Study on the Anti-oxidant and Cosmeceutical Activity of Taraxacum platycarpum (포공영(浦公英)의 항산화 및 화장품약리학적 활성 연구)

  • Objectives : In this study, anti-oxidant activity of Taraxacum platycarpum is confirmed to investigate cosmeceutical activity for utilization as a cosmetic ingredients. Methods : In the anti-oxidant and whitening effect experiment, the electron donating ability of Taraxacum platycarpum extracts, their SOD-like activity, and the inhibitory activity of xanthine oxidase, tyrosinase were examined. Results : Radical-scavenging activity of water and ethanol extract was examined using ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$ (DPPH). Water and ethanol extract from Taraxacum platycarpum showed 62.9% and 54.7% at 500 ppm concentration in DPPH radical inhibition, respectively. In the test of superoxide dismutase (SOD)-like activity, water extract showed 63.4% at 1,000 ppm concentration, while ethanol extract showed 49% at 1,000 ppm concentration. In the test of xanthine oxidase inhibition, ethanol extract showed 66.8% at 1,000 ppm concentration. Tyrosinase inhibition effect related to whitening effect showed 36.3% and 54.2% in ethanol extract and water extract at 1,000 ppm concentration, respectively. Conclusion : According to these results, it is possible that the extract of Taraxacum platycarpum can be used as a new natural material of cosmetic industry.

Antioxidative and Antibrowning Effects of Taraxacum platycarpum and Chrysanthemum indicum Extracts as Natural Antibrowning Agents (천연 갈변저해제로서의 민들레 및 감국 추출물의 항산화 및 갈변억제 효과)

  • This study was conducted to investigate the effects of Taraxacum platycarpum and Chrysanthemum indicum extracts on the browning of apple slices. The inhibition of the PPO activities, total phenolic contents, and DPPH free-radical-scavenging activities was measured from water and from the 80% EtOH extracts of Taraxacum platycarpum and Chrysanthemum indicum as natural antibrowning agents. Apples were cut into 15-mm-thick slices, and each slice was dipped for 1 min in different treatment solutions (1% Taraxacum platycarpum and Chrysanthemum indicum extracts, 1% citric acid) and was stored at room temperature. The inhibition of the PPO activity in the Chrysanthemumin indicum extract was better than that in the Taraxacum platycarpum extract. The highest DPPH free-radical-scavenging activity (76.10%) was found in the 80% EtOH extract of Chrysanthemum indicum. After 24 h, the ${\Delta}E$ value of the apple slices that were treated in the 80% EtOH extract of Chrysanthemum indicum was at the lowest level (2.22). The total phenolic content was 94.07 mg/g, and the total flavonoid content was 102.60 mg/g, in the 80% EtOH extract of Chrysanthemum indicum. The antibrowning effect of Chrysanthemum indicum was higher than that of Taraxacum platycarpum in the apple slices.

  • For the investigation of medicinal resources from Taraxacum species, the studies were carried out to evaluate the pharmaco-constituents in the aerial part of Taroxacum hallaisanensis, an endemic plant of Korea. From BuOH fraction of the MeOH extract, compound 1 (protocatechuic acid, $C_7H_6O_4,\;3,4-dihydroxy\;benzoic\;acid)$, compound 2 $[C_{22}H_{31}O_6,\;luteolin-7-O-{\alpha}-_L-rhamnopyranosyl(1{\rightarrow}6)-{\beta}-_D-glucopyranoside]$, and compound 3 $[C_{15}H_{20}O_6,\;luteolin-7-O-{\beta}-_D-glucopyranoside]$ were isolated by column chromatographic separation using polyamide and ODS-gel. The structures were elucidated by means of physico-chemical evidences($^1H-NMR,\;{12}^C-NMR$, IR, EI-Mass, FAB-Mass and GC).

Ameliorating Effect of Taraxacum platycarpum Extract in the Scopolamine-induced Cholinergic Blockade Mouse Model (Scopolamine으로 유도된 콜린성 신경계 차단 동물모델에서 포공영(Taraxacum platycarpum) 추출물의 기억력 개선효과)

  • Taraxacum platycarpum H. Dahl. (Compositae) has been used as an anti-inflammatory or anti-cancer agent in the clinic. Although its antidepressant effect has been reported, however, its cognitive function is not investigated until yet. In the present study, we investigated whether the water extract of T. platycarpum (WETP) could improve cognitive function in cholinergic blockade-induced amnesia mouse model using the passive avoidance or Y-maze task. WETP (12.5, 25 or 50 mg/kg) significantly ameliorated the scopolamine-induced cognitive impairment both in the passive avoidance test and the Y-maze test. In addition, WETP significantly inhibited acetylcholinesterase (AChE) activity measured by an ex vivo study using the mouse whole brain. These results suggest that WETP alleviates the cognitive dysfunction caused by the cholinergic blockade, in part, via AChE inhibition, and that it may be a useful for treating cognitive dysfunction.

Relationship of diploid East Aisan Taraxacum Wiggers using the capitulum morphological character (형태형질에 의한 동아시아산 민들레속 2배체 식물의 유연관계)

  • Genus Taraxacum propagated through diploid sexual reproduction and polyploid agamospermy. The cluster analysis of Korean(2 species, 4 population), Japanese (2 species 4 taxa 6 population) and Taiwanese (1 species, 3 population) Taraxacum species using 15 measured capitulum morphological characters was conducted to study the speciation of diploid Taraxacum in East Asia. We measured 15 capitulum morphological characters including length of capitulum, length and shape of outer-involucre, corniculate appandage. Within one population, these characters were very various and were overlapped. The result of cluster analysis using morphological character showed that all species were clustered into four groups (Korean species & T. japonicum group, T. platycarpum subspecies group, T. platycarpum subsp. hodense group, T. formosanum group). Korean species, T. hallaisanense (diploid) and T. ohiwanum (diploid, triploid) were clustered into T. japonicum that was occurred in Kansai provinces of Japan. Therefore, we could infer that Korean species was closely related to Japanese T. japonicum of Japanese species.

In Vitro Free Radical Scavenging and Hepatoprotective Activities of Taraxacum mongolicum (포공영의 자유라디칼 소거 및 간세포 보호활성)

  • The methanol (MeOH) extract and ist fractions of Taraxacum mongolicum (Compositae) were examined for their scavenging effects on 1,1-diphenyl-2- phenylhydrazyl (DPPH) and superoxide radicals, and hepatoprotective effects on tacrine-induced cytotoxicity in human hepatoma cell line, Hep G2 cells. Both methylene chloride $(CH_2Cl_2)$ and butanol (n-BuOH) soluble fractions of the MeOH extract showed the free radicals scavenging and hepatoprotective effects. From these results, it is suggested that hepatoprotective effect of these fractions partly relies on their free radical scavenging activity.

  • The methanol extract of Taraxacum coreanum Nakai (Compositae) was examined on the in vitro oxidation of human low density lipoprotein (LDL). It is well known that LDL oxidation induced artherosclerosis, if we can protect LDL oxidation process, excess plasma lipoprotein accumulation into the arterial lesion prone areas can be blocked. The methanol extract was treated with oxidized LDL which was incubated with $16\;{\mu}M$ $Cu^{2+}$ for metal catalyzed oxidation and TBA value, mobility on agarose gel and formation of conjugated diene and change of vitamin E were determined for the evaluation. The extract showed antioxidative effect at concentration $200\;{\mu}g/ml$ on LDL oxidation.

Dandelion (Taraxacum officinale) Flower Ethanol Extract Inhibits Cell Proliferation and Induces Apoptosis in Human Ovarian Cancer SK-OV-3 Cells

  • This study investigated the proapoptotic effect of ethanol extracts obtained from dandelion (Taraxacum officinale) flower on human ovarian cancer SK-OV-3 cells. Cells were treated with dandelion flowers ethanol extract (DFE) ranging from 1.5625 to $100{\mu}g/mL$ for 24 hr. Significant antiproliferative effects of DFE were first observed from at $6.25{\mu}g/mL$ (p<0.05), and this inhibition showed in a dose-dependent manner. When cells were treated with more than $6.25{\mu}g/mL$ DFE, cell-cycle analysis showed that DFE caused an increase in the percentage of sub-G0/G1 cells and arrested at the S and G2/M phase in a dose-dependent manner. Moreover, apoptosis induction by DFE involved p53 activation and bax upregulation as well as downregulation of bcl-2. Our findings indicate that DFE resulted in apoptotic cell death, suggesting that DFE possesses potential anticancer properties.
