Antibacterial and Radical Scavenging Activities of 1-C-(p-Hydroxyphenyl)-Glycerol from Trichosanthes kirilowii

  • 1-C-(p-Hydroxyphenyl)-glycerol (1) was isolated and purified by column chromatography and recrystallization from the rhizome of Trichosanthes kirilowii, firstly in this species. Isolated compound showed inhibitory effects on the growth of five bacteria (Bacillus cereus, Escherichia coli, Streptococcus faecalis, etc) completely at the concentration higher than 10 ppm as well as delayed the growth of three bacteria (Bacillus substilis, Staphylococcus aureus, Pseudomonas aeroginosa) at the concentration of 25 ppm for 60 h. Moreover, this compound showed potent antioxidant activity against DPPH radical ($IC_{50}$ = 56.0 ppm)

Diurnal Changes of Chlorophyll Fluorescence and Antioxidative Enzyme Activity of the Leaves from Four Subtropical Plants (아열대성 식물 4종의 엽록소형광과 항산화효소 활성의 일주기적 변화)

  • The diurnal changes of chlorophyll fluorescence and antioxidative enzyme activity were investigated in the leaves from four subtropical plant species (Crinum asiaticum var. japonicum Bak., Osmanthus insularis Koidz., Asplenium antiquum Makino and Chloranthus glaber Makino) under the natural habitats in summer and winter. The intensity of chlorophyll fluorescence was lower in O-, I-, J-, P-steps of O-J-I-P transient in winter than summer, and prominent diurnal change was not found in the fluorescence intensity of four subtropical plant species in winter. The activity and isoenzyme pattern of SOD and catalase did irregularly change seasonally and diurnally in four subtropical plant species. In contrast, the peroxidase activity and isoenzyme pattern was different depending on plant species and growth seasons; The activity increased slightly more in winter than in summer in four subtropical plant species, and several isoenzymes appeared in the leaves from C. asiaticum var japonicum, O. insularis and A. antiquum in winter.

  • This study was carried out in the Tropical Plant Resources Research Center of Korea National Arboretum to determine the optimum edaphic environment for the growth of tropical and subtropical plants. The physicochemical properties of artificial substrates and the growth characteristics of tropical and subtropical plants were investigated. Subtropical plants exhibited a high growth rate when cultivated in a substrate of Dry Zone that had physical properties similar to those of arid native soil. Mediterranean plants showed a low growth rate when grown in a substrate of Subtropical Zone that required changes in acidity. The substrate of Tropical Zone had high organic matter and mineral contents and therefore had good physical properties:this substrate has a good environment for the stimulation of the growth of tropical plants. Our results indicate that the chemical properties such as pH and mineral contents of most artificial substrates need to be more urgently improved than their physical properties in order to ensure better growth of tropical and subtropical plants. Initial management strategies for the construction of new tropical greenhouses were formulated, and data from monitoring studies will be continuously gathered and incorporated in the manual to keep it updated.

Intramuscular Administration of Zinc Metallothionein to Preslaughter Stressed Pigs Improves Anti-oxidative Status and Pork Quality

  • This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.

Raising Seedling at Hallasan Area of Sub-Alpine Improved Fruiting Rate of Squash (Cucurbita maxima) (단호박 착과율 향상을 위한 한라산 중산간지 육묘효과)

  • This study was carried out to determine the effect of raising seedling at Hallasan area of sub-alpine (altitude of 600m above sea level) to improve fruiting.ate of Squash (Cucurbita maxima) in retarding culture. 'Ebis' cultivar was seeded in plug tray of 32 cells and the seedlings were grown for 25 days. They were transplanted on August 26, 2004, following L-stem training method under rain-shielding condition. Seedling height, number of nodes and leaf area were higher in lowland than in sub-alpine area. T/R ratio of seedling in sub-alpine was much lower as compared with that in the lowland. The first fruiting was on the 19th node in sub-alpine area, and on the 26th node in the lowland area(control). The succeeding fruiting nodes were lower by 3 to 5 node than those of control. Fruiting rate of second flower was improved by 17.2% compared with the 1.4% in control. The marketable yield was increased by 27% by raising seedling in sub-alpine area (4,460 kg/10a). This also brought out 20% labour saving effect. The environmental condition for raising seedling in the sub-alpine area of Hallasan was effective for the improvement of Squash (Cucurbita maxima) fruiting rate compared with lowland area.

Use of In vitro Gas Production Technique to Investigate Interactions between Rice Straw, Wheat Straw, Maize Stover and Alfalfa or Clover

  • Measurement of gas produced during in vitro fermentation was used to investigate the fermentation characteristics and interactions of rice straw, wheat straw or maize stover mixed with alfalfa or clover at proportions of 100:0, 75:25, 50:50, 25:75 and 0:100, respectively. Cumulative gas production was recorded at 2, 4, 8, 12, 16, 24 and 48 h of incubation, and the Gompertz function was used to describe the kinetics of gas production. In vitro dry matter and organic matter disappearances (IVDMD and IVOMD) were determined after 48 h incubation. The rate of gas production of clover was higher (p<0.05) than that of rice straw, wheat straw, maize stover and alfalfa when straws and hays were incubated separately. Increasing the proportion of alfalfa in the straw-alfalfa mixtures increased (p<0.05) the rates, but not the maximum volume of gas production. However, both rate and the maximum volume of gas production were increased (p<0.01) as the proportions of clover increased in the straw-clover mixtures. The cumulative gas production at 48 h, IVDMD and IVOMD showed no consistent interaction effects between different mixtures of cereal straws and hays. The extent of interactive effects was affected by the types of cereal straw, legume hay and their proportions in the mixture. The appropriate combination for the mixture of rice straw or maize stover with leguminous hays was 75:25 and 25:75, respectively. The better combination occurred at a proportion of 50:50 for the mixture of wheat straw and alfalfa. We conclude that the suitable proportion of low-quality straw and high quality legume hay combination should be considered in the ration formulation system of ruminants according to the extent of positive interactive effects.

Effect of Apex Removal at Different Leaf Ages on the Growth and Yield of Broccoli. (엽령이 다른시기의 생장점 제거가 브로콜리의 생육 및 수량에 미치는 영향)

  • This experiment was carried out to investigate the effect of apex removal at different leaf ages on the growth and yield of broccoli in pinching cultivation. Apex was pinched when 2, 3 or 4 true leaves were spread during nursery bed, and non-pinching was conventional method. The plant height was not different among treatments but the fresh weight was decreased as the pinching days were delayed. The numbers of leaves and lateral shoots were increased compared to those of non-pinching by pinching. Days to central budding were shortended by 3 days when pinched at 2 true leaves spreading stage than those of non-pinching. Central (1st) head weight was heaviest when pinched at the time of 2 true leaves spreading (157g). Total head weight was heaviest when pinched at the time of 2 true leaves spreading (378g), and lowest at 4 true leaves spreading stage (196g). Marketable yields were increased by 48% when pinched at the time of 2 true leaves spreading (1,754kg) as compared with those of non-pinching (1,184kg) and lowest when pinched at the time of 4 true leaves spreading (820kg). Also, first harvest was shortended by 6 days by pinching at 2 true leaves spreading stage.

Effect of Planting Distance on Quality and Productivity in Staking Cultivation of Squash(Cucurbita maxima) under Rain-shielding Condition (단호박 입체재배시 재식거리가 품질 및 생산성에 미치는 영향)

  • This study was conducted to determine the optimum planting distance under rain-shielding condition for improving quality and productivity of squash by staking cultivation. The squashes were grown by 3 planting distances, such as 120${\times}$30, 120${\times}$40 and 120${\times}$50 cm during 3 months under non heat condition. The vine length, fresh weight and leaf area were reduced as planting distance was decreased until main branch was pinching. The first female flowering day and first fruit setting nodes were not different among treatments. The first fruit length and second fruit weight were increased in 120${\times}$50 cm planting distance, but the soluble solid contents were not different among treatments. In 120${\times}$30 cm plant distance, total yield was highest (6,510 kg/10a), but marketable yie이 was lowest. When the planting distance was between 120${\times}$40 cm and 120${\times}$50 cm, the marketable rate was similar (over 98%), but marketable yield was highest in 120${\times}$40 cm planting distance (5,012 kg/10a). The results of this study indicated that the optimum planting distance of staking cultivation in squash seemed to be 120${\times}$40cm.

Effects of different culture systems on the culture of prepuberal buffalo (Bubalus bubalis) spermatogonial stem cell-like cells in vitro

  • Currently, the systems for culturing buffalo spermatogonial stem cells (SSCs) in vitro are varied, and their effects are still inconclusive. In this study, we compared the effects of culture systems with undefined (foetal bovine serum) and defined (KnockOut Serum Replacement) materials on the in vitro culture of buffalo SSC-like cells. Significantly more DDX4- and UCHL1-positive cells (cultured for 2 days at passage 2) were observed in the defined materials culture system than in the undefined materials system (p < 0.01), and these cells were maintained for a longer period than those in the culture system with undefined materials (10 days vs. 6 days). Furthermore, NANOS2 (p < 0.05), DDX4 (p < 0.01) and UCHL1 (p < 0.05) were expressed at significantly higher levels in the culture system with defined materials than in that with undefined materials. Induction with retinoic acid was used to verify that the cultured cells maintained SSC characteristics, revealing an SCP3+ subset in the cells cultured in the defined materials system. The expression levels of Stra8 (p < 0.05) and Rec8 (p < 0.01) were significantly increased, and the expression levels of ZBTB16 (p < 0.01) and DDX4 (p < 0.05) were significantly decreased. These findings provided a clearer research platform for exploring the mechanism of buffalo SSCs in vitro.
