Anti-wrinkling Effects of Juniperus rigida Sied (노간주나무(Juniperus rigida Sieb.)의 주름개선 효과)

  • Human skin is constantly exposed to environmental conditions such as UV rays, polluted air, and chemical products. UV rays, in particular, affect skin in many ways causing wrinkles, fine wrinkles, rough skin, and xeroderma through a skin aging process. The purpose of this study was to investigate the anti-wrinkling effect of Juniperus rigida Sieb., derived from a common cedar tree found the world over. Measuring the elastase to investigate wrinkling efficacy, it was shown that at a concentration level of $1,000{\mu}g/ml$ of the two extracts, the water extract exhibited a lower than 10% inhibition activity, while the ethanol extract exhibited a 68.5% inhibition activity. Collagenase inhibition activity in the water extract and ethanol extract were 44.9% in the former and 97.2% in the latter extract, which in the case of the ethanol extract, is similar to ascorbic acid (99.6%). Moreover, measuring the biosynthesis of collagen by fibroblast, a concentration level of $50{\mu}g/ml$ of ethanol extract produced 151.52% of biosynthetic promotion, proving that the ethanol extract acts as a superb anti-wrinkling agent. The result of an investigation conducted on the influence of the ethanol extract on MMP-1 caused by UVA showed that at a concentration level of $1,00{\mu}g/ml$ of the ethanol extract of J. rigida Sieb a 67.1% inhibition activity was noted. At a concentration level of $50{\mu}g/ml$ of the ethanol extract of J. rigida Sieb a 35% and 39% inhibition ratio to MMP-1 protein and mRNA were observed respectively, thereby restraining the appearance of the collagen breakdown enzyme MMP-1 and wrinkle creation by skin photo-aging.

Skin-Whitening and UV-Protective Effects of Angelica gigas Nakai Extracts on Ultra High Pressure Extraction Process (초고압 추출 공정에 의한 당귀 추출물의 미백 및 자외선 차단 효과)

  • This study was performed to investigate the enhancement of UV-protection activities and skin-whitening effects from Angelica gigas Nakai extracts on ultra high pressure extraction process. Extraction at $60^{\circ}C$ treated by ultra high pressure for 15 minute and associated with ultrasofication (HPE15) was showed more than double yield, compare conventional extraction, as 12.24% (w/w) from A. gigas. Extracts of HPE15 reduced expression of MMP-1 on UV-irradiated CCD-986sk cells as 122.2% and revealed high inhibitory potency on tyrosinase as 69.4% by adding samples. Extracts of HPE15 from A. gigas showed strong inhibition effect on melanin production test by Clone M-3 cells as 82.4% by adding extracts. From the preliminary observations, we considered that the extracts from A. gigas could be potent natural materials for skin-whitening agent, and could be used as a potential anti-aging agent for the photo-damaged skin.

  • Carbonylated proteins (CPs) are synthesized by the chemical reaction of basic amino acid residues in proteins with aldehyde compounds yielded by lipid peroxidation. CPs are excited by a range of light from UVA to blue light, and resulted in the generation of superoxide anion radicals ($^{\cdot}O_2{^-}$) by photosensitizing reaction. Then, they CPs induce new protein carbonylation in stratum corneum through ROS generation. Furthermore, the superoxide anion radicals produce CPs in the stratum corneum (SC) through lipid peroxidation and finally affects skin conditions including color and moisture functions. The purpose of this study was to investigate the relationship between the production of stratum corneum carbonylated protein (SCCP) and the skin elasticity. 46 healthy female Koream at the ages of 30 ~ 50 years old were participated in this study for 8 weeks. The skin test was experiment conducted into two groups; placebo group (N = 23) used cream that did not contain active ingredients, and the other group (N = 23) used cream containing the elasticity improving ingredients. Test areas were the crow 's feet and the cheek. Various non-invasive methods were carried out to measure biophysical parameters on human skin indicating that dermis density and skin wrinkle were measured by using DUB scanner and Primos premium, respectively. Skin elasticity were measured using dermal torque meter (DTM310) and balistometer (BLS780). SCCP was assessed in a simple and non-invasive method using skin surface biopsy on the cheek of the subject. The amount of SCCP was determined using image analysis. All measurements were taken at 0, 4 and 8 8week. Results revealed that the amount of CP in SC was reduced when the skin wrinkle and skin elasticity related parameters were improved. This indicates that the correlation between the elasticity improvement and the amount of CP can be used as a anti-aging indicator and applicable to the skin clinical test for the measurement of skin aging in the future.

Physiological impact on layer chickens fed corn distiller's dried grains with solubles naturally contaminated with deoxynivalenol

  • Objective: An experiment was conducted to investigate the response of laying hens fed corn distiller's dried grains with solubles (DDGS) that are naturally contaminated with deoxynivalenol (DON). Methods: One hundred and sixty 52-week-old Lohmann Brown Lite hens were randomly allotted to five dietary treatments with 8 replicates per treatment. The dietary treatments were formulated to provide a range of corn DDGS contaminated with DON from 0% to 20% (i.e., 5% scale of increment). All laying hens were subjected to the same management practices in a controlled environment. Body weight, feed intake and egg production were measured biweekly for the entire 8-week experiment. The egg quality was measured biweekly for 8 weeks. On weeks 4 and 8, visceral organ weights, blood metabolites, intestinal morphology, and blood cytokine concentrations were measured. Results: The inclusion of corn DDGS contaminated with DON in the diet did not alter (p>0.05) the body weight, feed intake, hen-day egg production, egg mass and feed efficiency of the laying hens. No difference was found (p>0.05) in the egg quality of hens that were fed the dietary treatments. Furthermore, hens that were fed a diet containing corn DDGS contaminated with DON showed no change (p>0.05) in the visceral organ weights, the blood metabolites, and the cytokine concentrations. The crypt depth increased (p<0.05) as the amount of corn DDGS contaminated with DON increased. Proportionately, the villus height to crypt depth ratio of the laying hens decreased (p<0.05) with the increasing level of corn DDGS contaminated with DON in the diet. Conclusion: The inclusion of corn DDGS contaminated with DON up to 20% in layer diets did not cause changes in egg production performance and egg quality, which indicates that DON is less toxic at the concentration of 1.00 mg DON/kg.

Removal Characteristics of Organic Matters in Pretreatment and Reverse Osmosis Membrane Processes for Seawater Desalination (해수담수화 전처리 및 역삼투막여과 공정의 유기물 제거특성)

  • This study investigated removal characteristics of organic matters in pretreatment and reverse osmosis (RO) membrane processes for seawater desalination. Also, the influence of the changes in characteristics of organic matters on the membrane fouling was assessed. The pretreatment processes included dual media filtration (DMF), pressurized membrane filtration (MF), and submerged membrane filtration (SMF). Turbidity, UV absorption at 254 nm, dissolved organic carbon, size exclusion chromatography (SEC), fluorescence excitation emission matrix (FEEM), and transparent exopolymer particles (TEP) in raw and processed waters were analyzed. Ions and minerals were not removed by any pretreatment process tested, but were removed over 99% through the RO membrane process. Hydrophobic organics, which can play major role in organic membrane fouling, were relatively readily removed compared with hydrophilic ones. Membrane based pretreatment such as MF and SMF exhibited better removals of organics than conventional DMF. As the levels of organics in pretreated water decreased, the silt density index (SDI) decreased. MF treated water exhibited the lowest SDI value; this is possibly due to the lowest TEP ($0.1-0.4{\mu}m$) concentrations.

Studies on Transparent Sunscreen of Water-in-Oil Emulsion Type (유중수 타입의 외관이 투명한 자외선 차단 화장료에 관한 연구)

  • This study is related to the developing method of a transparent sunscreen cosmetic which has waterproofing property and no white turbidity when applied to skin. The transparent sunscreen is prepared by exploiting refractive index difference between oil-phase and water-phase of water-in-oil(W/O) emulsion. The sunscreen according to this study is prepared as a W/O type emulsion so that it is water-stable and water resistance. Also, the stability of W/O type emulsion is developed by adjusting the content of oil phase part and water phase part. As a result of this studying, the transparent W/O emulsion is prepared by adjusting the refractive index of oil-phase and water-phase within 0.004 and it is found that the stability of the transparent sunscreen is increasing when the water phase part is over 75% (w/w) of the W/O emulsion. Through clinical test of transparent sunscreen, the value of sun protection Factor(SPF) and Protection Factor of UVA(PFA) were determined. SPF and PFA values of transparent sunscreen were indicated $30.99{\pm}1.65$ and $3.01{\pm}0.30$.

  • Ultra-violet rays are very harmful to eye health care. The blocking of ultra-violet rays and a reduction of optical reflection in the visible light range, which is to increase the share of transmitted light, and avoid the formation of ghost images in imaging, are important for the applications of polymer eyeglasses lenses. In this study, the high-refractive index polymer lenses, n=1.67, were fabricated by injection-molded method with the xylene diisocyanate monomer, 2,3-bis-1-propanethiol monomer, and benzotriazol UV absorber (SEESORB 709) mixture. To reduce the reflection of the polymer lens surfaces, multi-layer anti-reflection (AR) coatings were coated for both sides of the polymer lens using an E-beam evaporation system. The optical properties of the UV blocking polymer lens were characterized using a UV-visible spectrometer. The material properties of the thin films, which were composed AR coating layers, refractive index, and surface roughness, were analyzed by ellipsometry and atomic force microscopy. As a result, the fabricated polymer lens perfectly blocked ultra-violet rays below 395 nm with a blocking rate greater than 99%.

  • Fructose-1, 6-diphosphate (FOP), a glycolytic metabolite is reported to ameliorate inflammation and inhibit the nitric oxide production in murine macrophages stimulated with endotoxin. It is also reported that FOP has cytoprotective effects against hypoxia or ischemia/reperfusion injury in brain and heart. In this study, we examined whether FDP has protective effects on UV-induced oxidative damage in skin cell culture system and human skin in vivo. FDP had a protective role in UVB-induced LDH release and ROS accumulation in HaCaT although it did not show direct radical scavenging effect in the experiment using 1, 1-diphenyl-2-picrylhydrazyl (DPPH). FDP also preserved cellular GSH content after UV irradiation in HaCaT and normal human fibroblast culture system. Cellular oxidative stress induces multiple downstream signaling pathways that regulate expression of multiple gene including MMP-1 and collagen, we examined the effects of FDP on UV-induced alteration of these protein expression in fibroblast culture and human skin in vivo. The increased MMP-1 expression in fibroblast and human skin by UV irradiation was significantly decreased by FDP. FDP also prevented the UV-induced decrease of collagen expression in fibroblast and human skin. Moreover, the decreasing the intracellular levels of reducing equivalents in human fibroblast by glutathione (GSH) depletion lowered the UVA dose threshold for reduction of procollagen expression, indicating that the differences of glutathione contents define the susceptibility of fibroblasts towards UV-induced reduction of procollagen expression. FDP also preserved cellular GSH content after UV irradiation, indicating that FDP has protective effects on UV-induced reduction of procollagen expression, which are possibly through maintaining intracellular reducing equivalent. Based on these premises, we examined the effect of daily use of a moisturizer containing FDP on facial wrinkle in comparison with vehicle moisturizer lacking FDP. In the clinical study, FDP significantly decreased facial wrinkle compared with vehicle alone after 6 months of use. Our results suggest that FDP has anti-aging effects in skin by increasing cellular antioxidant system and preventing oxidative signal and inflammatory reaction. Therefore FDP may be useful anti-aging agent for cosmetic purpose.

Susceptibility of Spodoptera exigua to UVA Insecticides Using Agricultural Multi-copter on Cabbage Field (농업용 멀티콥터를 활용한 무인항공기용 작물보호제에 대한 배추 파밤나방의 약제감수성)

  • We investigated the control efficacy and phytotoxicity of unmanned aerial vehicle-applied pesticides against the insect Spodoptera exigua, a major emerging pest in Chinese cabbage. Phytotoxicity was investigated in cabbage and 7 crops cultivated in the perconducted on 8 surrounding crops including Chinese cabbage at 1 to 2 times the recommended pesticide dosage. We treated cabbage fields with spinetoram suspension concentrate (16×), methoxyfenozide, sulfoxaflor suspension concentrate (16×). Then, we used water-sensitive paper to measure the distribution pattern of falling pesticide particles and the degree of coverage. Two of the pesticides showed 97% control efficacy, however, control efficacy might differ in resistant populations. Phytotoxicity was not observed in Chinese cabbage and the 7 surrounding crops treated with 1 to 2 times the recommended pesticide dosage. Analysis of the distribution pattern of falling pesticide particles revealed that breeze caused particle diffusion. Thus, wind is an important factor affecting the uniform treatment and diffusion of multicopter-applied pesticides. It follows that setting optimal conditions is necessary for effective control and treatment.

Preparation and Characteristics of Multilayer Lamellar Vesicle Using Phosphate Ester Surfactant (Phosphate ester 계면활성제를 이용한 다중층 라멜라 베시클의 제조 및 특성)

  • Various vehicles have been studied in order to protect skin ageing and sustain constantly moisturization. Recently, in relation to maintain of moisture, absorption and penetration of active materials, there has been introducing many preparing methods such as liposome, liquid crystal and multilamellar emulsion. We developed multilayer lamellar vesicle using cetearyl alcohol/ceteth-20 phosphate/dicetyl phosphate as analogy of phospholipid according to variation of shear rate and pH. These multilayer lamellar vesicles were confirmed by cross polarizing microscope. As results, morphologies of lamellar vesicle were not uniformed at low shear rate and pH. Also, stabilities for encapsulation of retinol were observed at $42^{\circ}C$ during two months. As a result, quantitative content of retinol decreased at low pH. Multilayer lamellar vesicle decreased 14% of transepidermal water loss compared with O/W emulsion. We compared multilayer lamellar sun cream to O/W sun cream using in vitro SPF test of water resistance and concluded that multilayer lamellar sun cream is similar to O/W sun cream in water resistance.
