Twelve clones of monoclonal antibodies (mAbs) against thyroglobulin were produced and characterized. Among them, three mAbs (TN-1, TN-2 and TN-3) showed high binding affinity to thyroglobulin. from ELISA inhibition assay, TN-2 showed considerable reactivity with soluble thyroglobulin. TN-2 also reacted with phosphatidylserine which has a negative charge in aqueous condition. These results suggest that TN-2 has characteristics of autoantibody concerned with thyroiditis.

Background : Cardiac troponin I (cTnI) is a specific marker of myocardial injury. It is known that a higher level of cTnI is associated with a poor clinical outcome in patients with acute coronary syndrome. An elevation in cTnI is also observed in various noncardiac critical illnesses. This study evaluated whether cTnI is useful for predicting the prognosis in noncardiac critically ill patients. Methods : From June 2003 to July 2004 at Gyeongsang National University Hospital, we enrolled 215 patients (male:142, female:73, mean age:$63{\pm}15$ years ) who were admitted for critical illness other than acute coronary syndrome at the medical intensive care unit(ICU). The severity score of critical illness (SAPS II and SOFA) was determined and serum cTnI level was measured within 24 hours after admission to the ICU. The mortality rate was compared between the cTnI-positive (${\geq}0.1{\mu}g/L$) and cTnI-negative ($cTnI<0.1{\mu}g/L$) patients at the $10^{th}$ and $30^{th}$ day after admission to the ICU. The mean cTnI value was compared between the survivors and non-survivors at the $10^{th}$ and $30^{th}$ day after admission to the ICU in the cTnI-positive patients. The correlation between cTnI and the severity of the critical illness score (SAPS II and SOFA) was also analyzed in cTnI-positive patients. Results : 1) The number of cTnI-negative and positive patients were 95(44%) and 120(56%), respectively. 2) The mortality rate at the $10^{th}$ and $30^{th}$ day after admission to the ICU was significantly higher in the cTnI-positive patients (29%, 41%) than in the cTnI-negative patients (12%, 21%)(p<0.01). 3) In the cTnI-positive patients, the mean value of the cTnI at the $10^{th}$ and $30^{th}$ day after admission to the ICU was significantly higher in the non-survivors ($4.5{\pm}9.2{\mu}g/L$, $3.5{\pm}7.9{\mu}g/L$) than in the survivors($1.8{\pm}3.6{\mu}g/L$, $2.0{\pm}3.9{\mu}g/L$) (p < 0.05). 4) In the cTnI-positive patients, the cTnI level was significantly correlated with the SAPS II score (r=0.24, p<0.001) and SOFA score (r=0.30, p<0.001). Conclusion : The cTnI may be a useful prognostic marker in noncardiac critically ill patients.
We tested gentamicin - resistant bacteria isolated from hospital sewage to confirm the presence of aac(3)II encoding aminoglycoside- (3)-N- acetyltransferase by dot-blot hybridization. A probe from the internal fragment of aac(3)II was hybridized to DNA from 41 % (39/95) of gentamicin resistant isolates. PCR was performed with primers from aac(3)II and Tn3. Of 39 strains, 13 strains had Tn3-aac(3)II structure. Minimal inhibitory concentration (MIC) test demonstrated that 18 strains containing Tn3-aac(3)II showed higher resistance to gentamicin than those of other strains. Thirteen strains were identified as 5 Escherichia coli, 3 Acinetobacter johnsonii, 2 Enterobacter agglomerans, 2 Micrococcus luteus, and 1 Pseudomonas facilis. These results suggest that gentamicin-resistant determinant of Tn3-aac(3)II structure was widely distributed in the gentamicin-resistant bacteria.

This paper presents results from a study of the effects of age and sex on body and carcass composition of different goat genotypes. A completely randomized $3{\times}3{\times}2$ factorial design was used. Factors were genotype (Thai Native; TN, $50%TN{\times}50%$ Anglo-Nubian (AN) and $25%TN{\times}75%AN$ goats), age ($206{\pm}8$, $349{\pm}9$ and $428{\pm}8days$ or 6.9, 11.6 and 14.3 months, respectively) and sex (male and female). It was shown that TN kids had significantly higher (p < 0.05) hind percentage (7.9%) compared with $50%TN{\times}50% AN$ (7.2%) and $25%TN{\times}75%AN$ (7.1 %) kids. TN kids had a significantly (p < 0.05) higher heart percentage (0.44%) than that of $25%TN{\times}75%AN$ kids (0.38%). However, there was no significant difference in heart percentage between TN and $50%TN{\times}50%AN$ and between $50% TN{\times}50%AN$ and $25%TN{\times}75%AN$ kids. TN and $50%TN{\times}50%AN$ kids had significantly higher (p < 0.05) carcass muscle content (63.1 and 63.7%, respectively) than did $25%TN{\times}75%AN$ kids (59.9%). Kids at 6.9 months of age had significantly higher percentages of hide, feet, intestinal tract, liver, heart, kidneys and carcass bone than those of kids at 11.6 and 14.3 months of age. There were no significant differences in these characteristics between kids at 11.6 and 14.3 months of age. However, kids at 11.6 and 14.3 months of age had significantly higher percentages of omental fat, dressing carcass, muscle, total fat (subcutaneous + intermuscular + kidney and pelvic fat), muscle to bone ratios (MBR), and muscle plus fat to bone ratios (MFBR) than those of kids at 6.9 months. Males had significantly higher gut contents, but lower percentages of liver, omental fat and heart. Prediction equations were developed relating empty body weight (EBW) to hot carcass weight and some dissectible carcass fractions.
Tetranectin (TN), an adipogenic serum protein, enhances adipocyte differentiation, however, its functional mechanism has yet to be elucidated. In the present study, we investigated the adipogenic function of TN by using medium containing TN-depleted fetal bovine serum (TN-del-FBS) and recombinant mouse TN (mTN). The adipocyte differentiation of 3T3-L1 cells was significantly enhanced by mTN supplementation essentially at differentiation induction, which indicated a potential role of the protein in the early differentiation phase. The adipogenic effect of mTN was more significant with insulin in the differentiation induction cocktail, implicating their close functional relationship. mTN enhanced not only the proliferation of growing cells, but also mitotic clonal expansion (MCE) that is a prerequisite for adipocyte differentiation in the early phase. Consistently, mTN increased the phosphorylation of ERK in the early phase of adipocyte differentiation. Results of this study demonstrate that the adipogenic function of mTN is mediated by enhancing MCE via ERK signaling.
This paper presents results from a study of the age and weight at puberty of Thai does. A randomized block $3{\times}3$ factorial design was used. Factors were genotype (Thai Native; TN, 75% TN $\times$ 25% Anglo-Nubian; AN and 50 TN $\times$ 50% AN does), year of kidding (1989, 1990 and 1991) and birth type (single or twin) as a block. It was shown that there were no significant (p > 0.05) difference in age at puberty among genotypes ($167.6{\pm}6.0$, $157.6{\pm}7.0$ and 160.0 = 4.7 days for TN, 75% TN $\times$ 25% AN and 50% TN $\times$ 50% AN does, respectively) and between birth types. However, does kidded in 1991 had significantly (p < 0.01) longer age at puberty ($194.0{\pm}7.4$ days) than did does kidded in 1989 ($143.0{\pm}4.5$ days) or in 1990 ($148.1{\pm}5.7$ days). There was an interaction effect between genotype and year. Fifty % TN $\times$ 50% AN does had significantly (p < 0.05) higher weights at pubery ($17.2{\pm}0.4kg$) than those of TN($14.3{\pm}0.5kg$ and 75% TN $\times$ 25% AN ($14.4{\pm}0.6kg$) does. There was no significant difference in weights at puberty between TN and 75% TN $\times$ 25% AN does. There was no effect of birth type on weight at puberty. There was an interaction effect between genotype and year of kidding on age and weight at puberty and on growth rate from birth to weaning.
Two gentamicin resistance R plasmids, pGM5 and pGM6, containing aacC2 gene were selected from environmental isolates. The gentamicin resistance determinants of R plasmids were cloned into the BamHI site of pUC18. Restriction enzyme map of inserted region of recombinant plasmids, pSYS and pSY6, and PCR results indicated that Tn3 sequence was located upstream of gentamicin resistance gene. Based on the restriction maps and susceptibility tests, it was concluded that the sequence of bla and 3' inverted repeat of Tn3 play a important roles in the expression of gentamicin resistance gene.
The bacteriophage Mu and transposon Tn5 containing plasmid pJB4JI-transferred transposon Tn5 to Caulobuter crescentus. When several thousand of transposon Tn5 insertion mutants were examined, we found auxotrophic and motility mutants at frequencies of 2% and 3%, respectively. Transposition of transposon Tn5 was analyzed by the reverse field electrophoresis and Southern hybridization. The results indicated that transposon Tn5 was randomly inserted to Caulobuter crescentus chromosome but the plasmid vector, pJB4JI, was not maintained.
Mobile genetic segments, or transposons, are also referred to as jumping genes as they can shift from one position in the genome to another, thus inducing a chromosomal mutation. According to the target site-specificity of the transposon during a transposition event, the result is either the insertion of a gene of interest at a specific chromosomal site, or the creation of knockout mutants. The former situation includes the integration of conjugative transposons via site-specific recombination, several transposons preferring a target site of a conserved AT-rich sequence, and Tn7 being site-specifically inserted at attTn7, the downstream of the essential glmS gene. The latter situation is exploited for random mutagenesis in many prokaryotes, including IS (insertion sequence) elements, mariner, Mu, Tn3 derivatives (Tn4430 and Tn917), Tn5, modified Tn7, Tn10, Tn552, and Ty1, enabling a variety of genetic manipulations. Randomly inserted transposons have been previously employed for a variety of applications such as genetic footprinting, gene transcriptional and translational fusion, signature-tagged mutagenesis (STM), DNA or cDNA sequencing, transposon site hybridization (TraSH), and scanning linker mutagenesis (SLM). Therefore, transposon-mediated genetic engineering is a valuable discipline for the study of bacterial physiology and pathogenesis in living hosts.
Background: Trigeminal neuralgia (TN) is one of the most painful disorder in the orofacial region, and many patients have suffered from this disease. For the effective management of TN, fundamental epidemiologic data related to the target population group are essential. Thus, this study was performed to clarify the epidemiological characteristics of TN in the Korean population. This is the first national study to investigate the prevalence of TN in Korean patients. Methods: From 2014 to 2018, population-based medical data for 51,276,314 subscribers to the National Health Insurance Service of Korea were used for this study. Results: The incidence of TN was 100.21 per 100,000 person-years in the year of 2018 in Korea, and the male to female ratio was 1:2.14. The age group of 51-59 years had the highest prevalence of TN. Constant increases in medical cost, regional imbalance, and differences in prescription patterns by the medical specialties were showed in the management of TN. Conclusions: The results in this study will not only help to study the characteristics of TN, but also serve as an important basis for the effective management of TN in Korea.
