Study of Characterization for Lean NOx Trap Catalysts Utilizing a Bench-Flow Reactor System (Bench-Flow Reactor System을 이용한 Lean NOx Trap 촉매의 특성 연구)

  • The performance of Lean NOx Trap (LNT) based on the catalysts of Pt/K/Ba/$\gamma-Al_2O_3$ with proprietary washcoat formulation is studied using a bench flow reactor system. To investigate the effect of temperature and gas hourly space velocity (GHSV) on the nitrogen oxides (NOx) trapping capacity as well as NOx breakthrough time and final ratio of $NO_2$ to NO of LNT, series of adsorption isotherms are carried out with simulated exhaust gases of the lean burn engines. Since typical operation of LNT requires periodic regeneration with a short rich excursion, where the stored or trapped NOx is released and subsequently reduced to $N_2$, the effect of the duration of lean and rich phase and type of reductants on the NOx conversion is investigated. NOx storage capacity and breakthrough time obtained from adsorption isotherms shows a volcano-type dependence on the temperature with a maximum NOx storage capacity occurring $350^{\circ}C$ and with a maximum breakthrough time occurring $400^{\circ}C$ at all GHSVs investigated in this study. Also, maximum ratio of $NO_2$ to NO is obtained at $400^{\circ}C$ with a GHSV of $75,000\;hr^{-1}$ Lean/rich cycle of 100 s lean and 5 s rich used with a concentration of 1.33% of $H_2$ and 4% of CO in the rich phase is found to be optimum at operating temperature of $350^{\circ}C$ and a GHSV of $50,000\;hr^{-1}$.

Determination of Mequitazine in Human Plasma by Gas-Chro-matography/Mass Spectrometry with Ion-Trap Detector and Its Pharmacokinetics after Oral Administration to Volunteers

  • The objective of this study was to develop an assay for mequitazine (MQZ) for the study of the bioavailability of the drug in human subjects. Using one mL of human plasma, the pH of the sample was adjusted and MQZ in the aqueous phase extracted with hexane; the organic layer was then evaporated to dryness, reconstituted and an aliquot introduced to a gas chromatograph/mass spectrometer (GC/MS) system with ion-trap detector. Inter- and intra-day precision of the assay were less than 15.1 and $17.7{\%}$, respectively; Inter- and intra-day accuracy were less than 8.91 and $18.6{\%}$, respectively. The limit of quantification for the current assay was set at 1 ng/mL. To determine whether the current assay is applicable in a pharmacokinetic study for MQZ in human, oral formulation containing 10 mg MQZ was administered to healthy male subjects and blood samples collected. The current assay was able to quantify MQZ levels in most of the samples. The maximum concentration ($C_{max}$ was 8.5 ng/mL, which was obtained at 10.1 h, with mean half-life of approximately 45.5 h. Under the current sampling protocol, the ratio of $AUC_{t{\rightarrow}last}$ to $AUC_{t{\rightarrow}{\infty}}$ was $934{\%}$, indicating that the blood collection time of 216 h is reasonable for MQZ. Therefore, these observations indicate that an assay for MQZ in human plasma is developed by using GC/MS with ion-trap detector and validated for the study of pharmacokinetics of single oral dose of 10 mg MQZ, and that the current study design for the bioavailability study is adequate for the drug.

  • We studied the dependence of hot-tarrier-induced degradation characteristics on nitrogen concentration in NO(Nitrided-Oxide) gate of nMOS, under ac and dc stresses. The $\Delta$V$_{t}$ and $\Delta$G$_{m}$ dependence of nitrogen concentration were observed, We observed that device degradation was suppressed significantly when the nitrogen concentration in the gate was increased. Compared to $N_2$O oxynitride, NO oxynitride gate devices show a smaller sensitivity to ac stress frequency. Results suggest that the improved at-hot carrier immunity of the device with NO gate may be due to the significantly suppressed interface state generation and neutral trap generation during stress.ess.

Effects of Leptin on Osteoclast Generation and Activity

  • Leptin, the product of the obese gene, is a circulating hormone secreted primarily from adipocytes. Several results suggest that leptin is important mediators of bone metabolism. The present study was undertaken to determine the effects of leptin on anti-osteoclastogenesis using murine precursors cultured on Ca-P coated plates and on the production of osteoprotegerin (OPG) in osteoblastic cells. Additionally, this study examined the possible involvement of prostaglandin $E_2\;(PGE_2)$/protein kinase C (PKC)-mediated signals on the effect of leptin on anti-osteoclastogenesis to various culture systems of osteoclast precursors. Osteoclast generation was determined by counting tartrate-resistant acid phosphatase positive [TRAP (+)] multinucleated cells (MNCs). Osteoclastic activity was determined by measuring area of resorption pits formed by osteoclasts on Ca-P coated plate. The number of 1,25-dihydroxycholecalciferol $(1,25[OH]_2D_3)$- or $PGE_2$-induced TRAP (+) MNCs in the mouse bone marrow cell culture decreased significantly after treatment with leptin. The number of receptor activator of NF-kB ligand (RANKL)-induced TRAP (+) MNCs in M-CSF dependent bone marrow macrophage (MDBM) cell or RAW264.7 cell culture decreased significantly with leptin treatment. Indomethacin inhibited osteoclast generation induced by $1,25[OH]_2D_3$ and dexamethasone, however, no significant differences were found in the leptin treated group when compared to the corresponding indomethacin group. Phorbol 12-myristate 13-acetate (PMA), a PKC activator, inhibited osteoclast generation induced by $1,25[OH]_2D_3$. The number of TRAP (+) MNCs decreased significantly with treatment by PMA at concentrations of 0.01 and $0.1{\mu}M$ in culture. Leptin inhibited PMA-mediated osteoclast generation. Isoquinoline-5-sulfonic 2-methyl-1-piperazide dihydrochloride (H7) had no effect on osteoclast generation induced by $1,25[OH]_2D_3$. Cell culture treatment with leptin resulted in no significant differences in osteoclast generation compared to the corresponding H7 group. Indomethacin showed no significant effect on TRAP (+) MNCs formation from the RAW264.7 cell line. PMA inhibited TRAP (+) MNCs formation induced by RANKL in the RAW264.7 cell culture. H7 had no effect on osteoclast generation from the RAW264.7 cell line. There was no difference compared with the corresponding control group after treatment with leptin. $1,25[OH]_2D_3$- or $PGE_2$-induced osteoclastic activity decreased significantly with leptin treatment at a concentration of 100 ng/ml in mouse bone marrow cell culture. Indomethacin, PMA, and H7 significantly inhibited osteoclastic activity induced by $1,25[OH]_2D_3$ in mouse bone marrow cell culture. No significant differences were found between the leptin treated group and the corresponding control group. The secretion of OPG, a substance known to inhibit osteoclast formation, was detected from the osteoblasts. Treatment by leptin resulted in significant increases in OPG secretion by osteoblastic cells. Taken these results, leptin may be an important regulatory cytokines within the bone marrow microenvironment.

A Dielectrophoresis Microfluidic Device for Trapping Bioparticles at Low Voltage and Frequency

  • Purpose: The necessity for precise manipulation of bioparticles has greatly increased in the fields of bioscience, biomedical, and environmental monitoring. Dielectrophoresis (DEP) is considered to be an ideal technique to manipulate bioparticles. The objective of this study is to develop a DEP microfluidic device that can trap fluorescent beads, which mimic bioparticles, at the low voltage and frequency of the sinusoidal signal supplied to the microfluidic device. Methods: A DEP microfluidic device, which is composed of polydimethylsiloxane (PDMS) channels and interdigitated electrode networks, is fabricated to trap fluorescent beads. The geometry of the interdigitated electrodes is determined through computational simulation. To determine the optimum voltage and frequency of the sinusoidal signal supplied to the device, the experiments of trapping beads are conducted at various combinations of voltage and frequency. The performance of the DEP microfluidic device is evaluated by investigating the correlation between fluorescent intensities and bead concentrations. Results: The optimum ratio of the widths between the negative and positive electrodes was 1:4 ($20:80{\mu}m$) at a gap of $20{\mu}m$ between the two electrodes. The DEP electrode networks were fabricated based on this geometry and used for the bead trapping experiments. The optimum voltage and frequency of the supplied signal for trapping fluorescent beads were 15 V and 5 kHz, respectively. The fluorescent intensity of the trapped beads increased linearly as the bead concentration increased. The coefficient of determination ($R^2$) between the fluorescent intensity and the bead concentration was 0.989. Conclusions: It is concluded that the microfluidic device developed in this study is promising for trapping bioparticles, such as a cell or virus, if they are conjugated to beads, and their concentration is quantified.

  • The oil trap in oil separator is one of the most important characteristics for normal operation of compressor. In this study, oil separation characteristics has been investigated for $CO_2$/PAG mixture using a gravity type of oil separator. The experimental study has been carried out in the range of oil concentration 0 to 5 weight-percent and the mixture temperature range of $0^{\circ}C$ to $15^{\circ}C$. The results obtained indicate that oil separation ratio in oil separator is increased with an increase in the oil concentration and mixture temperature.

  • We carry out an experimental investigation to analyze the basic performance of NO(nitric oxide) storage in a lean phase and also analyze the NO reduction achieved by the spraying of reducing agents in the rich phase of the exhaust gas in an LNT(Lean NOx Trap). This is an after-treatment system used to reduce the NOx emissions from a diesel engine. If the stored NO is reduced, we measure the outlet concentration downstream of the LNT. The test LNT material used in the experiments is commercial LNT. After being canned into stainless-steel(SUS304), it was built in a micro bench-flow reactor. Compositions of feed gases, three heated and three no heated gases were sprayed upstream of the LNT to analyze the characteristics. We use various temperatures and space velocities as response variables.

  • Liquid-chromatography coupled with electrospray-ion trap mass spectrometry was applied to the analysis of the eleutherosides B and E in the Eleutherococcus senticosus cortexes. The optimum ESI/MS results were obtained in the positive ion mode using extracted ion chromatogram targeting Na-adduct molecular ion of each compound. This method allowed rapid and simple gradient separation of underivatized eleutherosides B and E without pre-purification steps at very low concentration.

Protective Effect of Yellow-Green Vegetable Juices on DNA Damage in Chinese Hamster Lung Cell Using Comet Assay (Comet Assay를 이용한 케일, 명일엽, 당근, 돌미나리 녹즙의 Chinese Hamster Lung 세포 DNA 손상 보호 효과)

  • The present study was attempted to investigate the antioxidant capacity of popular yellow-green vegetable juices (kale, Angelica keishei, carrot, small water dropwort) and to investigate the effect of vegetable juices on protecting oxidative damage to DNA in cultured Chinese hamster lung (CHL) cells. Antioxidant capacity was analyzed by TRAP assay (Total radical-trapping antioxidant potential). Cellular DNA dmamage was measured by SCGE (single-cell gel electrophoresis, also known as comet assay. Cells incubated in medium with PBS (negative control) or with various concentration of the freeze dried green juices (25, 50, 100, 250 $\mu\textrm{g}$/$m\ell$) resuspended in PBS were treated with $H_2O_2$ (200 ${\mu}{\textrm}{m}$) as an oxidative stimulus for 5 min at 4$^{\circ}C$. The physiological function of each vegetable juice on oxidative DNA damage was analyzed and expressed as tail moment (tail length X percentage migrated DNA in tail) . Kale juice had the highest TRAP value suggesting that kale has the highest antioxidant capacity followed by Angelica keishei, small water dropwort and carrot. Cells treated with $H_2O_2$ had extensive DNA damage compared with cells treated with PBS or pre-treated with vegetable juice extracts. All green juices inhibited $H_2O_2$-induced DNA damage with kale being the most effective juice among the tested juices. These results indicate that green juice supplementation to CHL cells followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species. A Study on the Mercury Emission Characteristic and Comparison Tests for Applicability of Latest Mercury Measuring Methods - Focus on the Cement Kiln - (시멘트 소성시설에서의 수은 배출특성 및 최신 측정방법 적용성 평가 연구)

  • Recently, there has been growing interest in the emission characteristics and behavior of anthropogenic mercury compounds from emission sources. It is required to establish a standard for reliable mercury measurement method. Therefore, this study has evaluated the applicability of the new measurement method; Continuous Emission Monitoring (US EPA 30A, CEM). In addition, the reliability evaluation was conducted through Ontario Hydro Method (ASTM D6784, OHM) and Sorbent trap method (US EPA Method 30B). As a monitoring result for three months via CEM from cement kiln, the maximum mercury compounds concentration was about $600{\mu}g/Sm^3$. This is because of the various of raw materials and fuel, and the absence of mercury-control device. The mercury compounds concentrations of OHM, Sorbent trap and CEM were 13.64 $(3.33{\sim}32.41){\mu}g/Sm^3$, $13.94(5.97{\sim}23.44){\mu}g/Sm^3$ and $14.68(6.19{\sim}26.75){\mu}g/Sm^3$, respectively. The relative standard deviations (% RSD) of the three methods were 5.1~40.9%. The result of this study suggest that it is possible to apply the CEM in the cement kiln when, QA/QC such as calibration is verified.
