  • The interaction of extremely low frequency magnetic field (ELF-MF) on the frequency of micronuclei (MN) induced by benzo(a)pyrene (BP) in human lymphocytes was examined. A 60 Hz ELF-MF of 0.8 mT field strength was applied for 24 hours either alone or with the tumour initiator, BP. The frequency of MN induced by BP increased in a dose-dependent manner.(omitted)

개의 육아종성 뇌수막척수염 증례 보고 (A case of granulomatous meningoencephalomyelitis in a dog)

  • Granulomatous meningoencephalomyelitis (GME) is a sporadic, idiopathic, non suppurative inflammatory disease of the canine central nervous system. GME appears to have a worldwide distribution and to occur mostly in young to middle-age dogs of small breeds. A 6-year-old female mixed dog with wry neck, ataxia and rolling was submitted to the Cheju National University for diagnosis. Grossly, in the brain, cerebrospinal fluid was mildly increased and dilatation of the subarachnoid blood vessels was observed. Histopathologically, the lesions were characterized by perivascular cuffs of lymphocytes, various numbers of macrophages and plasma cells in the brainstem and cerebral white matter. Numerous granuloma composed of lymphocytes and histiocytes were scattered throughout the brainstem. Two malacic foci characterized by axonal swelling and gitter cell infiltration with hemorrhage were noted in the medulla oblongata and cerebellum. Special stains failed to demonstrate any infectious agents. Immunohistochemically, the infiltrated cells demonstrated strong positive reactions for CD3, a marker for T lymphocytes origin. Based on the clinical signs, histopathology, and immunohistochemistry, this case was diagnosed as GME in a mixed dog.

소자강기탕의 면역조절효과 (Effect of Sojagangqi-tang on the Immunomodulatory Action)

  • The purpose of this research was to investigate the effect of Sojagangqi-tang(SJGQT) on the immune cell activity. The addition of SJGQT enhanced the proliferation of cultured-mice splenocytes and thymocytes. Administration of SJGQT(250 mg/kg) accelerated the subpopulation of splenic T lymphocytes especially CD/sup 4+/-TH cells in BALB/c mice. But high concentration(500 mg/kg) of SJGQT decreased the splenic T, B lymphocytes and thymic Tc (CD/sup 8+/) lymphocytes. Oral administration of SJGQT(250 mg/kg) significantly enhanced the production of IFN-γ and IL-4 in mice serum. And also, the addition of SJGQT(100 ㎍/ml) inhibited the proliferation of cultured-Jurkat leukemia cells in vitro. These results suggest that SJGQT have a cellular immuno-modulatory effect and anti-cancer property action

보기약류(補氣藥類)의 면역약리학적(免疫藥理學的) 고찰(考察) (The immunopharmacologic study of drugs for replenishing Qi)

  • In the result of investigating the drugs for replenishing Qi in many herbal books, we could get consistent relation in their immunopharmacologic effects as follows: 1. The effects of drugs for replenishing Qi was shown in the Shen Nong's Herbal, but its classification was natural such as trees and plants, insects and beasts, fruits, vegetables, and we think that trial about classification of drugs for replenishing Qi was accomplished in the ben-cao-jiu-zhen(本草求眞). 2. Main drugs for replenishing Qi was Gingseng Radix, Codonopsis pilosulae Radix, Astragali Radix, Atractylodis macrocephalae Rhizoma, Dioscoreae Rhizoma, Glycyrrhizae Radix. 3. Drugs for replenishing Qi commonly contain polysaccharides as much, and it was thought that they have also immunopharmacolologic effects by means of activating T and B cells, secreting cytokines, making lymphocytes, activating NK cells, increasing abilities of voracity and generating antibodies. 4. Drugs for replenishing Qi mainly replenish lung-Qi and Wei-Qi so that they have also effects of activating T and B cells, secreting cytokines, making lymphocytes, activating NK cells, increasing abilities of voracity and generating antibodies. 5. Drugs for replenishing Qi modulate content of cAMP and inhibit Na(+)-K(+) ATPase, so that they have effects of treating indistinctive pulse from Qi deficiency by means of inhibition delivery of chemical substances, activating lymphocytes. promoting contraction of myocardium.

알러지성 접촉피부염 유발 피부 주변 림프절에서의 세포성 면역 활성에 관한 면역조직화학적 연구 - T 림프구와 IL-2 수용기의 분포 변화를 중심으로 - (Immunohistochemical Study on the Activation of Cell mediated immunity in Murine Lymph node on Allergic Contact Dermatitis by DNCB -Based on the change of T lymphocytes and Il-2 receptors-)

  • 본 연구는 알러지성 접촉피부염(allergic contact dermatitis) 유발 피부 주변 림프절에서의 세포성 면역활성으로 나타나는 면역조직화학적 형태변화를 조사하기 위해 DNCB로 인위적인 알러지성 접촉피부염을 BALB/C계 생쥐의 샅바위부분 피부에서 유발시킨 후 시간 경과에 따른 샅바위 림프절(inguinal lymph node)에서의 T 림프구와 IL-2 수용기의 분포변화를 관찰하였다. 대조군에서는 L3T4(CD4)에 양성반응을 보이는 도움 T 림프구, Ly2(CD8)에 양성반응을 보이는 세포독성 T 림프구 그리고 CD25R에 양성반응을 보이는 IL-2 수용기를 가진 세포는 곁피질(paracortex)과 수질동(medullary sinus)에서 분포하였다. DNCB에 의한 알러지성 접촉피부염 유발후 24시간부터 도움 T 림프군, 세포독성 T 림프구 그리고 IL-2 수용기를 가진 세포가 곁피질과 수질동에서 증가하기 시작하여 48시간에 이르러서는 그 분포와 양성반응성이 최고에 달했다. 48시간의 이러한 분포는 수질동에서 잘 나타났으며, 특히 세포독성 T 림프구가 많은 증가를 보였다. 72시간에 이르러서는 양성반응세포가 서서히 감소되는 것으로 나타났지만, 대조군에 비해서는 여전히 증가된 분포양상으로 나타났다. 이상의 결과로 미루어보아 DNCB에 의한 접촉성 피부염 유발시 림프절에서는 도움 T림프구의 분열 활성 증대로 인한 IL-2 생성 분비 증가의 결과 세포독성 T 림프구의 분열 활성을 유도하는 일련의 세포성 면역연쇄반응의 활성이 일어나게 된다. 이러한 세포성 면역연쇄반응의 활성은 주변 피부에서 일어나는 알러지성 접촉피부염로 인한 피부손상을 주도하는 것으로 사료된다.

한우 혈절에 관한 형태학적 연구 (Morphological studies on the hemal node in the Korean native cattle)

  • The hemal nodes of the Korean native cattle were studied by gross anatomy and light microscopy. Hemal nodes were spherical or ovoid in shape, and red or black in colors. A hemal node consisted of a thick capsule and a hilum, and had extensive subcapsular and deep sinuses distended by a great number of erythrocytes. Although a few lymphatic nodules and tissues were seen in the parenchyma, no typical cortex and medulla was defined. Blood vessels occurred, but lymph vessel was not observed in nodes. The stroma of the hemal node was composed of reticular cells and fibers. The parenchyma consisted of many erythrocytes and lymphocytes, and a few macrophages and megakaryocytes. The capsule and trabecula was a collagenous connective tissue with smooth muscle cells. B-lymphocytes were principally located in the lymphatic nodules of the hemal node. T-lymphocytes were scattered in the diffuse lymphatic tissues of the hemal node.

벼메뚜기(Oxya japonica japonica Thunberg) 물 추출물 LCT-CT의 항암면역 활성에 관한 유세포 분석학적 연구 (A Flow Cytometrical Analysis of the Antitumor and Immunostimulatory Effects of LCT-CT, a Cold-water Extract Prepared from Rice Grasshopper Oxya japonica japonica Thunberg)

  • Water extracts of rice grasshopper (Oxya japonica japonica Thurnberg) were prepared and their antitumor and immunostimulatory activities were investigated using a flow cytometer. When LCT-CT was ip injected into ICR mice at the dose of 33.3 mg/kg before and after the implantation of $4{\times}10^5$ cells/mouse of sarcoma 180 tumor cells, it inhibited the growth of the tumor cells by 96.6%, showed lymphoblstogenic activities on the splenic lymphocytes and increased the expression of CD25 molecules on the splenic T lymphocytes. When co-cultured with the splenic lymphocytes of a BALB/c mouse, LCT-CT showed strong immunostimulatory activities at the concentration of $25{\sim}100{\mu}g/ml$ by significantly increasing lymphoblasts ratio and CD25 expression.

PHA 및 conditioned medium 이 소의 순환혈액 림프구의 유약화와 rosette 형성에 미치는 영향 (Effect of PHA and conditioned medium on blastogenesis and rosette formation of bovine circulating blood lymphocytes)

  • This study was planned to estimate the activity of bovine circulating blood lymphocytes using phytohemagglutinin-M(PHA) known as T cell mitogen. Bovine circulating blood mononuclear cells(MNCs) was separated, and cultured with or without macrophage($PHA^+/M{\phi}^+$ or $PHA^+/M{\phi}^-$) in conditioned medium which stimulated with various concentration of PHA(0, 5, 10, 15 and $20{\mu}g/ml$ in medium), and then investigated the blastogenic response and rosette formation of lymphocytes. Blastogenic rate(BR) was especially increased in PHA concentration(10 and $15{\mu}g/ml$) of $PHA^+/M{\phi}^+$ group and their BR were $41.5{\pm}6.8%$ and $44.4{\pm}8.9%$, respectively and BR in PHA concentration(15 and $20{\mu}g/ml$) of $PHA^+/M{\phi}^-$ group was $32.8{\pm}6.2%$ and $31.4{\pm}4.6%$, respectively. BR of lymphocytes was more increased in $PHA^+/M{\phi}^+$ than $PHA^+/M{\phi}^-$ group when these cells were stimulated by PHA. Rosette forming rate(RFR) of lymphocytes to SRBC highly increased when SRBC was treated with AET and/or dextran, respectively. On the orther hand, RFR significantly increased more in $PHA^+/M{\phi}^+$ and $PHA^+/M{\phi}^-$ group than in control group, but when compared with two groups, statistical significancy was recognized only in PHA concentration($15{\mu}g/ml$, p<0.026) of $PHA^+/M{\phi}^+$ group.

베타-아밀로이드 단백질 유전자 발현 연구 (Gene Expression of Beta-Amyloid Precursor Protein)

  • Lowered immune function in the senile dementia patients may be related to the abnormal metabolism of amyloid precursor protein(APP). To investigate the passibility of an abnormal metabolism of APP in lymphocytes and the possible role of APP in the activation of lymphocytes in senile dementia patients, immunohistochemical study of rat spleen and fluorescence activated cell sorter analysis(FACS) of human lymphocytes with the specific antigen far each lymphocyte and double fluorescent marker with antibody to APP were performed. After stimulating lymphocyte with phytohemagglutinin(PHA), APP mRNA and protein were extracted and quantitfied and the influence of ${\beta}$-amyloid protein($A{\beta}$) specific antibody on lymphocyte division was investigated. In spleen, the majority of cells showing $A{\beta}$ immunoreactivity was found in the T-sell dependent zone. FACS indicated that around 90% $CD_4(+)$ T-cells and 60% of $CD_8(+)$ T-sell were immunoreactive to $A{\beta}$ specific antibody(mAb 4G8). Northern blot analysis shows that lymphocyte APP mRNA was gradually increased to reach a maximum at 3 days after activation with lectin mitogen PHA. However, the $A{\beta}$ immunoreactivity an cell surface remained constant during stimulation with PHA, indicating that the release of APP(secreted farm of APP) might be increased. A very large increase in soluble APP secretion was observed in T-lymphocyte upon activation, but only law levels in the resting stale. Immunoblot was carried out an the protein obtained from cell lysate after stimulating lymphocyte by applying PHA to the cultured lymphocyte, and the result was that $A{\beta}$ band of immature farm under 116 KDa marker decreased as the duration of culture was increased after PHA stimulation. The monoclonal $A{\beta}$ specific(4G8) and polyclonal APP antibodies did not inhibit the [$^3H$]-thymidine uptake of mitogen-treated lymphocytes significantly, suggesting that mitogenesis can not be inhibited by specific $A{\beta}$ and polyclonal APP antibody. These results suggest that APP is expressed in T-cell and might be closely associated with the function of T-cells.

폐흡충(Paragonimus westermani) 감염이 흰쥐의 IgE 생성 및 비장림프구 아군분포에 미치는 영향 (Changes of IgE production, splenic helper and suppressor T lymphocytes in mice infected with Paragonimus westermani)

  • 폐흡충 피낭유충을 BALB/c 마우스에 경구 감염시킨 후 마우스를 시기별(3일, 1주, 2주, 3주, 4주, 5주, 7주, 9주, 13주 및 23주)로 희생시켜 말초혈액내 호산구수와 혈청내 IgE 항체가를 측정하는 한편 비장 림프구를 분리하여 림프구 아군(helper T, cytotoxic / suppressor T, pan T)에 대한 단세포군 항체를 결합시켜 EPICS C fluorocytometer로 분석하였다. 폐흡충에 대한 혈청내 총 IgE 항체가는 감염 3주부터 증가되어 감염 4주에 높게 증가되었으며 13주 이후 23주까지 높은 항체가가 유지되었다. 말초혈액내 호산구수는 감염 2주 부터 증가되어 감염 9주에 $664.1/\textrm{mm}^3$으로 가장 증가 되었으며 그 이후에는 감소하여 23주에는 정상보다 약간 증가되었다. 폐흡충을 감염시키고 감염 3일, 1주, 2주, 3주, 4주, 7주 및 23주에 T 림프구 아군을 분석한 결과 L3T4 helper T 림프구와 Lyt-2 cytotoxic / suppressor T 림프구의 비율은 감염 군에서 감염 초기에 대조군에 비해 약간 감소된 경향을 보였으나 대조군과 유의한 차이를 보이지 않았다. Thy 1.2 pan T 림프구의 비율은 감염 2주와 4주에 대조군에 비해 유의하게 감소되었고(P<0.05) 7주 이후에는 대조군과 비슷하였다. 이러한 결과들로 보아 마우스의 폐흡충감염에서 특이 항원이나 마이토젠의 자극이 없이는 비장킬프구의 아군분포에 큰 변화가 없는 것으로 생각된다. 생각된다.
