  • Dihydroxyacetone (DHA) has been used as a self tanning agent and many emulsion formulations containing DHA have been studied. In an emulsion, many factors which have negative effect on DHP and the resultant DHA decomposition can destabilize the emulsion base. In this study, two kinds of emulsion with 5% DHA were prepared, O/W type emulsion and Multilamellavesicle (MLV) type emulsion to compare the stabilization effects of both emulsions on the DHA. The OHA concentration was analyzed quantitatively by high performance liquid Chromatography (HPLC), also the pH and viscosity of both emulsions were measured for stability. This process was carried out over 4 months. For HPLC, a bondaclone $C_{18}$ column with a mobile phase of distilled water and UV detector were used. The results of these experiment showed that DHA is more stable in an MLV emulsion than it is in an O/W type emulsion.

O/W 유제의 물리적 안정성에 대한 PVP의 영향 (Effect of PVP on the Physical Stability of O/W Emulsion)

  • To make a stable o/w emulsion, the effects of egg lecithin as an emulsifier and polyvinylpyrrolidone (PVP) as an auxiliary emulsifier on the physical stability of emulsion were investigated. The oil-in-water emulsion system was manufactured by microfluidizer and evaluated the physical stability. Average particle size and size distribution of emulsion was measured by dynamic light scattering analyzer and interfacial tension was measured. From the interfacial tension tested, critical micelle concentration of the egg lecithin was 0.1 %w/v and optimal concentration for the preparation of emulsion was 1.0 %w/v. The mean particle size was about $0.2\;{\mu}m$ which was suitable for injections. The short-term accelerated stability studies were conducted by centrifugation, freeze-thaw method and shaking of the emulsion samples. The addition of PVP was caused the reduction in the particle size and improved the physical stability of emulsion. These results suggested that a mixed interfacial film comprising the egg lecithin and PVP was formed at the o/w interface and it was effective in preventing phase separation under thermic or mechanical stress. We used antineoplaston A10 (A10) as a model drug which is peptide and amino acid derivative having a action to the living organism against the development of neoplastic growth by a nonimmunological progress. It has a poor solubility in water and there may be a difficulty in formulation of A10. Emulsion formulation study about A10 was performed. Solubility of A10 in emulsion was about five times as high as that in water. From the results of solubility and partition coefficient, almost A10 molecules in o/w emulsion exist in the interface between oil and water.

  • The O/W and W/O emulsions containing Scutellariae Radix extract(SRE) which is very slightly soluble in oil phases and sparingly soluble in water phases, were prepared by homogenizing water and oil phases with emulsifier. The diameters of emulsion were ranged from 100 to $300\;{\mu}m$. The viscosity of W/O emulsion was higher than that of O/W emulsion. W/O emulsion was more stable than O/W emulsion which was gradually degraded when tested by centrifuge method and temperature tolerance method at $50^{\circ}C$. The antibacterial activity of two emulsions was not significantly different from that of aqueous solution of SRE, and showed similar MIC and bacterial growth inhibition rate.

THE STABILITY OF ALL-TRANS-RETINOL IN NOVEL LIQUID CRYSTALLINE OW EMULSION

  • We investigated the stability of all-trans-retinol on the liquid crystalline O/W emulsion composed of mainly alkyl polyglycerine, alkyl polyglucose and glycerine, and compared the activity of all-trans-retinol in the various forms of liquid crystal. Under certain conditions, novel liquid crystalline gel was formed around oil droplets, and layers of this liquid crystalline gel were very wide and rigid. (SWLC; Super Wide Liquid Crystal) SWLC was very helpful to stabilize retinol in O/W emulsion. After storage at 45 C for 4 weeks, all-trans-retinol in O/W emulsion composed of SWLC retained above 85% of the activity upon HPLC analysis, whereas those within no liquid crystalline emulsion gave 47% and normal liquid crystalline emulsion composed of fatty alcohols gave 40 60%. Retinol in oil phase is nealy insoluble in pure water, but in cosmetic emulsion systems can be slightly solubilized into water because emulsifiers and polyols in emulsion systems function as solubilizers. In this case, water in outer phase acts as a media for oxygen transporation$.$and thus destabilizes retinol. As a result, retinol in O/W emulsion has a tendency to become unstable. SWLC surrounding oil droplet which contains retinol is wide and rigid, therefore reduces contact between inner phase and outer phase To make SWLC, properties of emulsifiers are very important phase transition temperature should be high, and the structure of surfactants should be bulky, and their ratio should be suitable to make rigid and wide liquid crystalline gel layer in order to reduce contact between retinol in inner phase and water in outer phase.

계면활성제를 이용한 수분산 Tocopherol 유화물의 개발 (Development of Water Soluble Tocopherol Emulsion Using Surfactants)

  • 지용성의 토코페롤이 갖고 있는 많은 생리활성 기능을 수용액 상태에서도 발현할 수 있도록 유지 용해성을 갖는 토코페롤을 친수성 및 친유성 계면활성제를 이용하여 수분산 기법으로 최적의 토코페롤 유화액을 제조하고 최적의 유화 critical micellization concentration(CMC)을 선정하고자 표면장력 및 유화액의 저장안정성을 측정하였다. 실험에 사용한 토코페롤의 분자량은 $340{\sim}360$ 정도였으며 유화모델은 O/O/W/W형으로 선정하였다. 유화안정성과 표면장력과의 상관관계를 살펴보면, 유화액의 표면장력이 $40{\sim}46\;dyne/cm$에서 안정성을 확인할 수 있었으며, 표면장력이 40 dyne/cm이하로 떨어졌을 경우에는 유화안정성이 낮아졌다. 토코페롤의 유화안정성 실험 결과, 20%, 30%, 40% 수용성 토코페롤 유화액 모두에서 hydrophobic 계통의 유화제로 polyglycerine polyricinoleate를 적용한 유화 에멀젼에서 유화안 정성이 높았으며, hydrophilic 계통의 유화제에서는 polyglycerine monostearate> polyglycerine monooleate> polyoxyethylene(20) sorbitan monooleate$\geq$ polyoxyethylene(20) sorbitan monolaurate 순으로 유화안정성을 나타내었다.

Hansen Solubility Parameter 를 통한 W/O 형 자외선차단 제형의 유화 안정성에 관한 연구 (A Study on the Emulsifying Stability of W/O Type Sunscreen Cream by the Hansen Solubility Parameter)

  • 자외선차단 제형은 내수성이 중요하므로 수상이 유상에 분산되는 water-in-oil (W/O) 에멀젼으로 만드는 것이 바람직하다. 이 경우에는 오일 특유의 무겁고 끈적이는 사용감 때문에 이를 보완하는 방법으로 실리콘계 오일을 사용한다. 이러한 오일은 유기 자외선차단제와의 상용성이 낮아 에멀젼의 안정성에 문제가 되고 있다. 본 연구에서는 자외선차단 제형의 W/O 에멀젼에서 다양한 오일들 간의 상용성을 Hansen solubility parameter(HSP)를 이용하여 수치적으로 도출하였다. HSP는 물질의 분산, 극성 및 수소 결합을 나타내는 지표로 물질간의 상용성을 판단하는데 유용하다. 본 연구에서는 HSP가 다른 다양한 오일을 선정하여 W/O 에멀젼을 제조하고 제형의 시간에 따른 점도 변화 및 안정성을 조사하여 HSP 수치와의 상관관계 또는 일치성을 고찰하였다. HSP를 이용하면 상용성이 높은 오일 선택을 용이하게 할 수 있었다. 또한, W/O 에멀젼에서 유상의 상용성이 높으면 점도의 경시변화가 작았으며 freeze-thaw cycle (-15 ℃ ~ 45 ℃) 조건에서 안정성이 향상된 것을 확인하였다. 향후 HSP를 통해 성분 구성을 최적화한다면 사용감이 우수하면서 안정성을 확보한 W/O형 자외선차단 제형 개발에 도움이 될 것이라 기대된다.

Formula Optimization of a Perilla-canola Oil (O/W) Emulsion and Its Potential Application as an Animal Fat Replacer in Meat Emulsion

  • The formulation of an oil/water (o/w) emulsion made up of a mixture of perilla oil and canola oil (30/70 w/w) was optimized using a response surface methodology to find a replacement for animal fat in an emulsion-type meat product. A 12 run Plackett-Burman design (PBD) was applied to screen the effect of potential ingredients in the (o/w) emulsion, including polyglycerol polyricinoleate (PGPR), fish gelatin, soy protein isolate (SPI), sodium caseinate, carrageenan (CR), inulin (IN) and sodium tripolyphosphate. The PBD showed that SPI, CR and IN showed promise but required further optimization, and other ingredients did not affect the technological properties of the (o/w) emulsion. The PBD also showed that PGPR played a critical role in inhibiting an emulsion break. The level of PGPR was then fixed at 3.2% (w/w total emulsion) for an optimization study. A central composite design (CCD) was applied to optimize the addition levels of SPI, CR or IN in an (o/w) emulsion and to observe their effects on emulsion stability, cooking loss and the textural properties of a cooked meat emulsion. Significant interactions between SPI and CR increased the cooking loss in the meat emulsion. In contrast, IN showed interactions with SPI leading to a reduction in cooking loss. Thus, CR was also removed from the formulation. After optimization, the level of SPI (4.48% w/w) and IN (14% w/w) was validated, leading to a perilla-canola oil (o/w) emulsion with the ability to replace animal fat in an emulsion-type meat products.

유중수적형 유화계내에서의 유화안정지수 산출법의 확립 (Determination of Emulsion Stability Index in W/O Emulsion)

  • 유중수적형 유화계의 유화안정지수를 산출하는 방법을 확립하기 위하여 간편하면서도 예민한 "부피 직접 측정법"을 개발하였는 바, 먼저 강력한 혼합기인 일종의 Ultra-turrax T25를 이용하여 유지방, 유화제 및 물로 구성되면서 유지방이 물보다 많이 함유되는 유중수적형의 유화계를 형성시킨 후 상온에서 시간이 경과함에 따라 분리되어지는 수분층의 부피를 측정하였으며, 유화안정지수는 $(1-V_s/V_a){\times}100$(단, $V_a$는 유화계내에 함유되어 있는 총 수분의 부피이며, $V_s$는 시간이 경과함에 따라 유화계로부터 분리되어지는 수분층의 부피를 의미한다)의 계산식에 대입하여 산출하였다. 한편, sorbitan trioleate, span 60 및 tween 20을 첨가하여 유중수적형 유화계를 형성시킨 후 90분이 경과한 다음 "부피 직접 측정법"에 의하여 유화안정지수를 산출하였는바, 각각 $95.4{\pm}1.8$, $56.1{\pm}2.8$$41.6{\pm}2.2$였으며, 이러한 값은 유화안정지수의 산출방법으로서 일반적으로 사용하고 있는 "Titus의 방법"과 비교해 볼 때 5.0 이내의 차이-유화안정지수가 평형에 도달하였을 때의 차이 값-를 나타냄을 알 수 있었다.

Parenteral Docetaxel Emulsion System and Its Stability

  • Docetaxel is an anticancer agent with low aqueous solubility. More extensive clinical use of this drug is somewhat delayed due to lack of appropriate delivery vehicles. An attempt was made to adopt an o/w emulsion as the drug carrier which incorporated docetaxel in the propyleneglycerol stabilized by a mixed-emulsifier system. A suitable formulation was found in this study: 10 mg/mL docetaxel, 10% (w/v) oil blend, 4% (w/v) PG, 3% (w/v) Solutol HS 15 in 2.25% (w/v) glycerol solution. The formulated emulsion has very good stability when stored at $40^{\cird}C$, and the docetaxel containment efficiency can be maintained above 95% and the mean emulsion diameter around $10{\mu}m$ for at least 3 months. The formulated emulsion is a promising carrier for docetaxel and other lipophilic drugs.

Effects of HLB value on oil-in-water emulsions: Droplet size, rheological behavior, zeta-potential, and creaming index

  • Using mixed nonionic surfactants Span/Tween, we investigated the effects of HLB value on the O/W emulsion stability and rheological behaviors. In this study, MS-01 (Span 60 & Tween 60) and MS-02 (Span 80 & Tween 80) was used as mixed nonionic surfactants. We considered required HLB value 10.85 and selected corresponding HLB value range 8-13. The droplet size distributions, droplet morphology, rheological properties, zeta-potential and creaming index of the emulsion samples were obtained to understand the mechanism and interaction of droplets in O/W emulsion. The results indicated that optimal HLB number for O/W emulsions was 10.8 and 10.7, while using MS-01 surfactant and MS-02 surfactant respectively. MS-01 (HLB = 10.8) sample and MS-02 (HLB = 10.7) sample showed smallest droplet size and highest zeta-potential value. Rheological properties are measured to understand rheological behaviors of emulsion samples. All emulsion samples showed no phase separation until 30 days storage time at $25^{\circ}C$.
