Antioxidative and Cellular Protective Effects of Lysimachia christinae Hance Extract and Fractions (금전초 추출물 및 분획물의 항산화 활성 및 세포 보호 효과)

  • In the present study, we investigated the antioxidative properties, cellular protective effects and component analyses of 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Lysimachia christinae Hance (L. christinae Hance). In the evaluation of antioxidative properties, the free radical scavenging activities ($FSC_{50}$) of 50% ethanol extract, ethyl acetate fraction and aglycone fraction were 146.8, 22.2 and $27.2{\mu}g/mL$, respectively and total antioxidant capacities ($OSC_{50}$) were 29.3, 2.9 and $4.5{\mu}g/mL$, respectively. The ethyl acetate fraction showed the highest free radical scavenging activity and total antioxidant capacity. Also, the cellular protective effects (${\tau}_{50}$) of 50% ethanol extract, ethyl acetate fraction and aglycone fraction on $^1O_2$ induced photohemolysis of human erythrocytes were 26.9, 57.5 and 103.9 min at $5{\mu}g/mL$, respectively. In particular, ${\tau}_{50}$ of the aglycone fraction exhibited a higher cellular protective effect than that of (+)-${\alpha}$-tocopherol (37.7 min). The cell viability of the ethyl acetate fraction on the UVB-induced cell damage increased up to 90.1%. In addition, the ethyl acetate fraction ($5-25{\mu}g/mL$) showed cellular protective effects on the $H_2O_2-induced$ cell damages in a dose-dependent manner. TLC, HPLC, UV-vis spectroscopy and LC-MS were used to analyse components of the ethyl acetate fraction and the main components were quercetin, kaempferol and their glycosides. In conclusion, L. christinae Hance extract/fraction can function as antioxidants to protect the skin exposed to UV radiation and may also be used as a novel functional cosmetic material, for example, an antioxidant against skin photoaging.

Antimicrobial, Antioxidant and Cellular Protective Effects of Houttuynia cordata Extract and Fraction (어성초 추출물 및 분획물의 항균, 항산화 및 세포보호활성)

  • This study was conducted to investigate the physiological activities of Houttuynia cordata extracts and fractions. H. cordata extracts were extracted with 50% ethanol and the ethyl acetate fractions were obtained from the extracts. Minimum inhibitory concentration (MIC) values of the ethyl acetate fraction for S. aureus and B. subtilis were $78{\mu}g/mL$ and $312{\mu}g/mL$, respectively, indicating the high activity against gram-positive bacteria. The free radical scavenging activity ($FSC_{50}$) for 1,1-diphenyl-2-picrylhydrazyl (DPPH) was higher in the ethyl acetate fraction with $12.00{\mu}g/mL$ compared to that of $27.15{\mu}g/mL$ for 50% ethanol extract. The total antioxidant activity ($OSC_{50}$) values for reactive oxygen species (ROS) produced in $Fe^{3+}-EDTA/H_2O_2$ system by a luminol-dependent chemiluminescence method were 2.91 and $0.983{\mu}g/ml$ for the 50% ethanol extract and ethyl acetate fraction, respectively. To investigate cellular protective effects on the HaCaT cell, the intracellular ROS scavenging activity was measured after UVB irradiation and the ethyl acetate fraction of H. cordata showed the activity in a concentration-dependent from $1.6{\mu}g/mL$ and a reduction rate of 54.3% at a maximum concentration of $12.5{\mu}g/mL$. Also, HaCaT cell protective effect against $H_2O_2$-mediated decreased the cell viability of the ethyl acetate fraction of H. cordata which significantly increased the cell viability from $0.8{\mu}g/mL$ and the maximum cell viability showed 86.9%. The ethyl acetate fraction of the H. cordata extracts was analyzed by TLC and HPLC. As a result, quercitrin, isoquercitrin, hyperoside, chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid, rutin and afzelin were identified. From the above results, it was suggested that the extracts and fractions of H. cordata have a potential to be applied in the field of cosmetics as a natural antioxidant/preservative capable of protecting the cell membrane from the oxidative stress by eliminating ROS and exhibiting the antimicrobial effect.

Improving the efficacy of Lespedeza cuneata ethanol extract on ultraviolet-induced photoaging (야관문 에탄올 추출물의 자외선 노출에 의한 피부 광노화 개선 효과)

  • This study evaluated the improving efficacy of Lespedeza cuneata ethanol extract on skin photoaging induced by ultraviolet (UV) irradiation. The total polyphenol and flavonoid contents of the extract were respectively $134.98{\pm}1.70$ and $16.20{\pm}0.05$ mg/g, respectively. The superoxide anion radical scavenging activity and electron-donating ability of the extract were shown to be dependent on concentration, and the antioxidant ability was shown to be more effective in superoxide anion radical scavenging activity than in electron-donating ability under the same concentration conditions. In the in vivo test conducted using hairless mouse with skin photoaging induced by UVB irradiation, the skin erythema of the groups treated with the extract (AS) reduced to 28% of the control, and the skin moisture content increased to 131%.. The extract treatment of the UV-damaged skin improved the morphological and histopathological state of the skin. Furthermore, the SOD, GST and CAT activities in the skin tissue of the AS group increased, and the XO activity and TBARS generation decreased. With regard to the genes related to the photoaging skin, the expression of PAK, p38, c-Fos, c-Jun, TNF-${\alpha}$ and MMP-3 in the skin of the AS group were found to have decreased. It was therefore concluded that Lespedeza cuneata ethanol extract can reduce wrinkle formation in the skin due to the regulation of the gene expression caused by the exposure to UVB light.

Antioxidant and Cellular Protective Effects of Moringa oleifera Leaves Extract (드럼스틱 잎 추출물의 항산화 및 세포보호 효과)

  • In this study, we investigated the antioxidative and cellular protective effects on HaCaT cells and erythrocytes of Moringa oleifera (M. oleifera) leaves extract and its fractions. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction of M. oleifera leaves. The free radical scavenging activity ($FSC_{50}$) of the extract and fractions of M. oleifera leaves were in the following order: 50% ethanol extract ($77.10{\mu}g/mL$) < ethyl acetate fraction ($20.63{\mu}g/mL$) < aglycone fraction ($17.00{\mu}g/mL$) by using the 1, 1-diphenyl-2-picrylhydrazyl. In $Fe^{3+}-EDTA/H_2O_2$ system using the luminol, reactive oxygen species (ROS) scavenging activities (total antioxidant capacity, $OSC_{50}$) of aglycone fraction ($OSC_{50}=0.63{\mu}g/mL$) was the strongest among all extracts, which was much higher than L-ascorbic acid ($1.50{\mu}g/mL$). In the $^1O_2$-induced cellular damage of erythrocytes, the cellular protective effects of 50% ethanol extract (${\tau}_{50}=46.9min$) and aglycone fraction (${\tau}_{50}=122.1min$) were higher than (+)-${\alpha}$-tocopherol (${\tau}_{50}=37.7min$), known as a lipophilic antioxidant at $10{\mu}g/mL$. After cell damage induced by $400mJ/cm^2$ UVB irradiation, the cellular protective effects of ethyl acetate and aglycone fraction of M. oleifera leaves extract were showed on the concentration from 0.20 to $1.56{\mu}g/mL$. These results suggest that M. oleifera leaves extract and its fractions can function as a natural antioxidant agent in cosmetics on skin exposed to UV radiation by protecting cellular membrane against ROS.

Study on the Expression of Matrix Metalloproteinase-1 by Promoter Polymorphism in Human Dermal Fibroblast (섬유아세포에서 프로모터 다형성에 의한 Matrix Metalloproteinase-1의 발현에 관한 연구)

  • The skin fibroblasts of different origins showed different expression levels of MMP-1 in response to TNF-α treatment or UV irradiation. We hypothesized that this is caused by polymorphism in the MMP-1 promoter region. To elucidate it, first of all, we analyzed and classified the genotype of the -1607 site of the MMP-1 promoter in 23 commercially available primary fibroblasts, and then we examined the expression of MMP-1 by TNF-α or UVB stimulation for each classified genotype. As a result of the analysis, fibroblasts with 6 1G/1G genotypes, 10 1G/2G genotypes, and 7 2G/2G genotypes were identified. Hs68 and Detroit 551 cell lines were confirmed to have 1G/2G genotypes. In the 1G/1G genotype, MMP-1 was expressed twice as high as that of the control group by TNF-α treatment, and was hardly expressed by UV light. In the case of the 1G/2G genotype, MMP-1 was expressed 2.45 fold higher by TNF-α treatment, and 1.4 fold by UV light than the control. In the case of the 2G/2G genotype, MMP-1 was expressed 1.35 fold by TNF-α treatment, and was highly expressed by 2.5 fold by ultraviolet rays compared to control. It can be estimated that MMP-1 expression is better induced in the 1G genotype by TNF-α and in the 2G genotype by UV light. In addition, it can be presumed that MMP-1 expression is increased by creating a site where the Ets transcription factor can bind by another G inserted at the -1607 position. These studies have not been conducted at all in fibroblasts in relation to skin aging, so it is an area that needs to be further studied in the future. In conclusion, since the skin is an organ that is affected by both intrinsic aging and photoaging at the same time, when analyzing the expression of MMP-1 as a target for improving skin aging, it is necessary to select cells with a genotype suitable for the experimental conditions of the study.

Antioxidant, Antimicrobial and Cellular Protective Effects of Lycopus lucidus Extract and Fraction (택란 추출물 및 분획물의 항산화, 항균 및 세포 보호 효과)

  • In this study, antioxidative, antibacterial and cytoprotective effects of the ethanol extract and ethylacetate fraction of Lycopus lucidus (L. lucidus) were compared and analyzed. Free radical scavenging activities ($FSC_{50}$) of the L. lucidus extract and fraction were found to be 65.1 and $64.9{\mu}g/mL$ respectively. In the $Fe^{3+}-EDTA/H_2O_2$ system, the reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) for the extract and fraction were 6.6 and $6.3{\mu}g/mL$, respectively which showed excellent total antioxidant abilities. The extract showed antibacterial activity against S. aureus, while the fraction showed in all the bacteria except for A. niger. The cytoprotective effect of L. lucidus extract was compared to that of the fraction and the effect against $^1O_2$-induced cellular damage of human erythrocytes (${\tau}_{50}$) was 51.3 and 73.7 min at $50{\mu}g/mL$, respectively. For the cytoprotective effect of keratinocytes damaged by $H_2O_2$ and UVB, the extracts did not show any efficacy but showed efficacy at $1-2{\mu}g/mL$, respectively. The fraction increased the cell viability up to 85.8 and 81.9%, respectively. As a result of intracellular ROS scavenging activity, the scavenging activity was observed at $1-2{\mu}g/mL$ of the fraction. From the results comparing the physiological activities of L. lucidus extract and the fraction, the ethylacetate fraction of L. lucidus has antioxidative effect similar to that of the extract whereas superior antimicrobial and cytoprotective effects than that of the extract. Overall, the ethylacetate fraction of L. lucidus protects cells from an external stress which can be used as a potential cosmetic material.

Antimicrobial and Antioxidant Activities of Perilla frutescens var. acuta Extract and Its Fraction and Their Component Analyses (자소엽 추출물의 항균 및 항산화 효과와 성분분석)

  • In this study, antimicrobial and antioxidative activities of Perilla frutescens var. acuta were investigated with 50% ethanol and the ethyl acetate fraction and also the components were analyzed. The minimum inhibitory concentration (MIC) of the ethyl acetate fraction for both Staphylococcus aureus and Pseudomonas aeruginosa were $78{\mu}g/mL$, indicating high antimicrobial effects. The free radical scavenging activity ($FSC_{50}$) and the reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system values of the ethyl acetate fraction were $25.90{\mu}g/mL$ and $1.40{\mu}g/mL$, respectively. After the cell damage induced by $400mJ/cm^2$ UVB irradiation, the cytoprotective effect of the ethyl acetate fraction of P. frutescens var. acuta showed the concentration dependent manner ranging from 2.0 to $16.0{\mu}g/mL$. The intracellular ROS inhibitory activity in HaCaT cells decreased to 28.6% and 40.7% for the 50% ethanol extract and ethyl acetate fraction, respectively at the concentration of $32{\mu}g/mL$. Components of rosmarinic acid, luteolin, apigenin, caffeic acid and ethyl caffeate were identified in the ethyl acetate fraction. These results suggest that the extract and fraction of P. frutescens var. acuta may be applied to the field of cosmetics as a natural material that protects the skin from an external environment by having antimicrobial and antioxidative activities.

Antimicrobial, Antioxidant and Cellular Protective Effects against Oxidative Stress of Anemarrhena asphodeloides Bunge Extract and Fraction (지모 뿌리 추출물과 분획물의 항균활성과 항산화 활성 및 세포보호 연구)

  • Extracts and fractions of Anemarrhena asphodeloides Bunge were prepared and their physiological activities and components were analyzed. Antimicrobial activities of the ethyl acetate and aglycone fractions were $78{\mu}g/ml$ and $31{\mu}g/ml$, respectively, for Staphylococcus aureus and $156{\mu}g/ml$ and $125{\mu}g/ml$, respectively, for Pseudomonas aeruginosa. 1,1-Diphenyl-2-picrylhydrazyl free radical scavenging activities ($FSC_{50}$) of 50% ethanol extract, ethyl acetate fraction, and aglycone fraction of A. asphodeloides extracts were $146.2{\mu}g/ml$, $23.19{\mu}g/ml$, and $71.06{\mu}g/ml$, respectively. The total antioxidant capacity ($OSC_{50}$) in an $Fe^{3+}$-EDTA/hydrogen peroxide ($H_2O_2$) system were $17.5{\mu}g/ml$, $1.5{\mu}g/ml$, and $1.4{\mu}g/ml$, respectively. The cytoprotective effect (${\tau}_{50}$) in $^1O_2$-induced erythrocyte hemolysis was 181 min with $4{\mu}g/ml$ of the aglycone fraction. The ${\tau}_{50}$ of the aglycone fraction was approximately 4-times higher than that of (+)-${\alpha}$-tocopherol (${\tau}_{50}$, 41 min). Analysis of $H_2O_2$-induced damage of HaCaT cells revealed that the maximum cell viabilities for the 50% ethanol extract, ethyl acetate fraction, and aglycone fraction were 86.23%, 86.59%, and 89.70%, respectively. The aglycone fraction increased cell viability up to 11.53% at $1{\mu}g/ml$ compared to the positive control treated with $H_2O_2$. Analysis of ultraviolet B radiation-induced HaCaT cell damage revealed up to 41.77% decreased intracellular reactive oxygen species in the $2{\mu}g/ml$ aglycone fraction compared with the positive control treated with ultraviolet B radiation. The findings suggest that the extracts and fractions of A. asphodeloides Bunge have potential applications in the field of cosmetics as natural preservatives and antioxidants.

Protective Effects of Complex Diet of Korean red ginseng and fish oil on UV-induced Photoaging (홍삼과 어유(魚油) 복합식이에 의한 UV-유도 광노화 억제효과)

  • Objective : UV irradiatiion causes skin-aging involving coarse wrinkles, thickning, dyspigmentation, and rough skin surface. This study was aimed to elucidate the anti-winkle activity of complex diet of Korean red ginseng (RG) and fish oil (FO) on UV-induced Photoaging. Methods : To investigated photo-protective effects of Korean red ginseng and fish oil on UV-induced damaged skins, SKH hairless female mice were randomly divided into six groups [control, UV, UV/RG, UV/FO, UV/RG/FO(low), UV/RG/FO(high)]and orally administered three times a week respectively. UV radiation was applied to the backs of the mice three times a week for 8 weeks. Expressions of matrix metalloproteinase (MMP)-3, MMP-13 and tissue inhibitor matrix metalloproteinase (TIMP)-1 in skin were measured by immunohistochemical staining. Results : In this study, UVB-induced epidermal hypertropy was diminished by RG group or FO group or complex group of RG and FO. Expression levels of MMP-3 and MMP-13 were reduced and expression level of TIMP-1 was increased by RG group or FO group or complex group of RG and FO. Especially MMP-3 and MMP-13 were markedly reduced by diet of FO and complex diet of RG and FO compared with untreated group. Conclusions : This results suggest that complex diet of RG and FO have a anti-wrinkle activity on UV-induced photo-aging and intrinsic aging.

Anti-Oxidant Effect and Anti-Inflammatory of Fermented Citrus unshiu Peel Extract by using Schizophyllum commune (치마버섯을 이용한 진피 발효 배양물의 항산화 및 항염 효과)

  • Citrus unshiu (C. unshiu) Markovich were dried peel of mandarin orange, of which fresh fruit was one of the famous foods in Korea and Eastern Asia. In the oriental medicine, C. unshiu peel was known to have a diuretic effect and to strengthen spleen function. Recently, natural flavonoids of C. unshiu peel have been investigated. In this study, C. unshiu peel extract containing flavonoid-glycosides was cultured with Schizophyllum commune (S. commune) mycelia producing ${\beta}$-glu- cosidase and its biological activities were investigated. ${\beta}$-glucosidase of S. commune mycelia converted the flavonoid-glycosides (rutin and hesperidin) into aglycones (naringenin and hesperetin). Fermented C. unshiu peel extract compounds were analyzed by HPLC system. The photoprotective potential of fermented C. unshiu peel extract was tested in human dermal fibroblasts (HDFs) exposed to UVA. Fermented C. unshiu peel extract extract also showed notable in vitro anti-inflammatory effect on cellular systems generating cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) metabolites. Also, UVB-induced production of interleukin-$1{\alpha}$ in human HaCaT cells was reduced in a dose-dependent manner by treatment with fermented C. unshiu peel extract. These results suggest that fermented C. unshiu peel extract may mitigate the effects of photoaging in skin by reducing UV-induced adverse skin reaction.
