Analysis of Periodontitis Biomarker Expression in Gingival Crevicular Fluids

  • Background: Periodontal disease, also known as gum disease, is a major dental inflammatory disease with a very high prevalence; it is the main cause of tooth loss. Therefore, diagnostic biomarkers that can monitor gum inflammation are important for oral healthcare. Since the gingival crevicular fluid (GCF) adequately reflects changes in the periodontal environment, they have become a target for the development of effective diagnostic biomarkers for periodontitis. In the present study, the level of the target molecules suggested as diagnostic biomarkers for periodontitis were analyzed in GCF samples collected from healthy individuals and periodontitis patients. In addition, useful targets for the diagnosis of periodontitis were evaluated. Methods: GCF samples were collected from healthy individuals and periodontitis patients using absorbent paper points. SDS-PAGE and Coomassie staining were performed for protein analysis. The protein concentrations of GCF specimens were determined using the Bradford method. The levels of the target molecules appropriate for diagnosing periodontal disease were measured by ELISA, according to the manufacturer's protocol. Results: The protein concentration of GCF collected from periodontitis patients was 3.72 fold higher than that in an equal volume of GCF collected from healthy individuals. ELISA analysis showed that the level of interukin-6 (IL-6), IL-8, metalloproteinases 2 (MMP-2), MMP-9, tumor necrosis factor-alpha (TNF-α), azurocidin, and odontogenic ameloblast-associated protein (ODAM) were higher in the GCF samples from the periodontitis patients than in those from the healthy individuals. However, the level of IL-6 and TNF-α were relatively low (> 5 pg/ml). The prostaglandin E2 (PGE2) levels were not significantly different between the two GCF samples. Conclusion: These results indicate that IL-8, MMP-2, MMP-9, azurocidin, and ODAM are potentially useful diagnostic biomarkers for periodontitis; combining multiple biomarkers will improve the diagnostic accuracy of periodontitis.

A Study of the Antioxidant and Anti-Inflammatory Effects of Dusokohwaeum

  • Background: The aim of this study is to determine the antioxidant and anti-inflammatory effects of Dusokohwaeum (DOE). Methods: To measure the antioxidant and anti-inflammatory effects of DOE, the total flavonoid and polyphenol contents and radical scavenging activity were measured. Furthermore, reactive oxygen species (ROS), nitric oxide, and cytokine production were measured by treating lipopolysaccharide-induced RAW264.7 cells with DOE, and gene expression levels of inducible cyclooxygenase-2, nitric oxide synthase, and cytokines were evaluated. Results: Radical scavenging experiments revealed a significant concentration-dependent increase in scavenging capacity. The production of ROS, nitric oxide, and cytokines in the cells showed a significant concentration-dependent decrease when compared with the control group. The gene expression levels of inducible cyclooxygenase-2, nitric oxide synthase, and cytokines also showed a significant concentration-dependent decrease when compared with the control group. Conclusion: Interestingly, the antioxidant and anti-inflammatory effects of DOE were 23.42 ± 0.64 mg GAE/g and 20.83 ± 0.98 mg QE/g, respectively. The administration of DOE resulted in a concentration-dependent increase in scavenging ability in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging ability experiments. The production of intracellular ROS and nitric oxide was significantly reduced in the presence of DOE. The production of inflammatory cytokines (prostaglandin E2, tumor necrosis factor-alpha [TNF-α], interleukin-1 beta [IL-1β], and IL-6) was significantly reduced in the presence of DOE. Finally, the expression levels of inducible nitric oxide synthase, cyclooxygenase-2, TNF-α, IL-1β, and IL-6 were significantly decreased in the presence of DOE.

Immunological Analysis of Postoperative Delirium after Thoracic Aortic Surgery

  • Background: Delirium is a recognized neurological complication following cardiac surgery and is associated with adverse clinical outcomes, including elevated mortality and prolonged hospitalization. While several clinical risk factors for post-cardiac surgery delirium have been identified, the pathophysiology related to the immune response remains unexamined. This study was conducted to investigate the immunological factors contributing to delirium in patients after thoracic aortic surgery. Methods: We retrospectively evaluated 43 consecutive patients who underwent thoracic aortic surgery between July 2017 and June 2018. These patients were categorized into 2 groups: those with delirium and those without it. All clinical characteristics were compared between groups. Blood samples were collected and tested on the day of admission, as well as on postoperative days 1, 3, 7, and 30. Levels of helper T cells (CD4), cytotoxic T cells (CD8), B cells (CD19), natural killer cells (CD56+CD16++), and monocytes (CD14+CD16-) were measured using flow cytometry. Results: The median patient age was 71 years (interquartile range, 56.7 to 79.0 years), and 21 of the patients (48.8%) were male. Preoperatively, most immune cell counts did not differ significantly between groups. However, the patients with delirium exhibited significantly higher levels of interleukin-6 and lower levels of tumor necrosis factor-alpha (TNF-α) than those without delirium (p<0.05). Multivariate analysis revealed that lower TNF-α levels were associated with an increased risk of postoperative delirium (p<0.05). Conclusion: Postoperative delirium may be linked to perioperative changes in immune cells and preoperative cytokine levels. Additional research is required to elucidate the pathophysiological mechanisms underlying delirium.

Immune Regulatory Effect of Newly Isolated Lactobacillus delbrueckii from Indian Traditional Yogurt

  • Lactic acid bacteria (LAB) are microorganisms that are believed to provide health benefits. Here, we isolated LAB from Indian fermented foods, such as traditional Yogurt and Dosa. LAB from Yogurt most significantly induced TNF-α and IL-1β production, whereas LAB from Dosa induced mild cytokine production. After 16S rRNA gene sequencing and phylogenetic analysis, a Yogurt-borne lactic acid bacterium was identified and classified as Lactobacillus delbrueckii subsp. bulgaricus, and it was renamed L. delbrueckii K552 for the further studies. Our data suggest that the newly isolated L. delbrueckii can be used for the treatment of immune deficiency disorders.

Immune-Enhancing Effects of Lactobacillus plantarum 200655 Isolated from Korean Kimchi in a Cyclophosphamide-Induced Immunocompromised Mouse Model

  • In this study, we evaluated the immune-enhancing activity of kimchi-derived Lactobacillus plantarum 200655 on immune suppression by cyclophosphamide (CP) in ICR mice. Animals were fed distilled water or 1×109 colony-forming unit/kg B.W. 200655 or Lactobacillus rhamnosus GG as a positive control for 14 days. An in vivo model of immunosuppression was induced using CP 150 and 100 mg/kg B.W. at 7 and 10 days, respectively. Body weight, spleen index, spleen weight, and gene expression were measured to estimate the immune-enhancing effects. The dead 200655 (D-200655) group showed an increased spleen weight compared to the sham control (SC) group. Similarly, the spleen index was significantly higher than that in the CP-treated group. The live 200655 (L-200655) group showed an increased mRNA expression of tumor necrosis factor-alpha (TNF-α) and interleukin (IL)-6 in splenocytes. Also, the iNOS and COX-2 mRNA expression was upregulated in the L-200655 group compared to the CP-only (SC) group. The phosphorylation of ERK and MAPK was also upmodulated in the L-200655 group. These results indicate that L. plantarum 200655 ameliorated CP-induced immune suppression, suggesting that L. plantarum 200655 may have the potential to enhance the immune system.

작약, 감초 및 작약감초탕의 투여가 궤양성 대장염 동물 모델에 미치는 영향: 대사산물 변화를 포함하여 (Effects of Paeoniae Radix, Glycyrrhiza Uralensis and Jakyakgamcho-tang Treatment on Ulcerative Colitis Animal Model: Including Changes in Metabolites)

  • Objectives To investigate the effect of Jakyakgamcho-tang (JGT) on Dextran sulfate sodium (DSS) induced ulcerative colitis. Methods Experimental animals were divided into six groups; group 1, normal group; group 2, DSS-induced colitis group; group 3, 5-aminosalicylic acid (5-ASA)-treated group; group 4, Glycyrrhiza Uralensis (Gamcho, GC)-treated group; group 5, Paeoniae Radix (Jakyak, JY)-treated group; group 6, JGT-treated group. Inflammatory cytokines and their metabolites were detected. Result In the JGT group, the levels of tumor necrosis factor alpha (TNF-α), prostaglandin E2 (PGE2) were significantly decreased, whereas that of Interleukin-10 (IL-10) were significantly increased. In addition, the metabolite profile changed in the JGT group with most metabolites increasing. Conclusion This study demonstrates that the therapeutic potential of JGT in ulcerative colitis. Further studies should be conducted to confirm our findings.

버섯류 추출물의 피부각질세포(HaCaTcell) 내 염증성 사이토카인 억제효과 (Inhibitory effect of mushrooms extract on TNF-α/INF-γ induced-cytokine in human keratinocytes, HaCaT)

  • 식용버섯인 팽이버섯, 꽃송이버섯, 참송이버섯, 느타리 버섯, 녹각영지, 상황버섯, 눈꽃 동충하초, 아가리쿠스, 잎새버섯, 석이버섯, 노랑느타리버섯, 분홍느타리버섯, 편각영지버섯, 현미 동충하초, 번데기 동충하초, 표고버섯의 물 및 에탄올 추출물에 대한 피부염증완화 효능을 확인한 결과 열수추출물에서는 잎새버섯과 번데기 동충하초가 피부염증완화 효능이 탁월하였으며, 에탄올 추출물에서는 영지버섯, 표고버섯, 번데기 동충하초와 팽이버섯에서 피부염증완화 효능이 탁월하였다. 특히, 각각의 추출물에서 피부염증완화 효능이 높은 것은 번데기 동충하초 임을 확인하였다.

산후우울증 환자에서 혈장 Cytokine의 농도변화에 대한 전향적 연구 (Plasma Levels of Cytokines in Patients with Postpartum Depression)

  • 연구목적 산후우울증은 일반적으로 산모의 10~15%에서 발병하는 질환으로 그 원인으로는 정신사회적인 요인과 산과적인 요인이 모두 관계 있다고 알려져 있다. 감염, 손상, 악성종양, 자가면역질환, 스트레스에 의해 면역체계가 자극되면 proinflammatory cytokine과 anti-inflammatory cytokine이 생성되고 분비된다고 알려져 있다. 본 연구에서는 산후우울증이 있는 경우 산후우울증이 없는 군에 비해 혈중 Transforming growth factor-β1(TGF-β1), Insulin-like growth factor-1 (IGF-1), β-Nerve growth factor (β-NGF), Interleukin-2 (IL-2), IL-4, IL-6, Tumor necrosis factor-α (TNF-α), Interferon-γ (IFN-γ)의 농도가 상승되어 있을 것이라 가정하고, 임산부에서의 우울증의 경과에 따라 cytokine의 농도의 변화를 알아보고자 한다. 방 법 본 연구는 총 104 명의 임산부와 60명의 임신을 하지 않은 정상 대조군을 대상으로 하였다. 우울 증상은 임신 24주, 출산 1주, 출산 6주에 에딘버러 산후 우울 척도(EPDS)를 사용하여 평가하였다. EPDS의 총 점수가 10 이상인 경우, 우울증상이 있는 것으로 판단하였으며 EPDS 점수 변화에 따라 임산부 정상대조군, 출산 후 우울-회복군, 출산 후 우울군 세 그룹으로 나누었다. 결 과 임신군과 비임신 정상대조군을 비교하였을 때 TGF-β1, IGF-1의 혈장 농도는 임신군에서 비임신 정상대조군보다 더 높았다(TGF-β1 ; p<0.01, IGF-1 ; p=0.026). 그러나 β-NGF, IL-2, IL-4, IL-6, IFN-γ, TNF-α는 비임신 여성대조군에 비해 임산부 정상대조군에서 그 농도가 낮게 측정되었다(β-NGF ; p=0.001, IL-2 ; p<0.01, IL-4 ; p<0.01, IL-6 ; p<0.01, IFN-γ ; p<0.01, TNF-α ; p<0.01) 임신 24주에 TGF-β1, IGF-1, β-NGF, IL-2, IL-4, IL-6, IFN-γ, TNF-α의 농도를 살펴보면 임산부 정상대조군, 출산 후 우울-회복군, 출산 후 우울군 세 군간에 유의한 차이가 없었으며 출산 6주에도 역시 유의한 차이가 관찰되지 않았다. 또한 임신 24주, 출산 6주에 시간에 따른 농도 차이를 비교해 보았을 때 세 군에서 모두 그 통계적으로 유의한 차이는 없었다. 결 론 본 연구는 비임신 정상대조군과 임신군간의 혈장 cytokine 농도에서 유의한 차이를 발견했으나 산후우울증군과 정상 임신대조군간의 혈장 cytokine 농도는 유의한 차이를 밝혀내지는 못했다.

Rutin alleviated lipopolysaccharide-induced damage in goat rumen epithelial cells

  • Objective: Rutin, also called vitamin P, is a flavonoids from plants. Previous studies have indicated that rutin can alleviate the injury of tissues and cells by inhibiting oxidative stress and ameliorating inflammation. There is no report on the protective effects of rutin on goat rumen epithelial cells (GRECs) at present. Hence, we investigated whether rutin can alleviate lipopolysaccharide (LPS)-induced damage in GRECs. Methods: GRECs were cultured in basal medium or basal medium containing 1 ㎍/mL LPS, or 1 ㎍/mL LPS and 20 ㎍/mL rutin. Six replicates were performed for each group. After 3-h culture, the GRECs were harvested to detect the relevant parameters. Results: Rutin significantly enhanced the cell activity (p<0.05) and transepithelial electrical resistance (TEER) (p<0.01) and significantly reduced the apoptosis rate (p<0.05) of LPS-induced GRECs. Rutin significantly increased superoxide dismutase, glutathione peroxidase, and catalase activity (p<0.01) and significantly decreased lactate dehydrogenase activity and reactive oxygen species and malondialdehyde (MDA) levels in LPS-induced GRECs (p<0.01). The mRNA and protein levels of interleukin 6 (IL-6), IL-1β, and C-X-C motif chemokine ligand 8 (CXCL8) and the mRNA level of tumor necrosis factor-α (TNF-α) and chemokine C-C motif ligand 5 (CCL5) were significantly increased in LPS-induced GRECs (p<0.05 or p<0.01), while rutin supplementation significantly decreased the mRNA and protein levels of IL-6, TNF-α, and CXCL8 in LPS-induced GRECs (p<0.05 or p<0.01). The mRNA level of toll-like receptor 2 (TLR2), and the mRNA and protein levels of TLR4 and nuclear factor κB (NF-κB) was significantly improved in LPS-induced GRECs (p<0.05 or p<0.01), whereas rutin supplementation could significantly reduce the mRNA and protein levels of TLR4 (p<0.05 or p<0.01).   • 알츠하이머 병은 인지 기능 저하로 인한 치매 발생으로 설명할 수 있는 만성 및 진행성 신경 퇴행성 질환이다. 알츠하이머 병의 특징은 세포 외 및 세포 내 아밀로이드 플라크의 형성이다. 아밀로이드 베타는 알츠하이머 병의 특징이며 미세아교세포는 아밀로이드 베타의 존재하에 활성화될 수 있다. 활성화된 미세아교세포는 전 염증성 사이토카인을 분비한다. 게다가, S100A9는 염증의 중요한 선천성 전 염증 기여자이며 알츠하이머 병에 잠재적인 기여자로 알려져 있다. 이 연구는 아밀로이드 베타 및 S100A9이 처리된 BV-2 세포에서 염증반응 및 NLRP3 인플라마솜 활성화에 대한 메트포르민 및 알파리포산의 효과를 조사했다. 메트포르민과 알파-리포산은 종양 괴사 인자-알파 및 일터루킨-6와 같은 염증성 사이토카인을 약화시킨다. 또한 메트포르민과 알파-리포산은 JNK, ERK, p38의 인산화를 억제하고, NF-kB 경로 및 NLRP3 인플라마솜의 활성화를 억제했다. 또한 메트포르민과 알파-리포산은 M1 표현형인 ICAM1의 수준을 감소시킨 반면 M2 표현형인 ARG1은 증가시켰다. 이러한 발견은 메트포르민과 알파-리포산이 아밀로이드베타 및 S100A9에 의한 신경 염증 반응에 대한 치료제가 될 수 있음을 시사한다.
