Optical Characteristics of Plasmonic Nano-structure Using Polystyrene Nano-beads (폴리스티렌 나노 비드를 이용한 플라즈모닉 나노 구조체의 광학 특성)

  • We proposed and demonstrated the double layered metallic nano-hole structure using polystyrene beads process to enhance the sensitivity of surface plasmon resonance (SPR). The double layered SPR structures are calculated using the finite-difference time-domain (FDTD) method for the width, thickness, and period of the metallic nano-hole structures. The thickness of the metal film and the metallic nano-hole is 30 and 20 nm in the 214 nm wide nano-hole size, respectively. The double layered SPR structures are fabricated with monolayer polystyrene beads of 420 nm wide. The sensitivities of the conventional SPR sensor and the double layered SPR sensor are obtained to 42.2 and 52.1 degree/RIU, respectively.

  • Exosomes, known as nanoscale extracellular vesicles in the range of 30-150 nm, are known to contain clinically significant information. However, there is still insufficient information on exosomal membrane proteins for cancer diagnosis. In this work, we investigated the characteristics of the membrane proteins of exosomes shed by cultured breast cancer cell lines using a surface plasmon resonance (SPR) spectroscopy and pre-activated alkanethiols modified sensor chips. The antibodies of breast cancer biomarkers such as MCU-16, EpCAM, CD24, ErbB2, and CA19-9 were immobilized on the pre-activated alkanethiols surfaces without any activation steps. The purified exosomes were loaded onto each antibody surface. The affinity rank of the antibody surfaces was decided by the relative capture efficiency factors for the exosomes. In addition, an antibody with a relative capture efficiency close to 100% was tested with exosome concentration levels of 104/µl, 105/µl, and 106/µl for quantitative analysis.

  • In this paper, we have theoretically analyzed and designed a dielectric multi-layer sensor with a SPR (surface plasmon resonance) using analytical calculation and FDTD (finite difference time-domain) methods. The proposed structure is composed of periodic layer and thin metal film. It has many advantages. One of that is a high sensitivity of the SPR. Another is a high Q-factor of the characteristics in the PhC (photonic crystals) micro-cavity structure. The incident light has double resonance characteristics, because the filtered light by PhC structure, dielectric multi-layer, is met the thin metal film for SPR effect. We have also observed the change of resonance characteristics according to the variation of effective index on the metal film.

Development of Thermo-Cosmetics Using Photothermal Effect of Gold Nanoparticles (금 나노입자의 광열효과를 이용한 온열화장품 개발)

  • Many applications of nanoparticles have been developed since 1970s. Surface plasmon resonance (SPR) effect can be generated at the surface of nanoparticles by illumination. SPR is the resonant oscillation of conduction electrons at the surface material stimulated by incident light. The collisions between excited electrons and metal atoms can cause the production of thermal energy (photothermal effect). Here, we presented the development of thermo-cosmetics using photothermal effect of gold nanoparticles. Gold nanoparticles (GNPs) were chosen for it's low toxicity. We also and investigated the cell biocompatibility and heating effectiveness for photothermal effect of GNPs. Synthesized GNPs were verified by UV-vis spectrophotometer, where GNP has a characteristic absorbance spectrum. Concentration of GNP was measured by atomic absorption analyzer. The cytotoxicity was confirmed by MTT assay and double staining assay. Photothermal effect of GNP was demonstrated by the thermal increasing properties depending on GNP concentration, which was taken by an IR-thermal camera with a xenon lamp as the light source. If the thermal effect of GNP is applied for thermo-cosmetics, it can supply heat to skin by converting solar energy into thermal energy. Thus, cosmetics containing GNPs can provide benefits to people in the cold region or winter season for maintaining skin temperature, which lead to a positive effect on skin health.

  • We have formulated hexagonal-shaped gold nanoplates in a single-step for photothermal therapy that gold ions to gold particles using pyrenyl dextran as reducible stabilizer and template. They exhibit anisotropic structure with broad surface plasmon resonance (SPR) band into near-infrared (NIR) spectrum enabling photothermal therapy. These gold nanoplates are also confirmed biocompatibility and high uptake efficiency due to binding with dextran molecules on the surface of gold nanoplates and cells. From in vitro phtothermal ablation study under NIR laser, gold nanoplates have the potential to use as photothermal agents.

Minor Coat Protein pIII Domain (N1N2) of Bacteriophage CTXф Confers a Novel Surface Plasmon Resonance Biosensor for Rapid Detection of Vibrio cholerae

  • Bacteriophages are considered excellent sensing elements for platforms detecting bacteria. However, their lytic cycle has restricted their efficacy. Here, we used the minor coat protein pIII domain (N1N2) of phage CTXφ to construct a novel surface plasmon resonance (SPR) biosensor that could detect Vibrio cholerae. N1N2 harboring the domains required for phage adsorption and entry was obtained from Escherichia coli using recombinant protein expression and purification. SDS-PAGE revealed an approximate size of 30 kDa for N1N2. Dot blot and transmission electron microscopy analyses revealed that the protein bound to the host V. cholerae but not to non-host E. coli K-12 cells. Next, we used amine-coupling to develop a novel recombinant N1N2 (rN1N2)-functionalized SPR biosensor by immobilizing rN1N2 proteins on gold substrates and using SPR to monitor the binding kinetics of the proteins with target bacteria. We observed rapid detection of V. cholerae in the range of approximately 103 to 109 CFU/ml but not of E. coli at any tested concentration, thereby confirming that the biosensor exhibited differential recognition and binding. The results indicate that the novel biosensor can rapidly monitor a target pathogenic microorganism in the environment and is very useful for monitoring food safety and facilitating early disease prevention.

Development of SPR Gas Sensor for Small Molecules Using Molecularly Imprinted Polymer Thin Films

  • Molecularly imprinted polymer thin films were applied to develop a gas sensor based on the surface plasmon resonance phenomenon for small gaseous molecules such as toluene and xylene. The imprinted polymer films were synthesized via photo-polymerization method using various combination of templates, functional monomers and cross-linkers. The temperature of pre-polymerization solutions and the power of UV light were controlled for optimized performance of gas sensing. The morphology and porosity of the polymer films were controlled by varying the mixing ratios of the pre-polymerization solutions and confirmed by atomic force microscopy. By fitting the adsorption/desorption sensorgrams to conventional kinetic models, the effects of different templates and cross-linkers were interpreted in term of the structural differences of the polymer networks formed on the gold film. The sensitivity and selectivity of sensors were estimated for toluene and xylene, and also for humidity and other gaseous molecules such as formaldehyde and ammonia.

  • 미생물수를 측정하는 가장 정확하고 보편적인 방법은 배양법이다. 그러나 배양법은 많은 시간과 노력이 소요되기 때문에 현장에서 실시간에 준하는 측정법으로 사용하기에는 원천적으로 한계를 가지고 있다. 이러한 한계를 극복하기 위해서는 실시간(real-time) 또는 준실시간(near real-time)으로 미생물을 검출할 수 있는 기술이 요구된다. 표면플라즈몬공명(surface plasmon resonance: SPR)은 생물분자 또는 미생물을 실시간 또는 준실시간으로 검출할 수 있는 센서로서 특이성(specificity)과 정확도(accuracy) 측면에서 일찍이 큰 관심을 받아왔다. 1902년, Wood(1902)는 반사 회절격자를 사용하여 연속광원의 스펙트럼을 관찰한 결과, 회절광 스펙트럼에서 어두운 좁은 밴드를 발견하였으며, Fano(1941)는 이 현상이 surface plasma waves와 관련이 있음을 이론적으로 밝혔다.
