도로공사에서 생애주기비용을 사용한 지급조정모델 개발에 관한 연구 (LCCA-embedded Monte Carlo Approach for Modeling Pay Adjustment at the State DOTs)

  • 미국에서는 품질 관리 문제를 해결할 수 있는 아이디어로 지급(Pay) Factor가 사용되어왔으며 현재는 생애주기비용까지 고려한 한층 진보된 지급조정모델 개발에 많은 관심을 가지고 있다. 하지만 이러한 지급조정모델절차의 개발은 품질특성변수 선택의 문제, 품질특성변수의 확률적 분포와 도로 공용성간의 관계 분석의 문제, 그리고 하나의 전체 지급조정모델 개발의 문제 등으로 미 교통부에서 어려움을 가지고 있다. 본 논문에서는 이러한 점을 극복하기 위한 방법론으로 생애주기비용분석을 고려한 몬테카를로 시뮬레이션 접근 방식이 사용되었다. 미 여러 교통부에서 적용 가능한 견본이 될 수 있도록 현 위스콘신 교통국에서 사용중인 도로관리 관련 시스템들에서 데이터를 축출하여 지급조정을 격정하기 위한 분석 절차를 제시하고 이를 근간으로 지급조정모델 결정 지원 시스템을 개발하였으며 민감도 분석을 실행하여 실제 데이터를 사용하여 개발된 지급조정모델의 적정성을 평가하였다. 본 논문에서 사용된 지급조정모델 개발 절차는 한층 정확성을 높인 도로공용성예측모델과 생애주기비용모델을 사용함으로써 실제 프로젝트에 사용 가능할 것으로 판단된다.

  • This study examined the effects of gender, parental support and treatment type on the treatment outcome of adolescent substance abusers. Outcome variable was the successful graduation (or drop) from an Intensive Adolescent Outpatient Program. Adolescents with their parents' support were treated in one of three treatment models (2-weeks Inpatient plus 6 week Intensive Croup-Oriented Outpatient, 8-weeks Intensive Croup-Oriented Outpatient, and 8-weeks Individual-Family Therapy) within a private hospital-affiliated treatment center by managed care practice allowing their own choice (non-random natural assignments). Several hypotheses were tested for main effects by the Log-Linear Analyses for a multi-dimensional contingency table with 440 adolescents (284 boys and 156 girls treated during 1992-l997) from middle-class families with private health insurances. The following results were found. Odds of graduating versus dropping out of the treatment program among : (1) girls were 1.7 times higher than those among boys; (2) adolescents with two-parent were 2.2 times higher than those among adolescents with one-parent ; (3) adolescents with Inpatient plus Outpatient was 1.7 times higher than that of those with Outpatient; (4) adolescents with Individual-Family Therapy was 2.3 times higher than that of those with Outpatient Model. There was no statistically significant outcome difference between the Individual-Family Therapy and the Inpatient plus Outpatient. Implications from the results were discussed. Suggestions were made to improve the treatment components in the areas of gender sensitivity, securing more parental support, alternatives for separation from peer group and integrating new peer groups, and flexibility for the unique needs of individual family. Also, some research questions for future studies were suggested.

Growth and Decay of Alpha Tracks in a Large Scale Cloud Chamber after Injection of Radon

  • The recognition of the natural background radiation is important not only for radiological education but also for the promotion of people's scientific view about radiation. We made a "room" on the web showing natural background radiation as part of a VRM (Virtual Radiation Museum). The "room" shows the video images of the tracks of charged particles from natural background radiation, alpha and beta ray track from known sources using a Large Scale Diffusion Cloud Chamber. The purpose of this study is to make clear the origin of a kind of track (named A-track) which is thick and easy to recognize with the length less than several cm in the cloud chamber, and to make numerical explanation of its counting rate. The study was carried out using a Large Scale Diffusion Cloud Chamber (Phywe, Germany) installed in the Niigata Science Museum. The Model RNC (Pylon Electronics, Canada) was used as Rn-222 source. Ra-226 activity in RNC was 111.6 Bq calibrated with NIST protocol. Rn-222 gas was injected into the cloud chamber. Continuous video recording with use of Digital Handycam (SONY, Japan) was carried out for 360 min. after injection of Rn-222 gas. The number of alpha-ray track (alpha track) in the video images was analyzed. The growth and decay curve of the total activity of Rn-222 and its alpha emitting progeny were calculated and compared with the count of the alpha tracks. As a result the alpha tracks formed by Rn-222 injection resemble A-Tracks. The relationship between A-track in the cloud chamber and atmospheric Rn is discussed.

  • 과파안정도의 문제중의 하나가 발전기에 장거리 송전선으로 무한의 bus가 연결되었을 때이다. Wisconsin 전력의 Point Beach발전소에 원자력turbine-발전기 #1 그리고 #2가 한 Unit 또는 두 Unit가 밀워키(Milwaukee)의 하나 또는 두 line과 분리되었을대 과파적 불안정 상태가 발생하였다. 이러한 조건하에서 전력계통안정장치(Power System Stabilizer)가 안정작동을 위하여 excitation 계통에 상용되었다. 전력계통안정장치(PSS)의 산정치를 등가 machine0infinits bus성능의 root-locus 분석과 computer 방법에 의한 분석으로 먼저 계측되었다. 이 방법은 전력계통안정장치의 디자인에 그래프식 접근을 허용하고, 디자인 제약들이 변화되어서 speed stabilizer가 계통댐핑(damping) 향상이 요구될때 종합적인 방법으로 생각되어 exciter-generator 계통의 간단한 모델을 세워 분석하였다.

  • 도시하수와 관련된 비점원 오염에 대한 관심은 더 나은 수질계획을 위한 새로운 도구의 발전을 이끌어 왔다. 이 논문은 도시의 수질연구를 위해 지리정보체계가 어떻게 응용되는지를 보여준다. 지리정보체계는 비점원 오염 모형화를 위한 토지이용자료를 관리하기 위해 이용되고, 또한 여러 가지 형태의 지리적 집합체내에서 오염물질을 평가하는데 이용된다. 경험적인 수질모형은 토지이용에 근본을 둔 오염물질 축적을 평가하기 위해 이용된다. 토지이용범위는 최근 사진의 판독을 통해서 구범위를 갱신함으로써 결정된다. 이러한 토지이용범위는 각각의 토지이용 다각형에 대한 오염물질 축적을 기록하는데 이용된다. 도시하수 유역과 소하수 유역의 범위를 결정하기 위해 하수지도를 수치화 시키고 해석한다. 하수구 소유역층에서 오염물질 축적을 중첩시킴으로써 주요하수의 출구에 대한 오염 축적물이 계산된다. 축적 정보를 기본으로 과다 오염 축적물의 임계지역이 어디에 위치하고 있으며, 오염 축적물을 통제하기 위한 최선의 관리기법의 효율성이 평가된다.

The Forkhead Gene fkhB is Necessary for Proper Development in Aspergillus nidulans

  • The forkhead domain genes are important for development and morphogenesis in fungi. Six forkhead genes fkhA-fkhF have been found in the genome of the model filamentous Ascomycete Aspergillus nidulans. To identify the fkh gene(s) associated with fungal development, we examined mRNA levels of these six genes and found that the level of fkhB and fkhD mRNA was significantly elevated during asexual development and in conidia. To investigate the roles of FkhB and FkhD, we generated fkhB and fkhD deletion mutants and complemented strains and investigated their phenotypes. The deletion of fkhB, but not fkhD, affected fungal growth and both sexual and asexual development. The fkhB deletion mutant exhibited decreased colony size with distinctly pigmented (reddish) asexual spores and a significantly lower number of conidia compared with these features in the wild type (WT), although the level of sterigmatocystin was unaffected by the absence of fkhB. Furthermore, the fkhB deletion mutant produced sexual fruiting bodies (cleistothecia) smaller than those of WT, implying that the fkhB gene is involved in both asexual and sexual development. In addition, fkhB deletion reduced fungal tolerance to heat stress and decreased trehalose accumulation in conidia. Overall, these results suggest that fkhB plays a key role in proper fungal growth, development, and conidial stress tolerance in A. nidulans.

흰쥐의 분리 폐장 관류 모델에서 Nitroglycerin의 폐장 보존 효과 (Protective Effect of Nitroglycerin on the Ischemia-Reperfusion Model of the Isolated Rat Lung)

  • 체내에서 발생하는 산화 질소는 허혈-재관류에 따른 폐혈관 저항을 감소시키고 혈관의 미세 투과도를 줄이고 세포손상을 방지하여 이식 후 폐장 기능의 보전에 도움이 된다고 알려져 있으나, 산화질소 자체의 세포독성으로 인해 재관류 동안 폐 부종을 오히려 증가시킬 수도 있다고 한다. 저자들은 산화 질소의 공여물질인 nitroglycerin을 투여한 분리 폐장 관류 모델을 이용하여 폐장 보존에 이점과 단점을 동시에 가지고 있는 산화질소가 허혈-재관류 과정에서 폐장의 기능에 미치는 영향을 알아보기 위하여 본 연구를 시행하였다. 대상 및 방법: Sprague-Dawley종의 수컷 흰쥐 35마리를 사용하였다. Nitroglycerin (NTG)군(n=18)은 NTG를 정맥 주사하고 University of Wisconsin용액에 혼합하여 폐장 관류를 시행하였고, 대조군(n=17)은 NTG 대신 같은 양의 생리 식염수를 사용하였다. 구득한 심폐블록을 1$0^{\circ}C$에서 24시간 동안 보관한 후, 분리 폐장 관류 모델에서 인체 혈액을 Krebs-Hensleit용액으로 희석하여 60분간 재관류하였다. 재관류하는 동안 기도내압과 페동맥압을 지속적으로 측정하였고, 관류후 30분과 60분에 혈액 내 산소와 이산화탄소 분압을 측정하였다. 재관류가 끝난 후 기관지폐포세척을 통해 폐포 내 단백함량을 측정하였으며, 중성구 침착 정도를 알기 위해 myeloperoxidase (MPO) 활성도를 측정하였다. 걸과: 두 군 간의 기도내압과 폐동맥압은 통계적으로 유의한 차이는 없었으나, NTG군에서 관류기간 중 폐동맥압이 상대적으로 낮게 유지되는 경향이 있었다. 이산화탄소 분압, 기관지폐포세척액 내의 단백질 함량, MPO의 활성도는 두 군에서 유의한 차이는 없었으나, 산소 분압은 NTG 군에서 통계적으로 유의하게 높게 나타났다(p<0.05). 전자현미경 검사에도 대조군에 비해 NTG군에서 폐포구조, 폐포상피세포, 모세혈관배열 등 미세구조의 손상이 적게 나타났다. 결론. 폐장 구득전 NTG의 투여는 폐장의 기능과 미세구조의 유지에 도움이 된다고 판단되며, 임상 폐이식에도 적용이 가능하리라 생각된다.

단순 작업성 심관류 모델에서의 신생돈 심장의 보존 후 백혈구-제거 혈액을 이용한 재관류가 심근 VCAM-1 발현 및 심기능에 미치는 영향 (Effect of reperfusion with leukocyte-depleted blood on the expression of myocardial vascular cell adhesion molecule-1 (VCAM-1) and myocardial function in isolated working heart perfusion model)

  • Background: Adhesion of leukocytes to myocardium or vascular endothelium has been known as an importation initial step in the ischemia-reperfusion injury which may affect the cardiac function. Therefore, leukocyte-depleted reperfusion may inhibit ischemia-reperfusion induced functional and ultrastructural deterioration. In this study, we quantified the time-dependent expression of the vascular cell adhesion molecule-1(VCMA-1) on piglet myocardium and demonstrated its relation to functional recovery using isolated piglet heart perfusion model. Material and Method: Neonatal(1 to 3 day old) piglet heart was harvested with 4$^{\circ}C$ University of Wisconsin solution (UWS) and presrved in the same solution for 12 hours. Ex vivo model of an isolated working neonatal piglet heart perfusion consisting of membrane oxygenator and roller-pump was used (Fig. 1). Hearts were grouped into leukocyte-non-depleted (group A, n=8) and leukocyte-depleted group(group B, n=8). In group B, hearts were reperfused with leukocyte-depleted blood using a leukocyte filter (Sepacell R, Asahi Medical, Japan). Segments of right atrium were taken before and after 1, 2, 3, and 4 hours of reperfusion for the evaluation of expression of VCAM-1. The intensity of immunohistochyemical satining of the VCAM-1 on the myocardium were graded semiquantitatively (0 to 4). For the evaluation of myocardial stroke work indices were calculated as well at the same time-points. Result: Mean expressins of VCAM-1 on the myocardium at 0, 1, 2, 3, adn 4 hours of reperfusion were 0.63, 1.44, 1.64, 2.65, and 3.34 in group A, while 0.56, 1.40, 1.50, 1.88 and 2.14 in group B (Fig. 3). Mean stroke work indices at 0.5, 1, 2, 3, and 4 hours after reperfusion were 1.35$\times$104, 1.32$\times$104, 1.14$\times$104, 0.81$\times$104, 0.68$\times$104 erg/gm in group A, while 1.40$\times$104, 1.43$\times$104, 1.43$\times$104, 1.28$\times$104, and 1.12$\times$104 erg/em in group B(Fig. 4). Conclusion : In this study, we demonstrated that leukocyte-depletion attenuated the expression of VCAM-1 during reperfusion and the time-dependent functional deterioration of the myocardium was well correlated with the degree of VCAM-1 expression.

한정된 O-D조사자료를 이용한 주 전체의 트럭교통예측방법 개발 (DEVELOPMENT OF STATEWIDE TRUCK TRAFFIC FORECASTING METHOD BY USING LIMITED O-D SURVEY DATA)

  • The objective of this research is to test the feasibility of developing a statewide truck traffic forecasting methodology for Wisconsin by using Origin-Destination surveys, traffic counts, classification counts, and other data that are routinely collected by the Wisconsin Department of Transportation (WisDOT). Development of a feasible model will permit estimation of future truck traffic for every major link in the network. This will provide the basis for improved estimation of future pavement deterioration. Pavement damage rises exponentially as axle weight increases, and trucks are responsible for most of the traffic-induced damage to pavement. Consequently, forecasts of truck traffic are critical to pavement management systems. The pavement Management Decision Supporting System (PMDSS) prepared by WisDOT in May 1990 combines pavement inventory and performance data with a knowledge base consisting of rules for evaluation, problem identification and rehabilitation recommendation. Without a r.easonable truck traffic forecasting methodology, PMDSS is not able to project pavement performance trends in order to make assessment and recommendations in the future years. However, none of WisDOT's existing forecasting methodologies has been designed specifically for predicting truck movements on a statewide highway network. For this research, the Origin-Destination survey data avaiiable from WisDOT, including two stateline areas, one county, and five cities, are analyzed and the zone-to'||'&'||'not;zone truck trip tables are developed. The resulting Origin-Destination Trip Length Frequency (00 TLF) distributions by trip type are applied to the Gravity Model (GM) for comparison with comparable TLFs from the GM. The gravity model is calibrated to obtain friction factor curves for the three trip types, Internal-Internal (I-I), Internal-External (I-E), and External-External (E-E). ~oth "macro-scale" calibration and "micro-scale" calibration are performed. The comparison of the statewide GM TLF with the 00 TLF for the macro-scale calibration does not provide suitable results because the available 00 survey data do not represent an unbiased sample of statewide truck trips. For the "micro-scale" calibration, "partial" GM trip tables that correspond to the 00 survey trip tables are extracted from the full statewide GM trip table. These "partial" GM trip tables are then merged and a partial GM TLF is created. The GM friction factor curves are adjusted until the partial GM TLF matches the 00 TLF. Three friction factor curves, one for each trip type, resulting from the micro-scale calibration produce a reasonable GM truck trip model. A key methodological issue for GM. calibration involves the use of multiple friction factor curves versus a single friction factor curve for each trip type in order to estimate truck trips with reasonable accuracy. A single friction factor curve for each of the three trip types was found to reproduce the 00 TLFs from the calibration data base. Given the very limited trip generation data available for this research, additional refinement of the gravity model using multiple mction factor curves for each trip type was not warranted. In the traditional urban transportation planning studies, the zonal trip productions and attractions and region-wide OD TLFs are available. However, for this research, the information available for the development .of the GM model is limited to Ground Counts (GC) and a limited set ofOD TLFs. The GM is calibrated using the limited OD data, but the OD data are not adequate to obtain good estimates of truck trip productions and attractions .. Consequently, zonal productions and attractions are estimated using zonal population as a first approximation. Then, Selected Link based (SELINK) analyses are used to adjust the productions and attractions and possibly recalibrate the GM. The SELINK adjustment process involves identifying the origins and destinations of all truck trips that are assigned to a specified "selected link" as the result of a standard traffic assignment. A link adjustment factor is computed as the ratio of the actual volume for the link (ground count) to the total assigned volume. This link adjustment factor is then applied to all of the origin and destination zones of the trips using that "selected link". Selected link based analyses are conducted by using both 16 selected links and 32 selected links. The result of SELINK analysis by u~ing 32 selected links provides the least %RMSE in the screenline volume analysis. In addition, the stability of the GM truck estimating model is preserved by using 32 selected links with three SELINK adjustments, that is, the GM remains calibrated despite substantial changes in the input productions and attractions. The coverage of zones provided by 32 selected links is satisfactory. Increasing the number of repetitions beyond four is not reasonable because the stability of GM model in reproducing the OD TLF reaches its limits. The total volume of truck traffic captured by 32 selected links is 107% of total trip productions. But more importantly, ~ELINK adjustment factors for all of the zones can be computed. Evaluation of the travel demand model resulting from the SELINK adjustments is conducted by using screenline volume analysis, functional class and route specific volume analysis, area specific volume analysis, production and attraction analysis, and Vehicle Miles of Travel (VMT) analysis. Screenline volume analysis by using four screenlines with 28 check points are used for evaluation of the adequacy of the overall model. The total trucks crossing the screenlines are compared to the ground count totals. L V/GC ratios of 0.958 by using 32 selected links and 1.001 by using 16 selected links are obtained. The %RM:SE for the four screenlines is inversely proportional to the average ground count totals by screenline .. The magnitude of %RM:SE for the four screenlines resulting from the fourth and last GM run by using 32 and 16 selected links is 22% and 31 % respectively. These results are similar to the overall %RMSE achieved for the 32 and 16 selected links themselves of 19% and 33% respectively. This implies that the SELINICanalysis results are reasonable for all sections of the state.Functional class and route specific volume analysis is possible by using the available 154 classification count check points. The truck traffic crossing the Interstate highways (ISH) with 37 check points, the US highways (USH) with 50 check points, and the State highways (STH) with 67 check points is compared to the actual ground count totals. The magnitude of the overall link volume to ground count ratio by route does not provide any specific pattern of over or underestimate. However, the %R11SE for the ISH shows the least value while that for the STH shows the largest value. This pattern is consistent with the screenline analysis and the overall relationship between %RMSE and ground count volume groups. Area specific volume analysis provides another broad statewide measure of the performance of the overall model. The truck traffic in the North area with 26 check points, the West area with 36 check points, the East area with 29 check points, and the South area with 64 check points are compared to the actual ground count totals. The four areas show similar results. No specific patterns in the L V/GC ratio by area are found. In addition, the %RMSE is computed for each of the four areas. The %RMSEs for the North, West, East, and South areas are 92%, 49%, 27%, and 35% respectively, whereas, the average ground counts are 481, 1383, 1532, and 3154 respectively. As for the screenline and volume range analyses, the %RMSE is inversely related to average link volume. 'The SELINK adjustments of productions and attractions resulted in a very substantial reduction in the total in-state zonal productions and attractions. The initial in-state zonal trip generation model can now be revised with a new trip production's trip rate (total adjusted productions/total population) and a new trip attraction's trip rate. Revised zonal production and attraction adjustment factors can then be developed that only reflect the impact of the SELINK adjustments that cause mcreases or , decreases from the revised zonal estimate of productions and attractions. Analysis of the revised production adjustment factors is conducted by plotting the factors on the state map. The east area of the state including the counties of Brown, Outagamie, Shawano, Wmnebago, Fond du Lac, Marathon shows comparatively large values of the revised adjustment factors. Overall, both small and large values of the revised adjustment factors are scattered around Wisconsin. This suggests that more independent variables beyond just 226; population are needed for the development of the heavy truck trip generation model. More independent variables including zonal employment data (office employees and manufacturing employees) by industry type, zonal private trucks 226; owned and zonal income data which are not available currently should be considered. A plot of frequency distribution of the in-state zones as a function of the revised production and attraction adjustment factors shows the overall " adjustment resulting from the SELINK analysis process. Overall, the revised SELINK adjustments show that the productions for many zones are reduced by, a factor of 0.5 to 0.8 while the productions for ~ relatively few zones are increased by factors from 1.1 to 4 with most of the factors in the 3.0 range. No obvious explanation for the frequency distribution could be found. The revised SELINK adjustments overall appear to be reasonable. The heavy truck VMT analysis is conducted by comparing the 1990 heavy truck VMT that is forecasted by the GM truck forecasting model, 2.975 billions, with the WisDOT computed data. This gives an estimate that is 18.3% less than the WisDOT computation of 3.642 billions of VMT. The WisDOT estimates are based on the sampling the link volumes for USH, 8TH, and CTH. This implies potential error in sampling the average link volume. The WisDOT estimate of heavy truck VMT cannot be tabulated by the three trip types, I-I, I-E ('||'&'||'pound;-I), and E-E. In contrast, the GM forecasting model shows that the proportion ofE-E VMT out of total VMT is 21.24%. In addition, tabulation of heavy truck VMT by route functional class shows that the proportion of truck traffic traversing the freeways and expressways is 76.5%. Only 14.1% of total freeway truck traffic is I-I trips, while 80% of total collector truck traffic is I-I trips. This implies that freeways are traversed mainly by I-E and E-E truck traffic while collectors are used mainly by I-I truck traffic. Other tabulations such as average heavy truck speed by trip type, average travel distance by trip type and the VMT distribution by trip type, route functional class and travel speed are useful information for highway planners to understand the characteristics of statewide heavy truck trip patternS. Heavy truck volumes for the target year 2010 are forecasted by using the GM truck forecasting model. Four scenarios are used. Fo~ better forecasting, ground count- based segment adjustment factors are developed and applied. ISH 90 '||'&'||' 94 and USH 41 are used as example routes. The forecasting results by using the ground count-based segment adjustment factors are satisfactory for long range planning purposes, but additional ground counts would be useful for USH 41. Sensitivity analysis provides estimates of the impacts of the alternative growth rates including information about changes in the trip types using key routes. Cytochrome P450 monooxygenase analysis in free-living and symbiotic microalgae Coccomyxa sp. C-169 and Chlorella sp. NC64A

  • Microalgae research is gaining momentum because of their potential biotechnological applications, including the generation of biofuels. Genome sequencing analysis of two model microalgal species, polar free-living Coccomyxa sp. C-169 and symbiotic Chlorella sp. NC64A, revealed insights into the factors responsible for their lifestyle and unravelled biotechnologically valuable proteins. However, genome sequence analysis under-explored cytochrome P450 monooxygenases (P450s), heme-thiolate proteins ubiquitously present in species belonging to different biological kingdoms. In this study we performed genome data-mining, annotation and comparative analysis of P450s in these two model algal species. Sixty-nine P450s were found in two algal species. Coccomyxa sp. showed 40 P450s and Chlorella sp. showed 29 P450s in their genome. Sixty-eight P450s (>100 amino acid in length) were grouped into 32 P450 families and 46 P450 subfamilies. Among the P450 families, 27 P450 families were novel and not found in other biological kingdoms. The new P450 families are CYP745-CYP747, CYP845-CYP863, and CYP904-CYP908. Five P450 families, CYP51, CYP97, CYP710, CYP745, and CYP746, were commonly found between two algal species and 16 and 11 P450 families were unique to Coccomyxa sp. and Chlorella sp. Synteny analysis and gene-structure analysis revealed P450 duplications in both species. Functional analysis based on homolog P450s suggested that CYP51 and CYP710 family members are involved in membrane ergosterol biosynthesis. CYP55 and CYP97 family members are involved in nitric oxide reduction and biosynthesis of carotenoids. This is the first report on comparative analysis of P450s in the microalgal species Coccomyxa sp. C-169 and Chlorella sp. NC64A.
