Solvent Free Solid Injector (SFSI)를 이용한 수삼, 홍삼, 백삼의 향기성분 분석방법 (The Analysis of Volatile Components of Fresh Ginseng, Red Ginseng and White Ginseng by Solvent Free Solid Injector (SFSI) Techniques)

  • 인삼의 휘발성 성분 분석 시 SFSI를 이용한 분석법의 최적화를 일하여 실험을 수행하였다. 최적화를 위하여 세 가지 시료조제방법인 생체시료, oven건조와 동결건조를 검토하였고, GC injector 온도는 100, 150, 200, 250, $300^{\circ}C$에서의 휘발성분 분석효율을 검토하였으며, 모세관에서 시료의 기화 시간인 preheating time은 3, 5, 7, 10, 15 min의 조건에서 검토한 결과, 시료 조제방법은 생체시료를 그대로 사용 시, GC injector 온도는 $250^{\circ}C$일 때 preheating time은 10min일 때 가장 분석효율이 높았다. 휘발성분 분석을 위해 사용된 인삼은 금산에서 재배한 6년근 삼으로 수삼, 백삼, 홍삼을 사용하였으며, SFSI가 장착된 CC/MS에 의해 휘발성분을 분석, 동정하였다. 분석결과 수삼에서는 33종, 백삼에서는 36종, 홍삼에서는 38종의 휘발성분이 분석되어져 SFSI에 의한 분석법이 인삼의 휘발성분을 분석, 동정하는 방법으로 이용될 수 있음을 보여주고 있다.

인삼의 가공현황과 전망 (Present and Future on the Processing of Ginseng)

  • In this review quality improvement, new products and processing of ginseng are discussed. Ginseng products are generally classified into two types; the dried product without significant change in original shape of fresh ginseng and various processed ginseng products in liquid or solid types prepared by addition of either ginseng extract of ground powder. The dried ginsengs are generally made 4 years old fresh ginseng roots for production of white ginseng and 6 years old ones for red ginseng. The processed ginseng products, such as ginseng drinks, extracts, teas, powders, capsules or tablets are prepared by addition of extract or powder of the ginseng roots which contain relatively high amount of saponin. At present, more than 200 items of 40 types of products are commercially available in over 70 countries in the world, Since consummers preference on the quality of ginseng products as an health food differs with their cultural background of each country, new products development and quality improvement should be investigated with concerning the particular preference of the consummers of various country. It has been generally found that the Orientals has higher product acceptance on strong ginseng flavor while the Westerners generally prefers the products having mild ginseng odor and taste. Recently consummers are asking for supplemented type of ginseng products with various medical herbs and vital materials instead of ginseng alone. Therefore future work on product development should be emphasized to meet the consummers demand and preference.

Taxonomy and Fermentation of Kitasatosporia kimorexae Producing New Thiopeptide Antibiotics, Kimorexins

  • An isolate, 90-GT-302, was found to produce antibiotics inducing typical mycelial swelling in Magnaporthe grisea and Fusarium solani. This isolate formed yellow substrate and white rectiflexbiles aerial mycelia in the early stages of growth. The aerial mycelium gradually changed its color to white and finally formed a gray spore mass. Analysis of the cell wall acid hydrolysate revealed the presence of LL- and meso-diaminopimelic acids, glycine, and galactose, which indicated cell wall type X. This result placed our isolate in genus Kitasatosporia. A comparison of isolate 9O-GT-302 with reference strains of Kitasatosporia spp., which not only demonstrated several differences in their physiological properties but also novelty of the active compounds produced by this isolate, led us to designate the isolate as Kitasatosporia kimorexae.

고려인삼이 암독소의 지방분해작용에 미치는 저해활성 (Inhibitory Activity of Korean Ginseng on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid)

  • 한국 홍삼과 백삼의 조산성 다당체 성분과 수용성 추출물, 그리고 ginsenoside 성분이 암독소 호르몬-L에 의해 유도된 지방 분해를 저해하는 효과를 검토하고자 본 연구를 수행하였다. 지방분해 및 식욕억제 인자로 알려진 암독소 호르몬-L은 saroma-180을 접종한 mouse의 복수액으로부터 부분정제하여 사용하였다. 한국 홍삼과 백삼으로부터 정ㅇ제하여 얻은 조산성 다당체 각 분획의 성분은 10$\mu$g/ml 농도 이상에서 암독소 호르몬-L이 유도하는 지방 분해에 억제효과가 있었고, 이들 중 PG1과 PG4에서 지방 분해 저해 활성이 가장 높았으며, 이를 더 정제한 결과 PG4-3분획에서 활성이 가장 높았다. 고려인삼 중 백삼과 홍삼의 각 4, 5, 6 년근별로 얻은 수용성 추출물의 암독소 호르몬-L의 지방분해 억제에 미치는 영향을 검토한 결과 억제율은 1,000$\mu$g/ml 농도에서 백삼 4, 5 및 6년근이 각각 56.3, 59.7 및 59.4% 였고, 홍삼은 각각 78.6, 79.1 및 82.5%로 백삼보다 홍삼의 억제율이 높았다. 인삼 시료 g당 총저해활성은 백삼의 경우 100$\mu$g/ml 농도에서는 4년근이, 1,000$\mu$g/ml 농도에서는 5년근에서 가장 높았으나, 홍삼의 경우 다같이 6년근에서 가장 높았다. 또한 ginsonoside 성분의 경우 지방 분해 억제율과 총저해활성이 모두 RB2에서 가장 높았다.

국내 유통 중인 인삼 및 인삼 제품류의 미생물 오염도 평가 (Microbial Contamination Levels of Ginseng and Ginseng Products Distributed in Korean Markets)

  • 본 연구는 시중에 유통되고 있는 인삼 및 인삼 제품류의 미생물 오염실태를 조사하고자 수행하였다. 충남 금산과 진주 인근지역에서 유통되고 있는 인삼 및 인삼 제품류 81점에 대해 미생물 오염도를 조사한 결과 일반세균과 대장균군은 수삼이 3.19~7.02 log CFU/g로 오염도가 가장 높았고, 백삼에서는 0.25~7.31 log CFU/g, 홍삼에서는 0~2.89 log CFU/g, 홍삼음료에서는 0~1.70 log CFU/mL 범위의 오염도를 나타내었다. 병원성 미생물은 홍삼음료를 제외한 수삼, 백삼 및 홍삼에서 B. cereus가 검출되었다. 그 오염 수준은 수삼의 경우 91.9%(34/37)에서 2.41 log CFU/g 수준으로 검출되었고, 백삼은 53.3%(8/15)에서 0.98 log CFU/g, 홍삼은 13.3%(2/15)에서 0.50 log CFU/g 수준으로 검출되었다. 곰팡이의 경우 수삼에서만 0.72~5.54 log CFU/g범위로 검출되었고, 나머지 백삼(73.3%), 수삼(73.3%), 홍삼음료(21.4%)는 1 log CFU/g or mL 이하의 매우 낮은 수준의 오염도를 나타내었다. 유통 중인 인삼 및 인삼 제품류의 미생물 오염도는 대체적으로 양호한 수준이었으나 일부 제품은 인삼류 제품 검사기준을 초과한 경우도 존재하기 때문에 미생물 오염 방지를 위한 위생관리에 주의를 기울일 필요가 있는 것으로 판단된다.

광 및 UV 조사가 인삼 모상근의 생장 및 사포닌 생합성에 미치는 영향 (Effects of White Light and UV Irradiation on Growth and Saponin Production from Ginseng Hairy Root)

  • To investigate the effect of culture conditions on growth and ginsenosides accumulation, we cultured the ginseng hairy root under three different media, white light or ultra-violet irradiation. The MS/B5 medium containning MS basal salt and B5 vitamin was good for the growth and ginsenoside accumulation. The light during the culture period of ginseng hairy root was irradiated. The growth was abundant in the ginseng hairy root cultured in dark. But the ginsenosides accumulation was higher than in the ginseng hairy root cultured in the light irradiation. When the ginseng hairy root was cultured in 20 L bioreactor, the ginsenosides accumulation was observed at 34% higher than the hairy root cultured in dark. UV irradiated the ginseng hairy root during the culture period. The long time irradiation of UV was caused decreasing the growth of ginseng hairy root, but the accumulation of ginsenosidess was increased as to the irradiated time.

고려인삼에서 분리된 Penicillium sp.의 동정 및 열저항성 (Identification and Thermal Resistance of Penicillium sp. Isolated from Korean Ginseng)

  • One kind of microorganism was isolated and identified from Korean fresh, white and red ginseng, and the effect of a preservative, sodium benzoate on the microorganism and its thermal resistant propertues were studied. The results obtained were as follows. The predominant strain on ginseng and ginseng products was identified as Penicillium sp. The strain showed perithecium structure producing ascospores. The growth of the strain was slightly inhibited at 0.0571 concentration of sodium benzoate. The minimal inhibitory concentration (MIC) of sodium benzoate against the strain was 0.26%. The D value of the strain at 56, 59, $62^{\circ}C$ were 9.9, 5.0 and 4.5 min, respectively.

Ginsenoside Rg1 promotes browning by inducing UCP1 expression and mitochondrial activity in 3T3-L1 and subcutaneous white adipocytes

  • Background: Panax ginseng Meyer is known as a conventional herbal medicine, and ginsenoside Rg1, a steroid glycoside, is one of its components. Although Rg1 has been proved to have an antiobesity effect, the mechanism of this effect and whether it involves adipose browning have not been elucidated. Methods: 3T3-L1 and subcutaneous white adipocytes from mice were used to access the thermogenic effect of Rg1. Adipose mitochondria and uncoupling protein 1 (UCP1) expression were analyzed by immunofluorescence. Protein level and mRNA of UCP1 were also evaluated by Western blotting and realtime polymerase chain reaction, respectively. Results: Rg1 dramatically enhanced expression of brown adipocyte-especific markers, such as UCP1 and fatty acid oxidation genes, including carnitine palmitoyltransferase 1. In addition, it modulated lipid metabolism, activated 5' adenosine monophosphate (AMP)-activated protein kinase, and promoted lipid droplet dispersion. Conclusions: Rg1 increases UCP1 expression and mitochondrial biogenesis in 3T3-L1 and subcutaneous white adipose cells isolated from C57BL/6 mice. We suggest that Rg1 exerts its antiobesity effects by promoting adipocyte browning through activation of the AMP-activated protein kinase pathway.

인삼의 품질 연구 현황 및 문제점 (Status of Research on Ginseng Quality and its Problem)

  • Ginseng has been used as a medicinal herb in the world for more than two thousand years. Inspection of the quality of ginseng was made since many hundred years ago. Ginseng quality has been graded by several methodes, based on saponin contents, number of ginsenosides, shape of root and tissue elaborateness. In present. ginseng products are usually evaluated by saponin contents and number of ginsenosides. On the other hand, fresh and manufactured ginseng roots such as red. white and semi-red ginseng, Taegeuk Sam, are mostly graded by root shape such as root development and skin (epidermis) color, and tissue elaborateness. which is a conventional grading method. However, the root shape grading method has a risk of overlooking real medicinal properties of ginseng. So. both the medicinal ingredients and the conventional grading method should be considered for the proper evaluation of ginseng quality. Therefore, for the establishment of better method in evaluating ginseng quality, the relationships of root shape and useful components are required to be studied.

인삼 Polyacetylene 성분의 추출방법 비교연구 (Comparative Studies on Methods of Extracting Polyacetylene Compounds from White Ginseng)

  • 백삼의 주요 polyacetylene 성분인 panaxynol과 panaxydol을 capillary-GC(FID)로 정량하고 몇 가지 추출용매(석유 ether, dichloromethane, ether, ethyl acetate, acetone, acetonitrile and methanol) 및 추출방법(진탕추출, Soxhlet 추출 또는 환류추출)에 따른 이들 성분의 추출율을 비교하였다. 이 때, GC column은 SPB-1 fused silica capillary (0.25 mm id $\times$30 m, Supelco)를 사용하였고, column oven 의 온도는 $200^{\circ}C$에서 $300^{\circ}C$까지 분단 $4^{\circ}C$씩 승온하였다. 추출용매에 따른 panaxynol과 panaxydol의 추출율은 methanol 에 의하여 환류추출하는 방법이 백삼 중 polyacetylene 성분을 최대로 추출하는 조건인 것으로 사료되었는데, 이 추출방법에 의하여 얻어진 4년근 백삼시료의 panaxynol과 panaxydol의 함량분석값은 각각 4.2mg/g과 6.4mg/g이었다.
