Immunohistochemical Analysis of TBX3 and $\beta$-catenin in Gastric Cancers

  • TBX3 has demonstrated oncogenic activity as a downstream target of the Wnt/$\beta$-catenin signaling pathway. In this study, the aim was to determine whether overexpression of the TBX3 protein is involved in the development and/or progression of gastric cancers. We analyzed the expression pattern of the TBX3 and $\beta$-catenin proteins in a series of 186 sporadic gastric cancers. Altered expression of the TBX3 and $\beta$-catenin proteins was observed in 54 (29.0%) and 48 (25.8%) of the 186 gastric cancers. Statistically, overexpression of the TBX3 and $\beta$-catenin proteins was not associated with the clinical and pathological parameters studied including: histological type, tumor location, tumor size, and the 5-year survival (P>0.05). However, TBX3 overexpression was closely associated with lymph node metastasis and aberrant $\beta$-catenin expression (P<0.05). In addition, overexpression of the TBX3 protein was confirmed by Western blot analysis of primary gastric cancer tissues and cell lines. These data suggest that TBX3 overexpression may play a role in the development and progression of sporadic gastric cancers.

Overexpression of Tbx3 Predicts Poor Prognosis of Patients with Resectable Pancreatic Carcinoma

  • Background: To determine the expressions of Tbx3, a member of subgroup belonging to T-box family, and its prognostic value in pancreatic carcinoma. Materials and Methods: We determined the expression levels of Tbx3 on both mRNA and protein levels in 30 pairs of fresh tumor tissues and paratumor tissues by quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting, respectively. In addition, protein level of Tbx3 were identified using immunochemistry in 80 pairs of paraffin-embedded specimen. The correlations between Tbx3 expression and various clinicopathological parameters as well as overall survival were evaluated. Results: Tbx3 mRNA and protein levels in tumor tissues were significantly higher than in the paratumor tissues by qRT-PCR ($0.05{\pm}0.007$ vs. $0.087{\pm}0.001$, p<0.001) and western blotting ($1.134{\pm}0.043$ vs. $0.287{\pm}0.017$, p<0.001). The statistical analysis based on immunohistochemical evaluation suggested that Tbx3 aberrant expression was significantly associated with several conventional clinicopathological variables, such as gender, age, tumor position, preoperative CA19-9 level, pathological T staging and N staging. Univariate and multivariate analyses revealed that Tbx3 expression was an independent prognostic factor for overall survival (<0.001). Conclusions: Our results suggest that overexpression of Tbx3 is associated with poor prognosis of pancreatic cancer patients. However, additional clinical trials are needed to accurately validate this observation.

Application of KOMPSAT-5 SAR Interferometry by using SNAP Software (SNAP 소프트웨어를 이용한 KOMPSAT-5 SAR 간섭기법 구현)

  • SeNtinel's Application Platform (SNAP) is an open source software developed by the European Space Agency and consists of several toolboxes that process data from Sentinel satellite series, including SAR (Synthetic Aperture Radar) and optical satellites. Among them, S1TBX (Sentinel-1 ToolBoX)is mainly used to process Sentinel-1A/BSAR images and interferometric techniques. It provides flowchart processing method such as Graph Builder, and has convenient functions including automatic downloading of DEM (Digital Elevation Model) and image mosaicking. Therefore, if computer memory is sufficient, InSAR (Interferometric SAR) and DInSAR (Differential InSAR) perform smoothly and are widely used recently in the world through rapid upgrades. S1TBX also includes existing SAR data processing functions, and since version 5, the processing capability of KOMPSAT-5 has been added. This paper shows an example of processing the interference technique of KOMPSAT-5 SAR image using S1TBX of SNAP. In the open mine of Tavan Tolgoi in Mongolia, the difference between DEM obtained in KOMPSAT-5 in 2015 and SRTM 1sec DEM obtained in 2000 was analyzed. It was found that the maximum depth of 130 meters was excavated and the height of the accumulated ore is over 70 meters during 15 years. Tidal and topographic InSAR signals were observed in the glacier area near Jangbogo Antarctic Research Station, but SNAP was not able to treat it due to orbit error and DEM error. In addition, several DInSAR images were made in the Iraqi desert region, but many lines appearing in systematic errors were found on coherence images. Stacking for StaMPS application was not possible due to orbit error or program bug. It is expected that SNAP can resolve the problem owing to a surge in users and a very fast upgrade of the software.

Comparative Analysis of Nkx2-5/GATA4/TBX5 Expression in Chicken, Quail and Chicken-quail Hybrids during the Early Stage of Cardiac Development in Embryos

  • The present study makes an investigation into expression of genes related to cardiac development in chicken, quail and chicken-quail hybrids during the early stage of embryogenesis. Real-time PCR was used to detect mRNA expressions of Nkx2-5, GATA4 and TBX5 in the heart of chicken, quail and chicken-quail hybrids embryos during the 3rd to 7th days of incubation. Results showed that NKX2-5 mRNA displayed a similar expression trend in chicken, quail and chicken-quail hybrids. The initial and highest expression of Nkx2-5 was focused on the 3rd day of incubation, then it declined till 5th day of incubation, thereafter, it fluctuated. Expression of Nkx2-5 gene in quail was significantly higher than in chicken and chicken-quail hybrids, and no significant difference was observed between the two latter species. GATA4 mRNA showed a similar expression trend between chicken and quail, which displayed a steady increase from 3rd to 6th d, then, the expression level decreased. However, GATA4 mRNA expression in chicken-quail hybrids was significantly higher than that in chicken and quail from 3rd to 5th d (p<0.01), but significantly lower than that in chicken and quail during the later stage of the experiment (p<0.05), due to the dramatic drop from 5th d onwards (p<0.01). TBX5 mRNA expression in chicken and quail showed the same trend as GATA4 expressed in the two species. Furthermore, TBX5 expression in chicken-quail hybrids was significantly higher than that in chicken and quail during the whole course of experiment, although relatively lower TBX5 expression was detected in the early stage. In conclusion, Nkx2-5, GATA4 and TBX5 genes showed dynamic changes during the process of cardiac development in chicken, quail and their hybrids embryos. In addition, the expression trend in chicken was similar to that in quail, and there was no significant difference for gene expression level, except NKX2-5. However, expression of these genes in chicken-quail hybrids was significantly different from their parents, the difference mechanism needs to be further explored.

Evaluation of enrichment broth and selective media for the detection of non-O157 enterohemorrhagic Escherichia coli (Non-O157 장출혈성대장균 검출을 위한 증균배지 및 선택배지 성능 평가)

  • In this study, specific and rapid enrichment and growth conditions for the most important, classic non-O157 enterohemorrhagic Escherichia coli (EHEC) serogroups were assessed. Three enrichment broth types, namely, EC medium with MUG broth, BRILA broth, and mTSB broth with novobiocin, were compared to identify the optimum enrichment broth for EHEC isolation. Four kinds of selective media, namely, ENDO agar, Chromocult agar, TBX agar, and CHROMagar$^{TM}$ STEC medium, were compared to identify the optimum one for non-O157 EHEC isolation. The results suggested that incubation in EC medium with MUG broth at $42^{\circ}C$ for 20 h is optimum for the enrichment of non-O157 EHEC. TBX agar was identified to have the highest specificity among the tested media. Consequently, a combination of complementary selective media according to serotype must be considered for comprehensive isolation of specific EHEC.

Effect of Linoleic Acid Content and $omega6/$omega3$ Ratio on the Induction of Gastric Ulcer in the Experimental Rat Model (Linoleic acid 함량과 $omega6/$omega3$비율이 침수속박 스트레스에 의한 위궤양의 발병에 미 치는 영향)

  • This study was designed to determine the effects of linoleic acid contents and $\omega$ 6/$\omega$3 ratios on the induction of gastric ulcer by water immersion and restraint stress. Sprague-Dawley rats were fed 5diets containing 7% fat(w/w) for 6weeks. These diet groups were Lh, Mh, Hh, Mm, Ml, : 3 different linoleic acid levels(0.3% of energy(L). 3.5(M), 10(H) and 3 different $\omega$6/$\omega$3 ratios (11(1), 33(m), 100(h) with beef tallow, sunflower or fish oil. The Lh group showed a significantly higher ulcer index (UI) than the Mh and Hh groups(p<0.05). At the same linoleic levels, the UI had no significant difference within the $\omega$6/$\omega$3 ratios. The Mh group showed significantly higher (p<0.05) PGE2 and TBX2 content than any other group. Pearson's correlation coeffcients between UI and PGE2 and TBX2 had a negatively significant correlation(p<0.05). Linoleic acid of gastric mucosal phospholipids was reflected by the diet, but was not significantly different. The most significant finding of this study is that not only the absolute amount of linoleic acid, but also the $\omega$6/$\omega$3 ratios are important factors for the prevention of gastric ulcer.

  • 본고에서는 TDX 시스팀의 신뢰도 보증 및 효율적 관리를 위하여 TDX-1A 시스팀에서 사용되고 있는 부품의 신뢰도 예측치와 현장운용데이터에서 나온 신뢰도 평가치를 비교 검토한후, TDX-1A 시스팀의 신뢰도 예측결과를 검증하고, 또한 외국의 고장율 예측방법과의 차이점을 도출하여 이를 TDX 시스팀의 신뢰도 예측방법에 적용한 후 TBX-1A 현장운용데이터로 보완된 TDX 환경에 적합한 부품 고장율 예측방법을 정립, 제시하고자 한다.

In silico Analysis of Downstream Target Genes of Transcription Factors (생명정보학을 이용한 전사인자의 하위표적유전자 분석에 관한 연구)

  • Objective: In the previous study, we complied the differentially expressed genes during early folliculogenesis. Objective of the present study was to identify downstream target genes of transcription factors (TFs) using bioinformatics for selecting the target TFs among the gene lists for further functional analysis. Materials & Methods: By using bioinformatics tools, constituent domains were identified from database searches using Gene Ontology, MGI, and Entrez Gene. Downstream target proteins/genes of each TF were identified from database searches using TF database ($TRANSFAC^{(R)}$ 6.0) and eukaryotic promoter database (EPD). Results: DNA binding and trans-activation domains of all TFs listed previously were identified, and the list of downstream target proteins/genes was obtained from searches of TF database and promoter database. Based on the known function of identified downstream genes and the domains, 3 (HNF4, PPARg, and TBX2) out of 26 TFs were selected for further functional analysis. The genes of wee1-like protein kinase and p21WAF1 (cdk inhibitor) were identified as potential downstream target genes of HNF4 and TBX2, respectively. PPARg, through protein-protein interaction with other protein partners, acts as a transcription regulator of genes of EGFR, p21WAF1, cycD1, p53, and VEGF. Among the selected 3 TFs, further study is in progress for HNF4 and TBX2, since wee1-like protein kinase and cdk inhibitor may involved in regulating maturation promoting factor (MPF) activity during early folliculogenesis. Conclusions: Approach used in the present study, in silico analysis of downstream target genes, was useful for analyzing list of TFs obtained from high-throughput cDNA microarray study. To verify its binding and functions of the selected TFs in early folliculogenesis, EMSA and further relevant characterizations are under investigation.

The Luminescence Properties of YAG:$Tb^{3+}$ Phosphor Prepared by Hydrothermal Synthesis (YAG:$Tb^{3+}$ 형광체 분말의 수열합성과 발광 특성)

  • YAG:Tb3+ as green phosphor were studied for the development of low voltage FED phosphor prepared by hydrothermal synthesis. We changed the concentration of luminescence center ion Tb3+ in hydrothermal reaction of which conditions were at 8M NH4OH as mineralizer, at 35$0^{\circ}C$ for 12hrs. As results, we could finally get the YAG:Tb3+ (Y3-xTbxAl5O12) powder of which particle size was about 0.2~1.0${\mu}{\textrm}{m}$. The excitation spectra and the green emitted spectra of YAG:Tb3+ phosphor powder were observed. When we doped 0.25 mol Tb to YAG, we could observe the maximum cathodoluminescence from YAG:Tb3+ phosphor and the chromaticity coordinate of the phosphor was shown x=0.35, y=0.56 in CIE1931 diagram.

A Potential Role of fgf4, fgf24, and fgf17 in Pharyngeal Pouch Formation in Zebrafish

  • In vertebrates, Fgf signaling is essential for the development of pharyngeal pouches, which controls facial skeletal development. Genetically, fgf3 and fgf8 are required for pouch formation in mice and zebrafish. However, loss-of-function phenotypes of fgf3 and fgf8 are milder than expected in mice and zebrafish, which suggests that an additional fgf gene(s) would be involved in pouch formation. Here, we analyzed the expression, regulation, and function of three fgfs, fgf4, fgf24, and fgf17, during pouch development in zebrafish. We find that they are expressed in the distinct regions of pharyngeal endoderm in pouch formation, with fgf4 and fgf17 also being expressed in the adjacent mesoderm, in addition to previously reported endodermal fgf3 and mesodermal fgf8 expression. The endodermal expression of fgf4, fgf24, and fgf17 and the mesodermal expression of fgf4 and fgf17 are positively regulated by Tbx1 but not by Fgf3, in pouch formation. Fgf8 is required to express the endodermal expression of fgf4 and fgf24. Interestingly, however, single mutant, all double mutant combinations, and triple mutant for fgf4, fgf24, and fgf17 do not show any defects in pouches and facial skeletons. Considering a high degree of genetic redundancy in the Fgf signaling components in craniofacial development in zebrafish, our result suggests that fgf4, fgf24, and fgf17 have a potential role for pouch formation, with a redundancy with other fgf gene(s).
