A Study on the Activity of Anti-Aging by Second Fermented Snail Extract with Hericium erinaceum Mycelium (노루궁뎅이버섯 균사체를 이용한 2 차 발효달팽이 추출물의 항노화 활성에 관한 연구)

  • This study is related to develop a snail extract through a snail secondary fermentation process, getting anti-aging activity with healthy and beauty skin care scientific applications. In order to obtain a primary fermentation was incubated with Hericium erinaceus mycelium. Through the secondary fermentation process using Leuconostoc mesenteroides, was deeply described a total process of obtaining second fermented extract using snail body. Mycelium is applied in this study was extracted using Hericium erinaceus mycelium and Leuconostoc mesenteroides. The final yield of the extract was 62 wt%. Experimental results of secondary fermentation snail extract were contained with 32 wt% water, 31.5 wt% total amino acid protein, 15.7 wt% polysaccharide, 12.3 wt% fatty acid and others 8.5 wt%. In addition, in order to study about skin beauty care and anti-aging activity, we evaluated antioxidant activity with DPPH, elastin enzyme (elastase) inhibitory activity, tyrosinase inhibition rate, collagen synthetic function, fibroblast synthetic activity. First; anti-oxidative activity of secondary fermentation snail extract (IC50%) was spent with 7.27 mg/mL, control samples were spent with green tea extract was 11.8 mg/mL, common snails extract was 15.7 mg/mL, DL-a-tocopherol was 9.25 mg/mL respectively. Second; elastin enzyme inhibitory activity of secondary fermentation snail extract (IC50%) was spent with 32.5 mg/mL, control samples were also spent with green tea extract was 45.9 mg/mL, general snail extract was 67.7 mg/mL. Third; tyrosinase inhibitory activity of secondary fermentation snail extract (IC50%) was spent with 140.3 mg/mL, control samples were also spent with green tea extract was 250.7 mg/mL, general snails extract was 389.5 mg/mL, niacineamide was 125.9 mg/mL. Forth; fibroblast synthetic activity of secondary fermentation snail extract was increased with 125.6%, control samples were also spent with green tea extract was 98.9%, general snails extract was 109.5%, niacineamide was 125.9 mg/mL, DL-a-tocopherol was 96.2%. Fifth; collagen synthetic activity of secondary fermentation snail extract was increased with 118%, control samples were also spent with green tea extract was 87.3%, general snails extract was 93.2%, adenosine was 127.9%. In conclusion, on the basis of this study, in the future it is expected to be applied to the skin beauty care application and development of Korean style cosmetic products.

Polyphenol Oxidase of Tea Leaf in Korea (국산 다엽의 Polyphenol Oxidase에 관한 연구)

  • Polyphenol oxidase was purified from an extract of tea leaf by ammonium sulfate fractionation followed by Sephadex G-150 column chromatography, which resulted in a 67-fold increase in specific activity. The enzyme had optimum pH 6.5 and was relatively heat stable. The substrate specificity of the tea leaf PPO showed high affinity toward pyrogallol and catechol. Potassium cyanide, sodium diethyldithiocarbamate, L-cysteine, 2-mercaptoethanol and ascorbic acid were potent inhibitors.

Quality Improvement of Chokochujang by the Addition of Green Tea Extract (녹차추출물을 첨가한 초고추장의 품질향상)

  • The effect on quality improvement of Chokochujang by the addition of green tea extract (GT) was evaluated on the basis of total cell numbers, pH, acidity, sugar content and sensory evaluation during its storage at $25^{\circ}C$ and $37^{\circ}C$ for 3 weeks. A dorminant strain was isolated from the Chokochujang and identified as Bacillus amyloliquefaciens ER282 based on the 16S rDNA sequence. The ethanol extract from GT was investigated for its antibacterial activity aid showed good activities enough to use for Chokochujang as a natural preservative. Growth of microorganisms was significantly inhibited by adding GT extract to Chokochujang but pH, acidity, sugar content and color was not much changed. Sensory evaluation and overall preference, however, was continuously declined as storage period increased, whereas the addtion of 3% GT extract could delay the quality loss of Chokochujang during storage at $25^{\circ}C$ and $37^{\circ}C$ for 3 weeks.

Beneficial Effect of Scutellaria Balicalensis Georgi Extract ont-Buthylhydroperoxide-Induced Inhibition of Organic Cation in Rabbit Renal Cortical Slices (황금약침액(黃芩藥鍼液)이 토끼의 신장절편에서 t-BHP로 유발된 유기양이온의 이동장애에 미치는 영향(影響))

  • Objective : This study was undertaken to determine whether Scutellaria balicalensis Georgi (SbG) extract exerts the protective effect against oxidant-induced alterations in organic cation transport in the renal proximal tubule. Methods : Organic cation transport was estimated by examining alterations in tetraethylammon - ium(TEA) uptake in rabbit renal cortical slices. The slices were treated with 0.2 mM tBHP for 60 min at $37^{\circ}C$. tBHP caused an inhibition in TEA uptake by renal cortical slices. Such an effect was accompanied by depressed Na+-K+-ATPase activity and ATP depletion. tBHP also induced a significant increase in LDH release. Results : SbG prevented tBHP-induced inhibition of TEA uptake in a dose-dependent manner at the concentration ranges of 0.05-0.1%. tBHP-induced inhibition of Na+-K+-ATPase activity and ATP depletion were significantly prevented by 0.05% SbG. tBHP-induced LDH release also was blocked by SbG. tBHP caused a significant increase in lipid peroxidation and its effect was prevented by SbG. Conclusion : These results suggest that SbG prevents oxidant-induced alterations in organic cation transport in rabbit renal cortical slices. Such protective effects of SbG may be attributed to inhibition of peroxidation of membrane lipid.

Green Tea Extract, not Epigallocatechin gallate Inhibits Catecholamine Release From the Rat Adrenal Medulla

  • The present study was designed to investigate the effects of green tea extract (CUMC6335) and epigallocatechin gallate (EGCG) on secretion of catecholamines (CA) in the isolated perfused rat adrenal gland. ill the presence of CUMC6335 (100 $\mu\textrm{g}$/mL) into an adrenal vein for 60 min, CA secretory responses evoked by ACh(5.32 mM), high $K^+$ (56 mM) and Bay-K-8644 (10$\mu$M for 4 min) from the isolated perfused rat adrenal glands were greatly inhibited in a time-dependent fashion. However, EGCG (8 $\mu\textrm{g}$/mL) did not affect CA release evoked by ACh, high $K^+$ and Bay-K-8644. CUMC6335 itself did fail to affect basal catecholamine output. Taken together, these results demonstrate that CUMC6335 inhibits greatly CA secretion evoked by stimulation of cholinergic nicotinic receptors as well as by the direct membrane deplarization from the isolated perfused rat adrenal gland. It is felt that this inhibitory effect of CUMC6335 may be due to blocking action of the L-type dihydropyridine calcium channels in the rat adrenal medullary chromaffin cells, which is relevant to the cholinergic nicotinic blockade. It seems that there is a big difference in mode of action between CUMC6335 and EGCG.

Combination of Green Tea Extract and L-Theanine Alleviates Electric Foot Shock Induced Stress by Modulating Neurotransmitters in Mice (녹차추출물과 테아닌 복합물의 신경전달물질 조절을 통한 항스트레스 효과)

  • Various neurotransmitters are involved in regulating stress systems. In this study, we investigated the combination relieving effect of green tea extract(GTE) and L-theanine on the stress induced by electric foot shock. Four week oral administration of GTE (24 mg/kg), L-theanine (4 mg/kg) or their combination reduced the levels of dopamine, noradrenaline and corticosterone in blood, brain cortex, hippocampus, and striatum, wherease increased serotonin level. The combination of GTE and L-theanine showed much greater effects than single treatment of each component, and the effects are comparable to diazepam (2 mg/kg). Therefore, this study suggests that the combination of GTE and L-theanine may act effective and be useful for stress relieving treatment.

DMfree®(Green Tea Extract) Inhibits IL-6 of Mycobacterium leprae Infected Mesenchymal Stem Cells (디엠프리(녹차추출물)에 의한 나균 감염 중간엽줄기세포의 IL-6 생산 억제)

  • Previous reports revealed that DMfree (green tea extract) inhibited expression of the IL-6 gene in Mycobacterium lepraeinfected MSCs (mesenchymal stem cells). This study aimed to measure IL-6, $IL-1{\beta}$, $TNF-{\alpha}$ and PGE2 production in M. leprae-infected MSCs using ELISA. To confirm the effect of DMfree on IL-6 and signal transduction, a western blotting test was performed. DMfree inhibited the expression of IL-6 in the MSCs and the heterodimer of STAT3, which also affects the expression of multiple genes. Though DMfree pre-treatment of control MSCs produced a baseline level of IL-6, it significantly inhibited the production of IL-6 in M. leprae-infected MSCs. There was no significant difference in IL-6 production between 1 and 7 day treatment groups. M. leprae-infected MSCs produced more $IL-1{\beta}$, $TNF-{\alpha}$ and PGE2, but DMfree could not inhibit their production at a physiological concentration. This is different from other reports that used higher concentration of EGCG treatment, resulting in significant inhibition of the cytokines. The inhibition appears to be related to the concentration of EGCG. These results indicate that DMfree can alleviate inflammation involving IL-6.

Anti-platelet Effect of Black Tea Extract via Inhibition of TXA2 in Rat

  • The aim of this work was to investigate the effect of black tea extract (BTE) on collagen -induced platelet aggregation. In this study, BTE (10~500 ㎍/mL) was shown to inhibit platelet aggregation via thromboxane A2 (TXA2) down-regulation by blocking cyclooxygenase-1 (COX-1) activity. Also, BTE decreased intracellular Ca2+ mobilization ([Ca2+]i). Additionally, BTE enhanced the levels of both cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP), which are aggregation-inhibiting molecules. BTE inhibited the phosphorylation of phospholipase C (PLC) γ2 and syk activated by collagen. BTE regulated platelet aggregation via cAMP-dependent phosphorylation of vasodilator-stimulated phosphoprotein (VASP) Ser157. The anti-platelet effects of BTE in high fat diet (HFD)-induced obese rats were evaluated. After eight weeks of BTE treatment (300 and 600 mg/kg), the platelet aggregation rate in the treated groups was significantly less than that in the HFD-fed control group. Also, BTE exhibited a hepatoprotective effect and did not exert hepatotoxicity. Therefore, these data suggest that BTE has anti-platelet effects on collagen-stimulated platelet aggregation and may have therapeutic potential for the prevention of platelet-mediated thrombotic diseases.

Improvement of Memory Impairment of Green Tea Extract/L-Theanine Through Inhibition of Secretase Activity and Cell Death In Vivo (녹차추출물/L-Theanine 혼합물의 Secretase 활성 억제 및 세포사 억제를 통한 기억력 회복능)

  • We examined the effect of green tea extract (GTE) and L-theanine combination on the $A{\beta}_{1-42}$-induced memory dysfunction. GTE and combination were administrated into mice for 3 weeks followed by injection of $A{\beta}_{1-42}$ to induce memory impairment. GTE and L-theanine administration significantly improved cognitive ability and reduced $A{\beta}_{1-42}$ level accompanied with the inhibition of neuronal cell death and activities of secretase. These results suggest that GTE and L-theanine combination may be a useful for preventing for the development or progression of Alzheimer's disease.

Comparison of Green Tea Extract and Epigallocatechin Gallate on Secretion of Catecholamines from the Rabbit Adrenal Medulla

In the presence of CUMC6335 $(200 {\mu}g/mL)$ into an adrenal vein for 60min, CA secretory responses evoked by ACh (5.32 mM), high $K^+$ (56 mM), DMPP $(100{\mu}M \;for\;2min)$, and Bay-K-8644 $(10{\mu}M\;for\;4min)$ from the isolated perfused rabbit adrenal glands were greatly inhibited in a time-dependent fashion. However, EGCG $(10{\mu}g/mL)$ did not affect CA release evoked by ACh, high $K^+$, and Bay-K-8644. CUMC6335 itself failed to affect basal catecholamine output.
