Lipoteichoic Acid Isolated from Lactobacillus plantarum Inhibits Melanogenesis in B16F10 Mouse Melanoma Cells

  • Lipoteichoic acid (LTA) is a major component of the cell wall of Gram-positive bacteria. Its effects on living organisms are different from those of lipopolysaccharide (LPS) found in Gram-negative bacteria. LTA contributes to immune regulatory effects including anti-aging. In this study, we showed that LTA isolated from Lactobacillus plantarum (pLTA) inhibited melanogenesis in B16F10 mouse melanoma cells. pLTA reduced the cellular activity of tyrosinase and the expression of tyrosinase family members in a dose-dependent manner. The expression of microphthalmia- associated transcription factor (MITF), a key factor in the synthesis of melanin, was also decreased by pLTA. Further, we showed that pLTA activated melanogenesis signaling, such as extracellular signal-regulated kinase (ERK) and phosphatidylinositol 3-kinse (PI3K)/AKT. In addition, the expression of heterogeneous nuclear ribonucleoprotein A1 (hnRNP A1) and HuR, which are important RNA-binding proteins (RBPs), was reduced. pLTA likely degrades MITF via regulation of melanogenic signaling and RNA stability of melanogenic proteins, resulting in the reduction of melanin. Thus, our data suggest that pLTA has therapeutic potential for treating hyperpigmentation disorders and can also be used as a cosmetic whitening agent.

Study on Cosmeceutical Activity of four Kinds of Korea Herb Medicine Materials (4가지 한방 소재(행인, 호장근, 자초, 강황)의 화장품약리활성에 관한 연구)

  • The aim of this study was to assess the cosmeceutical activies of four kinds of Korea herb medicine extracts using in cosmetics and related industries. The cosmeceutical activities of extracts were investigated by tyrosinase inhibition, astringent, anti-bacterial and MTT assay for cell viability. In the whitening effect, PA that the highest tyrosinase inhibition activity showed 56% at 10 ppm in ethanol extract. Also water and ethanol extract of RE showed 54%, 68% at 1,000 ppm, respectively, but LE and CA showed lower effect. Astringent effect of water and ethanol extract of PA appeared over 60% at 1,000ppm concentration but other extracts showed no astringent effect. In the anti-bacterial test, water and ethanol extract of PA showed no anti-bacterial effect against all microorganisms. But water and ethanol extract of RE showed anti-bacterial effect on Staphylococcus aureus, ethanol extract of showed on Staphylococcus aureus and Propionibacterium acnes, ethanol extract of CA showed on Candida albicans. The resUlt of stability test showed that the emulsion of containing PA were very stable at various temperature and sun-light test. Viscosity and pH of emulsion did not change. From the results of human patch test to assess the safety of cosmetics containing PA there was no negative reaction on skin was found.

Whitening Efficacy of Water Soluble Royal Jelly Removed Allergenic Protein (알러지 유발 단백질이 제거된 수용성 로열젤리의 피부미백 효과)

  • Royal jelly composes of many components, especially protein. Protein is a major factor which cause allergy. We focused on water soluble royal jelly (WSRJ) that was removed allergy - inducing protein. 10-hyroxy-2-decenoic acid content of WSRJ is 2.42 g/100 g, which is double compared to that of lypophilized RJ. To further access WSRJ as a cosmetic ingredient and potential external treatment for topical use, we investigated its ability to inhibit tyrosinase activity and melanin biosynthesis on melanogenesis in B16F1 melanoma cells. We found that WSRJ increased the cell viability in B16F1 melanoma cell and WSRJ (1~10 mg/ml) inhibited melanin synthesis in with 10 nM ${\alpha}$-melanocyte-stimulating hormone (${\alpha}$-MSH) for 48 h. WSRJ inhibited direct tyrosinase activity, which decreased melanin synthesis in ${\alpha}$-MSH stimulated B16F1 melanoma cells. Thease findings suggest that WSRJ induces the down regulation of melanogenesis by inhibiting tyrosinase activation.

Effects of Doinsenggitang on Melanin Synthesis and Gene Expression Inhibition in B16F10 Melanoma Cells (도인승기탕의 B16F10 세포주에서의 멜라닌 생성 및 유전자 발현 억제 효과)

  • This study involved observation of the inhibitory effect of 70% EtOH and water extracts from Doinseunggitang on melanin synthesis, tyrosinase activity, and western blotting using B16F10 melanoma cells. Doinseunggitang extracts inhibited melanin synthesis and tyrosinase activity in a dependent manner. As a result, it was found that Doinseunggitang 70% EtOH extracts inhibit melanin synthesis and tyrosinase activity, respectively, by 40% and 51%. In addition, western blotting analysis showed that 70% EtOH extracts inhibited tyrosinase, MITF, TRP-1, and TRP-2 expression. These results show that 70% ethanol extracts of Doinseunggitang could be developed as a skin whitening material in cosmetics.

Extracellular Vesicles from Korean Codium fragile and Sargassum fusiforme Negatively Regulate Melanin Synthesis

  • Although various marine ingredients have been exploited for the development of cosmetic products, no previous study has examined the potential of seaweed extracellular vesicles (EV) in such applications. Our results revealed that EV from Codium fragile and Sargassum fusiforme effectively decreased α-MSH-mediated melanin synthesis in MNT-1 human melanoma cells, associated with downregulation of MITF (microphthalmia-associated transcription factor), tyrosinase and TRP1 (tyrosinase-related proteins 1). The most effective inhibitory concentrations of EV were 250 ㎍/ml for S. fusiforme and 25 ㎍/ml for C. fragile, without affecting the viability of MNT-1 cells. Both EV reduced melanin synthesis in the epidermal basal layer of a three-dimensional model of human epidermis. Moreover, the application of the prototype cream containing C. fragile EV (final 5 ㎍/ml) yielded 1.31% improvement in skin brightness in a clinical trial. Together, these results suggest that EV from C. fragile and S. fusiforme reduce melanin synthesis and may be potential therapeutic and/or supplementary whitening agents.

Studies on Skin Anti-aging Efficacy of Hydrolysate from Phellinus igniarius (상황버섯 가수분해물의 피부 항노화 효능에 대한 연구)

  • Objective : The object of this study was to assess the efficacy of hydrolysate from Phellinus igniarius(HPI) on anti-aging activities in vitro measurement and mini clinical study performed on 5 subjects. Methods : To evaluate skin anti-aging efficacy of HPI, 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging activity, type I collagen synthesis, inhibition of nitric oxide(NO) production, inhibiton of tyrosinase, hyaluronan synthase(HAS)2, 3 mRNA expression were measured in vitro. Also, mini clinical study of skin hydration was performed on 5 subjects using HPI in distilled water(DW) and 1,3-butylene glycol diluted solution(30% in DW). Results : 1. DPPH radical scavenging activity of HPI was increased in a dose-dependant. 2. Type I collagen synthesis was increased in 50, 100 and 500㎍/㎖ of HPI. 3. NO production was not inhibited in all concentrations of HPI. 4. Tyrosinase was inhibited in 500, 1000, 2500 and 5000㎍/㎖ of HPI. 5. HAS2 mRNA expression was increased in 50, 100, 150 and 200㎍/㎖ of HPI, HAS3 mRNA expression was increased in 100, 150, and 200㎍/㎖ of HPI. 6. In the mini clinical study of 5 subjects, there was a difference in skin hydration over time for each solutions, but it was not statistically siginificant. Conclusions : HPI increased DPPH radical scavenging activity, type I collagen synthesis, and HAS2, HAS3 mRNA expression. HPI also suppressed tyrosinase. The findings of this study suggest that HPI can be used as an skin anti-aging material.

Inhibition of adipogenesis and melanogenesis by methanol extract of Codium fragile (Suringar) Hariot in 3T3-L1 adipocytes and B16F10 melanocytes (3T3-L1 및 B16F10 세포에서 청각 메탄올 추출물에 의한 지방 세포 분화 및 멜라닌 생성의 억제 효과)

  • Codium fragile (Suringar) Hariot, a green alga of the Codiales family, has been reported to have several bioactive properties, including antioxidant and anti-inflammatory properties. However, its antiobesity and whitening effects and their underlying mechanisms are unclear. This study aimed to evaluate the antiobesity and melanogenesis inhibitory effects of C. fragile using methanol extracts of C. fragile (MECF). The results of this study revealed that MECF inhibited the accumulation of lipid droplets and triacylglycerol in differentiated 3T3-L1 adipocytes, which was associated with the inhibition of the expression of adipogenesis-related transcription factors, such as peroxisome proliferator-activated receptor γ, CCAAT/enhancer-binding protein-α (C/EBPα), and C/EBPβ, which function as the key regulators of adipogenesis. Also, MECF reduced tyrosinase activity and melanin content in B16F10 cells as well as the expression of tyrosinase, tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-related transcription factor in the presence of α-melanocyte-stimulating hormone. Taken together, our findings suggest that the extract of C. fragile could be considered a promising functional ingredient for the prevention and treatment of obesity and skin pigmentation in the food and cosmetic industry.

Comparative evaluation on biological activities of Ziingiber officinale Roscoe extracted from different solvents (추출 용매를 달리한 생강 추출물에 대한 생리활성의 비교 평가 연구)

  • Objective : This study was designed to investigate anti-oxidative, anti-aging and anti-inflammatory activities of Zingiber officinale Roscoe water (ZD) and 50% ethanol extracts (ZE). Method : The antioxidant capacity of ZD or ZE was analyzed by DPPH, ABTS scavenging activities and reducing power, respectively. The anti-aging efficacy was performed with tyrosinase and collagenase inhibition activities. The anti-inflammatory activities were confirmed through inhibition of nitric oxide production and TNF-�� expression in LPS-treated Raw 264.7 cells. Result : As a result of measuring DPPH and ABTS radical scavenging activities, both ZD and ZE extracts increased in concentration-dependent manners. The ABTS radical scavenging activities of water and 50% ethanol extract were similar to that of positive control at high concentration. On reducing power, both ZD and ZE showed higher activities than the positive control at high concentration. On inhibition activities of tyrosinase and collagenase both ZD and ZE showed the large inhibition rates at low concentration. Also, the inhibitory efficacy of ZD on NO and TNF-�� production was better than ZE in RAW 264.7 cells. Conclusion : These results suggest that ZD and ZE have anti-oxidative, anti-aging and anti-inflammatory effects in vitro. Especially, ZD was more effective than ZE. Thus, Ginger extract is considered to be highly useful as a cosmetic or food material with anti-oxidative, anti-aging (prevention of whitening and wrinkle) and anti-inflammatory effects. Also these effects may vary depending on the extraction solvent.

Antioxidant and Anti-inflammatory Activity of Grateloupia filicina extract (지누아리(Grateloupia filicina) 추출물의 항산화 및 항염능)

  • In this study, the antioxidant ability, anti-inflammatory ability and whitening effect of the extracted Grateloupia filicina under optimal conditions were confirmed. The useful substances of Grateloupia filicina were extracted with various ethanol aqueous solutions to measure DPPH, ABTS, and nitrite radical elimination capabilities, and the polyphenol and flavonoid contents were analyzed. When extracted with a 20% ethanol aqueous solution, the radical elimination ability was the best, and the content of polyphenol and flavonoids was also the highest. From this, it was confirmed that the 20% ethanol aqueous solution efficiently extracted the antioxidant and anti-inflammatory substances of Grateloupia filicina. Meanwhile, the cytotoxicity and anti-inflammatory properties of the Grateloupia filicina extract were confirmed using RAW 264.7 cell lines. Grateloupia filicina extract did not show toxicity at a concentration of 200 ㎍/mL, and showed NO production inhibitory capacity of 50% or more at this concentration. As a result, it was confirmed that the Grateloupia filicina extract can be used as a functional cosmetic raw material having antioxidant and anti-inflammatory properties.

Inhibitory Effect of Citrus Unshiu Peel and Cimicifuga Dahurica Extracts on Melanogenesis accroding to the Various Extraction Methods (추출조건에 따른 진피(陳皮) 및 승마(升麻) 추출물의 미백 활성효과)

  • Objectives: Excessive melanin production leads to skin pigmentation, which causes various cosmetic and health problems. Citrus unshiu Markovich peel (CS) and Cimicifuga dahurica (CD) have long been widely used as a oriental medicinal plant because of their pharmacological properties including anti-ulcer, anti-oxidant, and anti-inflammatory properties. Methods: In this study, we investigated the inhibitory effects of CS, CD extracts or CS:CD=1:2 mixture on melanogenesis according to the various extraction methods. CS and CD extracted were prepared by ethanol extraction (EE), ultrasonification extraction (USE), Supercritical extraction (SCE), reflux extraction (RE), respectively. Results: DPPH radical scavenging and tyrosinase inhibitory activity of CD extracts or CS:CD=1:2 mixture were increased in dose-dependent manners. In addition, we evaluated the effect of CS, CD extracts or CS:CD=1:2 mixture on tyrosinase activity and melanogenesis in α-melanocyte stimulating hormone-induced B16-F10 melanoma cells. CS, CD extracts or CS:CD=1:2 mixture significantly inhibited tyrosinase activity and melanogenesis at 10-200 ㎍/mL. Conclusions: Therefore, our study suggests that CS and CD extracts have potential as a safe treatment for excessive pigmentation or as a natural ingredient in cosmetics.
