Metal pollution of aquatic ecosystems is a problem of economic and health importance. Information regarding molecular responses to metal exposure is sorely needed in order to identify potential biomarkers. To determine the effects of heavy metals on chironomids, the full-length cDNA of alcohol dehydrogenase (ADH3) from Chironomus riparius was determined through molecular cloning and rapid amplification of cDNA ends (RACE). The expression of ADH3 was analyzed under various cadmium and copper concentrations. A comparative and phylogenetic study among different orders of insects and vertebrates was carried out through analysis of sequence databases. The complete cDNA sequence of the ADH3 gene was 1134 bp in length. The sequence of C. riparius ADH3 shows a low degree of amino acid identity (around 70%) with homologous sequences in other insects. After exposure of C. riparius to various concentrations of copper, ADH3 gene expression significantly decreased within 1 hour. The ADH3 gene expression was also suppressed in C. riparius after cadmium exposure for 24 hour. However, the effect of cadmium on ADH3 gene expression was transient in C. riparius. The results show that the suppression of ADH3 gene by copper exposure could be used as a possible biomarker in aquatic environmental monitoring and imply differential toxicity to copper and cadmium in C. riparius larvae.

A biomarker is needed to monitor and manage the health of pigs from heat stress (HS). Therefore, we investigated the effects of HS on growth performance, nutrient digestibility, and blood profiles in finishing pigs. A total of 12 finishing pigs (n = 12) were raised in thermal neutral (TN; 25℃) conditions for a 3-d adaptation period. After the adaption, 6 pigs were exposed to HS at 33℃ (HS33) for 5 d. The pigs were fed the same diet based on corn and soybean meal. Chromic oxide was added to all the diets at a level of 2 g·kg-1 as an indigestible marker for the determination of the apparent total track digestibility (ATTD) of nutrients and amino acids. Blood samples were collected after the adaptation and heat treatment to verify the blood profiles. The HS33 pigs had a lower (p < 0.01) average daily feed intake (ADFI) and higher (p < 0.05) rectal temperature compared to the TN pigs. However, there was no difference in the ATTD of nutrients and amino acids. The HS33 pigs had reduced (p < 0.05) levels of serum glucose, non-esterified fatty acids (NEFA), total protein, albumin, and calcium compared to the TN pigs. However, the level of total bilirubin was increased (p < 0.05) in the HS pigs. In conclusion, HS reduced the feed intake and had an adverse effect on health. Altered blood profiles as a result of a negative energy balance are expected to be biomarkers of HS in finishing pigs.
IL-1RAcP는 일명 interleukin-1 receptor accessory protein이라 칭하며 interleukin-1 염증성 사이토카인과 interleukin-1 receptor I (IL-1RI) 결합체와 복합체를 형성하여 작용한다. IL-1RAcP는 면역반응, 스트레스 및 세포사멸과 관련이 있다. 본 연구의 목적은 참돔(Pagrus major)을 저수온(8℃, 33 psu) 및 저염분(20℃, 10 psu) 상태에 노출시킨 후, IL-1RAcP 유전자의 발현을 관찰하는 것이다. 연구결과, IL-1RAcP 유전자의 발현은 저수온(8℃, 33 psu) 및 저염분(20℃, 10 psu) 상태에서 유의적으로 증가하는 것으로 나타났다. 이 연구결과로서 IL-1RAcP 유전자는 저수온 및 저염분 등의 환경 스트레스에 대한 생체지표유전자로서 역할을 한다고 제의한다.

Objective: The objective of the present study was to investigate the comparative effects of dietary functional nutrients including glutamine (Gln), chromium picolinate (Cr picolinate), vitamin C (Vit C), betaine (Bet), and taurine (Tau) on growth performance, meat quality, immune responses, and stress biomarkers in broiler chickens raised under heat stress conditions. Methods: A total of 420 21-d-old Ross 308 male broiler chickens (initial body weight = 866±61.9 g) were randomly allotted to 1 of 7 treatment groups with 6 replicates. One group was kept under thermoneutral conditions and was fed a basal diet (PC, positive control). Other 6 groups were exposed to a cyclic heat stress condition. One of the 6 groups was fed the basal diet (NC, negative control), whereas 5 other groups were fed the basal diet supplemented with 0.5% Gln, 500 ppb Cr picolinate, 250 mg/kg Vit C, 0.2% Bet, or 1.0% Tau. The diets and water were provided ad libitum for 21 d. Results: Broiler chickens in NC group had decreased (p<0.05) growth performance and immune responses measured based on cutaneous basophil hypersensitivity (CBH), but increased (p<0.05) stress responses measured based on feather corticosterone concentrations and blood heterophil:lymphocyte than those in PC group. However, none of dietary functional nutrients had a positive effect on growth performance of broiler chickens. Dietary supplementation of 250 mg/kg Vit C improved (p<0.05) CBH responses of broiler chickens, but other functional nutrients had no such an improvement in CBH responses. All functional nutrients decreased (p<0.05) stress responses of broiler chickens. Conclusion: Functional nutrients including Gln, Cr picolinate, Vit C, Bet, and Tau at the supplemental levels used in this study decrease stress responses of broiler chickens to a relatively similar extent. However, this reduction in stress responses could not fully ameliorate decreased productive performance of broiler chickens raised under the current heat stress conditions.
Nitrosative modifications regulate cellular signal transduction and pathogenesis of inflammatory responses and neuro-degenerative diseases. Protein tyrosine nitration is a biomarker of oxidative stress and also influences protein structure and function. Recent advances in mass spectrometry have made it possible to identify modified proteins and specific modified amino acid residues. For analysis of nitrated peptides with low yields or only a subset of peptides, affinity 'tags' can be bait for 'fishing out' target analytes from complex mixtures. These tagged peptides are then extracted to a solid phase, followed by mass analysis. In this review, we focus on protein tyrosine modifications caused by nitrosative stresses and proteomic methods for selective enrichment and identification of nitrosative protein modifications.
The use of gltA gene, as a new biomarker for environmental stress biomonitoring, was investigated because of its key position as the first enzyme of the tricarboxylic acid (TCA) cycle. A recombinant bioluminescent Escherichia coli strain, EBJM2, was constructed using a plasmid carrying the citrate synthase (gltA) promoter transcribing the Photorhabdus luminescens IuxCDABE genes (gltA::luxCDABE). The responses from this strain were studied with five different classes of toxicants: DNA damage chemicals, phenolics, oxidative-stress chemicals, PAHs, and organic solvents. EBJM2 responded strongly to DNA damage chemicals, such as mitomycin C (MMC) and methyl-nitro-nitrosoguanidine (MNNG) and nalidixic acid with the strongest responses. In contrast, tests with several compounds from the other four classes of toxicants gave no significant response. Therefore, EBJM2 was found to be sensitive to DNA damage chemicals.

F$_2$-isoprostanes are derived in vivo principally from the formation of positional peroxyl radicals of arachidonic acid, endocyclization to prostaglandin G$_2$-like structures, and reduction to PGF$_2$-like compounds. F$_2$-isoprostanes have been proposed as biomarkers of lipid peroxidation, oxidative stress status, and oxidation of low density lipoprotein (LDL) in atherogenesis. Objectives: The questionnaire of occupational stress index (OSI) has been popular in the workplace, and it has been tailored for bus drivers in Taiwan. Nevertheless, its outcomes for participants are based on self-evaluations, thus validation by their physiological stress biomarker is warranted and this is the main goal of this study. Methods: A cross-sectional study of sixty-three city bus drivers and fifty-four supporting staffs for comparison was conducted. Questionnaire surveys, 24-hour urine cortisol testing, and blood draws for dehydroepiandrosterone-sulfate (DHEA-S) testing were performed. The measured concentrations of these biological measures were logarithmically transformed before the statistical analysis where various scores of stressor factors, moderators, and stress effects of each OSI domain were analyzed by applying multiple linear regression models. Results: For drivers, the elevated 24-hour urine cortisol level was associated with a worker's relationship with their supervisor and any life change events in the most recent 3 months. The DHEA-S level was higher in drivers of younger age as well as drivers with more concerns relating to their salary and bonuses. Non-drivers showed no association between any stressor or satisfaction and urine cortisol and blood DHEA-S levels. Conclusion: Measurements of biomarkers may offer additional stress evaluations with OSI questionnaires for bus drivers. Increased DHEA-S and cortisol levels may result from stressors like income security. Prevention efforts towards occupational stress and life events and health promotional efforts for aged driver were important anti-stress remedies.
기후변화는 현대 축산업이 직면한 가장 큰 위협 중의 하나이고, 특히 열 스트레스는 가금의 생산성 저하뿐만 아니라, 가축의 불편함과 고통을 증가시켜 동물의 복지에도 치명적 영향을 미칠 수 있다. 열 스트레스를 완화시키기 위한 다양한 전략들이 요구되는데, 가축의 열 스트레스에 대한 정확한 평가와 생물학적 평가지표의 개발은 기후변화에 대한 적응과 열 저항성 가금의 육종, 지속 가능한 가금 산업과 동물복지를 위해서 매우 중요하다. 스트레스호르몬, hetero- phils lymphocytes의 비율, 텔로미어 마모 정도 등 열 스트레스에 대한 다양한 바이오마커 중에 열충격단백질(HSPs)은 세포내 온도계로 여겨지며, HSPs 중에서도 HSP70은 가장 풍부한 단백질이면서 열 충격에 민감하게 반응하는 바이오 마커로 알려져 있다. HSP는 또한 열 충격으로부터 세포를 보호해주는 샤페론 활성을 지니고 있다. 따라서 HSP70 바이오마커는 가축의 열 스트레스에 대한 체계적 평가방법으로 주목받고 있으며, HSP70의 발현 변화의 이해는 기후변화에 대처하면서 지속가능한 가금 산업을 위해 매우 중요하고 판단된다. 이에 본 논문에서는 닭의 열 스트레스와 관련하여 HSP70을 중심으로 HSP의 작용기전과 역할, 열 스트레스와 HSP, 내열성 가축 선발을 위한 HSP70의 잠재성, 그리고 열 스트레스 바이오마커로서의 HSP70 등에 대한 최근까지의 연구들을 소개한다.
기후 변화로 인한 수온의 상승은 어류 서식지에 영향을 미친다. 수온의 변화는 어류 생리 거의 모든 부분에 영향을 미치는 것으로 알려져 있다. 기후 변화에 따른 수온의 상승은 산소 용해도의 감소 및 산소 운반 헤모글로빈의 결합 능력의 감소로 인해 저산소증을 초래할 수 있다. 본 연구는 대서양 연어(Salmo salar) 치어 성장의 최적수온($15^{\circ}C$)보다 고수온($20^{\circ}C$)에 사육 시, 대서양 연어 치어의 건강상태를 평가하기 위해 수행되었다. 평가 방법은 NGS RNAseq 분석방법을 이용하여 생체지표유전자를 개발하고, RT-qPCR 분석을 이용하여 생체지표유전자의 발현양상을 조사하는 것이다. 개발한 생체지표유전자로는 interferon alpha-inducible protein 27-like protein 2A transcript variant X3, protein L-Myc-1b-like, placenta growth factor-like transcript variant X1, fibroblast growth factor receptor-like 1 transcript variant X1, transferrin, intelectin, thioredoxin-like, c-type lectin lectoxin-Thr1-like, ladderlectin-like 및 calponin-1 등이다. 선택된 생체지표 유전자는 NGS RNAseq 분석을 통해 수온변화에 민감하게 발현한 유전자들이며, RT-qPCR 분석을 통한 이들 유전자의 발현 양상은 NGS RNAseq 분석을 통한 발현 양상과 매우 유사하게 나타났다.
