Postharvest Disease Control of Colletotrichum gloeosporioides and Penicillium expansum on Stored Apples by Gamma Irradiation Combined with Fumigation

  • To study the control of postharvest decay caused by Colletotrichum gloeosporioides and Penicillium expansum, gamma irradiation alone or in combination with fumigation was evaluated to extend the shelf life of apples in South Korea. An irradiation dose of 2.0 kGy resulted in the maximum inhibition of C. gloeosporioides and P. expansum spore germination. The gamma irradiation dose required to reduce the spore germination by 90% was 0.22 and 0.35 kGy for C. gloeosporioides and P. expansum, respectively. Microscopic observations revealed that when the fungal spores were treated with gamma irradiation (4.0 kGy), conidial germination was stopped completely resulting in no germ tube formation in C. gloeosporioides. Treatment with the eco-friendly fumigant ethanedinitrile had a greater antifungal activity against C. gloeosporioides and P. expansum in comparison with the non-treated control under in vitro conditions. The in vitro antifungal effects of the gamma irradiation and fumigation treatments allowed us to further study the effects of the combined treatments to control postharvest decay on stored apples. Interestingly, when apples were treated with gamma irradiation in combined with fumigation, disease inhibition increased more at lower (< 0.4 kGy) than at higher doses of irradiation, suggesting that combined treatments reduced the necessary irradiation dose in phytosanitary irradiation processing under storage conditions.

Protein Kinase C-$\beta$ Is Induced In Ionizing Irradiation Induced Pigmentation

  • Cutaneous hyperpigmentation is a well-known consequence of both acute and chronic X-irradiation, although the molecular mechanisms involved are not well understood. Recently, protein kinase C-$\beta$ (PKC-$\beta$) was shown to activate tyrosinase, a key and the rate-limiting enzyme in melanogenesis [1]. In this study, we have investigated its role in mediating ionizing radiation-induced pigmentation by exposing cultured human melanocytes to X-irradiation. Increased tyrosinase activity after the 4 Gys exposure was observed within 48 hrs and total melanin content doubled after 7 days. Interestingly, tyrosinase mRNA level was not affected by X-irradiation. However, there was a 2-3 fold increase in PKC-$\beta$ mRNA after 48 hours of irradiation, coinciding with the increase in tyrosinase activity. This induction was not due to non-specific heat generated during the irradiation because when melanocytes were incubated at 4$0^{\circ}C$, there was no induction of PKC-$\beta$ mRNA. Taken together, these data suggest that X-irradiation induces cutaneous hyperpigmentation, at least in part, by up-regulating the level of PKC-$\beta$.

Thoracic Re-irradiation for Locally Recurrent Lung Cancer

  • Background: Patients with recurrent or progressive lung cancer experience a significant symptom burden, negatively affecting quality of life and reducing life expectancy. Thoracic re-irradiation can be used for palliative treatment to relieve symptoms or as a curative treatment. Methods: Using patient charts, we identified and reviewed 28 cases that had received palliative thoracic re-irradiation for recurrent lung cancer. Results: Before re-irradiation, 32% of patients had stage III non-small cell lung cancer and six had small cell lung cancer. The median interval between treatments was 18.7 months. Median follow-up was 31.2 months from the initial radiotherapy and 5 months after re-irradiation. A better performance status before re-irradiation (<80 vs >80, p=0.09) and a lower overlap 90% isodose (<70 vs >70, p=0.09) showed trends toward improved survival. Grade 1-2 toxicity from re-irradiation was recorded in 12/28 patients, and no grade 3 or 4 acute toxicity was encountered. Conclusion: The role of palliative treatment in survival is not clear but it can provide symptomatic relief in patients, with no high grade toxicity. Further studies with greater patient numbers and longer follow-up times should facilitate determination of the role of this treatment in toxicity and effects on survival.

Low-Dose Gamma Irradiation as Means of Isolating Carotenoid-Hyperproducing Yeast Mutant

  • In order to isolate carotenoid-hyperproducing yeast, low-dose gamma irradiation was used as means of mutagenesis. Phaffia rhodozyma was treated by gamma irradiation of less than 10 kGy, which is considered to be a wholesome irradiation condition established by the Food and Drug Administration. Through repeated rounds of gamma irradiation and visual screening, mutant 3A4-8 was obtained. It produced a $3,824{\mu}g$ carotenoid/g yeast, 69% higher content than $2,265{\mu}g/g$ yeast of the unirradiated one. This result indicates that low-dose gamma irradiation could be used as means of mutagenesis to obtain carotenoid-hyperproducing strain of Phaffia rhodozyma, since only carotenoid-hyperproducing yeast survived gamma irradiation by scavenging oxygen radicals generated by radiolysis of water.

국내 기상 측정결과를 이용한 일사량 예측 방법 기초 연구 (A Basic Study to Predict Solar Insolation using Meteorological Observation Data in Korea)

  • To well design the solar energy system using solar energy, the correlation to calculate solar irradiation is basically needed. So, this study was performed to reveal the relationships between the solar irradiation and four meteorological observation data(dry bulb temperature, relative humidity, sunshine duration, and cloud cover) which are different from previous other researches. And then, we finally proposed the first order non-linear correlation from the measured solar irradiation using four meteorological observation data with MINITAB. To show the deviation of the solar irradiation between measured and calculated, this study compared using the daily total solar irradiance and the maximum peak value. From those results, the calculation error was estimated about maximum 25.4% for the daily total solar irradiance. The error of the solar irradiation between measured and calculated was made from the curve fitting error. So, solar irradiation prediction correlation with higher accuracy can be obtained using 2nd or higher order terms with four meteorological observation data.

  • This study was designed to investigate the effects of irradiation on the salivary ductal cells, especially on the intercalated ductal cells of the rat parotid glands. For this study, 36 Sprague-Dawley strain rats were irradiated on the head and neck region with absorbed dose of 15Gy by Co-60 teletherapy unit, Picker's model 4M60. The conditions irradiated were that field size, SSD, dose rate and depth were 12×5㎝m, 50㎝, 222 Gy/min. and 1㎝. respectively. The experimental animals were sacrificed 1, 2, 3, 6, 12 hours and 1, 3, 7 days after the irradiation and the changes of the irradiated intercalated duct cells of the parotid glands were examined under the light and electron microscope. The results were as follows: 1. Under the light and electron microscope, the nucleus, mitochondria and secretory granules showed severe changes in the early stage after irradiation and the most severe cellular de- generations were observed 2 hours after irradiation, but the repair processes began from 6 hours after irradiation. 2. Under the electron microscope, loss of the nuclear membranes, derrangement of the chromosomes, swelling and destruction of the secretory granules, and widening of the intercellular spaces were observed after irradiation. 3. Under the light microscope, atrophy and irregular proliferation of the ductal cells, cuboidal metaplasia, hyperchromatism, and the construction or obstruction of the lumen were observed after irradiation.

Effect of electron-beam irradiation on leakage current of AlGaN/GaN HEMTs on sapphire

  • This study examined the effect of electron-beam (E-beam) irradiation on the electrical properties of n-GaN, AlGaN and AlGN/GaN structures on sapphire substrates. E-beam irradiation resulted in a significant decrease in the gate leakage current of the n-GaN, AlGaN and HEMT structure from $4.0{\times}10^{-4}A$, $6.5{\times}10^{-5}A$, $2.7{\times}10^{-8}A$ to $7.7{\times}10^{-5}A$, $7.7{\times}10^{-6}A$, $4.7{\times}10^{-9}A$, respectively, at a drain voltage of -10V. Furthermore, we also investigated the effect of E-beam irradiation on the AlGaN surface in AlGaN/GaN heterostructure high electron mobility transistors(HEMTs). The results showed that the maximum drain current density of the AlGaN/GaN HEMTs with E-beam irradiation was greatly improved, when compared to that of the AlGaN/GaN HEMTs without E-beam irradiation. These results strongly suggest that E-beam irradiation is a promising method to reduce leakage current of AlGaN/GaN HEMTs on sapphire through the neutralization the trap.

자외선을 처리한 목재 플라스틱 복합재의 물리 및 역학적 성질 평가 (Evaluation on Physical and Mechanical Properties of Wood Plastic Composites Treated under Ultraviolet Irradiation)

  • In this study, we received each wood plastic composites (WPC) from three manufacturers. These WPCs were evaluated regarding their physical and mechanical properties of both before and after accelerated weathering by ultraviolet (UV) irradiation. The total time of exposure of the WPCs to UV irradiation was 1800 h. The water absorption, volumetric swelling and shrinkage of WPCs did not affected by UV irradiation. Among the mechanical properties, there was no significant differences in bending strength and screw withdrawal resistance of UV treated WPCs compared with those of reference WPCs. However, surface hardness of WPCs showed decrease under UV irradiation. Stereoscopic microscopy observation revealed deterioration of the surface layer polymer in all weathered WPCs by UV. Exposure of the WPCs to UV irradiation caused decomposition and disappearance of the polymer layer. From this result, we can estimate that damage of polymer by UV led to a decrease in the surface hardness of the WPCs. The wood flours retained original shape after accelerated weathering by UV irradiation.

Ultrastructural Changes in Cryptosporidium parvum Oocysts by Gamma Irradiation

  • Cryptosporidium parvum is known as one of the most highly resistant parasites to gamma irradiation. To morphologically have an insight on the radioresistance of this parasite, ultrastructural changes in C. parvum sporozoites were observed after gamma irradiation using various doses (1, 5, 10, and 25 kGy) following a range of post-irradiation incubation times (10 kGy for 6, 12, 24, 48, 72, and 96 hr). The ultrastructures of C. parvum oocysts changed remarkably after a 10-kGy irradiation. Nuclear membrane changes and degranulation of dense granules were observed with high doses over 10 kGy, and morphological changes in micronemes and rhoptries were observed with very high doses over 25 kGy. Oocyst walls were not affected by irradiation, whereas the internal structures of sporozoites degenerated completely 96 hr post-irradiation using a dose of 10 kGy. From this study, morphological evidence of radioresistance of C. parvum has been supplemented.

Control of Postharvest Bacterial Soft Rot by Gamma Irradiation and its Potential Modes of Action

  • Gamma irradiation was evaluated for its in vitro and in vivo antibacterial activity against a postharvest bacterial pathogen, Erwinia carotovora subsp. carotovora (Ecc). Gamma irradiation in a bacteria cell suspension resulted in a dramatic reduction of the viable counts as well as an increase in the amounts of DNA and protein released from the cells. Gamma irradiation showed complete inactivation of Ecc, especially at a dose of 0.6 kGy. In addition, scanning electron microscopy of irradiated cells revealed severe damage on the surface of most bacterial cells. Along with the morphological changes of cells by gamma irradiation, it also affected the membrane integrity in a dose-dependent manner. The mechanisms by which the gamma irradiation decreased the bacterial soft rot can be directly associated with the disruption of the cell membrane of the bacterial pathogen, along with DNA fragmentation, results in dose-dependent cell inactivation. These findings suggest that gamma irradiation has potential as an antibacterial approach to reduce the severity of the soft rot of paprika.
