The Effects of BunSimGiEumGami-Bang(Fenxinqiyinjiameifang) on Serotonin of P815 cell (분심기음가미방(分心氣飮加味方)의 항산화능과 serotonin 대사에 미치는 영향)

  • Objectives : This experiment was designed to investigate the effect of the BSGE hot water extract on serotonin biosynthesis of depression model. Methods : The cytotoxicity of the BSGE hot water extract was analyzed by MTT assay on P815 cell. The antioxidant activity was measured by DPPH free radical-scavenging activity and SOD activity on P815 cell. The quantity of 5-HT and expression of TPH-1, AAADC and MAOa mRNA was measured by of HPLC profle Analysis on P815 cell. Results : 1. The BSGE hot water extract increased DPPH free radical-scavenging activity and SOD activity on P815 cell. 2. The BSGE hot water extract increased significantly the quantity of 5-HT. 3. The BSGE hot water extract increased the expression of TPH-1 mRNA. Conclusions : This experiment shows that the BSGE hot water extract might be effective for the prevention and treatment of depression. Investigation into the clinical use of the BSGE hot water extract for depression is suggested for future research.

  • A new embryonic cell line (OFEC-17FEN) derived from olive flounder Paralichthys olivaceus was developed. OFEC-17FEN cells were subcultured for <30 passages over ~200 days. OFEC-17FEN cells had a doubling time of 114.34 h and modal diploid chromosome number was 48. The pluripotency genes POU5f1 and NANOG were expressed in OFEC-17FEN cells. However, the lack of several pluripotency-related genes expression indicates that OFEC-17FEN cells are not stem cells. OFEC-17FEN cells transfected with plasmid pEGFP-c1 exhibited a strong green fluorescent signal at 48 h after transfection. Accordingly, OFEC-17FEN cells may be useful for both basic research and biotechnological application.

  • The bioremediation is an economic technology to remove the organopollutants from soil. It is often found that the remediation could not remove the compounds below the levels determined by vigorous extractions as required by regulatory agencies. The reason for the reduced bioavailability with increasing time of aging has been accredited to the sequestration of the compounds in remote sites within or between soil particles. Then, the aging could be defined as the time-dependent sequestration. Partitioning and entrapment have been suggested as mechanism for aging. The rate and extent of the sequestration varies among dissimilar soils. The bioavailability of aged pollutants in soil could be measured by bioassays, mild solvent extraction, and soild-phase extractions. The sequestration could be affected by many factors including various soil properties, wetting and drying cycle, and the presence of cosolutes and NAPLs etc. The bioavailability and sequestration should be considered to determine the environmentally acceptable endpoint.

  • 토석류(Debris flow)는 산지사면이나 계곡 등에서 진흙과 돌덩어리 등을 포함하는 토석 그 자체 또는 토석과 물의 일체가 유체의 상으로 흘러내리는 흐름을 말하는 것으로 발생을 예측하기가 곤란하며, 하류에 도로가 존재하거나 인근에 민가가 있을 시 막대한 피해를 가져오는 자연현상이다. 또한 산지계곡에서 발생하는 토석류는 발생장소와 시기가 서로 떨어져 있어 연구를 진행하는데 어려움이 있다. 국내에서는 토석류피해의 저감을 위한 방법으로 주로 토석류 대책 사방댐의 설치를 선택하고 있다. 하지만 사방댐의 설치위치는 현재까지 모호한 결정기준에 의지하고 있어 이에 대한 개선이 필요한 실정이다. 본 연구에서는 3차원 정밀좌표를 손쉽게 취득할 수 있는 삼차원 광대역 레이저 스캐너를 이용하여 토석류 발생 가능성이 높은 지역의 지형자료를 취득하고 토석류를 해석할 수 있는 1차원 수치모형을 이용하여 토석류 유출량을 예측하였다. 또한 사방댐의 설치위치결정에 대한 참고자료로 활용하기 위해 사방댐의 토석류피해 저감효과를 설치위치에 따라 분석하였다. 모형의 적용결과 토석류 발생 저감을 위해 설치한 사방시설의 위치에 따른 토석류 저감효과를 비교 확인할 수 있었다.

Rate Effects of Swine Manure Fermented with Sawdust on Efficiency of Nitrogen Utilization of Silage Corn and Soil Fertility

  • This study was carried out to examine the effects of animal manure on efficiency of the nitrogen utilization of silage corn (Zea mays L.) and soil fertility. The experiment was conducted on the field plot at Gongiam, Kwangju, Kyunggi-Do for 3 years, from 1996 to 1998, and arranged in split-plot design with three replications. The main plots were two kinds of composts such as swine manure fermented with sawdust (SMFWS) and swine manure fermented without sawdust (SMF). Subplots were the nitrogen fertilization rate (0, 100, 200, 300 and 400kgNhalyear). The nitrogen (N) yield increased as the nitrogen fertilization rate increased up to a rate of 300 kg Nha, but decreased at rate of 400 kg Nlha. Nitrogen yield in SMF treatments was higher than that of SMFWS treatments. But there were no significant differences between SMFWS and SMF treatments. Organic matter (OM) content of the soils in SMFWS was higher than that of SMF, &d was not significantly different between SMFWS and SMF treatments. OM content increased with increasing the nitrogen fertilization rate. Total nitrogen (TN) content of the soils increased as the nitrogen fertilization rate increased. No difference of TN content was found between SMFWS and SMF treatments. (Key words : Swine manure, Compost, Corn, Sawdust, Nutritive value)

Growth Inhibition and Apoptosis Induction of Gastric Cancer Cells by Copper (II) Glycinate Complex

  • The in vitro cytotoxic effects of newly synthesized copper (II) glycinate complex were investigated in two gastric cancer cell lines of SNU484 and SNU638 cells. The complex inhibited the growth and decreased the viability of both gastric cancer cells in a dose-dependent manner. Gastric cancer tells treated with the complex exhibited the features of apoptosis, as demonstrated by fragmentation of chromosomal DNA, activation of caspase-3-like enzyme, and cleavage of poly[ADP-ribose] polymerase (PARP). With the treatment of copper (II) glycinate complex, the active form of caspase-3 was observed in SNU484 cells, but not in SNU638 cells, indicating that an alternative pathway of apoptosis might have been triggered in SNU638 cells. In conclusion, copper (II) glycinate complex induces apoptosis of SNU484 and SNU638 gastric cancer cells, and it is suggested that novel copper (II) glycinate complex is highly active against human gastric cancer cells.

  • To produce ergosterol Saccharomyces toke KBA No. 6 was used and various factors related to ergosterol accumulation were investigated. The most effective inorganic nitrogen source was ammonium chloride and 3.50 % of ergosterol was accumulated in the cell when the C/N ratio was 200/1. When Tween 80 and potassium nitrite were used simultaneously, ergosterol content and total amount of ergosterol were increased by 56 % and 45 %, respectively, compared to their control in which 0.2 % of Tween 80 and 0.1 % of potassium nitrite were used. Under the optimum conditions, ergosterol content increased from 1.73 % to 5.3 % and the total amount of ergosterol was increased from 65.2 mg/l to 135.15 mg/l.

  • An attitude control system using reaction thrusters for the upper stage of a launch vehicle is considered. The thruster configuration (position and direction) determines control system response, fuel consumption, effective torque and system fault tolerance. We propose a procedure for finding the optimal thruster configuration with desired control effectiveness over the range of selected torque commands. An optimization technique called Particle Swarm Optimization is used for the numerical experiments. The validity of the solution is checked through computer simulations.

  • Contrast to the 6-DOF nonlinear dynamic modeling of nonlinear tracking problem, 3-DOF point-mass modeling of flight mechanics is efficient and adequate for applying the trajectory optimization problem. There exist limitations to apply an optimal trajectory from point-mass modeling as a reference trajectory directly to conduct the nonlinear tracking control, In this paper, new matching trajectory optimization scheme is proposed to compensate those differences of mismatching. To verify performance of proposed method, full ascent three-dimensional flight trajectories are obtained by reflecting the real constraints of flight conditions and airship performance with and without jet stream condition. Then, they are compared with the optimal trajectories obtained from conventional method.

  • A Fourier Transform Spectrometer was used to study upper mesospheric thermodynamic by observing the hydroxyl(OH) emission. Rocket-born and satellited-born photometers place the peak emission near 87 km. The instrument was installed in February 1999 at King Sejong station ($62.22^{circ}S,301.25^{circ}E$), Antarctica and has been in routine operation since then. An intensive operational effort has resulted in a substantial data between April and June, 1999. A harmonic analysis was carried out to examine information on the tidal characteristics. The measured amplitudes of the 12-hour oscillation are in the range of 2.4-3.7 K, which are in resonable agreement with theoretical model outputs. The harmonic analysis also revealed 8-hour oscillation which is not expected from the traditional theoretical studies. In addition, the observed 8-hour oscillations are apparent and sometimes dominate the temperature variation in the upper mesosphere.
