Identification of a new marine bacterium Ruegeria sp. 50C-3 isolated from seawater of Uljin in Korea and production of thermostable enzymes (대한민국 울진 연안 해양에서 분리한 해양 미생물 Ruegeria sp. 50C-3의 동정 및 내열성 효소 생산)

  • A marine bacterium, designated as strain 50C-3, was isolated from a seawater sample collected from the East Sea of South Korea. The strain is a Gram-negative, aerobic, yellow colored polar-flagellated bacterium that grows at $20-50^{\circ}C$ and pH 5.5-8.5. Optimal growth occurred at $40-50^{\circ}C$, at pH 6.5-7.5, and in the presence of 2% (w/v) NaCl. Based on 16S rRNA gene sequence similarity, the isolate was considered to represent a member of the genus Ruegeria. The result of this analysis showed that strain 50C-3 shared 99.4% and 96.98% sequence similarity with Ruegeria intermedia CC-GIMAT-$2^T$ and Ruegeria lacuscaerulensis ITI-$1157^T$, respectively. Furthermore, strain 50C-3 showed clear differences from related strains in terms of several characteristics such as motility, carbon utilization, enzyme production, etc. The DNA G+C content was 66.7 mol%. Chemotaxonomic analysis indicated ubiquinone-10 (Q-10) as the predominant respiratory quinone. Based on phenotypic, chemotaxonomic, and phylogenetic characteristics, the isolate represents a novel variant of the Ruegeria intermedia CC-GIMAT-$2^T$, for which we named Ruegeria sp. 50C-3 (KCTC23890=DSM25519). Strain 50C-3 did not produce cellulase and agarase, but produced alkaline phosphatase, ${\alpha}$-galactosidase, and ${\beta}$-galactosidase. The three enzymes showed stable activities even at $50^{\circ}C$ and thus regarded as thermostable enzymes. Especially, the ${\beta}$-galactosidase activity enhanced by 1.9 times at $50^{\circ}C$ than that at $37^{\circ}C$, which may be very useful for industrial application.

Neutralization of Acidity and Ionic Composition of Rainwater in Taean (태안지역 강우의 산성도 중화 및 화학성 평가)

  • The issue of acid precipitation and related environmental problems in East Asia has been emerging. To evaluate the acidity and chemical characteristics of rainwater in Korea, its chemical properties during cultivation season from April to October were investigated at Taean in 2007. Also, to estimate the contribution of ions on its acidity, ion composition characteristics and neutralization effects by cation ions were determined. The ion balance between cations and anions values showed high correlation. The mean values of pH and EC were 4.9 and $32.9{\mu}S\;cm^{-1}$, respectively. The EC of rainwater showed seasonal characteristic, which was $91.4{\mu}S\;cm^{-1}$ with relatively low rainfall compared with other monitoring periods. $Na^+$ was the main cation followed by $NH_4{^+}$ > $Ca^{2+}$ > $H^{+}$ > $Mg^{2+}$ > $K^+$. Among these ions, $Na^{+}$ and $NH_4{^+}$ covered over 70% of total cations. In the case of anion, the order was $SO_4{^{2-}}$ > $NO_3{^-}$ > $Cl^{-}$. The mean value of sulfate, which is main anion component in the samples was $152.1{\mu}eq\;L^{-1}$. Also, 90% of soluble sulfate in rainwater was $nss-SO_4{^{2-}}$(non-sea salt sulfate). With fractional acidity and theoretical acidity of rainwater samples, $NH_4{^+}$ and $Ca^{2+}$ contributed greatly in neutralizing the rain acidity.

The Factors Controlling the Formation of Spring Population of Acartia hongi (Copepoda: Calanoida) in Incheon Coastal Water, Korea (인천 연안에서 요각류 Acartia hongi 춘계 개체군 형성의 영향 요인)

  • To investigate the factors controlling the spring population of Acartia hongi, egg production, hatching time of egg and predation pressure were measured. Egg production was maintained the superior position between winter and spring. Egg production was positively correlated with not only water temperature when water temperature was below $7^{\circ}C$ but also chlorophyll-a concentration when it was from $7^{\circ}C$ to $21^{\circ}C$. A regressive equation of development time$(D_e,\;day)$ of eggs derived from water temperature$(T,\;^{\circ}C)$ was obtained as $D_e=18.9(T-0.4)^{-1.0}$, showing longer development time at water temperature below $5^{\circ}C$. In Cross Correlation Analysis(CCA) to examine the time-lag relationship among abundances of developmental stages of A. hongi, egg production rate calculated by multiplying population egg production rate by hatching time showed more significant correlation with nauplii abundance than population egg production rate. Therefore, it suggests that hatching time is also recognized by a factor controlling formation of population and especially, in winter, high abundance of egg derived from high daily egg production rate and delayed hatch by low water temperature become the origin of initial spring population of nauplii. Egg predation by Noctiluca scintillans, suggesting a negative factor in formation of A. hongi spring population, was observed. During spring, A. hongi eggs were found in $2.9\sim21.1%$ of individuals of N. scintillans. It was deduced that $1.2\sim49.5%$ of the eggs produced by A. hongi was preyed on by N. scintillans. In conclusion, the factors controlling spring population of A. hongi were regarded as high egg production by winter generation, the delayed development time of egg by low temperature, and egg predation of N. scintillans.

Resistance of Vibrio parahaemolyticus Against the Environmental Factors (Vibrio parahaemolyticus의 환경인자(環境因子)에 대(對)한 저항성(抵抗性))

  • This study was carried out to investigate for resistance of V. parahaemolyticus that isolated from patients of food poisoning and fish and shellfish, captured in east coast of Kyungpook province of Korea from 1985 to 1986. VP ATCC 17802 and NAG V. ATCC 6538 were used as control. In fish, shellfish and seaweed, the more temperature increased, the shorter survival time was. In case of sea-water, the more temperature rose up, the longer survival time was, particularly in $37^{\circ}C$ and $25^{\circ}C$, the strains had survived after 6 months. And in tapwater, it was sterilized in 150 mins. and survived for 11.5 days on maximum in ground water. In kimchi, at room temperature, germicidal time was shorter more than 6 times compared with that which had been kept in refrigerator. It survived for 57.1 days in milk, 49.2 mins. in yougurt. Strains had been surviving in frozen condition at $-70^{\circ}C$ even after 6 months, present study time. In resistance test in water bath at several degrees of temperature, all the strains were sterilized in 20 mins. with $60^{\circ}C$. In resistance test to driness, number of surviving strains dropped rapidly in 10-11%) water contents. In UV $2538{\AA}$, strains were sterilized in 20 mins. In resistance test to alcohol, strains had survived for 0.1-4 mins. in fermentative wine of below than 25% and distilled wine of over than 25% in alcohol concentration. The bactericidal concentration of disinfectant was 1% in phenol and 3% in cresol. In 0.1M acetic acid and 0.1M lactic acid, number of surviving colonies decreased rapidly but not in citric acid. The more NaCl concentration rose up, the lower decreasing rate of number of surviving colonies was. The strains had showed sensitive response to vancomycin, chloramphenicol, gentamicin, and resisted to carbenicillin, ampicillin and kanamycin. When one day culture strain was cultured till 25th day, resistant strains to tetracycline and cephalothin were changed to sensitive.

Chemical Properties and Nutrient Loadings of Rainwater during Farming Season (영농기 강우의 화학적 특성 및 부하량 평가)

  • Acid rain and its problem to environment such as acid precipitation and environmental problems related to the air pollution in East Asia has been emerging. To evaluate the acidity and chemical characteristics of rainwater precipitated in western Korea, Suwon and Taean, its pH and ion concentrations were investigated during farming season (April to November) in 2009. Also, ion composition and cation-affected neutralization were determined to evaluate the contribution of cations on the acidity of rainwater. Ion and electrical conductivity between the measured and the estimated showed high correlation. The $Na^+$ in rainwater was the main cation followed by ${NH_4}^+>Ca^{2+}>H^+>Mg^{2+}>K^+$. Sum of $Na^+$ and ${NH_4}^+$ contents was over 65% of total cations contents. In the case of anions, the concentration was in order of ${SO_4}^{2-}>{NO_3}^->Cl^-$. The ${SO_4}^{2-}$ among anions in rainwater composed about 61%, which showed on average 130.2 ${\mu}eq\;L^{-1}$ and 121.3 ${\mu}eq\;L^{-1}$ during monitoring at Suwon and Taean, respectively. Also, 89.6 and 88.6% of soluble sulfate in rainwater at Suwon and Taean area was NSS-${SO_4}^{2-}$ (Non-Sea Salt sulfate). Especially, ${NH_4}^+$ and $Ca^{2+}$ contributed greatly in neutralizing the acid rain in dry season. Total nitrogen content flowed into soil from rain was around 1~2 kg $ha^{-1}$ in each month, but in July at Suwon, it reached 6 kg $ha^{-1}$ due to heavy rain (over 7.3 mm).

Quality Stability of Powdered Soup Using Powder from Oyster Wash Water (굴 세척액 유래 분말수프의 품질안정성)

  • This study was carried out to examine the quality stability of powdered soup using powder from oyster wash water (PSW). To compare the quality stability, powdered soup from oyster hot-water extracts (PSE) was also prepared by mixing hot-water extract powder (15 g), table salt (5 g), cream powder (19 g), milk replacer (12 g), wheat flour (20 g), corn flour (15 g), starch (5 g), glucose (7.5 g), and onion powder (1.5 g). In preparing PSW, powder from oyster wash water, instead of powder from oyster hot-water extracts, was added and other additives were the same proportion as PSE. The PSW and PSE were packed with laminated film bag (OPP,$20\;{\mu}m$; PE, $20{\mu}m$; paper, $45\;g/m^3$; PE, $20\;{\mu}m$; Al, $7\;{\mu}m$; PE, $20\;{\mu}m$), and then stored at ambient temperature for 12 months. The moisture content, water activity, peroxide value, and fatty acid composition showed little changes during storage of the PSW, The pH, volatile basic nitrogen content, and brown pigment formation increased slightly, while white index decreased slightly during storage of PSW. No significant difference was observed in the changes of food components between PSW and PSE during storage. According to a sensory evaluation, the change in quality of PSW was negligible during 12 months of storage. From the results of the chemical experiment and sensory evaluation, PSW packed with laminated film bag (OPP, $20\;{\mu}m$; PE, $20\;{\mu}m$; paper, $45\;g/{\mu}m$; PE, $20\;{\mu}m$) was revealed to be preserved in good quality during 12 months of storage.

Effect of Water Temperature on Ammonia Excretion of Juvenile Pacific Cod Gadus macrocephalus (대구 Gadus macrocephalus 치어의 암모니아 배설에 미치는 수온의 영향)

  • A study was carried out to examine the effect of water temperature on daily pattern and rate of total ammonia nitrogen (TAN) excretion in juvenile Pacific cod Gadus macrocephalus (mean body weight: $36.5{\pm}0.8\;g$) under fasting and feeding conditions. Fish were acclimated over 10 days under three different water temperatures (9, 11 and $13^{\circ}C$), and transferred to TAN measuring system under each water-temperature condition. After 72 hours of starving, fasting TAN excretion was measured at each temperature. To investigate post-prandial TAN excretion, fish were hand-fed with a commercial diet containing 40.6% crude protein for 7 days, two times daily at 08:00 and 16:00 h. Water was sampled from both the inlet and outlet of the fish chamber every 2 h over a 24-h period. Both fasting and post-prandial TAN excretion increased with increased water temperatures (p<0.05). Mean fasting TAN excretion rates at 9, 11 and $13^{\circ}C$ were 9.3, 11.0 and $11.9\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$, respectively. The value of $9^{\circ}C$ was lower than those of 11 and $13^{\circ}C$ (p<0.05), but there was no significant difference between $11^{\circ}C$ and $13^{\circ}C$. Mean post-prandial TAN excretion rates at 9, 11 and $13^{\circ}C$ were 23.0, 31.6 and $45.4\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$, respectively. A peak value of post-prandial TAN excretion rate occurred after 2 h from each feeding, and the second value is always higher than the first value. Maximum post-prandial TAN excretion rate occurred after 10 h from the first feeding at $9^{\circ}C$ (mean $38.0\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$), $11^{\circ}C$ ($52.9\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$) and $13^{\circ}C$ ($77.5\;mg\;TAN\;kg\;fish^{-1}\;h^{-1}$), respectively. The TAN loss for ingested nitrogen at $9^{\circ}C$ (43.9%) was lower than those of $11^{\circ}C$ (46.4%) and $13^{\circ}C$ (48.4%). The overall results indicate that water temperature exhibits a significant effect on the nitrogen excretion of juvenile Pacific cod.

Effect of Water Temperature and Body Weight on Oxygen Consumption Rate of Starry Flounder Platichthys stellatus (강도다리 Platichthys stellatus의 산소 소비율에 미치는 수온과 체중의 영향)

  • The effect of water temperature (T) and body weight (W) on oxygen consumption of fasted starry flounder Platichthys stellatus was investigated in order to assess the metabolic response of this species at given conditions. The oxygen consumption rate (OCR) was measured under six different water temperatures (4, 7, 10, 13, 16 and $19^{\circ}C$) and at two different body weights (mean weight of fry group : 1.5 g; fingerling group : 37.4 g) at an interval of 5 minutes for 24 hours using a continuous flow-through respirometer. In each treatment three replicates were set up and a total 540 fish in fry groups and 90 fish in fingerling groups were used. The OCRs increased with increase of water temperature in both groups (p<0.001). Mean OCRs at 4, 7, 10, 13, 16 and $19^{\circ}C$ were 1386.0, 1601.7, 1741.0, 1799.2, 2239.1 and $2520.3mg\;O_2\;kg\;fish^{-1}\;h^{-1}$ in fry groups, and 83.8, 111.4, 126.3, 147.1, 187.7 and $221.3mg\;O_2\;kg\;fish^{-1}\;h^{-1}$ in fingerling groups, respectively. The OCRs decreased with increasing body weights at six different water temperatures (p<0.001). The relationship between water temperature and body weight is described by the following equation : OCR=1520.91+40.85T-49.22W ($r^2=0.95$, p<0.001). The energy loss by metabolic response increased with an increase in water temperature and a decrease in body weight (p<0.001). Mean energy loss rates by oxygen consumption at 4, 7, 10, 13, 16 and $19^{\circ}C$ were 907.9, 1046.5, 1141.6, 1177.0, 1467.3 and $1650.1kJ\;kg\;fish^{-1}\;d^{-1}$ in fry groups and 54.8, 73.0, 82.9, 96.2, 122.9 and $144.6kJ\;kg\;fish^{-1}\;d^{-1}$ in fingerling groups, respectively. The $Q_{10}$ values of fingerling groups were higher than those of fry groups at given temperature ranges. The $Q_{10}$ values at $4{\sim}7^{\circ}C$, $7{\sim}10^{\circ}C$, $10{\sim}13^{\circ}C$, $13{\sim}16^{\circ}C$ and $16{\sim}19^{\circ}C$ were 1.62, 1.32, 1.12, 2.07 and 1.48 in fry groups, and 2.59, 1.52, 1.67, 2.25 and 1.73 in fingerling groups, respectively.

  • This study focuses on Korean-American writer Hong-Eun$(1880{\sim}1951)'s$ American travel gasaes and sis who played an active role under the rule of Japanese imperialism. This study also investigates Hong-Eun's experience and expression on American travel and culture and discusses his changes in stream of consciousness. According to American travel sigaes which were published in the New Koren Times in 1936. 1937, and 1949, his consciousness can be summarized as follows. First travel siga depicts his inner conflict as a refugee who lost one's home country. That is to say. by observing Indians' losing identity and their miserable labor conditions, he developed his own critical eyes on American society. Eventually he missed his country desperately and sought for the ways of his returning there. Second travel sijo reveals his own agony about not be able to return his home country where he could Possibly visit. In other words, after suffering from his agony, it is evident that he started to take positive attitude towards American society and establish his own identity. Based upon Hong-Eun's changes in consciousness as a writer, the researcher hypothesizes that there exists Diaspora-Consciousness in his work. His consciousness is strongly related with his attitude towards his home country whether it Is positive or vice versa. When his home country declared her independence. his attitude towards immigrant society was positively changed, which was quite contradictory from his previous one. In this transition period, not only he accepted American ideology and life, but he re-conceptualized them as a Korean mode. In sum, Hong-Eun's mental traces lie on the core of hybrid and diaspora which Post-Colonial literature values highly of.

Effects of Water Temperature on Oxygen Consumption in Starry Flounder Platichthys stellatus Reared in Seawater and Freshwater (해수 및 담수사육 강도다리 Platichthys stellatus의 산소소비에 미치는 수온의 영향)

  • The effects of water temperature on oxygen consumption (OC) of starry flounder Platichthys stellatus reared in seawater (SW) and freshwater (FW) was performed in closed water-recirculating system containing respiratory chamber. Fish acclimated in separate indoor tanks with SW (nine of fish used, $263.0{\pm}40.4$ g) or FW (nine of fish used, $265.8{\pm}34.8$ g) were sampled. The OC of starry flounder at $15^{\circ}C,\;20^{\circ}C$ and $25^{\circ}C$ were $74.4{\pm}17.0,\;85.9{\pm}15.8,\;98.3{\pm}11.4\;mg\;O_2\;kg^{-1}hr^{-1}$ in SW and $46.7{\pm}12.0,\;63.3{\pm}7.5,\;82.6{\pm}5.3\;mg\;O_2\;kg^{-1}hr^{-1}$ in FW, respectively, showing a linear increase in OC with water temperature. The OC of fish reared in both SW and FW clear diel rhythm, with lower values at daytime and higher values in the night, in accordance with light (09:00~21:00 hr) and dark (21:00~09:00 hr) phases of the diel cycle (12L : 12D) in water temperature at $15^{\circ}C$ and $20^{\circ}C$. However OC of fish reared in both SW and FW showed unclear diel rhythm with light and dark phases of the diel cycle in water temperature at $25^{\circ}C$. Starry flounder reared in FW had higher ventilation rates than those in SW, but SW had higher OC per breath than those in FW.
