Pitch canker of Pinus rigida, caused by Fusarium subglutinans f. sp. pini, was first noticed in Inchon, Korea, and is now being spread into other plantations of pines. Typical symptoms re resin flows from canders on the shoots, branches, and trunks, and resin-soaking of the sap wood under the bark. Pathogenic fungi were isolated from the infected shoots, branches, and sees on Fusarium-selective medium. Mycelial growth and microscopic characteristics were examined. Pathogenicity test was carried out by inoculating four common species of pines (P. rigida, P. densiflora, P. thunbergii, and P. koraiensis) in Korea. P. rigida and P. thunbergii showed symptoms identical to those of naturally infected trees, while P. densiflora and P. koraiensis remained free symptomless.
Dendropanax morbifera Leveille, an endemic species in Korea, is best known as a tree that produces a resinous sap. Although D. morbifera is used in folk medicine for various diseases, its active ingredients are largely unknown. In this study, we investigated antioxidative activities of ethanolic extracts of three parts of this plant including leaves, debarked stems, and roots. The root extracts exhibited strong 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activity compared with leaf or stem extracts. The root extracts showed hepatoprotective activity against t-butyl hydroperoxide-induced HepG2 cells, and reduced the ROS level in the cells. The root fractions lowered the mRNA level of COX-2 on lipopolysaccharide-stimulated Raw246.7 cells. These results suggest that ethanolic root extracts of D. morbifera are a source of antioxidant and hepatoprotective compounds, which indicate a potential for a botanical drug.
This is the basic study to investigate the amount of transpirational water loss in thrifty mature Quercus mongolica stand by the heat pulse method. The differences of heat pulse velocity by direction and depth, differences of heat pulse velocity by dominant, codominant and suppressed trees, diurnal changes of heat pulse velocity due to the change of leaf water potential, vapor pressure deficit and radiation, and sap flow path way in sapwood by dye penetration were measured in stems. Finally the amounts of daily and annual transpiration in stand were calculated by the heat pulse velocity. The results obtained were summarized as follows : 1. Relationship between heat pulse velocity(V) and sap flow rate(SFR) was obtained as a equation of SFR=1.37V. 2. The sap flow rate was high in the order of dominant, codominant, and suppressed trees. The daily heat pulse velocity changed with radiation, temperature and vapor pressure deficit. 3. The heat pulse velocity showed the similar diurnal variation as the leaf water potential change. 4. The heat pulse velocity showed the highest value in May(4.0cm/hr in average), the lowest one in July(2.9cm/hr in average). 5. The heat pulse velocity in the same stem presented the highest value in the northern direction, medium in western, and the lowest in southern and eastern. 6. The heat pulse velocity in stem was highest in 0.5cm, medium in 1.0cm, and lowest in 1.5cm depth from the surface of stem. 7. The sap flow path way in stem showed sectorial straight ascent pattern in four sample trees. 8. The amount of sap flow(SF) was presented as a equation of $SF=1.37A{\cdot}V$(A: the cross-sectional area of sapwood, V: heat pulse velocity), and especially SF was larger in dominant tree than codominant and suppressed trees. 9. The amount of daily transpiration was 5.6ton/ha/day, and its composition ratio was 72% at day and 28% at night. 10. The amount of stand transpiration per month was largest in May(168ton/ha/month), lowest in July(125ton/ha/month). The amount of stand transpiration per year was 839ton/ha/year.
Taxol contents in various parts of 15 years old yew tree were determined. The descending order of taxol content per unit mass was stem bark, root bark, needle and seed. In the seed, that order was seed coat, embryo and endosperm. The total amount of taxol extractable from a 15 years old yew tree was 1.68 gram. This amount was distributed in needle, stem bark, root bark and seeds as 48.0, 23.8, 27.9 and 0.4%, respectively. Altitudinal variation of taxol content was also observed. More taxol was observed in yew trees grown at high altitude over 1000m above sea level. Calli and suspension cultures were induced from various yew trees. The presence of taxo] in cultured cells was established bv HPLC. The taxol content in cultured cells were different according to the source of explants. These results may be useful for the goal of large scale taxol production by cultured yew tree cells.

In this study, a new method for extending storage-life and palatability of Betula platyphylla sap by applying lactic acid bacteria fermentation was developed. The fluids of saps were filtered through 0.22 ${\mu}m$ membrane filter and each fermented by 8 different lactic acid bacteria which are Lactobacillus acidophilus, Lactobacillus brevis, Leuconostoc mesenteroides, Leuconostoc lactis, Lactococcus lactis, Pediocossus pentosaceus, Pediococcus dextrinicus, Streptococcus thermophilus. All the tested lactic acid bacteria except P. dextrinicus grew fast up to $10^6{\sim}10^7cfu/mL$ levels and lowered pH down to about pH 4 levels in 48 hours in both saps. The produced organic acids and lowered pH level inhibited the growth of spoilage microorganisms almost completely for 2 weeks during storage at room temperature. Addition of xylitol in the saps before fermentation accelerated the growth of lactic acid bacteria and increased the sweetness and overall taste of final product. The filtration process did not affect the mineral compositions of Betula platyphylla saps. Also the compositions and amounts of minerals showed very minor differences before and after fermentation in Betula platyphylla saps inoculated with L. acidophilus. By applying lactic acid fermentation to extend storage-life of tree saps instead of heat treatment, it was possible to keep natural minerals in active forms without any modifications.

Targeted genome editing using the CRISPR/Cas9 system is a ground-breaking technology that is being widely used to produce plants with useful traits. However, for woody plants, only a few successful attempts have been reported. These successes have used Agrobacterium-mediated transformation, which has been reported to be very efficient at producing genetically modified trees. Nonetheless, there are unresolved problems with plasmid sequences that remain in the plant genome. In this study, we demonstrated a DNA-free genome editing technique in which purified CRISPR/Cas9 ribonucleoproteins (RNPs) are delivered directly to the protoplasts of a hybrid poplar (Populus alba × Populus glandulosa). We designed three single-guide RNAs (sgRNAs) to target the stress-associated protein 1 gene (PagSAP1) in the hybrid poplar. Deep sequencing results showed that pre-assembled RNPs had a more efficient target mutagenesis insertion and deletion (indel) frequency than did non-assembled RNPs. Moreover, the RNP of sgRNA3 had a significantly higher editing efficacy than those of sgRNA1 and sgRNA2. Our results suggest that the CRISPR/Cas9 ribonucleoprotein-mediated transfection approach is useful for the production of transgene-free genome-edited tree plants.
To inverstigate the boring insects in Pinus densiflora forests, bait logs were set up in healthy-looking forests of Chunchon Hongchon and in damaged forests(gall formation rate 70----) by Thecodiplosis japonensis of Pyongchang Jeongsun. The period of investigation was from April to August in 1999. Boring insects investigated were 35 species of 12 families. Five species of them were unrecorded species from Korea. Natural enemies investigated were 5 species of 2 families. The prefered parts of tree according to species of boring insects were lower trunk in Siphalinus gigas and 3 other species, middle-stem in Monochamus sutor and 7 other species, and top stem in Orthotomicus suturalis and 1 other species. Hylurgops interstitialis were found in all parts of tree. Sap wood was attacked by Xyleborus validus Cerambycidae, heart wood by Hylobitelus haroldi Siphalinus gigas, and cambium region by Pissodes nitidus P. obscurus Shirahoshizo insidiosus Scolytidae.
An isolate of Peanut stunt virus (PSV) isolated from black locust tree (Robinia pseudo-acacia L.) showing severe mosaic and malformation symptoms, was designated as PSV-Rp. PSV-Rp was characterized by the tests of host range, physical properties, RNA and coat protein composition and RT-PCR analysis. Nucleotide sequences of the cucumoviruses CP genes were also used for identification and differentiation of PSV-Rp. Six plant species were used in the host range test of PSV-Rp. PSV-Rp could be differentiated from each Cucumovirus strain used as a control by symptoms of the plants. The physical properties of PSV-Rp virus were TIP $65^{\circ}C$, DEP $10^{-3}$, and LIP $2{\sim}3$ days. In dsRNA analysis, PSV-Rp consisted of four dsRNAs, but satellite RNA was not detected. Analysis of the coat proteins by SDS-PAGE showed one major protein band of about 31 kDa. RT-PCR using a part of Cucumovirus RNA3 specific primer amplified ${\sim}950bp$ DNA fragments from the crude sap of virus-infected black locust leaves. RFLP analysis of the RT-PCR product could differential PSV-RP from CMV The nucleotide sequence identity between the PSV-Rp CP and the TAV-P CP genes and the PS-V-RP CP and CMV-Y CP genes were 61.6% and 40.5%, respectively. On the other hand, the nucleotide sequence identity of the PSV-Rp CP gene was $70.9%{\sim}73.4%$ in comparison with those of PSV subgroup I (PSV-ER and PSV-J) and 67.3% with that of PSV subgroup II(PSV-W). Especially, the nucleotide sequence identity of PSV-Rp CP gene and that of PSV-Mi that was proposed recently as the type member of a novel PSV subgroup III was 92.4%.
This study was carried out to investigate the identification of the woods of commercially important exotic tree species grown in Korea. The test trees used in this study were selected 21 species grown in Kwangyang and Suwon, Korea. The items of macroscopical features were observed and examined principally on the annual rings, transitions spring to the summerwood, pore types and arrangements, sap and heartwoods, wood colors, odor and tastes, resin ducts, parenchymas, and rays etc. The microscopical features observable in the elements, and their compositions such as vessels, tracheids, wood fibers, ray parenchyma cells, and intercellular cannals were observed and measured. The observed and measured results investigated were synthesized, and accordance with these results macroscopical and microscopical keys were prepared for the wood identification as seen in the text.

Synchrotron X-ray micro imaging technique was employed to non-invasively monitor the water flow inside xylem vessels in a bamboo leaf. The phase contrast X-ray images clearly visualized plant anatomy and the rise of a water front inside the vessels. Consecutive X-ray images taken for 60 seconds revealed water rise kinetics against gravity in the xylem of a cut dry leaf taken from a bamboo tree. For the first time, traces of water rise, variation of contact angle between water and xylem wall as well as the internal structure of xylem were obtained. In xylem vessels, a repeating flow pattern has a typical flow velocity of 30.7$\mu\textrm{m}$/s and faster flow is established intermittently. It is concluded that the transmission type of X-ray micro imaging can be used as a powerful tool to investigate the ascent of sap in the xylem vessels at a resolution higher than that of MRI.
