Isolation and Characterization of a Theta Glutathione S-transferase Gene from Panax ginseng Meyer

  • Plants have versatile detoxification systems to encounter the phytotoxicity of the wide range of natural and synthetic compounds present in the environment. Glutathione S-transferase (GST) is an enzyme that detoxifies natural and exogenous toxic compounds by conjugation with glutathione (GSH). Recently, several roles of GST giving stress tolerance in plants have demonstrated, but little is known about the role of ginseng GSTs. Therefore, this work aimed to provide further information on the GST gene present in Panax ginseng genome as well as its expression and function. A GST cDNA (PgGST) was isolated from P. ginseng cDNA library, and it showed the amino acid sequence similarity with theta type of GSTs. PgGST in ginseng plant was induced by exposure to metals, plant hormone, heavy metals, and high light irradiance. To improve the resistance against environmental stresses, full-length cDNA of PgGST was introduced into Nicotiana tabacum. Overexpression of PgGST led to twofold increase in GST-specific activity compared to the non-transgenic plants, and the GST overexpressed plant showed resistance against herbicide phosphinothricin. The results suggested that the PgGST isolated from ginseng might have a role in the protection mechanism against toxic materials such as heavy metals and herbicides.

전기침이 급만성 결박스트레스로 유도된 스트레스호르몬 반응에 미치는 영향 (Effects of Electroacupuncture on Plasma Stress Hormone Responses to Acute and Chronic Immobilization Stress)

  • 목적 : 침은 다양한 질환의 치료에 보편적으로 쓰이고 있으며 고혈압, 당뇨, 정신질환 등을 포함한 스트레스성 질환에도 활용되고 있다. 결박 스트레스는 스트레스 호르몬(코르티코스테론, 멜라토닌)의 혈장 농도를 크게 증가시키는 간단하고 효과적인 스트레스 요인이다. 본 연구는 결박 스트레스를 시행한 백서의 스트레스 호르몬의 혈중 농도에 대한 전기침의 효과를 조사하였다. 방법 : 결박 그룹은 2시간의 결박 스트레스를 받았으며 결박 스트레스 및 고주파수 전침그룹과 결박 스트레스 및 저주파수 전침그룹은 결박 스트레스와 고주파수 전침, 또는 결박 스트레스와 저주파수 전침을 동시에 각각 시행하였다. 급성 스트레스 유발 시에는 결박 스트레스를 1차례, 만성 스트레스 유발 시에는 7차례 시행하였다. 전기침 자극에는 우측 족삼리($ST_{36}$)를 사용하였다. 결박 스트레스 및 전기침으로 유도된 코르티코스테론과 멜라토닌의 농도를 측정하기 위해서 결박 스트레스 및 전기침 자극이 시작된 30분, 60분, 90분, 120분 후에 백서를 단두하여 혈액 샘플을 채취하였다. 결과 : 급성 스트레스 유발 시에는 고주파수 전침그룹의 코르티코스테론 혈장 농도가 증가하였고 멜라토닌 농도의 시간적 패턴을 변화시켰으나 저주파수 전침그룹에서는 유의한 변화가 없었다. 만성 스트레스 유발 시에는 고주파수 전침그룹의 혈장 코르티코스테론과 멜라토닌 농도가 유의하게 감소되었으나 저주파수 전침그룹에서는 변화가 없었다. 결론 : 이러한 결과는 전침이 결박 스트레스로 유도된 스트레스 호르몬의 혈장 농도 및 시간적 분비패턴을 변화시키는 효과가 있으나 스트레스 호르몬 반응을 변화시키는 데 있어서 주파수에 따른 유의한 차이가 있다는 것을 의미한다.

Antioxidant Activities and Melanogenesis Inhibitory Effects of Terminalia chebula in B16/F10 Melanoma Cells

  • To examine the potential of Terminalia chebula as a whitening agent, we measured antioxidant activity using DPPH$\cdot$, ABTS${\cdot}^+$ assays and ferric-reducing antioxidant power (FRAP) assays, and depigmenting activity using B16F10 melanoma cells. The intracellular reactive oxygen species (ROS) level was monitored by $H_2DCFDA$ fluorescence labeling, and melanin contents in B16F10 melanoma cells by 960 $J/m^2$ dose of UVA-induced oxidative stress. The radical-scavenging activities of T. chebula extract (TCE) were measured in terms of $EC_{50}$ values using DPPH$\cdot$, ABTS${\cdot}^+$ assays and FRAP value were 280.0 ${\mu}g/mL$, 42.2 ${\mu}g/mL$ and 113.1 ${\mu}mol$ $FeSO_4{\cdot}7H_2O/g$, respectively. We found that ROS and melanin concentrations were reduced by TCE treatments of 25 ${\mu}g/mL$ under UVA-induced oxidative stress. Tyrosinase activity and melanin contents in $\alpha$-melanocyte stimulating hormone (MSH)-induced melanoma cells both decreased dose-dependently in the treatment groups. TCE similarly reduced melanogenesis in B16F10 melanoma cells stimulated by $\alpha$-MSH as compared to arbutin as a positive control. T. chebula may prove to be a useful therapeutic agent for hyperpigmentation and an effective component in skin whitening and.or lightening cosmetics.

Effects of crocin and metformin on methylglyoxal-induced reproductive system dysfunction in diabetic male mice

  • Objective: This study investigated the effect of crocin in methylglyoxal (MGO)-induced diabetic male mice. Methods: Seventy 1-month-old male NMRI mice weighing 20-25 g were divided into seven groups (n=10): sham, MGO (600 mg/kg/day), MGO+crocin (15, 30, and 60 mg/kg/day), MGO+metformin (150 mg/kg/day), and crocin (60 mg/kg/day). MGO was administered orally for 30 days. Starting on day 14, after confirming hyperglycemia, metformin and crocin were administered orally. On day 31, plasma and tissue samples were prepared for experimental assessments. Results: Blood glucose and insulin levels in the MGO group were higher than those in the sham group (p<0.001), and decreased in response to metformin (p<0.001) and crocin treatment (not at all doses). Testis width and volume decreased in the MGO mice and improved in the crocin-treated mice (p<0.05), but not in the metformin group. Superoxide dismutase levels decreased in diabetic mice (p<0.05) and malondialdehyde levels increased (p<0.001). Crocin and metformin improved malondialdehyde and superoxide dismutase. Testosterone (p<0.001) and sperm count (p<0.05) decreased in the diabetic mice, and treatment with metformin and crocin recovered these variables. Luteinizing hormone levels increased in diabetic mice (p<0.001) and crocin treatment (but not metformin) attenuated this increase. Seminiferous diameter and height decreased in the diabetic mice and increased in the treatment groups. Vacuoles and ruptures were seen in diabetic testicular tissue, and crocin improved testicular morphology (p<0.01). Conclusion: MGO increased oxidative stress, reduced sex hormones, and induced histological problems in male reproductive organs. Crocin and metformin improved the reproductive damage caused by MGO-induced diabetes.

Relationship between saliva and blood cortisol in handled cows

  • Objective: The objective of the study was to determine the relationship between plasma and salivary cortisol concentrations in beef cattle that were subjected to handling prior to sampling. Methods: Twenty-one Nguni cows of three age categories; 5 to 7 yr (n = 7), 8 to 10 yr (n = 6), and 11 to 13 yr (n = 8) were handled for five consecutive weeks. In the pen, a human avoidance test was performed and cattle responses to restraint in the chute and crush were observed. In addition, rectal temperature readings were taken and, faecal samples were collected and analysed for glucocorticoid metabolites. Through the handling and restraint process, excretory and vocalisation behaviour, as a sign of stress were observed and recorded. Thereafter, six cows were randomly selected and subjected to an adrenocorticotropic hormone (ACTH) challenge. Blood and saliva samples were extracted to determine cortisol concentrations. Results: Repeated handling affected (p<0.05) faecal glucocorticoid metabolites, rectal temperatures, avoidance distance, crush scores as well as urination and defaecation behaviour. Acclimation to handling was variable based on each respective parameter. Saliva cortisol concentrations increased and decreased significantly (p<0.001). A peak value of $136.78{\pm}15.869nmol/L$ was observed 30min after administration of ACTH, from a baseline value of $8.75{\pm}15.869nmol/L$. Plasma cortisol concentrations did not differ (p>0.05) across the time of sampling. A low and insignificant correlation (r = 0.0131, p>0.05) between plasma and saliva cortisol was therefore observed. Conclusion: We conclude that if beef cows are subjected to handling prior to sampling, a weak relationship exists between plasma and salivary cortisol levels.

The changes of stresses and ecdysteroid biosynthesis gene expression levels in Kynurenine 3-monooxygenase mutant Bombyx mori

  • Silkworms have long been bred with human history to produce silk. It has been with humans for longer than other industrial insects, and the silkworm variety has been continuously improved. Silkworms have been developed into the optimal form for producing high quality silk and pupae. Recently, the production of transgenic silkworms has further expanded the possibility of industrial value of silkworms. Kynurenine 3-monooxygenase (KMO), which is a flavin enzyme, is known for its involvement in ommochrome pigment synthesis. In the field of mammals, including humans, previous studies have revealed the function and role of KMO, which is an important enzyme for various immune responses and cell protection. However, in the case of insects, the function of KMO has only been studied to be involved in the formation of pigment, and accordingly, KMO is used exclusively on screening for generation of transgenic insects as a marker. In this study, using KMO-edited silkworms, it was intended to discover the novel functions and roles of KMO in silkworms by identifying changes in the expression of various genes associated with stress and growth. The changes were observed in expressions of genes regulating on stresses to survive and those on ecdysteroid hormone between wild-type (WT) silkworms and kmo mutant silkworms. The loss of KMO, in particular, decreased the expression of the shadow (sad) gene, one of the Halloween genes in the synthesis of ecdysteroid. In conclusion, these results suggest that silkworm KMO is responsible for potential functions regarding stress response and ecdysteroid synthesis.

Effects of feather processing methods on quantity of extracted corticosterone in broiler chickens

  • Corticosterone is known as a biological stress index in many species including birds. Feather corticosterone concentration (FCC) has increasingly been used as a measure for chronic stress status in broiler chickens. As sample preparation is the first step of analytical process, different techniques of feather matrix disruption need to be validated for obtaining better result in analysing corticosterone extraction. The current study was a validation of pulverizing the feather by bead beater (BB) and surgical scissors (SS) processing prior to corticosterone extraction in feather of broiler chickens. The type of feather processing prior to the hormone extraction may alter the final output. Thereby, finding a standard method according to laboratory facilities is pivotal. This study carried out to determine the effects of feather pulverization methods on the extraction amount of corticosterone in broiler chickens. Feathers were sampled from four weeks old Ross 308 broiler chickens (n = 12 birds). All broiler chickens were kept under the same environmental condition and had access to feed and water. Feather samples were assigned to one of the following processing methods 1) using a BB for pulverizing and 2) using a SS for chopping into tiny pieces. Each sample was duplicated into two wells during enzyme immunoassay (EIA) analysis to improve the accuracy of the obtained data. The results showed lower standard errors and constant output of FCC by using the BB method compared with the SS method. Overall comparison of FCC showed a significantly higher (p < 0.001) amount of the FCC in the BB compared with the SS. Overall, using the BB method is recommended over the SS method for feather processing due to the ability to homogenize a large number of samples simultaneously, ease of use and greater extraction of feather corticosterone.

Heterologous Expression of the Hot Pepper ABA 8'-Hydroxylase in Escherichia coli for Phaseic Acid Production

  • The CYP707A family genes encoding ABA 8'-hydroxylase catabolize abscisic acid (ABA), a plant stress hormone that plays an important role in stress condition, such as drought, heat, cold and salinity. Phaseic acid (PA) is a catabolic product of ABA. Recent studies have shown that PA is important for the physiological functions in plants. It is also a neuroprotective molecule that protects against ischemic brain injury in mice. To obtain enzymes for the PA production, four CaCYP707A genes (CaCYP707A1, CaCYP707A2, CaCYP707A3 and CaCYP707A4) were isolated from hot pepper. They were heterologously expressed in Escherichia coli. Among them, CaCYP707A2 showed significantly higher expression levels in both the membrane fraction and the soluble fraction. Preferred redox partners were investigated to improve the efficiency of CaCYP707A2's catalytic reaction, and NADPH-cytochrome P450 reductase (CPR) from hot pepper (CaCPR) was preferred over other redox partners (i.e., rat CPR and ferredoxin reductase/ferredoxin). The production of 8'-hydroxy ABA and PA by ABA hydroxylation activity was confirmed in CaCYP707A2 from both membrane and soluble fractions. Therefore, CaCYP707A2 is the first identified plant CYP protein that is expressed a soluble form in cytosolic fraction having stable activity. Taken together, we propose a new CYP707A protein with industrial applications for PA production without additional modifications in E. coli heterologous expression.

Application of data fusion modeling for the prediction of auxin response elements in Zea mays for food security purposes

  • Food security will be affected by climate change worldwide, particularly in the developing world, where the most important food products originate from plants. Plants are often exposed to environmental stresses that may affect their growth, development, yield, and food quality. Auxin is a hormone that plays a critical role in improving plants' tolerance of environmental conditions. Auxin controls the expression of many stress-responsive genes in plants by interacting with specific cis-regulatory elements called auxin-responsive elements (AuxREs). In this work, we performed an in silico prediction of AuxREs in promoters of five auxin-responsive genes in Zea mays. We applied a data fusion approach based on the combined use of Dempster-Shafer evidence theory and fuzzy sets. Auxin has a direct impact on cell membrane proteins. The short-term auxin response may be represented by the regulation of transmembrane gene expression. The detection of an AuxRE in the promoter of prolyl oligopeptidase (POP) in Z. mays and the 3-fold overexpression of this gene under auxin treatment for 30 min indicated the role of POP in maize auxin response. POP is regulated by auxin to perform stress adaptation. In addition, the detection of two AuxRE TGTCTC motifs in the upstream sequence of the bx1 gene suggests that bx1 can be regulated by auxin. Auxin may also be involved in the regulation of dehydration-responsive element-binding and some members of the protein kinase superfamily.

Effects of an Agro-Healing Activity Program on the Physiological Condition of Adults with Chronic Metabolic Diseases

  • This study was conducted to find out the effects of agro-healing activities and strolling on the physiological conditions of patients with chronic metabolic diseases. A total of 11 subjects participated in agro-healing activities, and their average age was 49.6±11.3. Changes in their salivary cortisol were compared and analyzed before and after participating in the agro-healing activity program, and it was found that the stress hormone decreased after participating in the program. Differences in brain wave between before and after participating in agro-healing activities and strolling were measured, and the ratio of alpha to high beta (RAHB) of subjects increased in most of the measured points after participating in agro-healing activities. In addition, the increase in the value in the temporal lobe (T3) that controls mortor skills and occipital lobe (O2) that controls visual functions indicates that the activities were effective in improving relaxation and stability. The relative low beta (RLB) power spectrum of subjects also statistically significantly decreased in the right occipital lobe (O2) that controls visual functions, the relative mid beta (RMB) and relative high beta (RHB) power spectrum decreased in the left temporal lobe (T3) and the right occipital lobe (O2). These results indicate that the activities were effective in improving relaxation under no stress and tension. The systolic and diastolic blood pressure of subjects after participating in agro-healing activities decreased from the prehypertension level to the normal level. These results indicate that horticultural activities and visual elements in healing farms, such as agro-healing activities and strolling, have positive impacts on patients with chronic metabolic diseases accompanied with diabetes or high blood pressure by increasing the brain activity and psycho-physiological conditions of participants.
