Fusarium Wilt of Korean Blackberry Caused by Fusarium cugenangense

  • Wilt symptoms were frequently observed in Korean blackberry (Rubus coreanus) plants grown in farmers' fields located in Gochang and Jeongeup, Jeonbuk Province, Korea during disease surveys in July 2020. The disease occurred in 10 of the 13 fields surveyed in the two locations. The incidence of diseased plants in the fields was 5-80%. Seven isolates of Fusarium sp. were obtained from the diseased plants and examined for their morphological and molecular characteristics. All the isolates belonged to Fusarium oxysporum species complex based on the morphological characteristics but were identified as F. cugenangense based on the molecular characteristics. Two isolates of F. cugenangense were tested for pathogenicity on Korean blackberry plants by artificial inoculation. Pathogenicity of the two isolates on the plants was confirmed with the inoculation tests, which showed wilt symptoms similar to those observed in the diseased plants in the fields investigated. This is the first report of F. cugenangense causing Fusarium wilt in Korean blackberry.

Relationship between the Population of Ralstonia solanacearum in Soil and the Incidence of Bacterial Wilt in the Naturally Infested Tobacco Fields

  • The population of Ralstonia solanacearum (Rs) in soil is very important as a primary inoculum source of bacterial wilt in tobacco fields. To investigate the population of Rs, physical properties and chemical components during the tobacco growing season, soil samples were taken from the fifteen fields which were located in the flue-cured tobacco growing area, Ansung, Kyunggi province and Wonju, Kangwon province. Two fields of the fifteen were bacterial wilt free. Six fields had less than 10% plants being diseased and seven over 10%. The Rs population level determined by using SMSA medium generally showed an up-and-down pattern being low in May, high in Jun and July and low in August. The soil population in May and June showed a positive correlation with the incidence of bacterial wilt (r=0.571$^*$, r=0.688$^{**}$), but P$_2O_5$, content of soil was negatively correlated with the disease incidence (r=-0.539$^*$). These results suggest that Rs population in soil examined in May or in June, and the P$_2O_5$ content in soil should be key factors to determine the bacterial wilt potential of tobacco fields.

Biological Control of Strawberry Fusarium Wilt Caused by Fusarium oxysporum f. sp.fragariae Using Bacillus velezensis BS87 and RK1 Formulation

  • Two isolates, Bacillus sp. BS87 and RK1, selected from soil in strawberry fields in Korea, showed high levels of antagonism towards Fusarium oxysporum f. sp. fragariae in vitro. The isolates were identified as B. velezensis based on the homology of their gyrA sequences to reference strains. BS87 and RK1 were evaluated for control of Fusarium wilt in strawberries in pot trials and field trials conducted in Nonsan, Korea. In the pot trials, the optimum applied concentration of BS87 and RK1 for pre-plant root-dip application to control Fusarium wilt was $10^5$ and $10^6$ colony-forming units (CFU)/ml, respectively. Meanwhile, in the 2003 and 2005 field trials, the biological control efficacies of formulations of RK1 were similar to that of a conventional fungicide (copper hydroxide) when compared with a non-treated control. The RK1 formulation was also more effective than BS87 in suppressing Fusarium wilt under field conditions. Therefore, the results indicated that formulations of B. velezensis BS87 and RK1 may have potential to control Fusarium wilt in strawberries.

시금치 바이러스병에 관한 연구 II. 시금치에 발생하는 Broad Bean Wilt Virus (BBWV)의 분류동정 (Investigations on the Virus Diseases in Spinach (Spinacia oleracea L.) II. Identification of Broad Bean Wilt Virus Occuring Spinach)

  • 짙은 연색의 모자익병징을 나타내는 이병 시금치를 채집하여 Broad bean wilt virus(BBWV)를 분리동정 하였다. 분리된 Broad bean wilt virus (BBWV)를 지표식물에 즙액접종한 결과, 명아주(Chenopodium amaranticolor), 명아주(Chenopodium quinoa) ,잠두(Vicia faba)에서는 접종엽에 국부병반이 나타났고 접종상엽에서는 모자익(전신감염)병징이 나타났으며 꽈리(Physalis floridana), 시금치, 담배 (W.B), 담배 (Nicotiana glutinosa)에는 모자익 병징이 나타났다. 이병시금치로부터 분리한 병원바이러스와 BBWV의 항혈청을 한천내확산법(Agar gel-diffusion test)으로 반응시킨 결과 양성 반응이 나타났다. 이병엽을 Dip법으로 시료를 제작하여 전자현미경에서 검경한 결과 구형의 입자가 관찰되었으며 직경은 25nm이었다. 시금치 재배포장에서 BBWV의 발생분포는 수원, 안양, 대구 진주 등 거의 전국적으로 발생하였다.

Effect of Bacterial Wilt on Fungal Community Composition in Rhizosphere Soil of Tobaccos in Tropical Yunnan

  • Bacterial wilt, which is a major soil-borne disease with widespread occurrence, poses a severe danger in the field of tobacco production. However, there is very limited knowledge on bacterial wilt-induced microecological changes in the tobacco root system and on the interaction between Ralstonia solanacearum and fungal communities in the rhizosphere soil. Thus, in this study, changes in fungal communities in the rhizosphere soil of tobaccos with bacterial wilt were studied by 18S rRNA gene sequencing. The community composition of fungi in bacterial wilt-infected soil and healthy soil in two tobacco areas (Gengma and Boshang, Lincang City, Yunnan Province, China) was studied through the paired comparison method in July 2019. The results showed that there were significant differences in fungal community composition between the rhizosphere soil of diseased plants and healthy plants. The changes in the composition and diversity of fungal communities in the rhizosphere soil of tobaccos are vital characteristics of tobaccos with bacterial wilt, and the imbalance in the rhizosphere microecosystem of tobacco plants may further aggravate the disease.

담배세균성마름병[립고병(立枯病)]에 대한 담배품종의 저항성 검정법 (Screening Procedure of Tobacco Cultivars for Resistant to Bacterial Wilt Caused by Ralstonia solanacearum)

  • Bacterial wilt caused by Ralstonia solanacearum has become a severe problem on tobacco in Korea. No effective single control measure is available at present time. One of the most potential way for controlling the bacterial wilt on tobacco is growing tobacco cultivars resistant to the bacterial wilt. In this study, optimal conditions for screening tobacco cultivars resistant to the bacterial wilt were examined to provide reproducible and efficient methods in growth chamber testing and field experiments for evaluating plant disease resistance. For this, already-known inoculation methods, inoculum densities, and incubation temperature, and plant growth stages at the time of inoculation were compared using tobacco cultivars resistant (Nicotiana tabacum cv, NC95), moderately resistant (N. tabacum cv. SPG70), and susceptible (N. tabacum BY4) to the bacterial disease. It was determined that root-dipping of tobacco seedlings at six true leaf stage into the bacterial suspension with inoculum level of $10^8$ colony-forming units (CFU)/ml for 20 min before transplanting was simple and most efficient in testing for resistance to the bacterial wilt of tobacco caused by R. solanacearum, for which disease incidences and severities were examined at 2 weeks of plant growth after inoculation at $20{\sim}25^{\circ}C$ in a growth chamber. These experimental conditions could discriminate one tobacco cultivar from the others by disease severity better than any other experimental conditions. In field testing, the optimum time for examining the disease occurrence was late June through early July. These results can be applied to establishing a technical manual for the screening of resistant tobacco cultivars against the bacterial wilt caused by R. solanacearum.

코퍼 하이드록사이드를 이용한 토마토 풋마름병 방제 (Control of Bacterial Wilt of Tomato using Copper Hydroxide)

  • 국내에서 Ralstonia solanacearum에 의한 풋마름병은 토마토 재배에 심각한 피해를 주고 있다. R. solanacearum에 의한 풋마름병을 방제하기 위한 약제를 선발하기 위하여 5종 항생제를 이용하여 균에 대한 생장 억제 효과와 포장에서의 토마토 풋마름병 방제효과를 조사하였다 R. solanacearum에 대한 생장억제효과를 조사한 결과, streptomycin 수화제, oxytetracyclin streptomycin sulfate 수화제, oxolinic acid 수화제는 병원균에 대한 생육억제 효과가 우수하였다. 포장에서 토마토 풋마름병에 대한 방제효과 시험을 실시한 결과, copper hydroxide 수화제가 62.5%의 가장 높은 방제효과를 나타내었다. Copper hydroxide 수화제는 친환경유기농자재에 등록된 약제로서 관행 재배뿐만 아니라 토마토 친환경 재배시에도 풋마름병 방제에 사용할 수 있을 것이다.

Improvement of Biocontrol of Damping-off and Root Rot/Wilt of Faba Bean by Salicylic Acid and Hydrogen Peroxide

  • Rhizoctonia solani, Fusarium solani, F. oxysporum, and Macrophomina phaseolina were found to be associated with root rott and wilt symptoms of faba bean plants collected from different fieldes in New Valley governorate, Egypt. All the obtained isolates were able to attack faba bean plants (cv. Giza 40) causing damping-off and root rot/wilt diseases. R. solani isolates 2 and 5, F. solani isolate 8, F. oxysporum isolate 12 and M. phaseolina isolate 14 were the more virulent ones in the pathogenicity tests. Biocontrol agents (Trichoderma viride and Bacillus megaterium) and chemical inducers (salicylic acid [SA] and hydrogen peroxide) individually or in combination were examined for biological control of damping-off and root rot/wilt and growth promoting of faba bean plants in vitro and in vivo. Both antagonistic biocontrol agents and chemical inducers either individually or in combination inhibited growth of the tested pathogenic fungi. Biocontrol agents combined with chemical inducers recorded the highest inhibited growth especially in case SA + T. viride and SA + B. megaterium. Under green house and field conditions, all treatments significantly reduced damping-off and root rot/wilt severity and increased of survival plants. Also, these treatments increased fresh and weights of the survival plants in pots compared with control. The combination between biocontrol agents and chemical inducers were more effective than used of them individually and SA + T. viride was the best treatment in this respect. Also, under field conditions, all these treatments significantly increased growth parameters (plant height and number of branches per plant) and yield components (number of pods per plant and number of seeds per plant, weight of 100 seeds and total yield per feddan) and protein content in both seasons (2010~2011 and 2011~2012). Faba bean seeds soaked in SA + T. viride and SA + B. megaterium were recorded the highest growth parameters and yield components. Generally, the combination between biocontrol agents and chemical inducers recorded the best results for controlling damping-off and root rot/wilt diseases in greenhouse and field with addition improved plant growth and increased yield components in field.

Screening of Endophytic Fungal Isolates Against Raffaelea quercus-mongolicae Causing Oak Wilt Disease in Korea

  • Oak wilt disease caused by Raffaelea quercus-mongolicae has emerged obviously in Korea. We selected antifungal isolates against R. quercus-mongolicae among 368 endophytic fungal isolates from different parts of oak and pine trees. The experiment was conducted in the primary and secondary screenings by dual culture test. The antifungal activity of the selected isolates was assessed in culture filtrate test based on the inhibition rates in mycelial growth, sporulation, and spore germination of oak wilt fungus. Five isolates, E089, E199, E282, E409 and E415, showed strong antifungal activity in culture filtrate test, and their antifungal activity decreased on the culture media supplemented with heated culture filtrate. Higher mycelial growth inhibitions on the unheated media were recorded in E409 (Colletotrichum acutatum), E089 (Daldinia childiae), E415 (Alternaria alternata) and E199 (Daldinia childiae) with the inhibition rates of 79.0%, 70.1%, 68.9% and 64.5%, respectively. These isolates also had the higher sporulation inhibitions on unheated media with the rates of 96.8%, 84.2%, 82.8% and 80.5%, respectively. The spore germination of the oak wilt fungus was completely inhibited by E282 (Nectria balsamea) on both unheated and heated media. These results showed that a higher number of potent antifungal isolates against oak wilt fungus was isolated from the petiole compared to the other parts. This study could contribute to the development of biological control approaches for the management of oak wilt disease caused by R. quercus-mongolicae.

Occurrence of Root Rot and Vascular Wilt Diseases in Roselle (Hibiscus sabdariffa L.) in Upper Egypt

  • Roselle (Hibiscus sabdariffa L.) family Malvaceae is an important crop used in food, cosmetics and pharmaceutics industries. Roselle is cultivated mainly in Upper Egypt (Qena and Aswan governorates) producing 94% of total production. Root rot disease of roselle is one of the most important diseases that attack both seedlings and adult plants causing serious losses in crop productivity and quality. The main objective of the present study is to identify and characterize pathogens associated with root rot and wilt symptoms of roselle in Qena, Upper Egypt and evaluate their pathogenicity under greenhouse and field condition. Fusarium oxysporum, Macrophomina phaseolina, Fusarium solani, Fusarium equiseti and Fusarium semitectum were isolated from the natural root rot diseases in roselle. All isolated fungi were morphologically characterized and varied in their pathogenic potentialities. They could attack roselle plants causing damping-off and root rot/wilt diseases in different pathogenicity tests. The highest pathogenicity was caused by F. oxysporum and M. phaseolina followed by F. solani. The least pathogenic fungi were F. equiseti followed by F. semitectum. It obviously noted that Baladi roselle cultivar was more susceptible to infection with all tested fungi than Sobhia 17 under greenhouse and field conditions. This is the first report of fungal pathogens causing root rot and vascular wilt in roselle in Upper Egypt.
