Breeding on High Lycopene and Beta Carotene with Multi-Disease Resistance in Tomato

  • This study was carried out to breed and develop high quality and functional nutrient tomato with multi disease resistance as well as a stable growing adaptation for fresh market usage under protected plastic houses cultivation. The materials were used 5 inbred lines and their 6 hybrids of large tomato group, which have been bred and developed from 1999 to 2007 in Division of Plant Resource Department of Chungnam National University. Fruit weight showed hybrid vigor effect that $F_1$ hybrids weighed more than their parent lines, fruit shape formed three type of oblate, deep oblate and globe shape, in firmness and pericarp thickness have got a high significant correlation, inbred DN611 line was measured the most firm fruit with 6.04 mm pericarp thickness. In fruit color at maturity, pink color crossed to red color appeared all red fruit color in the $F_1$ hybrids, it means red skin color is a dominant gene compared to pink skin color is a recessive gene in tomato, while between fruit skin color and shoulder part color showed no any co-relationship. The sugar content and titratable acid of $F_1$ hybrids inherited an intermediate data of their parent lines, the flavor of KP543 inbred line and the hybrid (JB535 x KP543) revealed the better taste with high brix and proper titratable acid content$^{*}$. In beta carotene content DN611 line showed 2~3 times higher than other materials so that its 3 hybrids contained an increased level of beta carotene, lycopene content was not so much difference among inbred lines and $F_1$ hybrids, of them MD508 contained higher of 8.72 mg and hybrid (JB535 x JA517) had 8.05 mg lycopene content per 100 g fruit, overall pink skin color and red skin color measured a higher lycopene content than yellow and orange skin color at ripe stage. In disease resistance test by PCR marker for Fusarium race2 (I2), Nematode (Mi1), ToMV ($Tm2^2$), Cladosporium (Cf9), (JB535 x JA517) hybrid have got multi-resistance with homozygote band in Nematode, ToMV, Cladosporium and heterozygote band in Fusarium race2. Through this breeding program we could select high quality and functional nutrient with multi resistant $F_1$ hybrids and inbred lines in tomato which are two best hybrids (JB535 x MD508), (JB535 x JA517), additionally developed high beta carotene inbred line DN611 and increased the level of lycopene inbred line MD508. These results will be very useful to make a high quality tomato variety continuously.

Analysis of Lake Water Temperature and Seasonal Stratification in the Han River System from Time-Series of Landsat Images (Landsat 시계열 영상을 이용한 한강 수계 호수 수온과 계절적 성충 현상 분석)

  • We have analyzed surface water temperature and seasonal stratification of lakes in the Han river system using time-series Landsat images and in situ measurement data. Using NASA equation, at-satellite temperature is derived from 29 Landsat-5 TM and Landsat-7 ETM+ images obtained from 1994 to 2004, and was compared with in situ surface temperature on river-type dam lakes such as Paro, Chuncheon, Euiam, Chongpyong, Paldang, and with 10m-depth temperature on lake-type dam lake Soyang. Although the in situ temperature at the time of satellite data acquisition was interpolated from monthly measurements, the number of images with standard deviation of temperature difference (at-satellite temperature - in situ interpolated temperature) less than $2^{\circ}C$ was 24 on which a novel statistical atmospheric correction could be applied. The correlation coefficient at Lake Soyang was 0.915 (0.950 after correction) and 0.951-0.980 (0.979-0.997 after correction) at other lakes. This high correlation implies that there exist a mixed layer in the shallow river-like dam lakes due to physical mixing from continuous influx and efflux, and the daily and hourly temperature change is not fluctuating. At Lake Soyang, an anomalous temperature difference was observed from April to July where at-satellite temperature is $3-5^{\circ}C$ higher than in situ interpolated temperature. Located in the uppermost part of the Han river system and its influx is governed only by natural precipitation, Lake Soyang develops stratification during this time with rising sun elevation and no physical mixture from influx in this relatively dry season of the year.

Multi-carbohydrase application into energy and amino acid deficient broiler diets: A strategy to enhance performance of broiler chickens

  • The effect of Multi-Carbohydrase (MC) supplementation on growth performance, visceral organ weights, blood metabolites, jejunum morphology, nutrient digestibility, and carcass parameters of broiler chickens fed nutrient-deficient corn soybean-meal based diets containing high levels of non-starch polysaccharides from wheat and wheat by-products was investigated. A total of 378 one-day-old Ross 308 broiler chickens were randomly assigned to one of seven dietary treatments to give six replicates per treatment (nine birds per pen). Dietary treatments were as follows: (1) positive control (PC; commercial standard diet); (2) negative control 1 (NC-1; PC-120 kcal/kg metabolizable energy); (3) NC-2 (PC-3% standardized ileal digestibility [SID] amino acids). The remaining four dietary treatments were formulated with the addition of MC (MC; Superzyme-CSTM) into two negative controls along with two supplementation levels of MC (i.e., 0.025% and 0.05%, respectively). Improved body weight, average daily gain, and feed conversion ratio (p < 0.05) were observed in broiler chickens fed a reduced energy diet supplemented with MC compared to birds fed NC-1 diet from days 1-35. Additionally, birds fed a reduced energy diet with 0.05% MC showed comparable (p > 0.05) growth performance with birds fed PC for 35-day post-hatch. Furthermore, the addition of MC into reduced amino acid diets improved (p < 0.05) growth performance. Broiler chickens fed MC supplemented nutrient-deficient diets showed a greater (p < 0.05) villus height to crypt depth ratio than birds fed diets without MC on days 21 and 35. Similarly, improved (p < 0.05) nutrient digestibility was observed in birds fed reduced energy diets supplemented with MC compared to birds fed NC-1 on days 21 and 35. Our results suggest that MC supplementation into reduced energy or reduced amino acid diets containing wheat and wheat by-products has the potential to improve growth performance and nutrient digestibility while maintaining healthier gut morphology in broiler chickens from 1 to 35 days of age.

Rehabilitation of the edentulous patient with implant overdenture using CAD-CAM denture system: A case report (CAD-CAM으로 제작된 임플란트 피개의치를 이용한 무치악 환자의 보철 수복 증례)

  • This case report introduces a 74-year-old male patient who visited retention deficiency of the mandibular implant overdenture, which was fabricated 20 years ago. When the existing dentures were used, the vertical dimension was reduced, the maxillary complete denture lacked lip support and retention, and the mandibular overdenture lacked clip retention due to damage to the bar attachment. After removing the damaged bar attachment, it was replaced with a ball attachment, and impressions were taken using the DENTCATM Tray and then the vertical dimension was measured. The gothic arch tracing was performed to record the centric relation. Obtained impressions were scanned and the shape of final dentures was designed using software and try-in dentures were fabricated using 3D printer. After evaluating the occlusal plane, occlusal relationship, facial shape, and pronunciation using the try-in dentures, the bite registration was recorded, and the final denture was manufactured based on this. The inner surface of the denture was adjusted and bilateral balanced occlusion was formed, and the housing was connected to the mandibular denture by a direct method. This case reports have shown satisfactory resultin recovering improved retention and esthetic outcome by increasing the vertical dimension and the lip support using CAD-CAM technique and the ball attachment.

  • These studies were conducted to find out the polishing methods that improve yield and quality of the polished barley. Four varieties of hulled barley, Dongbori 1. Bunong, Kangbori and Suwon 182 which were produced in Suwon, Korea in 1979, were subjected to this experiment. The polishing machine, manufactured by Satake Co, was used as test mill. Increasing the roller speed of polishing machine causes more polished barley in a unit period. The speed influenced more in length than thickness or width of grain. Therefore the shape of grain became bold type as the speed increased. The optimum roller speed was 1,300rpm in ideal shape of polished barley. The lowest hardness was observed in the husk layer and the hardness was found in the decreasing order of the aleurone, testa, peri carp and the endosperm layer. The thickness of bran layer, weight of 1,000 kernel and hardness of polished barley were greatly different according to barley varieties. Also the length, thickness, width and the ratio of length to width of barley grain were significantly different in barley varieties. The ratio of length to width of the polished barley was 1.59 in Suwon 182, 1.53 in Bunong, 1.51 in Kangbori and 1.26 in Dongbori 1.

Carcinogenicity and mutagenicity of heterocyclic amines in transgenic models

  • 2-Amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) is a mutagenic and carcinogenic heterocyclic amino found in cooked meat. The in vivo mutagenicity and hepatocarcinogenicity of MeIQx were examined in mice harboring the lacZ mutation reporter gene ($Muta^{TM}$ Mice) and bitransgenic mice over-expressing the c-myc oncogene. C57B1/$\lambda$lacZ and bitransgenic c-myc (albumin promoter)/$\lambda$lacZ mice were bred and weaned onto an AIN-76 based diet containing $0.06\%$ (w/w) MeIQx or onto control diet. After 30 weeks on diet, only male bitransgenic mice on MeIQx developed hepatocellular carcinoma ($100\%$ incidence) indicating that there was synergism between c-myc over-expression and MeIQx. By 40 weeks, hepatic tumor incidence was $100\%$ ($17\%$) and $44\%$ ($0\%$) in male c-myc/$\lambda$lacZ and C57B1/$\lambda$lacZ mice given MeIQx (or control) diet, respectively, indicating that either MeIQx or c-myc over-expression alone eventually induced hepatic tumors. At either time point, mutant frequency in the lacZ gene was at least 40-fold higher in MeIQx-treated mice than in control mice of either strain. These findings suggest that MeIQx-induced hepatocarcinogenesis is associated with MeIQx-induced mutations. Elevated mutant frequency in MeIQx-treated mice also occurred concomitant with the formation of MeIQx-guanine adducts as detected by the $^{32}P$-postlabeling assay. Irrespective of strain or diet, sequence analysis of the lacZ mutants from male mouse liver showed that the principal sequence alteration was a single guanine-base substitution. Adenine mutations, however, were detected only in animals on control diet. MeIQx-fed mice harboring the c-myc oncogene showed a l.4-2.6-fold higher mutant frequency in the lacZ gene than mice not carrying the transgene. Although there was a trend toward higher adduct levels in c-myc mice, MeIQx-DNA adduct levels were not significantly different between c-myc/$\lambda$lacZ and C57B1/$\lambda$lacZ mice after 30 weeks on diet. Thus, it appeared that factors in addition to MeIQx-DNA adduct levels, such as the enhance rate of proliferation associated with c-myc over-expression, may have accounted for a higher mutant frequency in c-myc mice. In the control diet groups, the lacZ mutant frequency was significantly higher in c-myc/$\lambda$lacZ mice than in 057B1/$\lambda$1acZ mice. The findings are consistent with the notion that c-myc over-expression is associated with an increase in mutagenesis. The mechanism for the synergistic effects of c-myc over-expression on MeIQx hepatocarcinogenicity appears to involve an enhancement of MeIQx-induced mutations.

Seismic analysis and dynamic behavior characterization of rib-reinforced pre-cast tunnels (리브 보강 프리캐스트 터널의 내진 해석 및 동적거동 특성 파악)

  • The novel cut-and-cover tunnel construction method using rib-reinforced pre-cast arch segments has been recently developed and applied for practice to secure a structural stability of high covering and wide width section tunnels. Cut-and-cover tunnels are usually damaged by the seismic behavior of backfill grounds in case of a low covering condition. Seismic analyses are performed in this study to characterize the dynamic behavior of rib-reinforced pre-cast arch cut-and-cover tunnels. Seismic analyzes for 2 lane cast-in-place and rib-reinforced pre-cast arch cut-and-cover tunnels are carried out by using the commercial FDM program (FLAC2D) considering various field conditions such as the covering height embankment slope and excavation slope. It can be concluded that the amplification of seismic wave is reduced due to an increase in the structural stiffness induced by rib-reinforcement. The results show that the rib-reinforced pre-cast arch cut-and-cover tunnels are more effective against the seismic loading, compared to the cast-in-place cut-and-cover tunnels.

Seismic analysis of tunnel considering the strain-dependent shear modulus and damping ratio of a Jointed rock mass (절리암반의 변형률 의존적 전단탄성계수 및 감쇠비 특성을 고려한 터널의 내진 해석)

  • Contrary to an intact rock, the jointed rock mass shows strain-dependent deformation characteristics (elastic modulus and damping ratio). The maximum elastic modulus of a rock mass can be obtained from an elastic wave-based exploration in a small strain level and applied to seismic analyses. However, the assessment and application of the non-linear characteristics of rock masses in a small to medium strain level ($10^{-4}{\sim}0.5%$) have not been carried out yet. A non-linear dynamic analysis module is newly developed for FLAC3D to simulate strain-dependent shear modulus degradation and damping ratio amplification characteristics. The developed module is verified by analyzing the change of the Ricker wave propagation. Strain-dependent non-linear characteristics are obtained from disks of cored samples using a rock mass dynamic testing apparatus which can evaluate wave propagation characteristics in a jointed rock column. Using the experimental results and the developed non-linear dynamic module, seismic analyses are performed for the intersection of a shaft and an inclined tunnel. The numerical results show that vertical and horizontal displacements of non-linear analyses are larger than those of linear analyses. Also, non-linear analyses induce bigger bending compressive stresses acting on the lining. The bending compressive stress concentrates at the intersection part. The fundamental understanding of a strain-dependent jointed rock mass behavior is achieved in this study and the analytical procedure suggested can be effectively applied to field designs and analyses.

SonazoidTM versus SonoVue® for Diagnosing Hepatocellular Carcinoma Using Contrast-Enhanced Ultrasound in At-Risk Individuals: A Prospective, Single-Center, Intraindividual, Noninferiority Study

  • Objective: To determine whether Sonazoid-enhanced ultrasound (SZUS) was noninferior to SonoVue-enhanced ultrasound (SVUS) in diagnosing hepatocellular carcinoma (HCC) using the same diagnostic criteria. Materials and Methods: This prospective, single-center, noninferiority study (NCT04847726) enrolled 105 at-risk participants (71 male; mean age ± standard deviation, 63 ± 11 years; range, 26-86 years) with treatment-naïve solid hepatic nodules (≥ 1 cm). All participants underwent same-day SZUS (experimental method) and SVUS (control method) for one representative nodule per participant. Images were interpreted by three readers (the operator and two independent readers). All malignancies were diagnosed histopathologically, while the benignity of other lesions was confirmed by follow-up stability or pathology. The primary endpoint was per-lesion diagnostic accuracy for HCC pooled across three readers using the conventional contrast-enhanced ultrasound diagnostic criteria, including arterial phase hyperenhancement followed by mild (assessed within 2 minutes after contrast injection) and late (≥ 60 seconds with a delay of 5 minutes) washout. The noninferiority delta was -10%p. Furthermore, different time delays were compared as washout criteria in SZUS, including delays of 2, 5, and > 10 minutes. Results: A total of 105 lesions (HCCs [n = 61], non-HCC malignancies [n = 19], and benign [n = 25]) were evaluated. Using the 5-minutes washout criterion, per-lesion accuracy of SZUS pooled across the three readers (72.4%; 95% confidence interval [CI], 64.1%-79.3%) was noninferior to that of SVUS (71.4%; 95% CI, 63.1%-78.6%), meeting the statistical criterion for non-inferiority (difference of 0.95%p; 95% CI, -3.8%p-5.7%p). The arterial phase hyperenhancement combined with the 5-minutes washout criterion showed the same sensitivity as that of the > 10-minutes criterion (59.0% vs. 59.0%, p = 0.989), and the specificities were not significantly different (90.9% vs. 86.4%, p = 0.072). Conclusion: SZUS was noninferior to SVUS for diagnosing HCC in at-risk patients using the same diagnostic criteria. No significant improvement in HCC diagnosis was observed by extending the washout time delay from 5 to 10 minutes.

[ $^1H$ ] MR Spectroscopy of the Normal Human Brains: Comparison between Signa and Echospeed 1.5 T System (정상 뇌의 수소 자기공명분광 소견: 1.5 T Signa와 Echospeed 자기공명영상기기에서의 비교)

  • Purpose : To evaluate the usefulness and reproducibility of $^1H$ MRS in different 1.5 T MR machines with different coils to compare the SNR, scan time and the spectral patterns in different brain regions in normal volunteers. Materials and Methods : Localized $^1H$ MR spectroscopy ($^1H$ MRS) was performed in a total of 10 normal volunteers (age; 20-45 years) with spectral parameters adjusted by the autoprescan routine (PROBE package). In all volunteers, MRS was performed in a three times using conventional MRS (Signa Horizon) with 1 channel coil and upgraded MRS (Echospeed plus with EXCITE) with both 1 channel and 8 channel coil. Using these three different machines and coils, SNRs of the spectra in both phantom and volunteers and (pre)scan time of MRS were compared. Two regions of the human brain (basal ganglia and deep white matter) were examined and relative metabolite ratios (NAA/Cr, Cho/Cr, and mI/Cr ratios) were measured in all volunteers. For all spectra, a STEAM localization sequence with three-pulse CHESS $H_2O$ suppression was used, with the following acquisition parameters: TR=3.0/2.0 sec, TE=30 msec, TM=13.7 msec, SW=2500 Hz, SI=2048 pts, AVG : 64/128, and NEX=2/8 (Signa/Echospeed). Results : The SNR was about over $30\%$ higher in Echospeed machine and time for prescan and scan was almost same in different machines and coils. Reliable spectra were obtained on both MRS systems and there were no significant differences in spectral patterns and relative metabolite ratios in two brain regions (p>0.05). Conclusion : Both conventional and new MRI systems are highly reliable and reproducible for $^1H$ MR spectroscopic examinations in human brains and there are no significant differences in applications for $^1H$ MRS between two different MRI systems.
