Comparison of antibiotic residues on beef and pork carcass in Seoul (서울지역의 도축 소, 돼지 지육에서의 잔류물질 비교 조사)

  • This study was carried out to compare the antibiotic residues in the muscles of cattle and pigs from slaughter-houses in Seoul from 2000 to 2002 by EEC-4-plate, Charm II and HPLC methods. The results were summarized as follows; 1. Residual materials were detected from 41 samples(0.6%) by EEC-4-plate method from random sampling and 38 samples(12%) by Charm II method from directed sampling. 2. Violation rates were 0.3% by monitoring and 4.7% by surveillance program. 3. The 35 samples were classified as tetracyclines 30(86%), sulfonamides 4(11%), ${\beta}$-lactams 1(3%) and two samples simultaneously determined oxyteracycline plus sulfadimetoxine, and sulfamerazine plus sulfadimetoxine. 4. The highest residual concentration(ppm) of chlortetracycline, oxytetracyline, sulfamethazine, sufadimetoxine and penicillin were 0.5, 12.0, 6.4, 2.6 and 0.44, respectively.

A study on decomposition of sulfonamide during meat processing (육가공시 설폰아미드의 분해에 관한 연구)

  • 돈육의 가열과 냉동저장시 잔류된 설폰아미드가 증가 또는 감소되는 정도를 조사하였다. 설파메라진, 설파메타진, 설파모노메톡신, 설파디메톡신, 설파퀴녹사린을 돈육에 100ng/g씩 주사하고 30분간 $60^{\circ}C$로 가열하였을 때 설파메타진은 40%증가하였으나, $60-120^{\circ}C$로 가열한 다른 설폰아미드는 12-22% 감소하였다. 4주간 $-20{^{\circ}C}$이하로 냉동하였을 때, 설파메라진과 설파퀴녹사린은 각각 12%, 19% 감소하였으나 다른 설폰아미드는 변화가 없었다. 설폰아미드의 검출한계는 고상분산을 이용한 정제시 2.6-27ng/g이있으며, 액상분배를 이용한 정제시 25-36ng/g이었다. 본 실험의 결과 돈육중의 설폰아미드는 가열과 냉동저장에 따라 안정성이 변화될 수 있으며, 잔류물 정제방법으로는 고상분산과 액상분배방법을 이용할 수 있을 것으로 생각된다.

Monitoring of Selected Veterinary Antibiotics in Animal Carcass Disposal Site and Adjacent Agricultural Soil (가축매몰지 및 인근 농경지의 축산용 잔류 항생제 모니터링)

  • In Korea, over 4,700 animal carcass disposal sites were installed until 2011 due to the outbreak of foot and mouth disease. Due to the putrefaction of buried animals, the leachate containing veterinary antibiotics may release into surrounding environments. Antibiotic residues in the environment cause the formation of antibiotic resistance bacteria threatening human and ecosystem health. This study reports the concentrations of five antibiotics, including tetracycline (TC), chlortetracycline (CTC), oxytetracycline (OTC), sulfamethazine (SMZ), and sulfamethoxazole (SMX), in soils from animal carcass disposal site and adjacent agricultural field. Concentrations of antibiotics at animal carcass disposal sites (TC: $144.26-350.73{\mu}g/kg$, SMZ: $17.72-44.94{\mu}g/kg$) were higher than those at agricultural field (TC: $134.16-320.73{\mu}g/kg$, SMZ: $6.48-8.85{\mu}g/kg$) whereas the concentrations of CTC, OTC, and SMX were below detection limit in both sites. Results showed that the antibiotics in animal carcass site might leach to the soil and possibly contaminating the groundwater. Future studies will focus on the transfer of antibiotics residues into food crops.

Survey on residual antibiotics for beef, pork and chicken at slaughter house in Gyeongbuk province (경북지방 도축장의 지육에 대한 잔류물질 실태조사)

  • This survey was carried out to detect the residual antibiotics in beef (n = 1,071), pork (n=7,837) and chicken (n=1,536) from slaughter houses in Gyeongbuk province by EEC-4 plate method, Charm II and HPLC during 2005. Residues of antibiotic were detected from 9 beef (0.8%) and 119pork (1.52%) by EEC-4 plate method, and total positive rates were 1.23% (128). 126 samples of the 128 positive samples by the EEC-4 plate method were detected by charm II test. 128 samples were classified as tetracyclines 110 (95.5%), ${\beta}$-lactam 2 (1.6%), sulfonamide 22 (17.2%), quinolone 1 (0.8%). The highest residual concentration of oxytetracycline, tetracycline, chlor-tetracycline, penicillin, sulfamethazine, sulfadimethoxine, sulfaquinoxaline, sulfamerazine, sulfamonomethoxine and enrofloxacin were 7.57, 0.27, 0.40, 0.24, 14.24, 4.33, 8.59, 0.12, 0.09 and 1.98 ppm, respectively and 49 samples were exceeded legal admitted levels.

Investigation of residual levels and pertaining detection wavelength of 5 sulfonamides in the mouse muscle (마우스 근육내 설파제 잔류량조사 및 동시분석파장 연구)

  • This survey was carried out to determine 5 residual sulfonamides(sulfamerazine : SMR, sulfamethazine : SMT, sulfamonomethoxine : SMM:, sulfadimethoxine : SDM, sulfaquinoxaline : SQX) in muscle of mouse. For this investigation, pertaining detection wavelength, residual levels and residual times in muscle of mouse were summarized as follows ; 1. Pertaining detection wavelength of 5 residual sulfonamides(SMR, SMT, SMM, SDM, SQX) was 270nm by HPLC/UV detection. 2. After 1 day put a stop to sulfonamides administration, residual levels was 1∼l.5ppm, but were not detected at day 7 3. Withdrawal time of 5 sulfonamides were about 7 days suspectly.

Comparision of TTC-II and disk assay method for the detection of antibiotic residues in milk (우유내 잔류물질 검사방법인 TTC-II법 및 Disk assay법의 비교시험)

  • This test process on screening method for the detection of residual antibiotics in milk is simple, economic, sensitive to residual antibiotics and was given approval international organs. Thus, this study was carried out the comparison of Disk assay method and TTC-II method for sensitivity and minimum detectable range of antibiotics in raw milk. The results of this study was summarized as follows ; 1. The number of samples requested for treatment of mastitis was 198 samples. Comparison or analytical results among the methods of TTC-II, disk assay and Delve sp was that TTC-II 37 samples(18.6% ), Disk assay 125samples(63.1%), Delve SP 130 samples(65.7% ) reacted positively. Conformity rate of Delve SP and Disk assay was 70%. 2. Detectable limits of disk assay method in some antibiotics were more sensitive than those of official method(0.05-0.0025ppm in the $\beta$-lactams, 1ppm in two aminoglycoside, 0.2 ppm in one tetracycline, similar in one macrolide) 3. For sensitivity of residual sulfonamides TTC-II was much more sensitive than disk assay. Detectable limits of sulfamethazine and sulfadimethoxine were 30 to 50ppm levels. 4. The best medium preservation period is 1-2 days. 5. Concentration of brome cresol purple related to resistance for B stearothermophilus culture was 24ppm/ml. These results show that disk assay method for screening detection of antibiotics residuces in milk is worthy of use.

Study on Development of Enzyme-Linked Immunosorbent Assay for the Screening of Chloramphenicol Residues (잔류 Chloramphenicol 검사용 효소 면역측정법의 개발에 관한 연구)

  • The monoclonal antibody to chloramphenicol(CAP) was produced to develop an enzyme-linked immunosorbent assay(ELISA) for residual CAP. An immunogen(CAP-BSA) was prepared by immunogen, antibody titer was measured by indirect ELISA. Spleen cells form the immunized mouse were fused with SP2/OAg14 myeloma cells. Among hybridomas selected in HAT media, 6 clones shown high antibody titer to CAP were subjected to cloning by limit dilution, and all of the monoclonal antibodies(MCA1, 2, 3, 4, 5, 7 and 9) produced by each clone were identified as IgG1 by ELISA isotyping analysis. Competitive ELISA condition was established by using the purified monoclonal antibody MCA1 as primary antibody and CAP-HSA conjugate as coating antigen. Standard curve of CAP(n=28) showed that the lowest detection limit of CAP is 20ng/ml level. The cross-reactivities of the 6 monoclonal antibodies showed that CAP sodium succinate. CAP base, P-nitrophenol, and p-nitrobenzyl alcohol were 89∼178, 0.050∼2.237, 0.056∼0.794 and 0.013∼7.939%, respectively. No cross-reactivities were observed with phenylalanine, tyrosine, glutamine, thiamphenicol, neomycin, streptomycin, gentamicin, sulfamethazine, sulfathiazole, chlortetracycline and p-aminobenzoic acid.

Simultaneous Determination of Tetracyclines and Sulfonamides Residues in Pork and Flatfish Using Matrix Solid Phase Dispersion (MSPD) Extraction and HPLC (MSPD와 HPLC를 이용한 돼지고기 및 광어 중의 테트라싸이클린계 항생제와 sulfonamide의 다성분 잔류분석법 개발)

  • This study was conducted to develop more convenient simultaneous determination method by matrix solid phase dispersion (MSPD) and HPLC for sulfonamides such as sulfamerazine (SMR), sulfamethazine (SMT), sulfamonomethoxine (SMM), sulfadimethoxine (SDM), sulfaquinoxaline (SQX), and tetracyclines including oxytetracycline (OTC), tetracycline (TC) chlortetracycline (CTC) in prok and flatfish. The limits of detection were 0.047 $mg{\cdot}kg^{-1}$ for OTC, TC, SMR, SMT and SMM, and 0.033 $mg{\cdot}kg^{-1}$ for CTC, SDM, and SQX, respectively. So it is sufficiently possible to detect the eight tetracyclines and sulfonamides under their MRLs ($0.1{\sim}0.2mg{\cdot}kg^{-1}$). The average percentage recoveries of sulfonamides and tetracyclines from pig muscle and flatfish spiked standard solution were approximately $80.25{\sim}101.25%$ and $85.77{\sim}121.42%$, respectively. Therefore this method was efficient for simultaneous analysis of eight tetracyclines and sulfonamides.

Seasonal Monitoring of Residual Veterinary Antibiotics in Agricultural Soil, Surface Water and Sediment Adjacent to a Poultry Manure Composting Facility (계분 퇴비화 시설 인근 농경지 토양, 지표수 및 저질토의 계절별 잔류 항생물질 모니터링)

  • Concentration of antibiotics including a tetracycline group (TCs) of tetracycline (TC), chlortetracycline (CTC), and oxytetracycline (OTC), a sulfonamide group (SAs) of sulfamethoxazole (SMX), sulfathiazole (STZ), and sulfamethazine (SMT), an ionophore group (IPs) of lasalocid (LSL), monensin (MNS), and salinomycin (SLM), and a macrolide group (MLs) of tylosin (TYL) was determined from samples collected from the agricultural soil, stream water, and sediment. For the agricultural soil samples, the concentration of TCs had the highest value among all tested antibiotic's groups due to its high accumulation rate on the surface soils. The lower concentrations of SAs in the agricultural soils may be resulted from its lower usage and lower distribution coefficient (Kd) compared to TCs. The concentration of TCs in stream water was significantly increased through June to September. It would be likely due to soil loss during an intensive rainfall event and a reduction of water level after the monsoon season. A significant amount of TCs in the sediment was also detected due to its accumulation from runoff, which occurred by complexation of divalent cations, ion exchange, and hydrogen bonding among humic acid molecules. To ensure environmental or human safety, continuous monitoring of antibiotics residues in surrounding ecosystems and systematic approach to the occurrence mechanism of antibiotic resistant bacteria are required.
