Identification of the Vibrios Isolated from a Shellfish, Sunset Shell, (Soletellina olivacea) (빛조개(Soletellina olivacea)로부터 분리된 비브리오의 생화학적 성상)

  • This study was conducted to investigate the vibrio flora in an edible shellfish. sunset shelfish. Soletelliim olivacen. which were collected in the estuarine area. Dadaepo near Nakdong River in Korea lkoin January 1997 to November 1997. Including five pathogemc vibrios (Vibrio alginolyticus, Vibrio pamhaemol~~licz~s, Vibrio cholerae non-01. Vibrio vulnificus, and Vihrio jl~~vinlis), a lotal of eight species of vlbr~os (Vi61-io splendidrrs biovar I, Vibrio splendidus biovar 11, Vibrio snlrnonicida and Vibrio tr,~biasllii) were identified from the sunset shellfish by heir biochemical characters. The isolation of Vihrio pamhaemolyricns, which is known not to grow below $15^{\circ}C$, in winter season indicates that the sunset shelllish is one oT the natural owl.- wintering hosts for Vibrio parahuemolyticus.

Vibrios Isolated from Diseased Marine Culturing Fishes in Korea (질병의 증상을 보이는 해수 양식 어류에서 분리한 비브리오속 세균)

  • A bacteriological survey in maricultured fish farms was conducted in Korea from 2002 to 2004. A total number of 166 Vibrio isolates were collected from diseased fishes, including olive flounder (133 isolates), black rock fish (8 isolates), red sea bream (6 isolates), shrimp (5 isolates), black sea bream (4 isolates), abalone (3 isolates) and other fishes (7 isolates). All isolates were phenotypically characterized and then groups were obtained using the traditional biochemical test. Representative isolates of each group were genotypically characterized with sequencing the 16S rRNA genes or 16S-23S intergenic space genes. Above 14 species of Vibrio were identified as V. ichthyoenteri (45 strains), V. alginolyticus (34 strains), V. harveyi (32 strains), Ph. damselae subsp. damselae (Formerly V. damsela, 10 strains), V. campbellii (6 strains), V. costicola-like (6 strains), V. fisheri (5 stains), V. fluvialis (4 strains) and others Vibrio sp. (24 strains) by combining of biochemial and genetic characteristics.

Comparison of Automated Systems for Identification of Vibrio Species (Vibrio 속 세균 동정에 대한 자동화동정 시스템의 비교)

  • Twenty one Vibrio strains were isolated from costal water and their phenotypic properties were determined. We identified bacterial isolates by using automated identification systems, Vitek and MIDI, and compared their identification results with each other. The comparison of them provided identical species information for only one isolates, TL33( V. alginolyticus). From Vitek and MIDI,16 (76%), and 6 (29%) out of 21 isolates were identified to the species level, respectively. Vitek was more useful than MIDI for identification of Vibrio species.

  • The objectives of this study are to produce monoclonal antibodies (MAbs) against Vibrio parahaemolyticus and to develop an immuno-selective filtration (ISF) method for the rapid and sensitive detection of V. parahaemolyticus. The characterization of the MAb produced from HKVP 4H9-9 hybridoma cell was validated by enzyme-linked immunosorbent assay (ELISA) and western blot. The produced MAb was specific to V. parahaemolyticus and showed weak cross-reaction to V. alginolyticus, V. vulnificus and Staphylococcus aureus. After optimization of the method, $5{\times}10^1cell/mL$ of V. parahaemolyticus in a pure culture could be detectable. Although weak cross-reactivity to V. vulnificus, V. alginolyticus and Staphylococcus aureus was observed, the ISF was confirmed to be highly specific to V. parahaemolyticus. Especially, the ISF showed the most sensitivity compared to the immunoassays currently reported is easier to perform and quicker than ID-ELISA.

Rapid Identification of Vibrio Species Isolated from the Southern Coastal Regions of Korea by MALDI-TOF Mass Spectrometry and Comparison of MALDI Sample Preparation Methods

  • Vibrio species are generally recognized as pathogens predominant in seafood along coastal areas. The food industry has sought to develop efficient microbial detection methods. Owing to the limits of conventional methods, this study aimed to establish a rapid identification method for Vibrio isolated from Korea, based on matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Four different preparation procedures were compared to determine the appropriate means to pretreat Vibrio species, using 17 isolates and five reference strains. Extended direct transfer and full formic acid extraction methods using bacterial colonies on agar plates revealed very low identification rates. Formic acid and trifluoroacetic acid (TFA) extractions using bacterial broth cultures were also performed. All Vibrio isolates and reference strains prepared by TFA extraction were successfully identified to the species level (17/22, 77.3%) and to the genus level (5/22, 22.7%). Thus, TFA extraction was considered the most appropriate method to pretreat Vibrio species for MALDI-TOF MS. The remaining 33 isolates and two reference strains were prepared by TFA extraction and analyzed by MALDI-TOF MS. Overall, 50 isolates were identified to the species level (40/50, 80%) and to the genus level (10/50, 20%). All isolates were identified as 43 V. alginolyticus, six V. parahaemolyticus, and one V. vulnificus species. V. alginolyticus and V. parahaemolyticus were isolated from fish offal (87.5% and 12.5%, respectively), seawater (91.3%, 8.7%), and shellfish (62.5%, 37.5%), whereas V. alginolyticus and V. vulnificus were isolated from sediment (90.9% and 9.1%, respectively). This study established a reliable system of MALDI-TOF MS preparation and analysis for Vibrio identification.

High Concentration of Red Clay as an Alternative for Antibiotics in Aquaculture

  • The use of antibiotics in aquaculture raises environmental and food safety concerns because chronic exposure of an aquatic ecosystem to antibiotics can result in the spread of antibiotic resistance, bioaccumulation of antibiotics in the organisms, and transfer of antibiotics to humans. In an attempt to overcome these problems, high-concentration red clay was applied as an alternative antibiotic against the following common fish pathogens: Aeromonas salmonicida, Vibrio alginolyticus, and Streptococcus equinus. The growth of A. salmonicida and V. alginolyticus was retarded by red clay, whereas that of S. equinus was promoted. Phase contrast and scanning electron microscopy analyses confirmed the attachment of red clay on cell surfaces, resulting in rapid gravitational removal and cell surface damage in both A. salmonicida and V. alginolyticus, but not in S. equinus. Different cell wall properties of grampositive species may explain the unharmed cell surface of S. equinus. Significant levels of oxidative stress were generated in only the former two species, whereas significant changes in membrane permeability were found only in S. equinus, probably because of its physiological adaptation. The bacterial communities in water samples from Oncorhynchus mykiss aquacultures supplemented with red clay showed similar structure and diversity as those from oxytetracycline-treated water. Taken together, the antibiotic effects of high concentrations of red clay in aquaculture can be attributed to gravitational removal, cell surface damage, and oxidative stress production, and suggest that red clay may be used as an alternative for antibiotics in aquaculture.

  • Fifty four strains of pathogenic vibrios were isolated from the Gwangan Beach from May to October, 2002. The isolated vibrios were composed of 7 different species: Vibrio parahaemolyticus, V. cholerae non-O1, V. alginolyticus, V. vulnificus, V. hollisae, V. fluvialis, ane V. mimicus. In the detection rate, V. parahaemolyticus was most predominant as $46\%$(25/54). From the isolated strains, only 25 strains have hemolytic activity or 25 strains only proteolytic activity on agar plates. Eleven strains showed both hemolytic and proteolytic activity. No strains showed urease activity. All strains of V parahaemolyticus did not show hemolytic activity, while V. cholerae non-O1 strains showed $\beta$ hemolytic activity. Kanagawa phenomena of pathogenic vibrios did not accord with hemolytic activity of the culture supernatant at the late log phase. Some strains showed high hemolytic activity despite having proteolytic activity, but some weak hemolytic activities despite having no proteolytic activity.

Respiratory Chain-Linked Components of the Marine Bacterium Vibrio alginolyticus Affect Each Other

  • The aerobic respiratory chain of Vibrio alginolyticus possesses two different kinds of NADH oxidase systems, i.e., an $Na^{+}$-dependent NADH oxidase system and an $Na^{+}$-independent NADH oxidase system. When deamino-NADH, which is the only substrate for the $Na^{+}$-dependent NADH oxidase system, was used as a substrate, the maximum activities of $N^{+}$-dependent NADH: quinone oxidoreductase and $Na^{+}$-dependent NADH oxidase were obtained at about 0.06 M and 0.2 M NaCl, respectively. When NADH, which is a substrate for both $Na^{+}$-dependent and $Na^{+}$-independent NADH oxidase systems was used as a substrate, the NADH oxidase activity had a pH optimum at about 8.0. In cGntrastl when deamino-NADH was used as a substrate, the NADH oxidase activity had a pH optimum at about 9.0. On the other handle inside-out membrane vesicles prepared from the wild-type bacterium generated only a very small $\Delta$pH by the NADH oxidase system, whereas inside-out membrane vesicles prepared from Napl, which is a mutant defective in the $Na^{+}$ pump, generated $\Delta$pH to a considerable extent by the NADH oxidase system. On the basis of the results\ulcorner it was concluded that the respiratory chain-linked components of V. atginotyticus affect each other.

Assessment of the Effect of a Disinfectant to Control Vibrio Strain from White Leg Shrimps Cultured in Korea (국내산 양식 흰다리새우 유래의 비브리오균에 대한 소독제의 방제 효과 검증)

  • White leg shrimps which were cultured in a private aquaculture farm showed abnormal swimming behavior and appetite reduction in July 2023. Then, gradual mortality was observed in the aquaculture farm. During the diagnosis, bacterial strain KNUAFVaSHP08147 was isolated from the hepatopancreas of the dead shrimps. Based on the sequence of 16S rRNA gene, KNUAFVa-SHP08147 was proved to be Vibrio alginolyticus, showing 99.71 % nucleotide identity with that of V. alginolyticus MG54 and GS MYPK1. According to the result of the growth of KNUAFVa-SHP08147, the treatment a commercial disinfectant, VirkonTMS was not proved to be effective to prohibit the growth of Vibrio.
