선학초 추출물의 멜라닌합성 억제 및 항산화효과 (Study of Inhibitory Effect of Melanogenesis and Antioxidant Activity of Agrimonia pilosa Ledeb)

  • The purpose of this study was to investigate the mechanism of ethanol extract of Agrimonia pilosa Ledeb. (EAP)-reduced melanogenesis and diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity. Agrimonia pilosa Ledeb., a perennial herbaceous plant, has been used as an antihemorrhagic, anthelminntic, and antiinflammatory agents in Chinese herbal medicine. In the present study, we observed that melanin synthesis and tyrosinase activity of B16F10 cells were significantly decreased by EAP. However, EAP could not suppress tyrosinase activity in the cell-free system, whereas kojic acid directly inhibited tyrosinase activity. Furthermore, EAP decreased the protein expression of tyrosinase, tyrosinase-related prootein 1 (TRP-1), and tyrosinase-related prootein 2 (TRP-2). EAP scavenged DPPH radical up to 41% with 100 ${\mu}g/m{\ell}$ concentration. These results suggest that the hypopigmentary effect of EPA was due to regulation of tyrosinase protein.

미역쇠(Endarachne binghamiae)추출물의 멜라닌 생성 억제 효과 (Inhibitory Effect of Endarachne binghamiae Extract on Melanin Synthesis)

  • 본 연구에서는 동해안에서 채집된 88종의 해조류에 대하여 미백제로서 응용 가능성을 알아보기 위해 멜라닌 생성에 미치는 영향을 in vitro 실험을 통해 알아보았다. Melan-a 세포에 해조류 추출물을 처리하여 멜라닌 양을 측정한 결과, 고리매, 미역쇠, 왜모자반, 감태, 톳추출물에서 멜라닌 생성이 감소되었으며, 특히 미역쇠추출물을 처리하였을 때 가장 유효하게 농도의존적으로 멜라닌 생성이 감소되었다. 멜라닌 생성 억제 효과가 tyrosinase 활성 저해와 관련 있는지를 확인한 결과, 약한 활성을 가지지만 직접 작용하여 활성을 저해하지는 않으면서, 세포 내의 tyrosinase 활성을 농도의존적으로 저해하였다. 이는 세포 내 tyrosinase 활성저해가 직접적이 아닌 다른 기전에 의해 일어난다는 것을 의미하며, 그 기전 중의 하나인 ${\alpha}$-glucosidase활성을 측정한 결과 농도의존적으로 활성을 저해하였으며, 세포 내 tyrosinase 활성 저해와 높은 상관도를 보임을 확인하였다. 즉 미역쇠추출물은 N-linked glycoprotein인 tyrosinase의 glycosylation을 억제하여 tyrosinase의 세포 내 이동이나 활성을 억제할 수 있음을 알 수 있었다. 결론적으로, 미역쇠추출물은 피부 미백에 유효한 활성을 나타내는 천연 유용자원으로 활용할 가치가 있는 것으로 사료된다.

위령선 추출물의 항염활성 및 멜라닌 생성 억제효과 (Anti-inflammatory Effect and Inhibition of Melanin Biosynthesis of Clematis mandshurica)

  • Clematis mandshurica (Ranunculaceae) has traditionally been used as a remedy for antidiuretic, antifungal, rheumatic conditions and alleviate pain. We carried out to evaluate the anti-oxidative effect, anti-inflammatory effect and anti-melanogenic effect of ethanol extract and solvent fractions of Clematis mandshurica. The ethanol extract and the dichloromethane fraction of Clematis mandshurica showed an anti-oxidative effect in DPPH assay, the inhibitory activity of nitric oxide (NO) production in lipopolysaccharide (LPS) activated RAW 264.7 cell, and melanin synthesis and tyrosinase activity of B16F10 melanoma cells. They reduced NO production and melanin content in a dose-dependent manner at concentrations of $2.5{\sim}10{\mu}g/ml$. They also suppressed iNOS and tyrosinase protein and m-RNA expressions dose dependently, assayed by western blot analysis and RT-PCR experiment.

제주도 근해에 자생하는 참가죽그물바탕말 [Dictyota coriacea] 추출물의 멜라닌 억제 효과 및 항염증 효과 (Melanin Inhibitory Effect and Anti-inflammatory Effects of Dietyota coriacea Extracts Derived from Adjacent Sea of the Jeju Island)

  • 본 연구는 제주도 근해에 자생하는 참가죽그물바탕말 (Dictyota coriacea)의 에탄올 추출물 및 순차적 용매분획물을 얻어 B16F10 mouse melanoma 세포및 RAW 264.7 세포에 처리하여 멜라닌 생합성 및 tyrosinas 억제활성, NO 억제활성을 통하여 유용자원으로서 미백 및 기능성 화장료로 산업적 활용 가능성이 있는지 알아보고자 연구를 시행하였다. 실험한 결과 B16F10 세포에서 참가죽그물바탕말 에탄올 추출물과 헥산 분획물, 에틸아세테이트 분획물이 tyrosinase와 멜라닌 합성을 농도 의존적으로 뛰어나게 억제하였으며, 단백질 및 mRNA 발현양이 tyrosinase와 TRP-1 유전자에서 농도 의존적으로 감소함을 보였다. 하지만 TRP-2 단백질과 mRNA 유전자의 발현을 확인한 결과, TRP-2 단백질과 mRNA 발현이 농도에 관계없이 일정하였다. 또한 LPS로 자극한 RAW 264.7 세포에서 참가죽그물바탕말의 에탄을 추출물과 헥산 분획물, 에틸아세테이트 분획물이 가장 높은 NO 생성 억제 활성을 보였다. 따라서 본 연구결과는 참가죽그물바탕말 추출물이 항염증 활성을 가지는 미백 관련 기능성 소재로서의 활용가치가 있음을 보여준다.

B16/F10 생쥐 흑색종 세포에서 제주조릿대 추출물의 멜라닌 합성 저해 효과 (The inhibitory effect on the melanin synthesis in B16/F10 mouse melanoma cells by Sasa quelpaertensis leaf extract)

  • Effects of hot-water extract from Sasa quelpaertensis leaf (HWES) on melanogenesis were investigated in B16/F10 mouse melanoma cells. HWES inhibited cellular tyrosinase activity and melanin biosynthesis in a dose-dependent manner. Western blotting analysis showed that HWES dose-dependently inhibited tyrosinase and tyrosinase related protein-1 expression. Also, HWES suppressed sustained ERK activation in a concentration-dependent manner, suggesting that HWES inhibits the melanin biosynthesis through the suppressive effect against pathway involving sustained ERK activation.

동해 오징어 (Todarodes pacificus) 껍질 유래 콜라겐의 UV 보호능 및 미백효과 (UV Protection and Whitening Effects of Collagen Isolated from Outer Layer of the Squid Todarodes pacificus)

  • To develop a whitening agent, cytotoxicity of the soluble collagen isolated from Todarodes pacificus (CIT) was evaluated. CIT tested for cytotoxicity on human dermal fibroblast (CCD-986sk) was exhibited very low cytotoxicity. Because tyrosinase is the key enzyme for melanin biosynthesis, the use of various tyrosinase inhibitors is a common practice for whitening purpose in cosmetics. Tyrosinase inhibitory activity and melanin production assay were measured to confirm the whitening effect. The inhibitory effect of MMP-1 in UV-irradiated human dermal fibroblast was also performed. CIT showed strong inhibition potency on tyrosinase by 51.5% at 0.2 mg/mL which increased the inhibition by increasing the concentration of CIT, and showed 69.1% inhibition at 1.0 mg/mL. CIT showed strong inhibition effect on melanin production with 82% at 1.0 mg/mL. The CIT also reduced about 76% expression of MMP-1 in UV-irradiated CCD-986sk cell at 1.0 mg/mL. From the preliminary observations, we suggest that the collagen isolated from CIT could be a potential source of natural skin-whitening and anti-aging agents for the photo-damaged skin.

감초수추출물이 HM3KO 세포의 멜라닌 생성에 미치는 영향 (Effect of Glycyrrhizae Radix Water Extract on the Melanogenesis of Human Melanoma Cell)

  • This study was conducted to evaluate the effects of Glycyrrhizae Radix water extract, known as depigmenting agent, on melanin biosynthesis in the HM3KO human melanoma cells. The inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis was identified by mushroom tyrosinase assay in vitro. To determine whether Glycyrrhizae Radix water extract suppress melanin synthesis in cellular level, HM3KO cells were cultured in the presence of different concentrations of Glycyrrhizae Radix water extract and the effects on cell proliferation, melanin contents and tyrosinase activity were examined after 3 days. Treatment with Glycyrrhizae Radix at various concentrations did not exhibit any change of cell viability, and increased the cell proliferation. And the water extract of Glycyrrhizae Radix inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. These results suggest that the inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis is due to the suppression of tyrosinase in HM3KO cells.

AOT/이소옥탄/역미셀계에서의 Tyrosinase 반응 (Tyrosinase Reaction in AOT/Isooctane/Reverse Micelles)

  • 지용성 물질은 물에 용해되지 않아 수용액 효소반응계에서의 특정효과를 평가하기가 어렵다. 생체내에서 멜라닌을 합성하는 효소인 tyrosinase의 지용성 저해제를 탐색하고자 유기용매, 계면활성제 및 물로 구성된 광학적으로 투명한 RM을 도입하였다. Tyrosinase의 겉보기 반응이 물에서와 유사한 RM의 형성조건 중 유기용매는 isooctane, 계면활성제는 100 mM의 dioctyl sulfosuccinate(AOT)가 적절하였다. RM에서 물과 AOT의 몰 농도 비율이 15일 때 tyrosinase(103.5 units)는 기질인 3,4-dihydroxy-L-phenylalanine(0.18 mM)에 대하여 수용액에서와 유사한 거동을 나타내었다. 또한 tyrosinase 저해제인 cinnamic acid 존재 하에서도 tyrosinase 촉매 반응의 반응 생성물이 반응 시간에 비례하여 형성됨이 확인되어 지용성 물질도 RM에서는 tyrosinase 저해활성이 가능함을 시사하였다. 따라서 RM은 수용성인 효소와 기질 그리고 지용성인 저해제를 모두 하나의 상(phase)에 용해시킬 수 있어 지용성 물질의 tyrosinase 저해 효과를 분석할 수 있는 중요한 수단으로 활용할 수 있을 것으로 판단되었다.

MSH에 의해 자극된 B16F10세포에서 사간(射干)의 멜라닌 합성 억제 효과 (Inhibitory Effect of Belamcandae Rhizoma on the Melanogenesis in MSH-stimulated B16F10 cells)

  • Objective : The present study was designed to assess the potential inhibitory activity of an ethanol extract of Belamcandae Rhizoma (EBR) on the alpha-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis signal pathway in B16F10 melanoma cells. Methods : Several experiments were performed in B16F10 melanoma cells. We studied tyrosinase activity, melanin content, cell-free tyrosinase activity and DOPA stain, and performed Western blots and RT-PCR for proteins and mRNA involved in melanogenesis. Results : ${\alpha}$-MSH-induced tyrosinase activity and melanin content were inhibited significantly by EBR. EBR markedly suppressed the protein expression level of tyrosinase in B16F10 melanoma cells. On the other hand, the expression of tyrosinase-related protein-1 (TRP-1) and -2 (TRP-2; DCT) were not affected by EBR. To elucidate the mechanism of the depigmenting property of EBR, we examined the involvement EBR in cAMP response element binding (CREB) protein phosphorylation and microphthalmia-associated transcription factor (MITF) signalling induced by ${\alpha}$-MSH. EBR did not regulate CREB phosphorylation and MITF expression by ${\alpha}$-MSH. Nevertheless, the mRNA expression of tyrosinase was significantly attenuated by EBR treatment without changes in the expression of TRP-1 and -2 mRNA. Conclusion : Our study suggested that EBR inhibits ${\alpha}$-MSH-induced melanogenesis by suppressing tyrosinase mRNA.

Inhibitory Effect of Ceylon Black Tea Extract on the Melanogenesis in 𝛼-MSH Stimulated B16F10 Melanoma Cells

  • The desire to be light skinned is universal among women. Asia has a long history of using skincare formulations as whitening agents. There is an imperative need to develop novel cosmetics from herbal sources due to several unpleasant side effects and high costs. As a result, this study aims to investigate the effect of Ceylon black tea extracts on melanogenesis. Five different Ceylon black tea extracts were prepared and examined for total phenolic content (TPC), total flavonoid content (TFC), DPPH radical scavenging activity, and tyrosinase inhibitory activity. Furthermore, B16F10 melanoma cells were treated with these extracts and tested for cytotoxicity and protein suppression levels. According to the results of this study, the highest TPCs were obtained from ethanol and acetone extractions (240.303 ± 1.389 ㎍/g and 240.202 ± 4.700 ㎍/g, respectively), whereas the highest TFC was obtained from acetone extraction (57.484 ± 0.413 ㎍/g). Ceylon black tea extracted with ethanol exhibited the highest inhibitory activity on tyrosinase with an IC50 value of 0.277 ± 0.017 mg/mL and the highest DPPH radical scavenging activity with an EC50 value of 0.009 ± 0.000 mg/mL. Furthermore, western blot results revealed that tyrosinase, TRP-1, TRP-2, and MITF protein expression levels were dose-dependently suppressed, indicating the applicability of Ceylon black tea extract as a novel melanogenesis inhibitor.
