First Report on Hirsutella subulata, a Pathogen of Rice Stem Borer, Chilo suppressalis in Korea (국내 미기록 이화명충 병원사상균(Hirsutella subulata)에 관한 보고)

  • Hirsutella subulata (Hyphomycetes) was observed for the first time on diseased larvae of Chilo suppressalis in Korea. This fungus formed slender and slightly attenuated synnemata consisting of closely bound, longitudinal hyphae on the insect larvae. Typical characteristics of this species are ellipsoid or broadly obovoid phialides with slender, needle-like necks and asymmetric conidia. It is very similar to H. barberi but can be distinguished by its asymmetrical conidia.

Simultaneous Production System of Silkworm Dongchunghacho and Male Pupae Using Both Parent Sex-limited Larval Marking Variety (한성반문잠품종을 이용한 누에동충하초 및 숫번데기의 동시 생산체계)

  • This study was conducted to confirm the mass production of male pupae and sex-limited larval marking variety as a host for synnemata production of Isaria tenupes in RDA(Rural Development Administration). Silkworm pupation, infection rate and synnemate formation of I.tenuipes were examined. Among the silkworm varieties tested, male Hansaengjam showed the highest pupation rate at 98.7%. I. tenuipes infection rate of larvae of newly-exuviated 5th instar silkworm was 83.7 ~ 90.4% in the spring rearing season and 91.7 ~ 96.6% in the autumn rearing season. Synnemata production of I. tenuipes was execellent in female Yangwonjam with an incidence rate of 99.5% followed by male Yangwonjam(99.5%) and Baegokjam(99.4%) in the spring and autumn rearing season. Synnemata living weight ranged from 0.93 ~ 1.25 g in the spring rearing season. The female Hansaengjam had the heaviest synnemata weight(1.25 g). Synnemata dry weight ranged from 0.27 ~ 0.35 g in the spring rearing season. The female Yangwonjam had the heaviest synnemata weight(0.35 g).

Identification of Infectious Fungus in the Artificial Cultivation Rooms of Paecilomyces tenuipes (눈꽃동충하초 재배상 내에 발생된 오염균의 동정)

  • A infectious fungus (J131) recently isolated from artificial cultivation farms of Paecilomyces tenuipes is known to cause a great yield loss and deterioration of products. In this study, its occurrence and characterization were made. The isolate (J131) was identified as Paecilomyces farinosus (Holm ex S. F. Gray) Brown & Smith. Colonies of the isolate on PDA grew moderately fast, attaining a diameter of 19mm within 14 days at 24${\pm}$1$^{\circ}C$. Vegetative hyphae are smooth-walled and hyaline. Conidiophores mainly arose from the submerged mycelium, usually measuring 1.3-1.5$\mu\textrm{m}$ wide and consisted of verticillate branches with whorls of 2 to 4 phialides. Phialides of 3.8-5.8${\times}$1.5-1.8 $\mu\textrm{m}$, with a swollen basal portion tapered into a distinct neck of about 0.6-0.8 $\mu\textrm{m}$ wide. Conidia arisen from the phialides, were ellipsoidal to fusiform, smooth-walled, hyaline, 1.8-2.4${\times}$1.1-1.4$\mu\textrm{m}$. .When silkworm larvae were injected with concentration of 1${\times}$10$\^$8/ conidia/$m\ell$ of infectious fungus J131, infection rate was 41.3% and synnemata formation rate was 23.6%. Black spots were appeared on the surface of silkworm larvae in 5-9 days after injection. The surface of larvae was covered with hyphae which were gradually turned into yellow from white color during the incubation period of 3-4 days. 1 to several 4-6mm long synnemata of yellow or orange color were formed on the surface. The matured synnemata were ramified from its end and produced much conidia of white color.

  • The effects of LED irradiation on growth and functional components of crops are actively being studied. We investigated the effects of red LED on the cordyceps cultivation because Cordyceps is very difficult to find in the wild and to cultivation. The synnemata formation rate of P. tenuipes was 100% under red LED. Also weight, length, and number of the synnemata were significantly increased. We expect that the red LED treatment will be of help to improve farm income.

Asexual Stage and Fruit Formation of Cordyceps staphylinidaecola (유충노랑곰보동충하초(Cordyceps staphylindaecola)의 불완전세대와 자실체 형성)

  • One hundred fifty one specimens of Beauveria spp. from 19 different locations were collected from September 1 to August 31, 2002. Most of the isolates were identified as Beauveria. bassiana. Cordyceps staphylinidaecola collected from Mt. Obong in Chunchon City covered the host with mycelia which were produced 1 to 4 stromata along with asexual spores. The size of bright yellow ununiform stromata were about 45 mm and the head about $17mm{\times}4mm$. Perithecia completely immersed were $530{\sim}550{\times}290{\sim}300{\mu}m$ in size and mainly scattered on the head. Ascospore produced in asci in the size of $400{\sim}450{\times}4{\sim}5{\mu}m$ developed thread-like secondary spores, which were directly separated into secondary conidial spores. Conidia produced at apical portion of synnemata were $2.6{\sim}3.4{\times}1.2{\sim}1.9{\mu}m$ in size. High density of mycelium was observed at $25^{\circ}C$ ranged from pH 6.5 to 8.5 after 11 days of inoculation. It took 15 to 18 days after inoculation to fully grow on the medium mixed brown rice with pupa. Mycelium developed stromata on the medium 30 days after completion of mycelial growth, where perithecia were produced in 40 days.

The Medium Development for Entomopathogenic Fungi by Using Silkworm Powder (누에분말을 이용한 곤충기생균의 최적생장배지 개발)

  • This study was carried out to investigate the optimal medium and optimal dextrose concentration of 12 entomopathogenic fungi. The colonies of Cordyceps gracilioides, C. sp.(J23), Paecilomyces farinosus(J5), P tenuipes, Paecilomyces sp.(J4), Beauveria bassiana and B. brongniartii on SLA(silkworm larva agar) medium are similar to those on PDA(Potato dextrose agar) medium. On the contrary, the colonies of 5 fungi are very various according to its medium. The optimal medium for 1 1 fungi was SLA-A medium, which was better than PDA medium with regard to the mycelial growth. The optimal concentration on mycelial growth was superior to 6% for 6 fungi. When silkworm larvae was injected with P. tenuipes on SLA medium for fruiting body production. Infection rate, fruiting body formation rate, synnemata number and synnemata length were 91.3 $\pm$ 1.5%, 88.6 $\pm$ 2.3%, 56.6 $\pm$ 3.2 and 33.4 $\pm$ 2.88 ㎜ respectively. In economical analysis, the prototype medium can reduce 61.9% of production cost per 1 liter compared with PDA medium.

Distribution and Classification of Cordyceps Parasites in Cicada (매미를 기주로 한 동충하초의 분포와 분류 동정)

  • Purpose: The aim was to collect and classify the Cordyceps parasitized in cicadas from July to October every year from 1990 to 2016 in Korea. And they were frequently collected in Nepal, Vietnam, Japan, China, and Thailand. Methods: Cordyceps parasitizing cicadas collected in mountains and fields. Results: A total of 1,104 specimens were collected that belonged to 10 different species under nine genera. The highest number of samples belonged to Ophiocordyceps (563 specimens), followed by Isaria (361 specimens), Polycephalomyces (73 specimens), Cordyceps (70 specimens), Beauveria (25 specimens), Perennicordyceps (8 specimens), Metarhizium (2 specimens) and Purpureocillium (2 ones). Among Ophiocordyceps spp. O. longissima was most frequently collected with a total of 426 samples out of 563, followed by O. heteropoda with 120 ones and O. sobolifera with 17 specimens. The species mainly collected in Korea, but C. ishikariensis was collected in Nepal only. The new characteristic was that Isaria cicada-like synnemata were found growing together with C. ishikariensis stromata on the same host. In Korea, the collected 691 specimens in total out of 1,104 were found in Mt. Halla in Jeju Island. Other mountains in Korea where the samples were collected were Mountains Daeryong, Jiri, Yongmoon, Samag, Seolag, Gujeol, Duryun, Baegam, Chilgap, Chundeung, Naejang, Welchul, and Daeryong. The three samples were not identified. Conclusions: A total of 1,104 specimens belonged to 10 different species under nine genera, and the collected 691 samples were found in Mt. Halla in Jeju Island.

Formation of Teleomorph of the White Root Rot Fungus, Rosellinia necatrix, and the Potential Role of its Ascospores as Inocula

  • Stromata of the white root rot fungus, Rosellinia necatrix, were produced on diseased roots although they were reported to develop rarely in nature. Forty-two (42) out of 47 samples produced synnemata while 23 developed stromata. Forty-seven (47) isolates obtained from diseased root samples were divided into 24 mycelium compatibility groups (MCGs). Sixteen (16) out of 24 MCGs produced stromata. Single ascospore isolates from 10 stroma samples produced dsRNA-containing isolates from diseased tissue beneath stromata. The frequency of synnema production on axenic culture varied among isolates with different origin. The dsRNA was not transmitted vertically to the ascospore offspring despite the infection of various dsRNA in the parental isolates. The dsRNA was absent in 35 ascospore isolates in two stroma samples that originated from the isolates, in which dsRNA was not eliminated by hyphal tip isolation. Consequently, sexual reproduction in the white root rot fungus was suggested to produce propagules as a new infection source and to have the function to eliminate infectious factors such as mycoviruses.

Isolation and Culture of Entomopathogenic Fungus, Cordyceps sphecocephala

  • In this study, morphology of perithecia, asci, ascospores, etc. of C. sphecocephala were examined for its telemorphic characteristics. Its colony grew up to 32 mm in diameter on potato dextrose agar (PDA) for 30 days under the condition of $24{\pm}1^{\circ}C$. PDBLA and PDBAA media were selected as optimal media for C. sphecocephala, on which the growth was 1.5 times as fast as on PDA medium. Moreover, PDBLA medium induced successfully the synnemata of anamorphic state. C. sphecocephala was able to be proliferated in vitro on both larva and adult of honeybee drone as its substrate. After inoculated onto the drone larva, it produced mycelium at $24{\pm}1^{\circ}C$, with the maximum yield up to $67{\pm}3mg$ on the $50^{th}$ day.

  • Conidiomata of the Rosellinia necatrix were induced in axenic culture under near ultraviolet light radiation. Pieces of sterilized Japanese pear twigs were placed on the 7 days-old oatmeal agar culture in plates. The plates were further incubated far 5 days and then illuminated by under near ultraviolet light radiation. Syn-nemata were developed on twigs in 19 out of 20 isolates tested within 5 weeks, and conidia were observed in 12 out of the 19 isolates. The synnemata and conidia produced were morphologically identical to those of Dematophora necatrix.
