Organic matter is very important and essential factor to maintain productivity of paddy field. But as meat consumption and the demand of grain increase, the demand of forage also increased gradually in Korea. So the amount of organic matter in paddy field have been declined gradually by the reduction of return of rice straw for the forage use for cattle. There is not enough alternative resources for forage, we guess this trend of organic matter decline in paddy field would continue for the time being in Korea. So this study was performed to confirm the variation of organic matter and change of rice yield to select suitable rice cultivars which yield reduction is small in condition of organic matter decrease in paddy field. To confirm the change of rice growth and yield in condition of organic matter decrease in paddy field, we transplanted 10 rice cultivars which yield reduction are small in low fertilizer condition. We transplanted it Buan where double cropping of winter barley continues several years on June 10. Planting density were $70plants/3.3m^2$ Fertilizer amount was $N-P_2O_5-K_2O=9-4.5-5.7kg/10a$ and fertilizer split application of nitrogen was basal-tillering stage-panicle initiation = 50-20-30%. And in the other cultivation management, we observed rice standard culture of NICS. In paddy field where withdraw straws of barley and rice, the organic matter content showed tendency to decrease as the years go on. During rice cultivation season, organic matter decreased little by little, but it increased again after rice harvest season. Rice yield was more in order of Sodami, Chunghaejinmi and Saenuri. We judge that there is an advantage in rice yield of rice cultivars which have later heading date because of suitable ripening temperature. Although Sobibyeo and Shindongjinbyeo showed high yield, head rice yield decreased severely owing to chalky and cracked rice.
The individual production rate of Plecoglossus altivelis was studied from the 18th of June to the 25th of September (99 days) 1973, when the fish grow best at Gangjeong Stream, Jeju Do, as one of the ways to find out the reasons why the size of P. altivelis of Jeju Island is smaller than that of other places. The results obtained are as follows : 1) The indivudual production rate of the fish in the Gangjeong Stream during the summer 1973, was estimated as shown in Table 6. 2) The individual production rate of the fish and standing crop of algae on stones in the Gang-jeon Stream were no more than $7.2\%$ and $6.2\%$ respectively, when compared with those of the Ukawa River (which is located in Japan), thus the individual production rate of the fish at Gang-jeong Stream being very poor. 3) Population density was 5.2 individuals average per square meter during the research period. This is beyond the estimated density limit, i. e., 4.8 individuals per square meter, for the standing crop of algae on stones. 4) The period that the individual production rate of the fish was the most brisk during the summer in this stream was from August to September in 1973. 5) The poor individual production rate is primarily regarded due to insufficient gross ingestion. The reasons seemed to be insufficeient productivity of algae on the bottom stones, over-population density of the fish, and the lowered activity of ingestion due to low water temperature in daytime as compared with other places. Thus, the production rate of Plecoglossus altivelis in the Gangjeong Stream during the summer was greatly influenced by these factors.

The influences of protease on the removal of various protein soils from cotton fabrics were studied. The human epidermal stratum corneum, hemoglobin and casein were used as protein soils. The soiled fabrics were denatured by steaming for 30 min. before washing and laundered using Terg-O-Tometer under washing conditions. The removal efficiency was evaluated by analysis of protein on the fabrics before and after washing by means of copper-Folin method. The relations between the removal and the characteristics of protease were discussed. Also the degradation of protein were examined by microscopy. The seperation of human epidermal stratum corneum after hydrolysis was examined by SDS-PAGE. The results obtained were as follow : 1. The protein from the soiled cotton fabric was removed effectively by adding protease. The removal of protein was increased in proportion to increasing of the enzyme concentration up to a certain point, but it began to decrease above the point. The removal effect was high in the order of casein>human epidermal stratum corneum>hemoglobin. Especially the protein was more effectively removed in ADS solution(pH 9.5) containing enzyme. 2. When protease was used with ADS. the removal of protein was efficiently showed in relatively short time(5~15min.) compared to using ADS only. It is due to the properties of this enzyme that reacts with very short time. 3. Even at low temperature the removal efficiency of enzyme was relatively higher compared with the activity of enzyme. The removal of protein soil was increased up to a maximum near $50^{\circ}C$, and then decreased. 4. The removal of protein by protease was improved with the increase of alkalinity in the pH range from 9.5 to 11.0 but it began to decrease above pH 11.0. 5. According to the increase of mechanical agitation, the removal effect was increased. But the removal efficiency of protease was more effective compared with the agitation in detergency. 6. According to the SDS-PAGE separation and micrograph it was confirmed that the human epidermal corneum was effectively hydrolysed by the enzyme added. So the fragments of protein were removed more efficiently by means of the interfacial reaction of AOS.
Sn-Bi eutectic alloy has been widely used as one of the key solder materials for step soldering at low temperature. The Sn-58Bi solder paste containing chloride flux was adopted to compare with that using the chloride-free flux. The paste was applied on the electroless nickel-immersion gold (ENIG) surface finish by stencil printing, and the reflow process was then performed at $170^{\circ}C$ for 10 min. After reflow, the solder joints were aged at $125^{\circ}C$ for 100, 200, 300, 500 and 1000 h in an oven. The interfacial microstructures were obtained by using scanning electron microscopy (SEM), and the composition of intermetallic compounds (IMCs) was analyzed using energy dispersive spectrometer (EDS). Two different IMC layers, consisting of $Ni_3Sn_4$ and relatively very thin Sn-Bi-Ni-Au were formed at the solder/surface finish interface, and their thickness increased with increasing aging time. The wettability of solder joints was investigated by wetting balance test. The mechanical property of each aging solder joint was evaluated by the ball shear test in accordance with JEDEC standard (JESD22-B117A). The results show that the highest shear force was measured when the aging time was 100 h, and the fracture mode changed from ductile fracture to brittle fracture with increasing aging time. On the other hand, the chloride flux in the solder paste did not affect the shear force and fracture mode of the solder joints.
Glucosidic bond at the $C_{20}$ position of the sapogenins was hydrolyzed easily in the lower pH, higher temperature and longer time to give prosapogenins and sugars. The glucosidic bond of saponin at the $C_3\;of\; ginsenoside-Rb_1\;$, which is secondary carbon, was relatively stable due to the low electron density of -0.2. But the bond of saponin at the $C_{20}$ position, which is tertiary carbon with the relatively high electron density of -0.3, was liable to be hydrolyzed even in weak acidic solution by the increase of heating time. On the other hand, fresh and white ginseng contained 4.12 mg/g, 13.05 mg/g of citric acid, 0.68 mg/g, 2.18 mg/g of malonic acid, 1.13 mg/g, 3.68 mg/g of oxalic acid, 2.68 mg/g, 8.62 mg/g of malic acid and 0.13 mg/g, 0.46 mg/g of succinic acid, respectively. Ginseng saponins were very stable in ginseng extract neutralized with sodium carbonate or sodium bicarbonate corresponding to the equivalent amount of the total organic acid in the ginseng.
This study aims to provide the physicochemical properties of three kinds of organosmectites which can be diversely used in industries. Some properties of them were compared with Nasmectite. Three kinds of organosmectites such as Hexadecyltrimethylammonium(HDTMA), Benzyldimethyltetradecylammonium(BDTDA), and Cetylpyridinium(CP) exchanged smectites were manufactured for this study. Three types of organosmectites showed the alkaline character(pH 9), very low swelling property and viscosity, and a fast flocculation behavior because of strong hydrophobic property in contrast to hydrophilic Nasmectite. Three organosmectites showed the strong interlayer expansion with basal spacing from $19\AA$ to $23\AA$ compared with the Nasmectite of about 12 $\AA$. Organic cations such as HDTMA, BDTDA, and CP exchanged into smectite were completely decomposed in the temperature range from $250^{\circ}C$ to $400^{\circ}C$. Generally, three organosmectites showed the similar mineralogical, physicochemical and thermal properties. But their properties are quite different from Nasmectite. Considering economically, CP exchanged smectite would be used for the diverse utilization field in the future time.
This study is aimed to emphasize the significance of ore selection in lime manufacturing through the evaluation of applied-mineralogical impact factors of crude ores controlling calcination characteristics for some domestic limestones used currently for lime manufacturing. To do this work, systematic characterization and determination were carried out for the limestone ores and their calcination products in a fixed calcining condition (target temperature: $1000^{\circ}C$, retention time: 30 minutes, 2, 4, 10, 16 hours), and the results were correlated and discussed. Selected high-Ca limestones in this study are as much as > 98 wt%, but they are somewhat diverse in crystallinity, texture, and impurity composition. Synthesized quicklimes are varied depending on such a difference in ore characters. The Pungchon limestone has relatively very low calcination rate, and the limestones from the Gabsan formation and the Jeongseon formation exhibit good quality in calcination rate and decrepitation. Among these samples, the limestone ore from the Jeongseon formation is evaluated to be the best for crude ore in manufacturing of highly-reactive quicklime. In addition, it is characteristic that the Gabsan limestone comparative rich in Fe-bearing mineral such as pyrite and goethite is more conspicuous in sintering effect.
ERM proteins transfer the methyl group to $A_{2058}$ in 23S rRNA, which reduces the affinity of MLS (macrolide-lincosamide-streptogramin B) antibiotics to 23S rRNA, thereby confer the antibiotic resistance on micro-organisms ranging from antibiotic producers to pathogens and are classified into monomethyltransferase and dimethyltransferase. To investigate the differences between mono- and dimethyltransferase, tirD, a representative monomethylase gene was cloned in Escherichia coli from Streptomyces fradiae which contains ermSF, dimethylase gene as well to overexpress the TlrD for the first time. T7 promoter driven expression system successfully overexpress tlrD as a insoluble aggregate at $37^{\circ}C$ accumulating to around 55% of the total cell protein but unlike ErmSF, culturing at temperature as low as $18^{\circ}C$ did not make insoluble aggregate of protein into soluble protein. Coexpression of Thioredoxin and GroESL, chaperone was not helpful in turning into soluble protein either as in case of ErmSF. These results might suggest that differences between mono- and dimethylase could be investigated on the basis of the characteristics of protein structure. However, a very small amount of soluble protein which could not be detected by SDS-PAGE conferred antibiotic resistance on E. coli as in ErmSF which was expected from the activity exerted by monmethylase in a cell.
Bacteria in the viable but nonculturable (VBNC) state fail to produce colonies on routine bacteriological media, but are still alive in the state of very low metabolic activity. The aim of the present study was to induce the VBNC state of the Edwardsiella tarda using sea water microcosm under starvation conditions at $10^{\circ}C$ and to investigate resuscitation of the VBNC cells in temperatures changed from 10 to $25^{\circ}C$, with and without additives. E. tarda entered into the VBNC state within about 42-84 days of incubation in the microcosm. Throughout this period, the total cell counts as determined using acridine orange direct counting remained near the original inoculum level of ${\sim}10^8cells/ml$. The live cell counts measured with direct viable counting, on the other hands, declined to ${\sim}10^4cells/ml$. When the VBNC cells were incubated with addition of yeast extract, fish muscle extract or serum at $25^{\circ}C$, the ratios of resuscitated samples were 37%, 23%, and 37%, respectively. The characteristics of resuscitated E. tarda were consistent with those of the original E. tarda. When the resuscitated E. tarda were intraperitoneally injected into olive flounders, all fishes died within 5 days, indicating that the VBNC E. tarda might retain its pathogenic potential. Therefore, E. tarda under starvation conditions in the winter enter into the VBNC state and the VBNC E. tarda cells resuscitated at summer and autumn seawater temperature are considered to be pathogen continuously to olive flounder on the southern coast of Korea.
Some supported gold catalysts were prepared by impregnation and coprecipitation methods. Effect of gold addition and active sloe formation were studied by investigating particle sizes of gold, amounts of oxygen adsorbed, adsorption properties of CO and NO, and reduction and oxidation properties, etc.. The gold particles of the catalyst by impregnation were irregular and very large as 30~100 nm, but those by coprecipitation were uniform and ultra-fine as about 4 nm. On $Au/Al_2O_3$ catalyst, the addition of gold to inactive $Al_2O_3$ caused the decomposition of $N_2O$, and CO was not irreversibly adsorbed while $O_2$ was atomically and irreversibly adsorbed. The adsorption sites of oxygen were attributed to the active sites which were restricted to the circumference of hemispherical gold particle-support interface rather than all atoms on the surface of gold particle. Also, CO was reversibly and irreversibly adsorbed on $Al_2O_3$ at low temperature, and the addition of gold weakened both reversible and irreversible adsorptions. The affinity for CO on $Au/Co_3O_4$ catalyst decreased conspicuously compared to $Co_3O_4$. The effect of gold addition did not appear in reduction step but did remarkably in reoxidation step; the added gold promoted the reoxidation of the reduced cobalt atoms.
