  • A novel surface plasmon resonance(SPR) multisensing method, which does not require imaging apparatus such as CCD, has been proposed and implemented experimentally. The proposed method is based on the multichannel SPR and the separation of signals by use of additional layers whose thickness is controlled. SPR signals are influenced by the thickness of sensing layer as well as the optical condition of sensing surface. As the SPR signals from different ligands are usually positioned closely, the reflected light from sensing surface does not provide us with the clear differences of resonance signal depending on the kinds of ligands. It was found from our experiments that SPR signals from each ligand that is located on the additional layer with different thickness can be separated clearly enough to identify various signals from different ligands. Proposed method with theoretical design and simulation has been verified experimentally by using $SiO_2$ thin film layer as additional layer.

  • We have investigated the grating coupled surface plasmon resonance (GC-SPR) sensors using ZnO nano-grating structures to enhance the sensitivity of an SPR sensor. The GC-SPR sensors were analyzed using the finite-difference time-domain method. The optimum resonance angles of 49 degrees are obtained in the 150 nm wide grating structure with a period of 300 nm for the ZnO thickness of 30 nm. Then, the ZnO nano-grating patterns were fabricated by using laser interference lithography. The measured resonance angle of nano-grating patterns was around 49 degrees. Here, an enhanced evanescent field is obtained due to the surface plasmon on the edge of the bandgap when the ZnO grating structures are used to excite the surface palsmon.

고감도 단금속 및 쌍금속 표면 플라즈몬 공명 센서 설계를 위한 수치해석 연구 (Numerical Investigation on Surface Plasmon Resonance Sensor Design with High Sensitivity Using Single and Bimetallic Film Structures)

  • Surface plasmon resonance (SPR) has been widely used for biological and chemical sensing applications. The present study investigates numerically the optical characteristics for the single Au film and bimetallic Ag/Au film SPR configurations by using the multiple beam interference matrix (MBIM) method. We use the prism coupling method, especially Kretschmann configuration for excitation of surface plasmon wave (SPW). The estimated results of reflectance, phase shift and magnetic field intensity enhancement factor are provided for finding out the optimum configuration with high sensitivity for SPR measurement. As a result, the optimum thicknesses are found to be 52 nm for a single Au film and 5 nm to 36 nm for bimetallic Ag-Au film. From the comparison of full width half maximum (FWHM) values for reflectance, phase shift, and enhancement of magnetic field intensity, it is concluded that the highest sensitivity can be obtained when using the phase shift for SPR sensor.

A Portable Surface Plasmon Resonance Biosensor for Rapid Detection of Salmonella typhimurium

  • Here, the rapid detection of Salmonella typhimurium by a portable surface plasmon resonance (SPR) biosensor in which the beam from a diode laser is modulated by a rotating mirror is reported. Using this system, immunoassay based on lipopolysaccharides (LPS)-specific monoclonal anti-Salmonella antibody was performed. For the purpose of orientation-controlled immobilization of antibodies on the SPR chip surface, the cysteine-mediated immobilization method, which is based on interaction between a gold surface and a thiol group (-SH) of cysteine, was adopted. As a result, using the portable SPR-based immunoassay, we detected S. typhimurium in the range from 10^7 CFU/mL to 10^9 CFU/mL within 1 hour. The results indicate that the portable SPR system could be potentially applied for general laboratory detection as well as on-site monitoring of foodborne, clinical, and environmental agents of interest.

Detection of Multi-class Pesticide Residues Using Surface Plasmon Resonance Based on Polyclonal Antibody

  • The detection of carbamate (carbofuran, carbaryl, benfracarb, thiodicarb, and methomil) and organophosphate (diazinon, cadusafos, ethoprofos, parathion-methyl, and chlorpyrifos) pesticide residues with very low detection limits was carried out using surface plasmon resonance (SPR) based equipment. The capacity to develop a portable SPR biosensor for food safety was also investigated. The applied ligand for the immunoassays was polyclonal goat anti-rabbit immunoglobulin (IgG) peroxidase conjugate. Concentration tests using direct binding assays showed the possibility of quantitative analysis. For ligand fishing to find a proper antibody to respond to each pesticide, acetylcholinesterase (AChE), and glutathione-S-transferase (GST) were tested. The reproducibility and precision of SPR measurements were evaluated. With this approach, the limit of detection for pesticide residues was 1 ng/mL and analysis took less than 11 min. Thus, it was demonstrated that detecting multi-class pesticide residues using SPR and IgG antibodies provides enough sensitivity and speed for use in portable SPR biosensors.

  • We propose a novel micro surface plasmon resonance (SPR) sensor system based on polymer materials. The proposed SPR system consists of the incident medium with polymer waveguide and the gold thin film for sensing area. Using a polymer optical waveguide instead of a prism in SPR sensing system offers miniaturization, low cost, and potable sensing capability. The whole device performance was analyzed using the finite-difference time domain method. The optimum gold thickness in the attenuated total reflection mirror of polymer waveguide is around 50 nm and the resonance angle to generate surface plasmon wave is 68 degrees.

SPR-based Antibody-Antigen Interaction for Real Time Analysis of Carbamate Pesticide Residues

  • This research was conducted to develop a quick and sensitive method of detecting carbamate residues using the immobilization of antibody-antigen interactions with surface plasmon resonance (SPR). We have used commercialized surface plasmon resonance equipment (Biacore 3000). The antibody used for the immunoassay was specific for glutathione-s-transferase (GST) and the antigens included several carbamate pesticides (carbofuran, carbaryl, and benfuracarb). When antigens were applied to the protein GST, the detection limit was 2 ng/mL of carbamate pesticide. The fabricated protein GST maintained its activity for over 200 measurements. Thus we determined that the SPR biosensors could detect the specific reversible binding of a reactant in solution to a binding partner immobilized on the surface of the sensor and allow real-time detection and monitoring.

Characteristics of a Bimetal-Layer Chip of a Surface Plasmon Resonance Sensor in the Intensity Interrogation for Tumor Marker Detection

  • The characteristics of a bimetallic surface plasmon resonance (SPR) chip were investigated to detect a tumor biomarker, carcinoembryonic antigen (CEA). The linewidth and the tangential slope of the reflectance curve of the bimetallic SPR chip was compared with those of the reflectance curve of a conventional gold (Au) SPR chip. The changes in reflectance in response to the variation in CEA in the critical concentration range were analyzed at an angle where the tangential slope of the reflectance curve was maximum. From linear regression analysis, the sensitivity of the bimetallic SPR chip with respect to the CEA in critical concentration was obtained.

Development and evaluation of surface plasmon resonance imaging for the detection of antibodies against classical swine fever virus in swine

  • A protein chip based on surface plasmon resonance (SPR) imaging was developed for measuring classical swine fever virus (CSFV) antibody using a recombinant gp55 protein as an antigen. The diagnostic potential of SPR imaging for detecting antibodies to the CSFV gp55 protein was compared with that of a enzyme -linked immunosorbent assay (ELISA) using 70 pig sera. There was a strong positive correlation between the SPR imaging and ELISA (n=70, r=0.916, p<0.01). Therefore, the SPR imaging, which is a label-free and high-through put method, is expected to be a valuable tool in the serodiagnosis of CSFV.

표면플라즈몬공명 바이오센서를 이용한 살모넬라 검출 (Detection of Pathogenic Salmonella Using a Surface Plasmon Resonance Biosensor)

  • Rapid detection of foodborne pathogens has been a major challenge for the food industry. Salmonella contamination is well known in all foods including pasteurised milk. The possibility of specific detection of Salmonella Enteritidis by surface plasmon resonance (SPR) biosensor was explored using a commercially available portable SPR sensor. Self assembly technique was adopted to immobilize anti-Salmonella antibodies on the gold sensing surface of the SPR sensor. The concentration of polyclonal antibody for use in the SPR biosensor was chosen to 1.0 mg/mL. Experiments were conducted at near real-time with results obtained for one SPR biosensor assay within 1 hour. The limit of detection for Salmonella Enteritidis was determined to be $10^6$ CFU/mL in both PBS buffer and milk samples. The assay sensitivity was not significantly affected by milk matrix. Our results showed that it would be possible for employing the SPR biosensor to detect Salmonella Enteritidis in near real-time.
