노니 부정근으로부터 유효성분의 최적 추출조건 (Optimal Extraction Conditions of Active Components from the Adventitious Roots of Noni (Morinda citrifolia))

  • 노니 부정근 유효성분의 효율적인 추출조건을 규명하고자 용매 종류, 물과 메탄올의 비율(물, 20, 40, 60, 80, 100%), 추출시간 및 추출방법을 달리하여 실험을 진행하였다. 안트라퀴논, 페놀성 화합물 및 플라보노이드의 용매별 추출한 결과 1시간 동안 초음파 추출 시 60-80% 메탄올에서 효과적이었으며, 환류냉각 추출기로 2시간 추출 시 80% 메탄올에서 효과적이었다. 노니 부정근의 유효성분에 대한 추출방법 및 시간에 따른 추출효율을 비교하기 위하여 교반추출, 환류냉각추출, 초음파추출, 균질기추출, 초고압추출, 침지추출을 실시하였다. 페놀성 화합물은 초음파추출에서 가장 높은 함량을 보였으며 안트라퀴논과 플라보노이드 함량은 환류냉각추출에서 가장 높게 나타났다.

  • An advanced extraction method by ultrasonic extraction with applied solid phase extraction (SPE) has been developed for the determination of simultaneous eight major ginsenosides, namely ginsenosides Rg1, Re, Rf, Rb1, Rg2, Rc, Rb2, and Rd in the root of Panax ginseng. Four extraction methods including n-BuOH reflux extraction (Method A), 70% EtOH reflux extraction (Method B), 50% MeOH reflux extraction with SPE (Method C), and 50% MeOH ultrasonication with SPE clean-up process (Method D) were investigated for the determination of eight major ginsenosides. Total contents of ginsenosides were highest by extraction of Method C as $2.408{\pm}0.011%$. However, Method D was evaluated as relatively simpler and more efficient method due to short extraction time, small solvent consumption and less expensive, compared to conservative reflux method. Ginsenosides were also satisfactorily separated with good resolution and the accuracy range was between 1.05 and 4.06% as relative standard deviation (RSD) by Method D. SPE condition and HPLC condition were further optimized for determination of eight major ginsenosides by the ultrasonic extraction method. Conclusively, ultrasonic extraction of 2 g sample of ginseng using ultrasonic bath and 1 loading for SPE was evaluated as proper condition for extraction of ginseng.

6dB Drop법에 의한 용접 결함 초음파 신호의 카오스성 평가 (Chaoticity Evaluation of Ultrasonic Signals in Welding Defects by 6dB Drop Method)

  • This study proposes the analysis and evaluation method of time series ultrasonic signal using the chaotic feature extraction for ultrasonic pattern recognition. Features extracted from time series data using the chaotic time series signal analysis quantitatively welding defects. For this purpose analysis objective in this study is fractal dimension and Lyapunov exponent. Trajectory changes in the strange attractor indicated that even same type of defects carried substantial difference in chaoticity resulting from distance shills such as 0.5 and 1.0 skip distance. Such differences in chaoticity enables the evaluation of unique features of defects in the weld zone. In experiment fractal(correlation) dimension and Lyapunov exponent extracted from 6dB ultrasonic defect signals of weld zone showed chaoticity. In quantitative chaotic feature extraction, feature values(mean values) of 4.2690 and 0.0907 in the case of porosity and 4.2432 and 0.0888 in the case of incomplete penetration were proposed on the basis of fractal dimension and Lyapunov exponent. Proposed chaotic feature extraction in this study enhances ultrasonic pattern recognition results from defect signals of weld zone such as vertical hole.

  • In order to extract the curcuminoid such as curcumin, demethoxycurcumin (DMC), and bisdemethoxycurcumin (BDMC) in turmeric (Curcuma longa), solvent extraction methods (dipping and ultrasonic extraction method) and solid-phase extraction (SPE) were used. RP-HPLC (reverse-phase high-performance liquid chromatography) and TLC (thin-layer chromatography) were used for identification and analysis the three curcuminoid. From the experimental results, it is evident that the percentage of curcuminoid extracted from turmeric by ultrasonic extraction method was higher than dipping method. The percentage of curcumin extracted from turmeric by pure methanol was higher than any aqueous methanolic composition. Moreover, the total peak area of three curcuminoid was above 92% in RP-HPLC using solid-phase extraction. These results will form a database for investigating the constituents of natural products and the resources of pharmaceutical, nutrition, and cosmetic products.

  • 본 연구에서는 추출용매 물 100%을 이용하여 다양한 초음파 에너지(35, 72, 170 KHz)와 추출 시간(30, 60 min)에 의한 고삼으로부터 식물성에스트로겐 제니스테인 및 포르모노네틴의 추출량과 일반성분의 영향을 비교하였다. 전처리 단계는 초음파추출, 여과, 농축, 막분리로 구성되었다. 추출된 용액은 역상 고성능 액체크로마토그래피 (HPLC)를 사용하여 분석하였다. 이동상 조성은 A는 물/아세트산(99.9/0.1 vol%), B는 아세토니트릴/아세트산(99.9/0.1 vol%)이며, A/B를 80/20~65/35 vol%로 60분 동안 선형적으로 변화시켰다. 실험결과에 의하면 일반성분은 탄수화물(0.255~0.413%)을 제외한 나머지 성분들의 함량은 거의 비슷하게 확인되었다. 또한 주파수 170 KHz, 60 min에서 추출량이 3.17 g으로 추출 수율이 가장 우수하였고 천연물관련 화학 및 생물학 연구에 기초자료로 도움이 될 것이다.

  • Two extraction techniques, Ultrasonic and Microwave extraction method, were tested for the determination of arsenic in contaminated soil SRM (Montana Soil). The extraction mixture was prepared by mixing 1 M ortho-phosphoric acid and 0.1 M ascorbic acid. This extractant was known to preserve arsenic species. The appropriate extraction time was 10 min to 20 min and the recovery rate was about 80%. A coupled system, SPE-HG-ICP-AES, was used for the determination of inorganic arsenic species. The detection limit was around 2 $\mu\textrm{g}$/1 and the linearity of calibration curve was better than $R^2$=0.99.

추출조건이 노니 부정근에 함유된 rubiadin의 추출 효율에 미치는 영향 (Effects of Extraction Condition on Extraction Efficiency of Rubiadin in Adventitious Roots of Noni (Morinda citrifolia))

  • 노니($Morinda$ $citrifolia$) 부정근의 주요 성분인 rubiadin은 간 보호 효과가 있어 제약산업에서 높은 가치가 있다. 노니 부정근의 주요 유효성분인 rubiadin의 효율적인 추출조건을 규명하고자 용매 종류, 물과 메탄올의 비율(물, 20%, 40%, 60%, 80%, 100%), 추출시간 및 추출방법을 달리하여 추출하였다. 노니 부정근에서의 rubiadin의 함량분석은 HPLC 이용 분석조건을 확립하였으며, C-18 컬럼을 사용하여 280nm에서 메탄올과 물로 농도구배를 주어 분석하였다. 용매별 추출효율은 메탄올(0.08%) > 에탄올(0.05%) > 아세토니트릴(0.03%) > 아세톤(0.02%) 및 메틸렌클로라이드(0.02%) 순으로 증가하였다. 메탄올에 물의 혼합 비율을 달리하여 초음파 추출기로 1시간 동안 추출한 결과 60% 메탄올(0.21%) > 80% 메탄올(0.13%) > 100% 메탄올(0.07%) 및 40% 메탄올(0.07%) 순으로 효과적이었으며, 환류냉각 추출기로 2시간 동안 추출한 결과 60% 메탄올(0.21%) > 40% 메탄올(0.17%) > 80% 메탄올(0.14%) 용매처리구 순으로 효율이 좋았다. 추출방법 및 추출시간에 따른 rubiadin의 추출 효율을 비교하기 위해 환류냉각추출 및 초음파추출, 진탕추출법으로 추출하였다. 추출방법 및 추출시간에 따른 rubiadin의 추출효율은 초음파추출 및 진탕추출에서 각각 8 및 24시간 동안 추출했을 때 효율적이었다.

초음파를 이용한 오죽으로부터 Flavone C-glycosioes의 추출 및 항산화활성분석 향상 (Enhanced extraction and Antioxidant activity analysis of Flavone C-glycosides from Black bamboo using Ultrasonic wave)

  • 오죽으로부터 HPLC On-line $ABTS^{+}$ screening기법을 사용하여 Flavone C-glycosides의 특성중의 하나인 항산화활성을 빠르게 분석하였으며, 오죽으로부터 homoorientin와 orientin의 추출을 다양한 초음파 주파수와 시간의 추출방법을 적용하였다. 전처리한 추출액에 포함된 오죽의 Flavone C-glycosides을 분석하고 최적의 추출조건을 실험적으로 모색하였다. 실험결과에 의하면 전체 수율에서는 Frequency 72 KHz, 60분에서 3.37 mg으로 추출 효율이 높았고, homoorientin와 orientin의 항산화활성은 Frequency 35 KHz, 30분으로 추출한 시료가 73.8%와 14.8%로 가장 우수하였다.

추출방법에 따른 미숙사과 추출물의 유용성분 함량 및 항산화 활성 (Effective Component Contents and Antioxidative Activities of Unripe Apple by Extraction Methods)

  • The purpose of this study was to determine the optimum extraction method suitable for the availability of biological activities in unripe apples known to be rich in functional components. Unripe apples 'Hongro' picked on May 28th, 2019 were extracted by various extraction methods (hot water, ethanol, enzymatic pre-treatment, ultrasonic wave, and subcritical water) and their extracts were investigated yield, effective component contents, and antioxidant activities. Overall, the yields by the extraction solvent were higher in water than in organic solvent(ethanol) because water-soluble compounds were eluted from a polar solvent. Total phenol contents of the ultrasonic wave (ethanol) extracts were significantly higher in 6 times than hot water extract. Contents of flavonoid were highest in the ethanol extract at 29.14 mg QE/g. Contents of tannin and ursolic acid were also significantly higher in the ultrasonic wave (ethanol) extract. The DPPH radical and ABTS radical scavenging activities were the strongest in the ultrasonic wave (ethanol) extract. Correlation between effective components and antioxidant activities was high in the total phenol content with ABTS and the ursolic acid content with DPPH (p<0.01). The above results suggested that ultrasonic wave (ethanol) extract of unripe apples has the potential to act as a functional material.
