in 1996, 25.8%, 6.3% in 1997 and 18.1%, 7.6% in 1998 , rerspectively. Of Y. enterocolitica, 78% was resistant to ampicillin and carbenicillin. In the case of Y. aldovae, only 3 of 11 isolated were resistant to carbenicillin. Y. pseudotuberculosis were resistant to colistin. Also Y. frederiksenii to carbenicillin. There were many spring waters of Y. enterocolitical isolated from Nowon-Gu and Buk-han Mountain Park. So, it needs to clean the environment of those regions.

  • An epidemiological study was performed to evaluate the state of fish diseases between aquaculture fishes in the area of southern Kyeongnam, especially Tongyeong-si, Geoje-si, and Goseong-gun. Examination for a total of 91 cases was carried out for a variety of fishes, including Japanese flounder(Paralichthys olivaceus), rock fish (Sebastes schlegeli), sea bass (Lateolabrax japonicus), sea breams(Pagrus major), and red drum (Sciaenops ocellatus) from November, 1999 to October, 2001. The investigation resulted in exploring 155 infectious diseases, including 62 Vibriosis, 16 Edwasiella infection, 11 Streptococcosis, 7 Pseudotuberculosis, 15 Scuticociliatida infection, 20 Gill flukes, and 9 Trichodiniasis. When the infections were classified according to fish species, Japanese flounder was infected by 30 Vibrio spp., 16 Edwardsiella tarda, 6 Streptococcus spp., 12 Scuticociliatida infection. In case of sea bass, 10 Vibrio spp. was able to isolate out of 10 outbreaks, on the other hand, rock fish was dominantly infected by gill flukes since it was observed 12 out of a total of 20 cases. With reference to seasonal fluctuation, Vibrio spp. infection was continually observed through all seasons but Edwardsiella septicaemia, Streptococcosis and Pseudotuberculosis was mainly isolated in summer and autumn when the water temperature was raised high. Scuticociliatida infection was frequently occurred from late autumn to early spring but gill flukes were watched all the year regardless of temperature. As regards, multiple infection, 47 out of 91 cases was Identified to have involved in more than 2 pathogens, especially Vibrio spp. was highly mixed with other pathogens since 42 out of 47 cases was mixed with Vibrio spp.
