Expression Patterns of Genes Involved in Carotenoid Biosynthesis in Pepper

  • To study the regulatory mechanism of isoprenoid (carotenoid) biosynthesis, we have compared the expression patterns of nine isoprenoid biosynthetic genes in Korean red pepper (Capsicum. annuum cv. NocKaung). The expression of geranylgeranyl pyrophosphate synthase gene was initially induced at early ripening stage (I1) and was rather slightly decreased during pepper fruit ripening. The ex-pression of phytoene synthase gene was strongly induced at semi-ripening stage (I2) and the phytoene desaturase transcript was maximally induced at the fully ripened stage (R). Our results suggest that genes encoding two 3-hydroxy-3-methylglutaryl-CoA reductase isozymes (HMGR1 and HMGR2) and farnesyl pyrophosphate synthase might be not so critical in pepper carotenoid biosynthesis but three genes encoding geranylgeranyl pyrophosphate synthase, phytoene synthase and phytoene desaturase were induced in a sequential manner and coordinately regulated during the ripening of pepper fruit.

  • 인터넷포털 사이트에서 제공되고 있는 지식검색 질문응답서비스의 성능을 평가하기 위하여, 이용량이 많은 상위 네 가지 사이트를 대상으로 질문응답 데이터를 체계적으로 표집하였다. 디지털 참고봉사의 평가 틀을 적용하여 질문의 수와 유형, 응답률, 적시성, 정확성, 만족도를 측정하여 분석함으로써 서비스 현황을 진단하고 개선방안을 제시하였다.

Regulation of Expression of the Bacillus caldolyticus Pyrimidine Biosynthetic Operon by pyrR Gene, an Autogenous Regulator

  • The pyrR gene of the pyrimidine biosynthesis (pyr) operon of the thermophile Bacillus caldolyticus, encoding a uracil phosphoribosyltransferase (UPRTase), turned to rely as a pyr operon regulator. It has been proposed that PyrR mediates transcriptional termination-antitermination at three intercistronic regions of the par operon (S.-Y Ghim and J. Neuhard, J. Bacteriol.,176, 3698-3707, 1994). In this research, a plasmid carrying the pyrR region of B. caldolyticus could restore a pyrimidine regulation in a pyrR mutant of B. subtilis. Expression of pyrR was found to increase 6-7 fold during pyrimidine starvation. Additionally, a highly conserved nucleotide sequence which may constitute the binding site for a PyrR protein (PyrR-binding loop) in transcript was staggested. Alternative antiterminator and terminator structures involving three conserved motifs in front of the pyrR, pyrP and pyrB genes, respectively, are proposed to account for the observed regulation pattern.

또래 놀이에 나타난 유아의 맥락 및 탈맥락적 언어사용 (Use of Contextualized and Decontextualized Talk in Preschoolers' Peer Play)

  • The purpose of this study is to investigate low- and middle-income preschoolers' use of contextualized and decontextualized talk. The contextualized and decontextualized talk of 70 five-year-old children were analyzed by children's gender and their socioeconomic status. The participants, as dyads, participated in a peer play session for 10 minutes. The play sessions were videotaped and the videotaped data were transcribed. Each transcript was analyzed according to the modified version of Curenton et al. (2008)'s coding schemes. The results showed that children from middle-income families engaged more in both contextualized and decontextualized talk than children from low-income families. Also, girls from middle-income families engaged in more contextualized-decontextualized talk(psychological states) than girls from low-income families.

  • The 3' UTR (3' untranslated region) plays important roles in controlling gene expression through regulating 3' polyadenylation, mRNA export, subcellular localization, translational efficiency, and mRNA stability. Changes in the 3' UTR sequence in an expressed transcript can result in functional changes of the genes that are expressed in pathological conditions compared with those genes expressed in normal physiologic conditions. A genome-wide survey of 3' UTR variation was performed for the genes expressed during hematopoietic differentiation from CD34+ stem/progenitor cells to CD 15 + myeloid progenitor cells. Wide-spread differential usage of the 3' UTR was observed from the genes expressed during this cellular transition. This study implies that the 3' UTR can be a highly coordinated region for post-transcriptional regulation of the function of expressed genes.

Overriding Photoperiod Sensitivity of Flowering Time by Constitutive Expression of a MADS Box Gene

  • The majority of plants sense environmental signals, such as day length or temperature, to select their transition timing from vegetative growth t flowering. Here, we report the identification of a regulatory gene, OsMADS1, that controls the photoperiod sensitivity of flowering time. Constitutive expression of OsMADS1 in a long-day flowering plant, Nicotiana sylvestris, resulted in flowering in both short-day long-day conditions. Similarly, ectopic expression of the gene in a short-day flowering plant, N. tabacum cv. Maryland Mammoth, also induced flowering regardless of the day length. The transition time was dependent on the level of the OsMADS1 transcript in transgenic plants. These suggest that OsMADS1 is a key regulatory factor that determines the transition from shoot apex to floral meristem and that it can be used for controlling flowering time in a variety of plant species.

LAMMER Kinase Lkh1 Is an Upstream Regulator of Prk1-Mediated Non-Sexual Flocculation in Fission Yeast

  • The cation-dependent galactose-specific flocculation activity of the Schizosaccharomyces pombe null mutant of $lkh1^+$, the gene encoding LAMMER kinase homolog, has previously been reported by our group. Here, we show that disruption of $prk1^+$, another flocculation associated regulatory kinase encoding gene, also resulted in cation-dependent galactosespecific flocculation. Deletion of prk1 increased the flocculation phenotype of the $lkh1^+$ null mutant and its overexpression reversed the flocculation of cells caused by lkh1 deletion. Transcript levels of $prk1^+$ were also decreased by $lkh1^+$ deletion. Cumulatively, these results indicate that Lkh1 is one of the negative regulators acting upstream of Prk1, regulating non-sexual flocculation in fission yeast.

Regulation of Macrophage Ceruloplasmin Gene Expression: One Paradigm of 3'-UTR-mediated Translational Control

  • Ceruloplasmin (Cp) is a copper protein with important functions in iron homeostasis and in inflammation. Cp mRNA expression is induced by interferon (IFN)-${\gamma}$ in U937 monocytic cells, but synthesis of Cp protein is halted after about 12 h by transcript-specific translational silencing. The silencing mechanism requires binding of a 4-component cytosolic inhibitor complex, IFN-gamma-activated inhibitor of translation (GAIT), to a defined structural element (GAIT element) in the Cp 3'-UTR. Translational silencing of Cp mRNA requires the essential proteins of mRNA circularization, suggesting that the translational inhibition requires end-to-end mRNA closure. These studies describe a new mechanism of translational control, and may shed light on the role that macrophage-derived Cp plays at the intersection of iron homeostasis and inflammation.

  • White-rot fungi have the following enzyme systems for lignin degradation: laccase, lignin peroxidase and manganese peroxidase. There are other types of peroxidases related to lignin degradation, one of which we have cloned a cDNA gene of manganese-repressed peroxidase (MrP) in Trametes versicolor isolated in South Korea. The mrp transcript level has been decreased by $1{\mu}M\;of\;Mn^{2+}$.
