Modigication of host cells and Expression of Recombinant E. coli trp plasmids for the increased Production of Tryptophan in Klebsiella pneumoniae (Klebsiella pneumoniae에서 트립토판 생산증대를 위한 숙주개발 및 재조합 trp plasmid의 발현)

  • In order to increase the production of tryptophan by maximizing expression of recombinant trp plasmid, Klebsiella pneumoniae KC 105(pheA tyrA trpE trpR tyrR) was genetically modified. KC 107, inosine monophospate(IMP) auxotroph from KC 105 and KC 108, histidine(His) auxotroph from KC 107 were also derived respectively to increase phosphoribosylpyrophosphate(PRPP) production which is required for tryptophan biosynthesis. From KC 107 phosphoribosylpyrophosphate consumption which is required for tryptophan biosynthesis. From KC 107 and KC 108, KC 109 and KC 110, both arginine auxotrophs were derived respectively. To investigate the expression of recombinant trp plasmid in the selected K. pneumoniae mutants, the auxotrophic mutants were transformed with recombinant trp plasmids pSC 101-$trpE^{FBR}$, pSC 101-trpL(.DELTA.att) $trpE^{FBR}$ (pSC 101-trp-AF). Amount of tryptophan produced and activities of tryptophan synthase of $trpR^{-}$ mutant (KC 100) and $tyrR^{-}$ mutnat(KC 105) containing recombinant plasmid pSC 101-trp operon were increased by 30-40% as compared with KC 99(pheA tyrA trpE) containing recombinant plasmid pSC 101-trp operon. Activities of tryptophan synthase and production of tryptophan of KC 108 ($His^{-}$) and KC 109($Arg^{-}$) containing recombinant plasmid pSC 101-trp operon were increase by two-fold as compared with KC 107 containing pSC 101-trp operon.

Effects of $aroP^{-}$ mutation on the tryptophan excretion in escherichia coli ($aroP^{-}$변이가 E.coli에서 트립토판 방출에 미치는 영향)

  • As a part of the host cell development for a amplified recombinant trp operon, $aroP^-$ mutation was introduced in a E. coli host strain. $aroP^-$ mutation was induced by transposon Tn10 and transduced into the E. coli host cell by bacteriophage P1Kc. The effect of $aroP^-$ mutation on the excretion of tryptophan in E. coli $trpR^{-ts}/ColE_1 -trp^+$ cells was investigated. Mutant lacking the general aromatic transport system was resistant to ${\beta}-2-thienylalanine\;(2{\times}10^{-4}\;M)$, p-fluorophenylalanine $(2{\times}10^{-4}M)$, or 5-methyltryptophan $(2{\times}10^{-4}\;M.)[^3H]-tryptophan$ uptake of the $aroP^-$ mutant strain was reduced considerably as compared with $aroP^+$ counterpart. The rate of $[^3H]-tryptophan$ uptake of the $aroP^-$ mutant strain treated with $NaN_3(3{\times}10^{-2}\;M)$ was much less affected than that of $aroP^+$ counterpart. The $aroP^-$ transductants increased the tryptophan excretion from E. coli $trpR^{-ts}/ColE_1 -trp^+$ four times more than $aroP^+$ counterpart.

Media Optimization for L-tryptophan Production by Genetically Engineered Klebsiella pneumoniae (유전자 조작한 Klebsiella pneumoniae로부터 L-트립토판 생산을 위한 발효배지 조건)

  • The optimum medium composition for the production of L-tryptophan with Klebsiella pnuemoniae pheA tyrA trpE trpR/pSC 101-trp$^{+}$ and the effect of precusors in the optimum medium were studied. The specific growth rate in the optimum medium was almost the same as that in the basal medium, the former showing 1.01 and the latter 1.07 hr $$^{-1}$, but the produced tryptophan was increased 45% in the optimum medium. The maximum amount of produced tryptophan was 159 mg/l within 14 hours. Tryptophan production was ceased by casamino acid addition over 4 g/l in medium, but cell maSS increased with its addition. Indole and anthranilate as precusors had toxic effect on growth and tryptophan production at experimented concentration range (over 20 mg/l), but L-serine had good effect on tryptophan production, resulting in 175 mg/l tryptophan within 14 hours.

Functional roles of Tryptophan residues in diketoreductase from Acinetobacter baylyi

  • Diketoreductase (DKR) from Acinetobacter baylyi contains two tryptophan residues at positions 149 and 222. Trp-149 and Trp-222 are located along the entry path of substrate into active site and at the dimer interface of DKR, respectively. Single and double substitutions of these positions were generated to probe the roles of tryptophan residues. After replacing Trp with Ala and Phe, biochemical and biophysical characteristics of the mutants were thoroughly investigated. Enzyme activity and substrate binding affinity of W149A and W149F were remarkably decreased, suggesting that Trp-149 regulates the position of substrate at the binding site. Meanwhile, enzyme activity of W222F was increased by 1.7-fold while W222A was completely inactive. In addition to lower thermostability of Trp-222 mutants, molecular modeling of the mutants revealed that Trp-222 is vital to protein folding and dimerization of the enzyme.

Supply of Tryptophan and Tryptamine Influenced the Formation of Melatonin in Viola Plants (제비꽃속(Viola) 식물에서 tryptophan과 tryptamine 공급이 멜라토닌 생성에 미치는 영향)

  • Melatonin has been known as an animal hormone. However, melatonin exists in diverse organisms including higher plants. The biosynthesis and physiological roles for melatonin in plants is still largely unknown, although both dicot and monocot plants have melatonin and some medicinal plants even contain large amounts of melatonin. In this study we detected melatonin in diverse Viola plants, in which melatonin had not been examined so far, by reverse phase HPLC analysis, demonstrating the wide existence of melatonin in the genus of Viola. We then fed tryptophan (Trp) and tryptamine (TAM) to the incubation medium for Viola leaf sections to test their effects on melatonin formation. Trp is also the hypothesized starting material of melatonin in plants, and TAM is the following intermediate produced by the decarboxylation of Trp. Trp feeding did not affect the contents of melatonin. In contrast, TAM feeding clearly increased the level of melatonin in Viola leaves. Because TAM is derived from Trp, we concluded that the Trp-TAM pathway exists in Viola plants as well. Ineffectiveness of Trp feeding to the change of melatonin contents supports the hypothesis that the decarboxylation step from Trp to TAM is the rate-limiting step in plant melatonin biosynthesis.

Solvent Effects on Action Spectra for The Photodecomposition of N-Acetylphenylalanyl-L-Tryptophan and 3-Methyl Indole

  • The UV action spectra and quantum yields for photodestruction of tryptophan (Trp) in peptide such as N-acetylphenylalanyl-L-tryptophan (NAPT) and 3-methyl indole (scatole) were determined in aerated aqueous and organic solvents. The photodestruction of aqueous NAPT was shown to be initiated by photoionization without requirement of threshold energy, as demonstrated by the similarity of fluence effect curves obtained for the action at various wavelengths and the wavelength dependence of quantum yield comparable to that reported for the photoionization of L-Trp. N-formylkynurenine (NFK)-type photoproduct, which is a photodynamic sensitizer, was not found to be involved in the photodestruction of Trp in NAPT in aqueous solution. In contrast, the action spectra of NAPT and scatole in organic solvents have revealed evidences for the significant role of internal photosensitization by NFK-type photoproduct in photolysis of Trp in peptide.

The Effects of Tryptophan and Tyrosine-Enriched Diet on the Serum Cortisol, Glucose and Free Fatty Acid Levels of Stressed Rats (Tryptophan 및 Tyrosine 보강 식이가 Stress로 인한 혈청 Cortisol, Glucose 및 Free Fatty Acid의 농도 변화에 미치는 영향)

  • We made S.D male rats eat 3.6% tryptophan(trp) or tyrosine(thr)-enriched diet for 4 days and measured the trp or tyr use in serum of stressed rats as well as cortisol, glucose and free fatty acid(ffa) changes in serum. When control group had received stress treatment, their tyr level has dropped significantly but no changes in trp level and their cortisol, glucose, ffa levels in serum were increased significantly all together. Trp-enrichment alone can't change serum cortisol and ffa levels but trp pools in serum and brain enlarged by dietary enrichment. Trp-enriched group's serum glucose concentration was significantly lower than control-dieted group. When trp-enriched group received stress treatment, their responses to stress were different from control-dieted group. Serum trp concentration of trp-enriched-with-stress group has dropped significantly and cortisol level was increased significantly but not as much as control-dieted-with-stress group. Glucose and ffa levels of trp-enriched-with-stress group did not increase at all. Tyr-enriched group has also larger serum pool of tyr and lowest basal cortisol level in all three diet group. tyr-enriched group's and ffa levels were in normal range and those responses to stress were same pattern with control diet group. Most importantly in tyr-enriched-with-stress group, only slight but not significant increase of cortisol level was shown.

Electrochemical and Raman Spectroscopy Analysis for D- and L-Tryptophan-b-Cyclodextrin Inclusion Complexes

  • An enantioselective recognition of D- and L-tryptophan (Trp)-b-cyclodextrin (CD) inclusion complex was performed using electrochemical and FT-Raman spectroscopic analysis. From the electrochemical analysis, the selectivity coefficient ($K_{DL}$) of b-CD inclusion complexes was found higher than that of the D- and L-Trp in phosphate buffered saline (PBS, pH=7.0) solution. The percentage of enantioselectivity ($I_{%{ee}}$) for peak current of D-Trp-b-CD inclusion complexes was observed higher than that of L-Trp-b-CD inclusion complexes in PBS solution. From Raman spectroscopy, chemical shift difference (D, $cm^{-1}$) for the C=C stretch, ring vibration, and ring breathing of D-Try-b-CD inclusion complex were observed higher than that of L-Trp-b-CD inclusion complex. The electrochemical and Raman spectroscopic analyses were found very useful for chiral detection of racemic amino acid in the presence of b-CD.

  • The trp2 gene encoding trifunctional enzyme in Coprinus cinereus was investigated the expression in the heterothallic mushroom species. To identify the homology of trp2 gene to Schizophyllum commune and Pleurotus ostreatus, southern hybridization was performed with plasmid pHIONA8 containing C. cinereus trp2 gene as a probe, which resulted in a strong signal indicating an homologous sequence to the chromosomal DNA of S. commune. About 50 transformants per dish was appeared in the complementation test by pHIONA8 using S. commune tryptophan auxotroph as host. In the mating test between transformants and other mating type alleles, the fruiting body of S. commune was formed at $30^{\circ}C$ in $2{\sim}3$ weeks.

Isolation of Trp, Thr Overproducing Strain of Saccharomyces cerevisiae (Trp, Thr Analogue 복합 저항성 Saccharomyces cerevisiae 균주 개발)

  • To isolate a mutant which overproduces threonine and tryptophan, mutants of Saccharomyces cerevisiae were screened after UV and EMS mutagenesis. Hydroxynorvaline, a Thr analogue was used for selection of a Thr-overproducing mutant after UV mutagenesis. Among 31 mutants, TC 5-1 was selected as the strain candidate, based on amino acid analysis. TC 5-1 was then treated by EMS mutagenesis for Trp overproduction. Eight mutants were selected using fluorotryptophan for Thr and Trp overproducing strains. Amino acid analysis results showed that TC 6-1 was the best strain since it had the highest amount of Thr and Trp among mutants.
