  • A total of 119 raw milk samples from ten dairy farms were examined for total bacterial count, and 739 quarter milk samples of 118 dairy cows of 14 herds were examined for mastitis. The results obtained were as follows: 1. The mean of total bacterial counts of the 119 raw milk samples was 132,000 per ml. The total bacterial counts of 81 samples (68.1%) were under the standard of 100,000 per ml and those of the 38 samples (31. 9%) were over the standard. The number of bacteria showed a tendency to increase in summer. 2. One hundred and ninety five quarters (26.4%) of 98 cows (52.7%) were proved to be infected with mastitis. Clinical mastitis was found at 7 qtarters (3.5%) of 5 cows (5.0%). 3. Staphylococcus (44.9%) and Streptococcus (26.7%) were two main causative organisms of mastitis. Coliform bacteria (4.6%), Pseuedomonas spp. (4.6%), yeasts (1. 3%) and corynebacterium sp. (0.7%) were also isolated from the infected quarters. 4. The isolates were more sensitive to chloramphenicol ((96.1%), leukomycin (78.8%), streptomycin (75.5%) and tetracycline (72.4%). On the other hand, they were less sensitive to colistin (11.0%), oreandomycin (18.1%), sulfisoxazole (24.6%), penicilline (27.6%), kanamycin (43.3%) and erythromycin (49.7%). Especially the strains of Pseudomonas spp. isolated from the infected quarters were resistant to almost all the drugs examined.

한우(韓牛) 및 유우(乳牛)의 생식기내(生殖器內) 세균분리(細菌分離) 동정(同定) 및 약제감수성(藥劑感受性) (Isolation, Identification and Drug Susceptibility of Bacteria from Cow Genital Organs)

  • This study was conducted to determine the microorganisms inhabitating in cow genitalia and their antimicrobial drug susceptibility. During the period between July, 1985 and February, 1986, a total of 111 cow genitalia, 58 from Korean native and 53 from dairy cow, were sampled at three abattoirs. Gross pathological examination and bacterial isolation and identification were performed from the genital samples. In addition antimicrobial drug susceptibility test for the microorganisms isolated, some synergistic activity among drugs were examined on the major organism isolated from the cases of endometritis and pyometra. The results are summerized as follows: 1. Among the bacteria isolated from the genitalia, Staphylococcus spp., C. pyogenes, E. coli, Proteus spp., Streptococcus spp., Bacillus spp. were most frequently isolated whereas the genera of Pasteurella, Pseudomonas, Klebsiella and Yersina were detected far less frequently. 2. In Korean native cow the genera of Straphylococcus and Steptococcus were more isolated than dairy cow while in dairy cow the genera of Corynebacterium, Proteus, Escherichia were more of ten isolated than Korean native cow. 3. From cow genital organs showing lesions of endometritis and prometra, C. pyogenes was most frequently isolated, the isolation rate being 60 percent, and follow by Staphylococcus spp., Proteus spp., E. coli and Pasteurella spp. in the order. 4. Antimicrobial drug susceptibility test conducted on the major organisms isolated showed that all the isolates were susceptible to gentamicin, cephalosporin and sulfisoxazole, but resistant to tetracycline and penicillin. 5. Twenty-nine isolates of C. pyogenes were submitted to the synergistic activity test of cephalosporin, kanamycin and streptomycin with penicillin. Synergists were demonstrated in 90 percent, 31 percent and 27 percent of isolates examined by the combined use of penicillin and cephalosporin, penicillin and kanamycin, penicillin and streptomycin, respectively. About 10 percent of the isolates were found to be indifferent by the synergism test.

근관감염균에 대한 polyphosphate의 항균효과 (ANTIBACTERIAL EFFECT OF POLYPHOSPHATE ON ENDODONTOPATHIC BACTERIA)

  • This study was performed to observe the antibacterial effect of polyphosphate (polyP) with various chain lengths (P3~P75) on virulent. invasive strains of P. gingivalis A7A1-28 and W50, and multidrug resistant E. faecalis ATCC29212. P. gingivalis strains were grown in brain-heart infusion broth (BHI) containing hemin and vitamin K with or without polyP. PolyP was added at the very beginning of the culture or during the exponential growth phase of the culture. Inhibition of the growth of P. gingivalis was determined by measuring the absorbancy at 540nm of the grown cells. Viable cell counts of the culture and release of intracellular nucleotide from P. gingivalis were measured. E. faecalis was grown in plain BHI with antibiotics alone or in combination with polyP(calgon: 0.1~1.0%) and the bacterial absorbancy was measured. The overall results suggest that polyP has a strong antibacterial effect on the growth of the virulent strains of P. gingivalis and the antibacterial activity of polyP seems largely bactericidal. accompanying bacteriolysis in which chelation phenomenon is not involved. Although polyP does not exert antibacterial activity against E. faecalis, it appears to increase antibacterial effect of erythromycin and tetracycline on the bacterium. Therefore, polyP alone or in combination with antibiotics may be developed as a candidate for the agent controlling oral infections including endodontic infection.

건강에 영향을 주는 주변환경의 미생물 오염 실태 및 항생제 내성 (Bacterial Contamination and Antimicrobial Resistance of the Surrounding Environment Influencing Health)

  • 지역사회 획득 항생제 내성 세균은 전 세계에서 새로운 문제로 대두되고 있다. 특히 한국에서 항생제 내성 세균의 발생빈도는 다른 선진국들보다 높다. 본 연구에서는 서울지역의 다양한 주변 환경(가정집, 대학교, 대중교통, 소지품 등)으로부터 methicillin-resistant staphylococci (MRS), Pseudomonas, 그리고 대장균군(Enterobacteriaceae)의 분리율을 조사하였으며, 검출된 대장균군(Enterobacteriaceae)을 대상으로 신 항생제를 포함한 한국의 임상에서 널리 쓰이는 13종의 항생제에 대한 내성정도를 파악하였다. 총 239건의 시료에서 18건(7.5%)의 MRS, 10건(4.2%)의 Pseudomonas, 그리고 30건(12.6%)의 대장균군(Enterobacteriaceae)이 분리되었다. 또한 가정집에서 2건, 대학교에서 1건, 그리고 기타(공원 의자와 빌딩 문손잡이)에서 2건, 총 5건(2.1%)의 시료에서 메티실린 내성 황색포도상구균(methicillin-resistant Staphylococcus aureus, MRSA)이 검출되었으며, 대장균(Escherichia coli)은 가정에서 1건, 대중교통에서 3건, 그리고 기타(음식점 정수기 본체 상단)에서 1건, 총 5건(2.1%)의 시료에서 검출되었다. 분리된 대장균군(Enterobacteriaceae) 32균주에 대한 항생제 내성률은, cephalosporin 계열과 fluoroquinolone 계열에서 각각 71.9%, carbapenem 계열과 ${\beta}$-lactam 계열에서 각각 68.8%까지 나타났으며, tetracycline 계열과 aminoglycoside 계열에서는 각각 50.0%와 25.0%까지 내성을 나타냈다. 또한 trimethoprim/ sulfamethoxazole에 대한 내성률은 43.8%로 나타났다. 심지어 신 항생제인 tigecycline에 대한 내성률도 59.4%로 나타났으며, 시험한 모든 항생제에 내성을 나타내는 균주도 3건이나 검출되었다. 그리고 분리된 총 5균주의 대장균(E. coli)은 대부분의 시험 항생제에 내성을 나타냈으나, 일부는 ciprofloxacin과 gentamicin에 감수성을 보였다. 본 연구는 향후 주변환경의 항생제 내성 수준의 변화를 비교하기 위한 기초 자료를 제공하며, 이들 내성균의 발생과 확산 방지에 대한 다각적인 노력과 대책 마련이 필요함을 보여준다.

유산양 유즙으로부터 분리된 세균의 분포 및 항균제 감수성 검사 (Isolation and antimicrobial susceptibility of microorganisms from milk samples of dairy goat)

  • The aim of this study was to isolate microorganisms from half milk samples of dairy goats by California mastitis test (CMT) during the lactation period and to further investigate the susceptibility of isolated organisms to antimicrobial drugs. From a total of 235 half milk samples with CMT scores of 2 or above from 366 dairy goats distributed throughout Jeonnam province, microorganisms were isolated from 198 (83.5%) samples either singly (99.0%) or in combination (1.0%). The most prevalent microorganism was the coagulase-negative Staphylococcus spp., (44.4%, n=88) followed by Staphylococcus aureus (24.2%, n=48), Escherichia coli (11.1%, n=22) and Streptococcus spp. (7.6%, n=15). Isolated bacteria also included Bacillus spp. (2.5%, n=5), Pseudomonas spp. (2.5%, n=5), Micrococcus spp. (1.5%, n=3), Corynebacterium spp. (1.5%, n=3), Enterococcus facium (1.0%, n=2), Morganella morganii (0.5%, n=1) and Streptococcus agalactiae (0.5%, n=1). During the summer season, a high prevalence of all microorganisms were observed in which Staphylococcus spp. (30.8%), Escherichia coli (8.6%), and Streptococcus spp. (5.6%) were among the most prevalent bacteria isolated. Staphylococcus spp. was also shown to be high in the winter (21.7%). In most samples, the presence of bacterial pathogens in goat milk led to the increase in the total somatic cell count (SCC). Most of the half milk samples of dairy goats with bacterial contamination showed SCC of ${\geq}1{\times}10^6cells/ml$ (90.4%). Minor pathogens (11.4%) were more detected from milk samples with SCC of < $1{\times}10^6cells/ml$ than major pathogens (4.1%), while the major pathogens tended to be higher from samples with SCC of ${\geq}3{\times}10^6cells/ml$. Susceptibility of these bacteria to 12 antimicrobial agents was tested by the Kirby-Bauer disc diffusion method. Results indicated that more than 90% of bacteria isolated from CMT 2+ dairy goat half milk samples were susceptible to trimethoprim/sulfamethoxazole, amoxicillin/clavulanic, enrofloxacin and cephalothin while they were resistant to tetracycline (44.7%).

모돈 유즙으로부터 분리한 세균의 분포 및 항균제 감수성 조사 (Isolation and antimicrobial susceptibility of microorganisms from milk samples of healthy postpartum sows)

  • The growth rate of piglets will be decreased and the mortality of piglets will be increased in the postpartum sow with mastitis. The aim of this study was to isolate microorganisms from milk samples found in postpartum sows with suckling piglets and to further investigate antimicrobial susceptibility against isolated bacteria. Milk samples were collected from 567 udders of 42 lactating sows and the isolated bacteria was then identified. Isolated bacteria were tested with 12 antimicrobial agents. Bacteria were isolated from 260 milk samples (64.5%). Staphylococcus spp. were the most common microorganisms (74.5%) isolated from sow milk, followed by Streptococcus hyicus (53.9%), Staphylococcus epidermis (11.8%), Staphylococcus aureus (9.8%), and Staphylococcus haemolyticus (53.9%). In the Staphylococcus spp., Streptococcus pneumoniae (5.1%) was isolated as was Streptococcus sanguinis (3.8%) and Streptococcus ovis (2.9%). Results indicated that bacteria isolated from healthy sow milk were mostly susceptible to cephalothin (88.0%), amikacin (85.3%) and cefoxitin (82.4%), but were resistant to streptomycin (10.0%), kanamycin (33.1%) and tetracycline (19.6%).

수입 관상어에서 분리한 motile aeromonads의 특성 (Characters of motile aeromonads isolated from imported ornamental fish)

  • 현재 국내에 유통되고 있는 담수관상어의 대부분은 수입에 의존하므로 외래 병원체 및 항생제 내성균의 유입 가능성이 크다. 본 연구에서는 담수에 상재하는 균으로 인수공통 감염이 가능하며 어류에 조건성병원균으로 작용하는 Aeromonas hydrophila를 다양한 수입 담수관상어로부터 분리하여 특성을 분석하고자 하였다. 수입 후 1일 이내의 관상어를 수족관으로부터 수집하여 0.5% NaCl을 포함하는 TSA 배지에 간, 신장, 비장을 도말하여 세균을 분리하였다. 세균의 형태, 생화학적 특성 및 starch- ampicillin agar (SA)에서의 clear zone 형성 유무에 근거하여 분리 균의 약 70%인 226 균주가 Aeromonas spp.로 확인되어 이들의 분리 빈도가 매우 높음을 알 수 있었다. 이들 중 9균주를 선택하여 16S rRNA gene의 sequence를 분석한 결과 모두 A. hydrophila로 동정되었으며 API 20E test에서 대부분의 분리 균주는 amygdalin, D-melibiose, D-sucrose, trisodium citrate, L-arabinose 이용에서 표준 균주와 차이를 보였다. 10종의 항균제에 대한 분리 균주의 감수성을 디스크 확산법으로 확인한 결과 amoxycillin, ampicillin에 대해서는 모든 균주가 저항성이었고 florfecicol에 대해서는 모두 감수성이었다. 그러나 7균주는 tetracycline 계통의 항균제와 erythromycin, nalidixic acid 등에 복합내성을 보였다. A. hydrophila로 동정된 9개 분리 균주를 금붕어에 인위 감염을 실시한 결과, 3 균주는 5일 이내에 60~80%의 폐사를 나타내었다. 이러한 결과는 다약제내성이면서 독력이 있는 Aeromonas가 수입관 상어를 따라 국내로 유입되어 전파될 수 있으므로 수입상에서 소비처로 판매되기 전의 방어적 조치가 필요하다는 것을 시사한다.

시판 냉동식품에서 분리한 장구균의 항생제 내성 양상 (Antibiotic Resistance Patterns of Enterococcus spp. Isolated From Commercial Frozen Foods)

  • 2009년 4월부터 12월까지 서울시 보건환경연구원 미생물 관리팀에 의뢰된 냉동식품 100건을 대상으로 일반세균수, 대장균군수, 대장균과 장구균의 오염도를 조사였으며, 분리된 장구균을 대상으로 하여 항생제 감수성 검사를 실시하였다. 일반세균수는 평균 검출량이 $4.3{\times}10^4CFU/g$ 이고 대장균군수는 평균 검출량이 $4.3{\times}10^3CFU/g$ 이고 대장균은 100건 모두에서 불검출되어 0.0%의 검출률을 보였다. 100건의 검체 중 총 22건에서 장구균이 분리되었으며 이 가운데 12주는 E. faecium, 7주는 E. faecalis, 2주는 E. gallinarum, 1주는 E. hirae이었다. 분리균주의 항생제 내성률을 검사 한 결과, 내성을 나타내는 항생제들은 erythroycin, rifampin, teracycline ciprofloxacin, chlorampenicol, penicillin이며, vancomycin, ampicillin, gentamicin, strepomycin, linezolid에 대해서는 모두 감수성이었다. 또 22건의 분리 균주 중에서 2종 이상의 항생제에 다제 내성을 나타낸 균주가 15주나 되며 가장 많은 형태로는 6균주의 장구균에서 나타난 E-RA에 대한 것이었다.

A Gene-Tagging System for Monitoring of Xanthomonas Species

  • A novel chromosomal gene tagging technique using a specific fragment of the fatty acid desaturase-like open reading frame (des-like ORF) from the tox-argK gene cluster of Pseudomonas syringae pv. phaseolicola was developed to identify Xanthomonas spp.released into the environment as biocontrol agents. X. campestris pv. convolvuli FB-635, a pathogen of Convolvulus arvensis L., (bindweed), was chosen as the organism in which to develop and test the system. A 0.52 kb DES fragment amplified from P. syringae pv. phaseolicola C-199 was inserted into pGX15, a cosmid clone containing a 10.3 kb Eco RI-HindIII fragment derived from the xanthomonadin biosynthetic gene cluster contained in plasmid pIG102, to create a pigG::DES insertion. The 10.8 kb EcoRI-BamHI fragment carrying the pigG:: DES insertion was cloned into pLAFR3 to generate pLXP22. pLXP22 was then conjugated into X. campestris pv. convolvuli FB-635 and the pigG::DES insertion integrated into the bacterial chromosome by marker exchange. Rifampicin resistant, tetracycline sensitive, starch hydrolyzing, white colonies were used to differentiate the marked strain from yellow pigmented wild-type ones. PCR primers specific for the unique DES fragment were used for direct detection of the marked strain. Result showed the marked strain could be detected at very low levels even in the presence of high levels of other closely related or competitive bacteria. This PCR-based DES-tagging system provides a rapid and specific tool for directly monitoring the dispersal and persistence of Xanthomonas spp.released into the environment.

Comparison of biological and chemical assays for measuring the concentration of residual antibiotics after treatment with gamma irradiation

  • Antibiotic pollution is one of the factors contributing to the spread of antibiotic-resistant bacteria in the environment. Advanced oxidation and irradiation processes have been introduced to eliminate antibiotics from water and wastewater. However, few studies have reported the toxic effects of residual antibiotics and their byproducts induced by a treatment system. In this study, we compared the efficacies of chemical (high-performance liquid chromatography (HPLC)) and biological (antimicrobial susceptibility test) assays for measuring the concentrations of residual antibiotics after gamma irradiation for degrading amoxicillin, cephradine, lincomycin, and tetracycline. The concentrations of residual antibiotics estimated using the two assay methods were almost identical, except cephradine. In the case of cephradine, inhibited bacterial growth was observed that was equivalent to twice the concentration measured by HPLC in the samples subjected to gamma irradiation. The observed inhibition of bacterial growth suggested the generation of potentially toxic intermediates following antibiotic degradation. These results indicate that biological and chemical assays should be used in concert for monitoring antibiotic contamination and the toxic derivatives of antibiotic degradation. The results demonstrate that these four antibiotics can be decomposed by 2.0 kGy gamma-irradiation without toxic effects of their byproducts.
