장내 유해세균에 대한 가자(Terminalia chebula Retz.) 추출물의 항균 활성 (Antimicrobial Activity of terminalia chebula Retz. Extract of against Intestinal Pathogens)

  • 장내유해세균에 대한 세균성 장내질환치료와 관련된 한약재의 항균활성 검색을 통해 우수한 효과가 인정된 가자를 선정하고 혐기적 broth system에서 각 용매별 분획물의 6종의 장내유해세균에 대한 항균활성을 측정하였으며 그 항균물질을 column chromatography 법에 의해 부분 정제하였다. 가자 메탄올 추출물은 100-2,000g/ml 농도에서 시험균 Eubacterium limosum ATCC 10825, Escherichia coli ATCC 25922, Bacteroides fragilis KCTC 5013, Clostridium perfringens ATCC 3627, Staphylococcus aureus KFCC 11764 및 Salmonella typhimurium ATCC 14028 모두에 대해 항균활성을 보였으며 특히 2,000g/ml 농도에서는 Escherichia coli ATCC 25922 및 Staphylococcus aureus KFCC 11764을 제외하고는 거의 생육하지 못하였다. 각 분획물(n-hexane, ethylether, ethylacetate 및 water)들간의 항균활성은 Clostridium perfringens ATCC 3627을 제외하고는 유의적인 차이가 없었다. 가자의 ethylacetate 분획은 Sephadex G-200 column chromatography에 의한 30개 분획 중 12에서 21번까지의 분획에서 강한 항균활성을 나타내었으며 silica gel column chromatography에 의한 40개 분획 중에서는 22에서 34까지의 분획에서 항균활성을 보였다.

Anti-osteoarthritis Effects of the Combination of Boswellia serrata, Curcuma longa, and Terminalia chebula Extracts in Interleukin-1β-stimulated Human Articular Chondrocytes

  • In this study, extracts of Boswellia serrata gum resin, Curcuma longa rhizome, and Terminalia chebula fruit were combined in different ratios, and their anti-osteoarthritis effects were compared to determine which combination had the best synergistic effect. B. serrata, C. longa, and T. chebula extracts in a 2:1:2 ratio exhibited higher antioxidative activity in scavenging DPPH radicals than did the individual extracts alone or the other extract combinations. Additionally, the 2:1:2 combination significantly improved the levels of enzymatic antioxidants and antioxidant-related proteins. Moreover, this same combination ratio decreased the protein levels of matrix metalloproteinase (MMP) 3 and MMP13 in interleukin-1β-stimulated human articular chondrocytes (HCHs) and increased those of aggrecan and collagen type II alpha 1 chain (COL2A1). Analysis of the underlying mechanisms revealed that the 2:1:2 combination significantly inhibited the phosphorylation of nuclear factor kappa B (NF-κB) p65, extracellular regulated protein kinase (ERK), and p38 mitogen-activated protein kinase (MAPK). Therefore, the 2:1:2 combination of these three plant extracts has the best potential for use as an effective dietary supplement for improving joint health compared with the individual extracts and their other combination ratios.

Silkworm-food plant-interaction: search for an alternate food plant for tasar silkworm (Antheraea mylitta Drury) rearing

  • An experimental rearing of tropical tasar silkworm, Antheraea mylitta Drury was conducted to study silkworm-food plant-interaction and thereby to search for an alternate silkworm food plant. The silkworm-food-plant-interaction was studied with six different food plant species viz. Terminalia tomentosa, Terminalia arjuna, Terminalia belerica, Terminalia chebula of Combretaceae family and Lagerstroemia speciosa, Lagerstroemia parviflora of Lythraceae family. The rearing performance of silkworm on Lagerstroemia speciosa in terms of cocoons per DFL and silk ratio was found comparable with Terminalia tomentosa and Termonalia arjuna, the primary tasar silkworm food plant species. These three plant species also possessed better results in terms of physiological (leaf moisture content and net photosynthesis rate) and biochemical (Chlorophyll, protein, carbohydrate and crude fibre contents) characteristics to support silkworm rearing than Terminalia belerica, Terminalia chebula and Lagerstroemia parviflora. The correlation study between silkworm rearing performance and food plant's constituents indicates commercial perspective of Lagerstroemia speciosa as an alternate food plant for tasar silkworm rearing.

가자(Terminalia chebula Retz)추출물의 항산화 효과 (Antioxidative Effectiveness of Terminalia chebula Rets Extracts)

  • 탈지 가자박으로 부터 free, soluble 및 insoluble bound phenolic acids를 추출하여 식용대두유 기질에서 항산화효과률 0.02%(w/w)의 BHA, BHT의 항산화 효과와 비교하고자 각 기질과 대조구를 $60{\pm}1^{\circ}C$에서 35일간 저장하면서 매 7일 간격으로 과산화물가, TBA가를 측갱하였다. BHA, BHT와 free, soluble 및 insoluble hound phenolic acids를 첨가한 시험구와 대조구의 과산화물가는 21일의 경우 60, 30, 14, 11, 10, 100이었다. 같은 조건하에서 추출물의 TBA가는 0.150, 0.108, 0.105, 0.073, 0.078, 0.185이었다. 이것으로 보아 phenolic acids는 식용대두유 기질에서 우수한 항산화력을 나타내었다. 분리확인된 phenolic acids는 p-coumaric acid, ferulic acid, phloroglucinol, pyrogallol, vanillic acid, caffeic acid으로 확인되었다.

가자(Terminalia chebula) 추출물이 마우스의 생리활성에 미치는 영향 (Effect of Terminalia chebula on Physiological Activity in Mice)

  • 가자 추출물을 취하여 사람, 돼지, 쥐 및 개 등의 3% 적혈구용액으로 응집력시험을 행한 결과 7가지 적혈구 모두에 렉틴 활성이 나타났으며 $LD_{50}$는 390 mg/kg으로 측정되었다. 생리 활성에 대한 영향을 알아보고자 생체 내외인성 요인에 의한 친전자성 물질로 생체내에서 독작용, 노화, 발암 및 면역 억제작용을 유발하는 원인 물질인 free radical생성에 관여하는 효소인 XO 및 AO의 활성을 측정 한 결과, XO 에서는 일주일간 매일 300 mg/kg의 용량으로 가자 추출물을 투여한 군이 정상군보다 5배 증가되는 결과를 나타내었고 AO에서는 정상군보다 시료 투여군이 2배 증가되는 결과를 나타내었다 glutathione은 단백질이나 DNA합성, amino acid의 이동 반응 및 thiol기의 저장 등과 같은 생물학적으로 중요한 여러 가지 반응에 직접 관여하는 물질이다[16〕. 이에 간장 조직의 glutathione농도를 측정한 결과 간장 독성의 유발로 인한 효소 활성은 정상군에 비하여 1주간 매일 300 mg/kg의 용량으로 시료 투여한 군의 효소 활성이 79% 감소됨을 볼 수 있었다. 체내의 여러 가지 해독반응과정에 관여하는 GST효소활성을 측정 한 결과 정상군에 비해 1주간 매 일 300 mg/kg의 용량으로 시료 투여한 군이 66%정도 감소된다는 결과를 볼 수 있었다. glutathione 합성에 관여하는 $\gamma$-GCS의 활성과 산화형 glutathione을 환원형 glutathione으로 환원시키는 GR의 활성을 관찰한 결과 가자 추출물 투여군이 정상군보다 GR의 활성은 80% 감소되었고, 합성에 관여하는 $\gamma$-GCS의 활성은 정상군과 비교할 때 약간의 감소만을 나타내 glutathione함량 변동에 크게 영향을 미치지 않는 것으로 생각된다.

Anti-Arthritic and Analgesic Effect of NDI10218, a Standardized Extract of Terminalia chebula, on Arthritis and Pain Model

  • The fruit of Terminalia chebula Retzius has been used as a panacea in India and Southeast Asia but its biological activities have not been fully elucidated. Here we report anti-arthritic and analgesic effect of NDI10218, a standardized ethanol extract of Terminalia chebula, on collagen-induced arthritis and acetic acid-induced writhing model, respectively. Arthritis was induced in DBA/1J mice by immunizing bovine type II collagen and mice were treated with NDI10218 daily for 5 weeks after the onset of the disease. NDI10218 reduced the arthritis index and blocked the synovial hyperplasia in a dose-dependent manner. The serum levels of pro-inflammatory cytokines TNF-${\alpha}$, IL-6, and IL-$1{\beta}$ were significantly reduced in mice treated with NDI10218. Production of the inflammatory IL-17, but not immunosuppressive IL-10, was also inhibited in splenocytes isolated from NDI10218-treated arthritis mice. Administration of NDI10218 markedly decreased the number of T cell subpopulations in the regional lymph nodes of the arthritis mice. Finally, NDI10218 reduced the number of abdominal contractions in acetic acid-induced writhing model, suggesting an analgesic effect of this extract. Taken together, these results suggest that NDI10218 can be a new therapeutic candidate for the treatment of rheumatoid arthritis.

가자(訶子) 추출물과 그 유효성분 갈산이 근분화에 미치는 영향 (Investigation of the effect of Terminalia chebula fruit extract and its active ingredient, gallic aicd on muscle differentiation)

  • Objectives : Decrease in muscle mass and loss of muscle function due to aging are associated with various diseases. As interest in healthy aging increases, efforts to prevent and treat muscle hypoxia as an illness are increasing. Considering the physical limitations, a pharmacologic approach to the treatment of myopenia is needed. Methods : Terminalia chebula Rets has a wide range of pharmacological effects and is used as a medicinal product in traditional medicine. However, the drug effect on the treatment of muscle disorders has not been revealed. The purpose of this study was to evaluate the value of water extract of Terminalia chebula (WETC) as a therapeutic agent to relieve symptoms of muscle hypoxia. Results : WETC showed strong radical scavenging ability. In addition, WETC increased cell activity of myoblast, and we observed that WETC induces myoblast differentiation by immunoblot analysis using differentiation protein markers as well as cell morphology of myoblast. Based on these results, we examined the effect of chebulic acid, chebulagic acid, gallic acid, geraniin, and punicalagin on cell activity and differentiation of myoblasts. Gallic acid significantly increased cell activity of myoblast, and it was found to be an effective substance which not only induces myoblast differentiation but also promotes proliferation. Conclusions : We suggest that the WETC with antioxidant effect and its indicator gallic acid on cell activity, proliferation and differentiation of myoblast can be studied and developed as a food and medicine for prevention and treatment of various muscle diseases.

Antioxidant Activities and Melanogenesis Inhibitory Effects of Terminalia chebula in B16/F10 Melanoma Cells

  • To examine the potential of Terminalia chebula as a whitening agent, we measured antioxidant activity using DPPH$\cdot$, ABTS${\cdot}^+$ assays and ferric-reducing antioxidant power (FRAP) assays, and depigmenting activity using B16F10 melanoma cells. The intracellular reactive oxygen species (ROS) level was monitored by $H_2DCFDA$ fluorescence labeling, and melanin contents in B16F10 melanoma cells by 960 $J/m^2$ dose of UVA-induced oxidative stress. The radical-scavenging activities of T. chebula extract (TCE) were measured in terms of $EC_{50}$ values using DPPH$\cdot$, ABTS${\cdot}^+$ assays and FRAP value were 280.0 ${\mu}g/mL$, 42.2 ${\mu}g/mL$ and 113.1 ${\mu}mol$ $FeSO_4{\cdot}7H_2O/g$, respectively. We found that ROS and melanin concentrations were reduced by TCE treatments of 25 ${\mu}g/mL$ under UVA-induced oxidative stress. Tyrosinase activity and melanin contents in $\alpha$-melanocyte stimulating hormone (MSH)-induced melanoma cells both decreased dose-dependently in the treatment groups. TCE similarly reduced melanogenesis in B16F10 melanoma cells stimulated by $\alpha$-MSH as compared to arbutin as a positive control. T. chebula may prove to be a useful therapeutic agent for hyperpigmentation and an effective component in skin whitening and.or lightening cosmetics.

가자(訶子)의 급성 췌장염 보호 효과 (Protective effects of Terminalia chebula Retzins (TC) water extract on acute pancreatitis)

  • Objectives: Terminalia chebula Retzins (TC) has been used as a traditional remedy to treat gastrointestinal infectious and inflammatory diseases. However, the protective effect of TC on acute pancreatitis (AP) has not been studied. In this study, we tried to investigate the protective effect of TC water extract on cerulein-induced AP. Methods: To measure the protective effect of TC on AP, mice were injected with cerulein (50 ㎍/kg) hourly for 6 times. TC water extract (200 or 400 mg/kg) or saline was administered orally 1 h before the first injection of cerulein. The mice were sacrificed at 6 h after the last injection of cerulein. Pancreas tissues were taken for further analysis. Results: The administration of TC water extract showed an inhibitory effect on the increase of pancreas weight/body weight ratio and mitigated pancreatic damage in mice. Also, mRNA level of pro-inflammatory cytokines was inhibited by administration of TC water extract. Conclusion: Taken together, we found that administration of TC water extract ameliorates the severity of cerulein-induced AP, which suggests the potential to be an effective treatment on AP.

Cytoprotective Effect on Oxidative Stress and Inhibitory Effect on Cellular Aging of Terminalia chebula and Uncaria sinensis

  • The ethanol extract from the fruit of Terminalia chebula (Combretaceae) and the hook of Uncaria sinensis (Rubiaceae) exhibited significant inhibitory activity on oxidative stress and the age-dependent shortening of the telomeric DNA length. In the peroxidation model using t-BuOOH, human epidermal keratinocytes-neonatal foreskin (HEK-N/F) cells were treated with the T. chebula and U. sinensis extracts. The results showed a notable enhancing effect on the cell viability of 60.5 ${\pm}$ 3.8 and 65.0 ${\pm}$ 3.0%, respectively, by 50 $\mu\textrm{g}$/ml of the extracts. (omitted)
