Determination of Shelf-life of a Packaged Paralichthys Olivaceus for Super-chilled Distribution (슈퍼칠링 유통을 위한 포장광어 (Paralichthys Olivaceus)의 유통기한 설정 연구)

  • Maintaining steady and low temperature during distribution process is a key technique to inhibit microbial growth, one of the most important factors in determining the shelf life. It is possible to provide high-quality fresh products to consumers only when precise and adequate temperature control is guaranteed throughout the entire distribution process for fresh seafood. This study investigated the shelf-life of packaged fresh flounder (Paralichthys Olivaceus) in order to learn the feasibility of super-chilled distribution of fresh seafood. To estimate the shelf life, weight, number of bacteria such as E. coli, pH, sensory test and volatile basic nitrogen were investigated. As a result of the study, the difference in shelf life of 6 days at the super-chilling temperature (0±1℃) and 1 day at the general refrigeration temperature (8±2℃) (based on volatile base nitrogen) showed the market possibility of super-chilling distribution. Through additional empirical studies such as packaging methods and economic feasibility, it is expected to promote commercialization of super-chilling containers and packaging system developed in the future and secure customer reliability.

  • In order to improve the electrochemical performances of Li-ion batteries when spinel $LiMn_2O_4$ is employed as a cathode active material, binary conductive agents were prepared using two different particle-sized carbons like Super P Black and $Vulcan^{(R)}$ XC-72R. The electrochemical performances of the $LiMn_2O_4$ cell system using binary conductive agents were evaluated in terms of specific charge and discharge capacities and cycle life. The cell with binary conductive agent in the 3:7 weight ratios of Super P Black and $Vulcan^{(R)}$ XC-72R showed better electrochemical performances due to the proper combination of ionic diffusion rate and electric contact.

Biosynthesis of Polymyxins B, E, and P Using Genetically Engineered Polymyxin Synthetases in the Surrogate Host Bacillus subtilis

  • The development of diverse polymyxin derivatives is needed to solve the toxicity and resistance problems of polymyxins. However, no platform has generated polymyxin derivatives by genetically engineering a polymyxin synthetase, which is a nonribosomal peptide synthetase. In this study, we present a two-step approach for the construction of engineered polymyxin synthetases by substituting the adenylation (A) domains of polymyxin A synthetase, which is encoded by the pmxABCDE gene cluster of Paenibacillus polymyxa E681. First, the seventh L-threonine-specific A-domain region in pmxA was substituted with the L-leucine-specific A-domain region obtained from P. polymyxa ATCC21830 to make polymyxin E synthetase, and then the sixth D-leucine-specific A-domain region (A6-D-Leu-domain) was substituted with the D-phenylalanine-specific A-domain region (A6-D-Phe-domain) obtained from P. polymyxa F4 to make polymyxin B synthetase. This step was performed in Escherichia coli on a pmxA-containing fosmid, using the lambda Red recombination system and the sacB gene as a counter-selectable marker. Next, the modified pmxA gene was fused to pmxBCDE on the chromosome of Bacillus subtilis BSK4dA, and the resulting recombinant strains BSK4-PB and BSK4-PE were confirmed to produce polymyxins B and E, respectively. We also succeeded in constructing the B. subtilis BSK4-PP strain, which produces polymyxin P, by singly substituting the A6-D-Leu-domain with the A6-D-Phe-domain. This is the first report in which polymyxin derivatives were generated by genetically engineering polymyxin synthetases. The two recombinant B. subtilis strains will be useful for improving the commercial production of polymyxins B and E, and they will facilitate the generation of novel polymyxin derivatives.

  • The micro emulsion method has been successfully used for preparing perovskite LaCoO3 with uniform, fine-shaped nanoparticles showing high activity as electro catalysts in oxygen reduction reactions (ORRs). They are, therefore, promising candidates for the air-cathode in metal-air rechargeable batteries. Since the activity of a catalyst is highly dependent on its specific surface area, nanoparticles of the perovskite catalyst are desirable for catalyzing both oxygen reduction and evolution reactions. Herein, LaCoO3 powder was also prepared by sol-gel method for comparison, with a broad particle distribution and high agglomeration. The electro catalytic properties of LaCoO3 and LaCoO3-carbon Super P mixture layers toward the ORR were studied comparatively using the rotating disk electrode technique in 0.1 M KOH electrolyte to elucidate the effect of carbon Super P. Koutecky-Levich theory was applied to acquire the overall electron transfer number (n) during the ORR, calculated to be ~3.74 for the LaCoO3-Super P mixture, quite close to the theoretical value (4.0), and ~2.7 for carbon-free LaCoO3. A synergistic effect toward the ORR is observed when carbon is present in the LaCoO3 layer. Carbon is assumed to be more than an additive, enhancing the electronic conductivity of the oxide catalyst. It is suggested that ORRs, catalyzed by the LaCoO3-Super P mixture, are dominated by a 2+2-electron transfer pathway to form the final, hydroxyl ion product.

  • The purpose of this study was to observe the inital acidity of zinc phosphate cements(Confit and Super Cem), poly carboxylate cement(Unident and Poly F), zinc oxide engenol cement(Stailine Super EBA) and g1ass ionomer cement(Fuji ionomer) Each cement was manuplated in accordance with each manufacturer's direction. All cements were mixed at the room temperature of $23^{\circ}{\pm}$, $5^{\circ}C$, and the electrode of pH meter(Ionanalyzer) was inserted in the mixed cement, and the acidity of cement were measured for 20 minutes from begining of cement mixing at $23^{\circ}C$ and $37^{\circ}C$ Results were as follows 1. The acidity of all cements ranges from pH 3, 5 to 4, 5 at 2 minutes after the start of mixing 2. The value of pH at $23^{\circ}C$ was higher than the value of pH at $37^{\circ}C$ in all cements. 3. As the time elapsed, the pH in all cements rose. The 20 minutes after the start of mixing the range of acidity was from pH 5 to pH 6 except Poly F. 4. In polycarboxylate cement, the different value of acidity at $23^{\circ}C$ and $37^{\circ}C$ was greatest. 5. The curve pattern of acidity in Unident was similar to that in Poly F cement The pH value of Unident was higher then that of Poly F, and value of pH in the curve pattern of acidity in Confit were similar to those in Super cement.

The Flosing Properties of the Super Flowing Concrete using Class F Fly Ash (F급 플라이애쉬를 사용한 초유동 콘크리트의 유동특성)

  • This study dealt with the flowing properties of the super flowing concrete for class F fly ash producted thermal power plant and the application for concrete industry. For this purpose, fly ash is analyzed for confined water ratio($\beta_p$)and the super flowing concrete is tested the flowing properties including flowing velocity, funneling time, height difference of box test and compressive strength. As the result, in order to satisfy the flowing properties of the super flowing concrete using class F fly ash, the optimum mixing conditions are determined water-bindrer ratio 37$\pm$2%, volume ratio of fine aggregates(Sr) 47$\pm$2% and coarse aggregates(Gv) 51$\pm$1%.

  • This paper was developed and described core-process to implement low on resistance which was the most important characteristics of SJ (super junction) MOSFET. Firstly, using process-simulation, SJ MOSFET optimal structure was set and developed its process flow chart by repeated simulation. Following process flow, gate level process was performed. And source and drain level process was similar to genral planar MOSFET, so the process was the same as the general planar MOSFET. And then to develop deep trench process which was main process of the whole process, after finishing photo mask process, we developed deep trench process. We expected that developed process was necessary to develop SJ MOSFET for automobile semiconductor.

  • In Super Junction MOSFET, Charge Balance is the most important issue of the trench filling Super Junction fabrication process. In order to achieve the best electrical characteristics, the N type and P type drift regions must be fully depleted when the drain bias approaches the breakdown voltage, called Charge Balance Condition. In this paper, two methods from the fabrication process were used at the Charge Balance condition: Trench angle decreasing process and Bottom implantation process. A lower on-resistance could be achieved using a lower trench angle. And a higher breakdown voltage could be achieved using the bottom implantation process. The electrical characteristics of manufactured discrete device chips are compared with those of the devices which are designed of TCAD simulation.

The effect of surface treatment conditioning on shear bond strength between zirconia and dental resin cements (지르코니아 세라믹의 표면처리에 따른 치과용 접착제의 전단결합강도)

  • Purpose: The purpose of this study was to evaluate the effect of surface treatment on the shear bond strength of zirconia ceramic to 3 resin cements. Materials and methods: A total of 143 disk-shaped Zirconia blocks (HASS Co., Gangneung, Korea) were randomly divided into three treatment groups: (1) only 50 ${\mu}m$ $Al_2O_3$ sandblasting, (2) 50 ${\mu}m$ $Al_2O_3$ sandblast and zircona liner, (3) 50 ${\mu}m$ $Al_2O_3$ sandblasting and Rocatec (3M ESPE, Seefeld, Germany). Bistite II (Tokuyama Dental Co., Japan), Panavia F (Kuraray Medical, Japan), and Superbond C&B (Sun Medical, Japan) were used to cement onto the zirconia. After 24h of storage in distilled water, shear bond strength was evaluated. High value group was re-tested after thermocycling at 5,000 cycles(5-$55^{\circ}C$). Shear bond strength data were analyzed with one-way ANOVA, two-way ANOVA test and Post Hoc Test (${\alpha}$=.05). Shear bond strength data before and after thermocycling were analyzed with Independent sample T test (${\alpha}$=.05). Results: Super-bond C&B treated with Rocatec showed the most high shear bond strength. Super-bond C&B groups resulted in significantly higher than other cement groups (P<.05). Rocatec groups resulted in significantly higher than other surface treatment groups (P<.05). Shear bond strength has increased in Panavia F treated with Zirconia liner (P<.05). After thermocycling, shear bond strength was increased in Super-bond C&B treated with Rocatec but decreased in other groups (P<.05). Conclusion: Super-bond C&B cement resulted the highest shear bond strength and Rocatec system enhanced the shear bond strength. After thermocycling, shear bond strength has decreased in most resin cements except Super-bond C&B treated with Rocatec.

A GDD Model for Super Sweet Corn Grown under Black P. E. Film Mulch (흑색 P. E. Film 피복에서 초당옥수수의 생육기간을 표시하는 GDD모델 개발)

  • GDD models of corn were developed in bare soil, while sweet and super sweet corns are grown under black polyethylene (P. E.) film mulch in Korea. To develop a suitable GDD model under black P. E. film mulch, a super sweet com hybrid "Cambella-90" was planted from 1 April to 30 June in 2003 at the 10-day intervals under black P. E. film mulch and in bare soil. In bare soil the best GDD model was $GDD\;=\;{\sum}[H"+L')/2\;-\;10^{\circ}C]$, where H" was daily maximum temperature but is was substituted for $30^{\circ}C$ - (daily maximum temperature - $30^{\circ}C$) when higher than $30^{\circ}C$ and L' was daily minimum temperature, but it was substituted for $10^{\circ}C$ when lower than $10^{\circ}C$. The same GDD model could be adapted for com grown under black P. E. film mulch, but base temperature was substituted for $9^{\circ}C$. To determine planting date for the scheduled harvests, accumulated GDDs were calculated using 30-year average temperature data during the growing season. Under black P. E. film mulch planting dates were determined by subtracting GDD of the hybrid, $970^{\circ}C$, from accumulated GDD of scheduled harvest dates.
