Improved Therapeutic Profiles of PLA2-Free Bee Venom Prepared by Ultrafiltration Method

  • Bee venom (BV) has long been used in traditional Eastern and Western medicine for chronic inflammation, pain and skin therapy. Human exposure to BV, however, often causes unwanted adverse effects and is even fatal in some cases. Phospholipase $A_2$ ($PLA_2$) of BV is now suspected to play a key role in these adverse effects. We investigated the potential use of $PLA_2$-free bee venom (PBV) as a replacement for BV in cosmetic products. PBV prepared by molecular weight cut-off ultrafiltration exhibits a superior profile in comparison with regular BV, by inhibiting elastase activity and suppressing the induction of nitric oxide (NO) and metalloproteinase-9 (MMP-9), while retaining the effects of cell proliferation and protection against ultraviolet B (UVB)-induced damage in human dermal fibroblast cells. PBV thus appears to be more promising than BV as a cosmetic ingredient with a reduced potential for adverse reactions in the recipient.

Photoprotective Effects of Soybean Extract against UV-Induced Damage in Human Fibroblast and Hairless Mouse Model

  • Soy isoflavones have been reported to possess many physiological activities such as antioxidant activity and inhibition of cancer cell proliferation. This study investigated the photoprotective effects of soybean extract in human fibroblast cell line and hairless mice model. Human fibroblast was treated with soybean extract before and after ultraviolet B (UVB; 290-302 nm) irradiation. In the soybean extract treated group, the cells showed better resistance to ultraviolet (UV) than control group. The amount of type I collagen recovered from the soybean treated group was higher than the vehicle group exposed to UV-induced damage. Moreover, increased expression of metalloproteinases-1 as a result of UV irradiation was suppressed by the soybean extract. Female mice were orally administered soybean extract and irradiated with UVB light for 8 weeks. The effects of the soybean extract on the skin appearance, collagen deposition and epidermal thickness in the UV-damaged mouse skin were analyzed using histopathological methods. In soybean extract treated group, the skin had a better morphology than that of the control group. Furthermore, the amount of type I collagen was increased and overexpression of MMP-1 was reduced in the soybean extract group compared to vehicle group. Additionally, up-regulation of pro-inflammatory cytokines induced by UV irradiation was suppressed by dietary soybean extract treatment. It appears that soybean extract had a photoprotective effect, including anti-aging and anti-inflammatory effect, from UV-induced damage in not only human fibroblast, but also hairless mice. We confirmed that these effects were possibly due to promotion of collagen synthesis and inhibition of MMP-1 expression.

각질형성세포에서 ROS로 유도된 염증반응에 대한 능실 추출물 및 그 분획물의 항염 효과 (Fractionated Trapa japonica Extracts Inhibit ROS-induced Skin Inflammation in HaCaT keratinocytes)

  • 자외선은 외부적인 스트레스 자극인자로 작용하여 사람 각질형성세포에서 reactive oxygen species (ROS)와 비활성 코르티손을 활성 코르티솔로 전환시키는 효소인 $11{\beta}$-hydroxysteroid dehydrogenase type 1 ($11{\beta}$-HSD1)의 발현 및 활성을 증가시킨다고 알려져 있다. 또한, ROS가 증가된 피부에서는 염증 유발 사이토카인과 염증 매개 인자의 발현이 증가되어 결과적으로 염증반응을 일으키게 되는 원인이 된다. 본 연구에서는 각질형성세포(HaCaT)에서 $11{\beta}$-HSD1 억제제가 ROS 분해효소인 catalase의 생성을 회복시킴에 착안하여, $11{\beta}$-HSD1의 발현을 저해함과 동시에 ROS로부터 유도되는 염증 반응을 억제하는 천연물 소재를 발굴하고자 하였다. 그 중 능실 추출물과 그 분획물은 각각 $11{\beta}$-HSD1의 발현과 ROS 생성 증가를 억제하고, 염증성 사이토카인인 tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\alpha}$, $-1{\beta}$의 발현을 억제하였다. 또한, 자외선에 의해 유도되는 염증 매개인자인 cyclooxygenase (COX)-2, inducible nitric oxide synthase (iNOS), prostaglandin $E_2$ ($PGE_2$)의 생성을 저해하였다. 따라서 본 연구 결과로부터 능실 추출물 및 그 분획물은 $11{\beta}$-HSD1의 발현을 억제함과 동시에 ROS에 의해 유발된 피부 염증 반응을 효과적으로 저해함을 확인하였다.

Effects of Draconis Resina on the Collagenase Activities and the Procollagen Synthesis in Hs68 Human Fibroblasts, and Tyrosinase Activity

  • Objectives : Draconis Resina (DR), the resin of Daemonorops draco Bl., is used to circulate the blood and to stop bleeding. It also has been used to generate flesh including ulceration. The present study investigated the effects of DR extract on collagen metabolism in human fibroblasts and tyrosinase activity in mushroom tyrosinase.Methods : The effect of DR extract on type I procollagen production (collagen type I synthesis) and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts cell line. Hs68 cells after ultraviolet B (UVB, 312 nm) irradiation was measured using the enzyme - linked immunosorbent assay (ELISA). The tyrosinase activity was also measured to find out the whitening effects in mushroom tyrosinase by ELISA method.Results : There was no cytotoxicity at DR extract at concentrations of 10 μg/ml, 30 μg/ml, and 100 μg/ml. DR extract significantly inhibited the increase of collagenase activity, whereas it did not show on the reduction of type I procollagen in UVB damaged Hs68 cells. DR extract did not reduce the L - DOPA oxidation. However, it significantly reduced the tyrosinase activity by DR extract at concentraions of 0.1 mg/ml, 1 mg/ml and 10 mg/ml.Conclusions : In conclusion, DR showed the anti-wrinkle and whitening effects via the inhibition of collagenase production and the tyrosinase activity. These results suggest that DR may have potential as an anti-aging ingredient in cosmetic herb markets.

Alleviation of Ultraviolet B-Induced Photoaging by 7-MEGATM 500 in Hairless Mouse Skin

  • The purpose of this study was to investigate the effect of $7-MEGA^{TM}$ 500 on the improvement of skin aging in an UVB-induced photo-aging model of hairless mice. The dorsal skin of hairless mice was exposed to UVB three times a week for 12 weeks to induce skin wrinkle. After inducing the wrinkle, $7-MEGA^{TM}$ 500 was orally administered once a day for 4 weeks. Skin thickness, skin barrier function, and wrinkle indicators were improved by treatment with $7-MEGA^{TM}$ 500. Both gene and protein expression levels of MMP-3 and c-Jun in skin were significantly decreased by $7-MEGA^{TM}$ 500. Therefore, the intake of $7-MEGA^{TM}$ 500 is thought to have a positive effect on the improvement of skin aging, although further studies are needed.

자외선 B를 조사한 인간유래각질세포에서 두릅순 에탄올추출물의 산화적 스트레스 억제효과 (Suppressive effects of ethanol extract of Aralia elata on UVB-induced oxidative stress in human keratinocytes)

  • 두릅순에서 얻은 70% 에탄올 추출물의 in vitro 항산화 효과를 측정하고, UVB에 의한 피부광노화를 유도하는 주요 원인인 ROS의 생성을 억제하는 효과가 있는지 알아보기 위하여 인간유래각질세포 (HaCaT)를 이용하여 실험을 수행하였다. 두릅순의 총 폴리페놀과 플라보노이드 함량은 각각 20.15 mg tannic acid/g dry wt, 18.75 mg rutin/g dry wt 이었고, 70% 에탄올 추출물의 DPPH 라디칼을 소거능 ($IC_{50}$)은 $98.5{\mu}g\;AA\;eq./mL$이었으며, $1,000{\mu}g/mL$ 농도에서 ABTS 라디칼 소거능과 환원력 (FRAP)은 각각 $41.8{\mu}g\;ascorbic\;acid\;(AA)\;eq./mL$$29.7{\mu}g\;AA\;eq./mL$로 우수한 항산화효과를 보였다. 두릅순 추출물을 HaCaT 세포에 24시간 전처리했을 때 UVB 조사에 의한 ROS 생성이 유의하게 감소되었으며, 산화적 스트레스에 민감하게 반응하는 전사인자인 Nrf-2와 각질세포에서 ROS를 제거하는 역할을 하는 주요 항산화효소인 SOD-1의 단백질 수준의 발현은 증가한 반면, UVB 조사에 의하여 증가하였던 HO-1의 단백질 발현은 감소되었다. 그러나, catalase의 단백질 발현에는 영향을 주지 못하였다. 본 연구결과는 두릅순 70% 에탄올 추출물에 함유된 항산화효능이 우수한 어떤 페놀화합물들이 UVB 조사로 인하여 생성된 ROS를 직접적으로 제거할 뿐 아니라 각질세포에 존재하는 방어시스템의 활성화를 통하여 산화적 스트레스로 인한 악영향을 막아줄 가능성을 제시하고 있다. 따라서, 두릅순 70% 에탄올 추출물은 UVB에 의한 피부손상 및 피부광노화를 억제하는 기능성식품 및 화장품 소재로 이용될 수 있을 것이다.

Direct Analysis in Real Time Mass Spectrometry (DART-MS) Analysis of Skin Metabolome Changes in the Ultraviolet B-Induced Mice

  • Ultraviolet (UV) radiation is a major environmental factor that leads to acute and chronic reactions in the human skin. UV exposure induces wrinkle formation, DNA damage, and generation of reactive oxygen species (ROS). Most mechanistic studies of skin physiology and pharmacology related with UV-irradiated skin have focused on proteins and their related gene expression or single-targeted small molecules. The present study identified and analyzed the alteration of skin metabolites following UVB irradiation and topical retinyl palmitate (RP, 5%) treatment in hairless mice using direct analysis in real time (DART) time-of-flight mass spectrometry (TOF-MS) with multivariate analysis. Under the negative ion mode, the DART ion source successfully ionized various fatty acids including palmitoleic and linolenic acid. From DART-TOF-MS fingerprints measured in positive mode, the prominent dehydrated ion peak (m/z: 369, M+H-$H_2O$) of cholesterol was characterized in all three groups. In positive mode, the discrimination among three groups was much clearer than that in negative mode by using multivariate analysis of orthogonal partial-least squares-discriminant analysis (OPLS-DA). DART-TOF-MS can ionize various small organic molecules in living tissues and is an efficient alternative analytical tool for acquiring full chemical fingerprints from living tissues without requiring sample preparation. DART-MS measurement of skin tissue with multivariate analysis proved to be a powerful method to discriminate between experimental groups and to find biomarkers for various experiment models in skin dermatological research.

Effects of Porcine Placenta Extract Ingestion on Ultraviolet B-induced Skin Damage in Hairless Mice

  • The aim of our study was to evaluate the potential benefits of an oral supplement containing porcine placenta extract (PPE) on skin parameters related to cutaneous physiology and aging. PPEs were administered orally to hairless mice for 12 wk. The effects of oral PPE administration on skin water-holding capacity and Transepidermal Water Loss (TEWL) were similar to those of oral collagen (HYCPU2) administered as a positive control. Magnified photographs and replica images showed a reduction in UVB-induced wrinkle formation after collagen and PPE treatments. PPE treatments ameliorated the thicker skin surface that results from UVB exposure, based on a histological examination of skin tissue. The groups that were orally administered PPE (0.05%, OL; 0.1%, OH group) showed significantly reduced Matrix Metaloproteinase-2 (MMP-2) mRNA expression levels compared with the UVB control (Con), by 33.5% and 35.2%, respectively. The mRNA expression of another collagen-degrading protein, MMP-9, was also significantly lower in the groups that received oral administration of PPE (especially in the OH group) than in the control group. Additionally, oral administration of PPE significantly upregulated tissue inhibitor of metalloproteinase-1 (TIMP-1) and -2 mRNA expression levels compared with expression levels in the control group (p<0.05). This indicates that orally administered PPE activated the expression of Timp-1 and -2, inhibitors of MMP, which is responsible for collagen degradation in skin. Taken together, we propose that long-term oral administration of PPE might have a beneficial effect with respect to skin photo-aging.

Effects of Polygonati Rhizoma Extracts on the Collagenase Activity and Procollagen Synthesis in Hs68 Human Fibroblasts and Tyrosinase Activity

  • Objectives : This study was designed to investigate the collagen metabolism and tyrosinase activity of Polygonati Rhizoma extracts (PR). It's effects are to tonify spleen qi and augment the spleen yin. It enrichs the yin and moisten the lung. Methods : The effect of PR on type I procollagen production and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts Hs68 after ultraviolet B (UVB, 312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of PR was measured. Results : There were no cytotoxicity at concentrations of 10, 30, $100{\mu}g/ml$. The reduced type I procollagen production was recovered by PR in UVB damaged Hs68 cells at a concentration of $100{\mu}g/ml$ ($16.2{\pm}0.0$ ng/ml) from control group ($13.9{\pm}0.5$ ng/ml). However there was no statistical significance. PR reduced The increased MMP-1 activity after UVB damage at concentrations of $10{\mu}g/ml$, $30{\mu}g/ml$, and $100{\mu}g/ml$ in a dose dependent manner ($42.2{\pm}20.5%$, $44.8{\pm}8.5%$, and $22.0{\pm}5.8%$). PR $100{\mu}g/ml$ treatment showed the statistical significace (p < 0.05). PR significantly reduced the tyrosinase activity at a concentration of 10 mg/ml ($32.0{\pm}12.8%$, p < 0.05). However, the L-DOPA oxidation was not changed. Conclusion : PR showed the anti-wrinkle effects and whitening effects in vitro. Although more researches are needed to validate the efficacy, these results suggest that PR may have potential as an anti-aging ingredient in cosmetic herb markets.

Photo-protective and Anti-melanogenic Effect from Phenolic Compound of Olive Leaf (Olea europaea L. var. Kalamata) Extracts on the Immortalized Human Keratinocytes and B16F1 Melanoma Cells

  • Ethylacetate and butanol fractions of leaf extracts (OLE) showed the higher contents of total phenolic compounds than hexane and water fractions. Oleuropein contents were $4.21{\pm}0.57,\;3.92{\pm}0.43,\;0.32{\pm}0.03,\;5.76{\pm}0.32$, and $32.47{\pm}0.25mg$/100g for ethanol extract, and hexane, chloroform, ethyl acetate, and butanol fraction, respectively. Treatment of ultraviolet-B (UVB) irradiated cells with 3 OLEs prepared by using ethylacetate and butanol at concentrations 0.001, 0.005, and 0.01% respectively showed significant recovery of cell viabilities. Treatment of dexametason 1 mM reduced tumor necrotic factor (TNF)-${\alpha}$ secretion by about 40%. UVB irradiated immortalized human keratinocytes (HaCaT) cells were treated with 3 different OLEs at the same concentrations. Ethylacetate fraction showed the strongest inhibition activity with respect of reduction of the elevated (TNF)-${\alpha}$. Cytotoxicity of OLEs on the B16-F1 cells was evaluated through thiazolyl blue tetrazolium bromide (MTT) assay. Ethylacetate fraction has no cytotoxicity in the range of 0.005-0.01%. A slight cytotoxicity was observed at the concentration of 0.1% butanol fraction of OLE that caused 10% decrease in cell viability.
