Studies on the quantitative determination of Capsaicin in various species of the genus Capsicum (고추중의 Capsaicin 정량(定量)에 관(關)한 연구(硏究))

  • Various species of the genus Capsicum contain in their fruits an intensively sharp and pungent substance, Capsaicin, which was first isolated in an almost pure state by Thresh. Containing the pungent principle, Capsicums are used extensively in food as a spice and in medicine as a rubefacient and carminative. Numerous methods have been proposed for the isolation, the chemical structure and the quantitative determination of Capsaicin. Modifying the several methods described before, the Capsaicin contents in various species of the genus Capsicum were determined as follows. (1) The isolation of pure Capsaicin was the essential first step for the determination of Capsaicin contents. Powdered cayenne pepper was extracted with acetone. By the method of ether alkali partition extraction slightly modified at this laboratory and by the recrystallization with light petroleum ether that was repeated ten times, the pure crystalline Capsaicin was obtained. Using this Capsaicin, the standard absorption curve was drawn with Beckman spectrophotometer model DU for the quantitative determination of Capsaicin. 92) The powdered sample was extracted in a Soxhlet extractor with ether-acetone solvent system(3:1) for 25 hours. Capsaicin in this ether-acetone extracts was efficiently separated in a pure state by paper partition chromatography using 58% methanol solution as developing agent. It was found that 58% methanol was one of the most valuable solvent to separate Capsaicin from impurities such as sterols, fatty acids, waxes and carotenoid pigments. (3) The colorimetric method modifying the Schulte-Kruger's method which consists of measuring the red color produced with diazobenzenesulphonic acid was used. Capsaicin in various species of the genus Capsicum was determined quantitatively with use of Beckman spectrophotometer model DU at $480\;m{\mu}$.

Lipids of Fresh-Water Cockle, Corbicula elatior (재첩의 지질에 관한 연구)

  • Present investigation was carried out to clarify chemical characteristics, lipid composition, fatty acid components and the sterol composition of the lipids from the fresh-water cockle, Corbicula slatier. The results obtained are as follows :1) The oil content, iodine value and unsaponifiable matter of the total .lipids were 2.17, 119 and $16.6\%$ respectively. 2) The main components of the total lipids were phospholipids (ahout $43\%$), triglyceride and sterol. 3) The main fatty acids of the total lipids were $C_{16:0}\;(48.4\%),\;C_{14:0}\;(10.0\%),\;C_{18:3}\;(8.3\%),\;C_14:1\;(5.4\%)\;and\;C_{18:1}\;(5.3\%)$. 4) Sterols found were 24-methylenecholesterol ($14.1\%$), $\beta-sitosterol(22.5\%)$, fucosterol ($14.9\%$), isofucosterol (?) ($14.0\%$), $\Delta^7-avenasterol (?)(5.2\%)$ and cholesterol ($2.3\%$).

Cholesterol Biosynthesis from Lanosterol: Development of a Novel Assay Method, Characterization, and Solubilization of Rat Hepatic Microsomal Sterol Δ7-Reductase

  • A novel assay method is described for rapid quantitation of reaction rate of sterol ${\Delta}^7$-reductase (${\Delta}^7$-SR) which catalyzes reduction of the ${\Delta}^7$-double bond of sterols. Of six different organ tissues-liver, small intestine, brain, lung, kidney, and testis-. ${\Delta}^7$-SR activity was detected only in liver (2.30 nmol/min/mg protein) and testis (0.11 nmol/min/mg protein). Using a newly developed method which employs diet-induced enzyme proteins and ergosterol as substrate, we assessed both kinetics ($K_m=210\;{\mu}M$, $V_{max}=1.93\;nmol/min/mg$) and inhibition of the rat hepatic ${\Delta}^7$-SR against well-studied cholesterol lowering agents such as triparanol ($IC_{50}=16\;{\mu}M$). 3-$\beta$-[2-(diethylamino)ethoxy]androst-5-en-17-one (U18666A) ($IC_{50}=5.2\;{\mu}M$), and trans-1.4-bis(2-chlorobenzylaminomethyl)cyclohexane dihydrochloride (AY-9944) ($IC_{50}=0.25\;{\mu}M$). Of the three well-known AY-9944-sensitive cholesterogenic enzymes (i.e., ${\Delta}^7$-SR, sterol ${\Delta}^8$-isomerase, and sterol ${\Delta}^14$-reductase). ${\Delta}^7$-SR was found to be the most sensitive enzyme with a noncompetitive inhibition of this compound ($K_i=0.109\;{\mu}M$). Substrate specificity studies of the microsomal ${\Delta}^7$-SR indicate that the relative reaction rate for 7-dehydrocholesterol and ergosterol are 5.6-fold and 1.6-fold higher than that for lathosterol. ${\Delta}^7$-SR activity was also modulated by feeding rats a diet supplemented with 0.5% ergosterol (>2.6-fold) in addition to 5.0% cholestyramine plus 0.1% lovastatin ($\simeq$5.0-fold). Finally, microsomal ${\Delta}^7$-SR was solubilized by 1.5% 3-[3-(cholamidopropyl)-dimethylammonio]-1-propanesulfonate (CHAPS) and enriched on PEG (0~10%) precipitation, which should be suitable for further purification of the enzyme.

Comparative Studies on the Lipid Content and Neutral Lipid Composition in Nonglutinous and Glutinous Rice (멥쌀과 찹쌀중의 지방질 함량 및 중성 지방질의 조성에 관한 비교)

  • The lipids content and neutral lipid component of milled rice grain were studied for four nonglutionous and two glutinous varieties grown in Korea. The average total lipid contents in nonglutinous and glutinous varieties were 0.94% and 1.78% by dry weight basis, respectively. The ratios of neutral lipid, glycolipid and phospholipid in the total lipid were 76.5 : 14.8 : 8.6 in the nonglutionous and 82.4 : 11.1 : 6.5 in the glutinous, respectively. Main classes of the neutral lipids were triglycerides, free fatty acids, steryl esters, free sterols, monoglycerids and diglycerides in both of nonglutinous and glutinous, and no significance was observed in the content of the classes between both varieties. The major fatty acid composition of total and neutral lipids were linoleic, oleic and palmitic acids in both of nonglutinous and glutinous varieties.

  • Streptomyces griseorubiginosus var. soyoensis previously identified, produced two kinds of antifungal antibiotics, trans-cinnamamide and another new substance. The latter was identified to be a new substance of tetraene family by establishment of UV, IR, NMR, mass spectra and chemical reactions and rotatively named as Tetraene KM-A. Through an antimicrobial activity test using serial agar dilution method, Tetraene KM-A showed strong growth inhibitory activity against fungi and yeasts, but not against procaryotes tested. The inhibitory action of Tetraene KM-A on fungi was remarkably ineffective when some of sterols were added to the cultural media. $LD_{50}$ of the Tetrene KM-A to mice and rats by intravenous injection were 84.3 and 90.4 mg/kg respectively. $LD_{50}$ to mice by oral feeding was 1503mg/kg.

  • To Produce the modified Cheongiu that has high ethanol content, an ethanol-tolerant strain Saccharo-myces cerevislae SE2l1 was screened from Saccharomyces cerevisiae Kyokai No. 10 strain. The isolate showed faster growth than in the medium containing 10% ethanol compared with original strain. The isolate produced a higher concentration of ethanol and showed higher resistance to ethanol, high osmolarity and heat than the original strain. The analyses of yeast membrane components indicated that there were no significant changes in composition of sterols and phospholipids between the isolated and the original strain. However, during the fermentation, the iso-lated strain could change the fatty acid composition in the membrane more rapidly in the direction of decreasing membrane unsaturation and accumulate more trehalose in the cell than the original strain. These data suggest that the ability to change its membrane fatty acid composition and to accumulate trehalose may make the isolated strain easily adapt to changes in external condition.

Effect of Dietary Calcium and Fat on Plasma Cholesterol Level and Cholesterol Metabolism in 1, 2-dimethylhydrazine-treated Rats (Dimethylhydrazine으로 처리한 쥐에서 식이의 Calcium 함량과 지방종류에 따라 혈장 Cholesterol 수준과 Cholesterol 대사에 미치는 영향)

  • The study was designed to observe the effect of dietary calcium and fats on plasma cholesterol level, hepatic microsomal fluidity and HMG-CoA reductase activity as well as the excretion of fecal bile acids and neutral sterols in 1, 2-dimethylhydrazine(DMH)-treated rats. Male Sprague Dawley rats, at 7 weeks of age, were divided into 2 groups, 0.3% and 1.0% Ca levels and each group again subdivided into 2 groups of corn oil and perilla oil. Each rat was intramuscularly infused with DMH for 6 weeks to give total dose of 180mg/kg body weight and also fed experimental diet containing 15%(w/w) different fit and Ca(0.3% or 1.0%) for 20 weeks. High dietary calcium(1.0%) did not significantly influence on plasma cholesterol as well as hepatic microsomal fluidity and HMG CoA reductase activity, but significantly reduced the excretion of total bile acid per gram of faces and increased the excretion of total neutral sterol. However, high dietary Ca reduced the excretion of secondary bile acid(deoxycholic and lithocholic acids) which was known as promoter for colon cancer. Perilla oil rich in n-3 ${\alpha}$-linolenic acid significantly decreased plasma cholesterol by increasing hepatic microsomal fluidity compared with corn oil, but did not influence on HMG CoA reductase activity. Perilla oil did not influence on fecal excretion of total and primary bile acids, but reduced the excretion of secondary bile acids. Therefore, it could be recommended to consume more fish product and food rich in calcium and use more perilla oil in meal preparation to prevent from coronary hear disease and colon cancer especially when high fit diet has been practiced.   • Seven diterpenes, four polyacetylenes, a lipid glycerol, and two sterols were isolated from the methylene chloride fraction of the root of Aralia cordata. Their chemical structures were determined as (-)-pimara-8(14), 15-dien-19-oic acid (2), pimaric acid (3), (-)-kaur-16-en-19-oic acid (4), 17-hydroxy-ent-kaur-15-en-19-oic acid (9), $7{\alpha}$-hydroxy-(-)-pimara-8(14), 15-dien-19-oic acid (10), $16\alpha$, 17 -dihydroxy-(-)-kauran-19-oic acid (11), 16-hydroxy-17-isovaleroyloxy-ent-kauran-19­oic acid (12), falcarindiol (5), dehydrofalcarindiol (6), dehydrofalcarindiol-8-acetate (7), falcarin­diol-8-acetate (8), alpha-mono palmitin (13), stigmasterol (1), and daucosterol (14) by the spectral evidences. These compounds were tested with COX-1 and COX-2 inhibition assays. This study found that compounds 2, 4, 5, 6, 7, 8, and 10 inhibited COX-1 dependent conversion of the exogenous arachidonic acid to $PGE_2$ in a dose-dependent manner with $IC_{50}$ values of $134.2{\mu}M$, $121.6{\mu}M$, $170{\mu}M$, $50.4{\mu}M$, $11.7{\mu}M$, $99.6{\mu}M$, and $69.6{\mu}M$, respectively. But, most of these compounds weakly inhibited COX-2 dependent $PGE_2$ generation. Among them, only compound 4 showed relatively significant inhibitory activity $(IC_{50}\;:\;127.6{\mu}M)$.

Alteration of Lipid Metabolism by Ginseng Supplements With Different Levels of Vitamin E in High Cholesterol-Fed Rats

  • Ginseng may have antioxidant and pharmacologic effects similar to those of vitamin E. The interactive effect of ginseng and vitamin E was studied with respect to cholesterol metabolism and the antioxidant status. A ginseng supplement (0.1%, wt/wt) with comparable levels of vitamin E was provided with a high-cholesterol (1%, wt/wt) diet to rats for 5 weeks. The amount of vitamin E included in the ginseng-free and ginseng diets was either a low (low-E) or a normal (normal-E) level. The ginseng supplements significantly (p<0.05) altered the concentrations of plasma triglycerides in both the low-vitamin E group and normal-vitamin E group compared to the each ginseng-free group. The hepatic triglyceride and cholesterol content were not significantly (p>0.05) different between groups regardless of the vitamin E level in the diet. The hepatic HMG-CoA reductase activity was significantly (p<0.05) lowered by the ginseng supplement in both the low-vitamin E and the normal-vitamin E groups compared to the ginseng-free group. The HMG-CoA reductase activity was also significantly (p<0.05) lowered with in increase of the dietary vitamin E in the ginseng-free group. The excretion of fecal neutral sterol was significantly (p<0.05) lower in the normal-E ginseng group than th low-E ginseng-free group. Neither dietary ginseng nor vitamin E significantly changed the hepatic antioxidant enzymes activity. This data indicates that ginseng supplements lower the concentration of plasma triglyceride and hepatic HMG-CoA reductase activity regardless of eh dietary vitamin E level. This information may contribute to understanding the interactive effect of ginseng and vitamin E on cholesterol biosynthesis in high cholesterol-fed rats.

Sources and Distributions of Organic Wastewater Compounds on the Mokpo Coast of Korea

  • Surface water and sediment samples collected from the Mokpo coast of Korea were analyzed for molecular markers of organic municipal wastewaters, i.e., 11 fecal sterols including coprostanol (Cop) and nonylphenolic compounds (NPs), to characterize the main routes of these wastewaters to the coast and to assess contamination levels. Concentrations of Cop ranged from 94 to 7,568 ng/L in surface water and from 43 to 38,108 ng/g dry weight in sediments. Concentrations of NPs [nonylphenol (NP) and nonylphenol mono- and di-ethoxylates ($NP_{1-2}EOs$)] ranged from 123 to 4,729 ng/L in surface water and from 4 to 2,119 ng/ng dry weight in sediments. The levels of these compounds were much higher at stations near the rivers that pass through the urban center of Mokpo and the outfall of the wastewater treatment plant (WWTP). The spatial distribution of Cop levels was statistically similar to that of NPs (r=0.809 and 0.982 in surface water and sediments, respectively), indicating that these compounds may have similar discharge points, transport, mixing, and deposition in the study area. These results suggest that considerable amounts of organic wastewater compounds are discharged through rivers and WWTP effluent to the Mokpo coast.
