Effects of Several Korean Native Wild Perennial Flowers and Kinds of Basal Sheets on Sod Formation (자생 숙근초화 및 바닥재의 종류가 뗏장 형성에 미치는 영향)

  • 14 kinds of perennial flower sods, including 10 Korean native wild perennial flowers such as Lychnis cognata, Achillea sibirica, Sedum spectabile, Sedum takesimense etc. were made with basal sheets of nonwoven fabric or jute fabric. Growth of plants planted on bark was better than that of planted on soil of open area. The plants planted on bark also showed difference in growth rate by the kinds of used basal sheets. In general, the growth of plants was good when nonwoven fabric was used as basal sheet instead of jute fabric. When jute fabric was used as basal sheet, It was severely decayed during growth period resulting in great decreasing of root sticking capacity to basal sheet. It was shown that the procumbent plants such as Tymus quinquecostatus, Ixeris stolonifera, Sedum oryzifolium, Sedum polystichoides having lower plant height and better ground coverage ability compared with upright plants such as Sedum spectabile and Aster glehni were more suitable for floral sod production.

Seeding of the Woody Plants for the Quick-coverage of the Slopes (비탈면 조기수림화를 위한 녹화용 식물의 활용에 관한 연구)

  • This study was conducted to investigate the germination rates and ground coverage rates of the 16 native wild flower, herbaceous, shrub and woody plants according to temperature and seeding timing for the revegetation and rehabilitation of the roadside slopes. Also, this study was conducted to suggest design criteria to select revegetation plants and to decide proper seed mixture for ecological restoration of the disturbed manmade slopes as a environmentally friendly construction. The results are summarized as follows. 1. Most of the plants germinated after 10~14 days. Woody plants germinated more slowly than wild flowers and herbaceous plants because of the hard seed. 2. Most of the plants showed the highest percentage of the germination rates under $15/25^{\circ}C$ temperature. The next was in order of under the $20/30^{\circ}C$ temperature, normal temperature(October) and $25/35^{\circ}C$ temperature. 3. At the chamber experiment, the Sorbaria sorbifolia var. stellipila showed the highest germination rates. Most of the native wild flowers germinated well and showed high germination rates under the various temperature. 4. As the field germination experiments, the seeding at August shows higher germination rates than that of the seeding at September, but showed lower surviving percentage of the germinated seedlings and lower ground coverage percentage than those of the seeding at September. After one year, Chionanthus retusus, Acer palmatum, Albizzia julibrissin are germinated and showed 10% coverage rates. So, they can be used as revegetation plants for the restoration works. 5. As seed mixture experiment, it was not effective only to increase the proportion of the ratio of the tree seeds, relatively high price, for making woody vegetation. It would be more successful to make vegetation structure by natural competition among wild flowers, shrubs and trees. 6. The excessive dominance of trees in revegetation works may destroy the lower layer of vegetation and it will be undesirable on the species diversity. It is more important to recover the species diversity of the plant community by seed mixture with the considering the germination, the growth characteristics and the correlation effect among revegetation plants. 7. To recover the manmade slopes quickly, increasing the ratio of the wild flower was recommended.

Yeasts Diversity of Wild Flowers in Mountains of Korea and Their Physiological Functionalities (한국의 야생화에서 분리한 효모의 종 다양성과 생리기능성)

  • This study focused on isolation of wild yeasts from natural flowers and elucidation of yeast diversity. Wild yeasts were isolated from various flowers collected from mountains on the islands including Jejudo, Ulleungdo, Yokjido, and Seonyudo as well as inlands including Gyejoksan, Oseosan, Beakamsan, and Deogyusan in Korea. Isolated yeasts were identified by comparison of nucleotide sequences for polymerrase chain reaction-amplified D1/D2 region of 26S rDNA or internal transcribed spacer 1 and 2 including 5.8S rDNA using BLAST. 289 strains belonging to 134 yeast species were isolated. Cryptococcus genus strains were the most frequently isolated species among the identified yeasts. Metschnikowia reukaufii was also frequently isolated. Twenty three species including Cryptococcus aureus were overlapped between those of mountains on islands and inland. Physiological functionalities such as antioxidant activity, xanthine oxidase inhibitory activity, and tyrosinase inhibitory activity for the 289 identified yeast strains were investigated using their supernatant and cell-free extracts. The supernatants of Candida sp. 78-J-2 and Metschnikowia reukaufii SY44-6 showed antioxidant activity of 22.5%, and anti-gout xanthine oxidase inhibitory activity of 49.6%, respectively.

Study on the Sod Forming Effects of the Native Plants for the Erosion Control in Slopes (비탈면 침식방지를 위한 자생초본식물의 뗏장 형성에 관한 연구)

  • This study was conducted to suggest vegetation mat forming methods with native wildflowers, introduced wild flower, grasses and native herb plants which can be used for erosion control of the disturbed slopes. The main results are summarized as follows. In forming the vegetation mat at early stage, foreign grasses, native wild plants shows more effective than the other seed mixture types. But, after 12 weeks, using native wild flowers shows more effective than foreign grasses. Also, the mixture of the native grasses and kentucky bluegrasses (seed mixture type F) are the best in ground coverage among the seed mixtures types. In order to restore slopes more naturally, more diverse plants should be used. The native grasses such as Marrubium incisum Bentham, Lutos comiculatus L. var. japonicus Regel, and Artemisia princeps Pampan seems to be useful to forming the vegetation mat, also it will be more easy if mixing with foreign grasses such as Kentucky bluegrass. The mixture of native grasses and introduced wild flowers are not proper in forming the vegetation mat. Also, pot planting of the native grasses are not proper in forming the vegetation mat.

Isolation of Yeasts from Wild Flowers in Gyonggi-do Province and Jeju Island in Korea and the Production of Anti-gout Xanthine Oxidase Inhibitor (경기도와 제주도 야생화들로부터 효모의 분리, 동정 및 항통풍성 Xanthine oxidase 저해물질의 생산)

  • Several yeasts were isolated from flowers found in Gyonggi-do Province and Jeju island in Korea. They were then identified by a comparison of their PCR-amplified D1/D2 regions of 26S rDNA, internal transcribed spacer 1 and 2 inclusive of 5.8S rDNA, using the BLAST database. A total of fifty four yeast strains were isolated from wild flowers in Gyonggi-do and the genus Pseudozyma was noted as being dominant. A total of thirty two strains were isolated from Songaksan and Seongsan-ilchulbong in Jeju island and Sporobolomyces ruberrimus was seen to be dominant. The anti-gout xanthine oxidase inhibitory activities of the culture broths and cell-free extracts from eighty six yeast strains were then determined. The cell-free extracts of Pseudozyma hubeiensis 228-S-1 exhibited the highest xanthine oxidase inhibitory activity of 19.6%. The XOD inhibitor was also maximally produced when Pseudozyma hubeiensis 228-S-1 was cultured at $30^{\circ}C$ for 36h in YEPD medium.

  • Rhodosporidium fluviale 201-C-1 and Kluyveromyces thermotolerans 150-JE-2(1) were isolated from wild flowers in Geumsan-gun, Chungcheongnam-do and Wando-gun, Jeollanam-do, respectively. Rhodosporidium paludigenum 86-J-1 and Candida sp. 80-J-3 were also isolated from flowers in Jeju island, Korea. Characteristics for these yeasts were not reported before. Therefore, morphological and physiological characteristics for these yeasts were investigated in this study. R. fluviale 201-C-1 was halophilic asporogenous yeast and formed pseudomycelium. It also grew at $30^{\circ}C$ and pH 4.0-6.0. K. thermotolerans 150-JE-2(1) was sugar-tolerant and halotolerant asporogenous yeast. It formed pseudomycelium and showed positive urease activity. Candida sp. 80-J-3 and R. paludigenum 86-J-1 were asporogenous yeasts, and grew well in yeast extract-peptone dextrose(YPD) medium, potato-dextrose medium as well as vitamin-free medium and YPD medium containing 5% NaCl.

Note on Newly Isolated Yeasts from Wild Flowers in Daejeon City, Korea (대전지역 야생화에서 분리한 미기록 효모 균주들의 보고)

  • We report morphological and physiological characteristics of newly isolated six kinds of yeasts which are new in Korean mycoflora. The yeasts were isolated from wild flowers in Daejeon city, Korea and identified by molecular analysis of the amplified internal transcribed space rDNA sequence or partial 18S rDNA sequence. Species of isolates were identified as Kuraishia capsulate, Lodderomyces elongisporus, Pseudozyma antarctica, Starmerella bombicola.

  • From 182 non-pathogenic wild yeast isolates from flowers, Pichia silvicola UL6-1 and Sporobolomyces carnicolor 402-JB-1 were selected for potent ${\gamma}$-aminobutyric acid production and microbiological characteristics were investigated. Pichia silvicola UL6-1 formed ascospores and pseudomycelia. The strain was also halotolerant, growing well in 5% NaCl-containing yeast extract-peptone-dextrose (YPD) medium. Sporobolomyces carnicolor 402-JB-1 did not form ascospores or pseudomycelia and grew well on 10% glucose-yeast extract-peptone medium.

Absence of AVP1 transcripts in wild type watermelon scions grafted onto transgenic bottle gourd rootstocks

  • In this study we confirmed the stable integration of Arabidopsis AVP1 in the genomes of bottle gourd $T_3$ homozygous lines and its transcription, and additionally evaluated possibility of translocation of the AVP1 mRNA from transgenic bottle gourd rootstocks to wild type watermelon scions. Each AVP1 gene in two bottle gourd T3 lines is abundantly expressed under a field condition. Given the grafting between wild type watermelon scions and AVP1-expressing bottle gourd rootstocks, no translocation of the AVP1 mRNA was detected in leaves, both sexual flowers, and fruits of the scions.

Isolation and Physiological Functionality of Yeasts from Wild Flowers in Seonyudo of Gogunsanyeoldo, Jeollabuk-do, Korea (전라북도 고군산열도의 선유도 일대 야생화들로부터 효모의 분리 및 생리기능성)

  • Sixty one yeast strains of 21 species were isolated from wild flowers in Gogunsanyeoldo, including Seonyudo of Jeollabuk-do, Korea, and identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA using BLAST. Among them, Cryptococcus sp. including C. aureus SY1-4 were found to be dominant, and Metschnikowia sp. including M. reukaufii SY20-1 and Rhodotnula sp. such as R. ingeniosa SY1-1 were also abundantly isolated. Some physiological functionalities of the culture broth and cell-free extracts from 61 yeast strains were determined. Supernatant from Metschnilowia reukaufii SY44-6 showed anti-gout xanthine oxidase inhibitory activity of 49.6% and whitening tyrosinase inhibitory activity of 38.4%, respectively.
