포공영(蒲公英)에 의한 뇌 소교세포에서 산화질소 (NO)의 생성 (Nitric Oxide Production in Brain Microglial Cells by Taraxacum officinale)

  • Nitric oxide (NO) is now recognized as a mediator of several biological and immunological functions, but unlike classical neurotransmitters. NO simply diffuse of the postsynaptic cells and around affecting cells. Taraxacum officinale (Compositae) has been used for maintenance of vitality, and they still occupy an important place in the traditional Korean medicine. We have examined that the effect of Taraxacum officinale water extract on NO synthesis in microglial cells of murine's brain, using the Griess method. And this study was evident that Taraxacum officinale did not induce NO production without recombinant interferon gamma ($rIFN-{\gamma}$), whereas Taraxacum officinale (10-1000 g/ml) with $rIFN-{\gamma}$ effectively produced NO in microglial cells of brain. As result. NO production in microglial cells increased most significantly in dose of 100 g/ml of the Taraxacum officinale and the production of NO was dependent on the dose of Taraxacum officinale, NG-monomethyl-L-arginine, competitive inhibitor of NO synthase, reduced the NO production by Taraxacum officinale stimulation with $rIFN-{\gamma}$ in microglial cells of murine. The effect of Taraxacum officinale was mainly dependent on Taraxacum officinale-induced tumor necrosis factor- secretion. Conclusively, this study suggested that Taraxacum officinale stimulate NO production at microglial cells in brain, which may be an important factor for mediating immune and neuroendocrinologic regulation in nervous system.

서양민들레와 흰민들레 추출물의 Nitric Oxide 생성억제 및 소거 활성과 Tyrosinase 저해 활성 (Nitric Oxide Production Inhibitory and Scavenging Activity and Tyrosinase Inhibitory Activity of Extracts from Taraxacum officinale and Taraxacum coreanum)

  • The study was conducted to investigate functional materials as skin whitening and anti-inflammatory agent from Taraxacum officinale and Taraxacum coreanum. The total polyphenol and flavonoid content in the ethanol extract of Taraxacum officinale were found to be 64.07mg/g and 32.46mg/g, respectively. In tyrosinase inhibitory activity, the hot water extract of Taraxacum coreanum was higher than the other extracts. However, in nitric oxide (NO) scavenging ability, the ethanol extract of Taraxacum coreanum was higher than the other extracts. the ethanol extract of Taraxacum coreanum showed strong NO production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability measurement by MTT assay and the lactate dehydrogenase (LDH) assay against L929 cell, the extracts were exhibited fine cell viabilities and normal LDH release levels as nontoxic result in sample concentration of $250{\sim}1000{\mu}g/m{\ell}$. As a result, the ethanol extract and the hot water extract of Taraxacum coreanum could be applicable to functional materials for anti-inflammatory and skin whitening related fields, respectively.

국내 자생 민들레 5종의 항산화 활성 및 Nitric Oxide 생성억제 활성 (Effect of Five Korean Native Taraxacum on Antioxidant Activity and Nitric Oxide Production Inhibitory Activity)

  • The effect on the antioxidant activity and Nitric Oxide activity production inhibitory activity of Taraxacum has not been known. Therefore, phenolics and flavonoid contents were investigated from the ethanol extracts of five different Taraxacum species. The results showed that, among the five Taraxacum, T. hallaisanensis contains the highest total phenolic and flavonoid contents. When the antioxidant activity was measured by DPPH, $ABTS^+$ and reducing power activity, the free radical scavenging activity of T. hallaisanensis was also the highest among five Taraxacum species. However, measurement by CCK-8 assay in Raw264.7 cells indicated that the extracts of Taraxacum species have no effect on cell viability. Moreover, we also investigated the effect of Taraxacum species on NO scavenging activity in lipopolysaccharide (LPS)-stimulated Raw264.7 cells. The results clearly showed that Taraxacum species inhibited NO production, and the inhibitory effect of T. hallaisanensis was the strongest. The above results suggested that Taraxacum species affected the antioxidant and NO scavenging activity, and among the five species, antioxidant and NO scavenging activity assay of T. hallaisanensis was significantly higher than those of other four Taraxacum species. Therefore, T. hallaisanensis could be used as a potential drug with anti-oxidant and anti-inflammatory effect.

서양민들레의 항산화능이 저장 기간 동안 유과의 품질특성에 미치는 효과 (Antioxidant Effect of Taraxacum officinale on Quality Characteristics of Yukwa containing Taraxacum officinale during Storage)

  • This study investigated the antioxidant effect of Taraxacumofficinale on the qualitycharacteristics of Yugwa during storage. Total polyphenol content was higher than total flavonoid content. DPPH radical scavenging activity and antioxidative index of Taraxacum officinale ethanol extracts were significantly (p<0.05) lower compared with those of Vit.C, whereas they were similar with those of BHA. The L, a, and b levels of Yugwa significantly (p<0.05) decreased with increasing amount of Taraxacum officinale powder; 6% Taraxacum officinale powder showed the lowest L and b levels. Hardness was unaffected by Taraxacum officinale content during the storage period. Brittleness of Yugwa was significantly (p<0.05) lower in control, 1.5%, and 6% Taraxacum officinale powder, whereas 3% Taraxacum officinale powder showed no significant differences during storage. Peroxide value significantly decreased in 3% and 6% Taraxacum officinale powders compared to those of control and 1.5% Taraxacum officinale powder after 4 weeks of storage. Taste, flavor, and crispness showed significant (p<0.05) differences according to Taraxacum officinale concentration. In conclusion, addition of 3~6% Taraxacum officinale powder provides beneficial effects in terms of storage life of Yugwa without any detrimental effects on quality characteristics.

RAPD를 이용한 자생 민들레 종과 귀화 민들레 종간의 연관계 분석 (Analysis of Genetic Relationship Among Native Taraxacum and Naturalized Taraxacum species using RAPD)

  • RAPD를 이용하여 국내에 생육하는 4종의 자생 민들레와 2종의 귀화 민들레 종간의 유전적인 유연관계를 분석하였다 Primer Screening을 통해 선발된 30개의 Primer를 이용하여 RAPD를 행한 결과, Polymorphic band 수는 141개로 나타나 민들레 종간의 유전적인 차이 분석이 가능하였다. Primer OPC12, OPD16, OPK16, OPK17, OPK20, OPSI에서는 각 종마다. 특정 밴드가 있는 것으로 조사되었다. 특히 OPS8에서는 564bp에서 귀화종인 서양민들레 종에서만 특이 밴드가 나타났다. RAPD분석결과 자생종과 귀화종 민들레들은 명확히 2군으로 구분되었다. 1군에는 서양민들레. 붉은씨서양민들레 등의 귀화종 그룹이었으며, II군은 민들레, 산민들레, 좀민들레, 횐민들레 등의 자생종 민들레 그룹으로 나뉘어졌다. Bootstrap 방법으로 6종의 유연관계를 분석한 결과도 비유사계수를 통한 방법으로 분석한 결과와 매우 유사하였다 우리 나라에서 자생하는 민들레속 6종의 주요형질을 조사한 결과 I군에 속하는 민들레류는 II군에 속한 종들에 비해 개화일수가 길고, 외총포편의 방향이 다르며 털의 유무 또한 다른 특징이 있었다. ll군에 속하는 횐민들레는 다른 민들레종과는 화색에서 확연한 차이를 보였으며 자생종들 속에서도 잎의 방향과 발아의 생리적 특성 등 유전적으로 여러 다른 점이 복합적으로 작용함으로서 II군내에서도 유전적 거리가 가장 먼 것으로 나타났다.

  • 남성의 클라이맥스 증후군, 남성갱년기, 또는 테스토스테론 결핍 증후군은 고령화시대 남성노인들의 건강에 새로운 문제 중 하나이다. 이 현상은 나이가 들수록 남성에게서 나타나는 자연현상으로 혈중 테스토스테론 수치의 감소와 신체적 및 정신적 활동의 현저한 감소를 특징으로 한다. 이 연구의 목적은 해면체내 일산화질소(NO)와 혈액 내 남성호르몬의 수준을 비교하여 포공영(Taraxacum coreanum) 열수 추출물의 효과를 조사하였다. 포공영 추출물은 생체외 및 생체내에서 NO 생산량을 용량 의존적으로 증가시켰다. 포고영 추출물을 4 주간 섭취 한 나이든 쥐(22주)에서 다이하이드로 테스토스테론과 17-하이드록시 스테로이드 탈수소효소의 수준과 신경 인산화 질소 합성효소와 cGMP의 양은 유의하게 증가했다. 그러나 프로스타글란딘$E_2$, 테스토스테론 및 성 호르몬 결합 글로불린 수치는 모든 군에서 차이가 없었다. 또한, 총 정자 수 및 운동성 정자 수 또한 유의한 차이가 없었다. 전반적으로, 이러한 결과는 NO, cGMP 및 유리 테스토스테론을 향상시키기 위한 안전하고 효과적인 천연물질로서 Taraxacum coreanum 추출물의 가능성을 시사한다.

포공영 추출물이 흰쥐간 Xanthine Oxidase 활성에 미치는 영향 (Effect of Taraxacum herba Extract on the Hepatic Xanthine Oxidase Activity in Rats)

  • This study was undertaken to investigate the effect of Taraxacum herba extract on the hepatic xanthine oxidase activity as a oxygen free radical generating enzyme in vitro and in vivo. It was observed that partial purified hepatic xanthine oxidase (type O) activity was strongly inhibited by the addition of Taraxacum herba n-butanol extract in vitro. The Km value of xanthine oxidase without affecting the Vmax value for xanthine was significantly increased by the addition of ta-dase (type O) activity was significantly inhibited by the treatment of Taraxacum gerba n-butanol ex-tract for 5days(over 40mg/kg, i.p), whereas, xanthine oxidase (type D) activity was not changed by the injection of Taracacum herba n-butanol extract. Meanwhile, liver weight / body weight(%), serum alanine aminotransferase activity and hepatic lipid peroxide content in Taraxacum herba n-buta-nol extract-treated rat were not changed. These findings led us to conclude that Taraxacum herba n-butanol extract may regulate the hepatic xanthine oxidase type O activity to prevent toxic effect of oxidative stress by the oxygen free radicals.

포공영(민들레)의 동치미 발효에 의한 간세포 보호 및 면역증진 효과 (Effects of fermented Taraxacum mongolicum by Dongchimi on protection of liver cell and immune activity)

  • Objective : This study set out to combine the treatment efficacy of Taraxacum with Dongchimi fermentation and investigate Taraxacum's effects on protection of liver cell and controlling nitric oxide(NO) through experiments, thus checking whether it had values as a physiological active matter. The experimental materials include Taraxacum Dongchimi (TD) and Taraxacum fermented by Dongchimi (TDF). As for methodology, experiments were carried out to compare TD and TDF in components, protection effects for liver cells, anticancer effects on liver cells, and protection effects for brain cells in the aspects of liver function and immunity enhancement. Method : The experimental materials include Taraxacum Dongchimi (TD) and Taraxacum fermented by Dongchimi (TDF). As for methodology, experiments were carried out to compare TD and TDF in components, protection effects for liver cells, anti-cancer effects on liver cells, and protection effects for brain cells in the aspects of liver function and immunity enhancement. Results : As shown in the chromatogram results, each valid component content increased in Taraxacum fermented by Dongchimi (TDF) for each time section. Of them, the valid component content at 36.80 minutes was approximately 2.7 times higher in TDF at 21.8% than in Taraxacum Dongchimi (TD) at 8.28%. TDF generated more excellent protection effects against the toxicity that caused oxidative damage to the liver cell(HepG2) with t-BHP than TD. The survival rate was low in TD of $100{\mu}g/m{\ell}$ and $300{\mu}g/m{\ell}$ and increased to 23.3% in TDF of $100{\mu}g/m{\ell}$. The survival rate was the highest at $300{\mu}M$ with a significant difference of 68.1%(P<0.05). Both TD and TDF showed effects of controlling nitric oxide production according to concentration with TDF recording a higher rate of controlling nitric oxide production than TD. There were significant differences(P<0.05) in the effects of controlling nitric oxide production at 200 ug/ml, 400 ug/ml in both groups. Especially the result TDF of $400{\mu}g/m{\ell}$ was thus similar to those of butein, the positive control group. Conclusion : The result of this studies is that Taraxacum fermented by Dongchimi (TDF) increased the valid component content compared with the simple mixture(TD). The findings clearly show that it is a material with the effects of improving immunity and liver cell protection. If fermentation methods are further developed to use it as a functional material, it will be subject to more opportunities of being used in other functional foods and make a contribution to integrated medicinal food development.

포공영을 이용한 마이크로캡슐 에멀젼에 관한 연구 (Study on the Microcapsule Emulsion Containing Taraxacum platycarpum)

  • The aims of this study was assess to preparation and stability of emulsion containing Taraxacum platycarpum ethanol extract our team was found to shape and size of capsule by optical microscope and Scanning Electron Microscope (SEM). In the stability test of the emulsion including Taraxacum platycarpum ethanol extract, it was stable of pH, viscosity during 30days. According to all the test results that have been given, it is possible that the ethanol extract of Taraxacum platycarpum can be used as a new cosmetics material.

흰민들레와 서양민들레의 부위별 영양성분 및 항산화효과 (Nutritional Composition and Antioxidative Activity of Different Parts of Taraxacum coreanum and Taraxacum officinale)

  • This study compared the nutritional composition and antioxidative activities of different parts (aerial part and root) of Taraxacum coreanum and Taraxacum officinale. To identify the nutrient composition of different parts of Daedelion species, nutrient contents were analyzed. Crude ash, protein, and fat contents of T. coreanum and T. officinale were higher in the aerial part compared to the root. There was no significant difference in crude ash, protein, or carbohydrate content of the aerial part depending on the Daedelion species. Aerial part contents of vitamin A, E, and C were significantly higher in T. coreanum than T. officinale. The K content of the aerial part of T. coreanum and T. officinale was highest, although there was no significant difference among Daedelion species. Contents of oxalic acid, citric acid, and acetic acid in the aerial part of T. coreanum were highest. Contents of total polyphenols, total flavonoids, and DPPH radical scavenging activity of the aerial part were significantly higher in T. coreanum than T. officinale. In conclusion, the aerial part of T. coreanum and T. officinale could be used as antioxidative functional food sources.
