Crystal structure of unphosphorylated Spo0F from Paenisporosarcina sp. TG-14, a psychrophilic bacterium isolated from an Antarctic glacier

  • Spo0F is a response regulator that modulates sporulation, undergoes phosphorylation for phosphorelay signal transduction, and interacts with various regulatory proteins; however, the mechanisms through which phosphorylation induces structural changes and regulates interactions with binding partners remain unclear. Here, we determined the unphosphorylated crystal structure of Spo0F from the psychrophilic bacterium Paenisporosarcina sp. TG-14 (PaSpo0F) and established a phosphorylation-state structural model. We found that PaSpo0F underwent structural changes (Lys54 and Lys102) by phosphorylation and generated new interactions (Lys102/Gln10 and Lys54/Glu84) to stabilize the ${\beta}4/{\alpha}4$ and ${\beta}1/{\alpha}1$ loop structures, which are important target-protein binding sites. Analysis of Bacillus subtilis Spo0 variants revealed movement by BsSpo0F Thr82 and Tyr84 residues following interaction with BsSpo0B, providing insight into the movement of corresponding residues in PaSpo0F (Thr80 and Tyr82), with further analysis of BsSpo0F/BsRapH interaction revealing alterations in the ${\beta}4/{\alpha}4$ loop region. These results suggest that phosphorylation-induced structural rearrangement might be essential for PaSpo0F activation and expand the understanding of Spo0F-specific activation mechanisms during sporulation.

Cytochrome $C_{550}$ is Related to Initiation of Sporulation in Bacillus subtilis

  • The effect of cytochrome $c_{550}$ encoded by cccA in Bacillus subtilis during the event of sporulation was investigated. The sporulation of cccA-overexpressing mutant was significantly accelerated, while disruptant strain showed delayed sporulation in spite of the same growth rate. Activity of sporulation stage-0-specific enzyme, extracellular $\alpha-amylase$ of mutant strains was similar to that of the control strain, but cccA-overexpressing mutant exhibited higher activity of stage-II-specific alkaline phosphatase and stage-III-specific glucose dehydrogenase when compared to deletion mutant and control strain. Northern blot analysis also revealed that cccA-overexpressing mutant showed high level of spo0A transcripts, while the disruptant rarely expressed spo0A. These results suggested that although cytochrome $c_{550}$ is dispensable for growth and sporulation, expression of cccA may play an important role for initiation of sporulation through regulation of spo0A expression.

The Incidence and Management of Dural Tears and Cerebrospinal Fluid Leakage during Corrective Osteotomy for Ankylosing Spondylitis with Kyphotic Deformity

  • Objective : To present the incidence and management of dural tears and cerebrospinal fluid leakage during corrective osteotomy [Pedicle Subtraction Osteotomy (PSO) or Smith-Petersen Osteotomy (SPO)] for ankylosing spondylitis with kyphotic deformity. Methods : A retrospective study was performed for ankylosing spondylitis patients with fixed sagittal imbalance, who had undergone corrective osteotomy (PSO or SPO) at lumbar level. 87 patients were included in this study. 55 patients underwent PSO, 32 patients underwent SPO. The mean age of the patients at the time of surgery was 41.7 years (21-70 years). Of the 87 patients, 15 patients had intraoperative dural tears. Results : The overall incidence of dural tears was 17.2%. The incidence of dural tears during PSO was 20.0%, SPO was 12.5%. There was significant difference in the incidence of dural tears based on surgical procedures ( PSO vs. SPO) (p<0.05). The dural tears ranged in size from 12 to $221mm^2$. A nine of 15 patients had the relatively small dural tears, underwent direct repair via watertight closure. The remaining 6 patients had the large dural tears, consequently direct repair was impossible. The large dural tears were repaired with an on-lay graft of muscle, fascia or fat harvested from the adjacent operation site. All patients had a successful repair with no patient requiring reoperation for the cerebrospinal fluid leak. Conclusion : The overall incidence of dural tears during PSO or SPO for ankylosing spondylitis with kyphotic deformity was 17.2%. The risk factor of dural tears was complexity of surgery. All dural tears were repaired primarily using direct suture, muscle, fascia or fat graft.

Studies on the Toxic Activity of Bacillus sphaericus Spo -D1216 Asporogenic Mutant (Bacillus sphaericus Asporogenic Mutant Spo - D1216의 독성에 관한 연구)

  • The cell cultures and crude extracts of Bacillus sphaericus 1593 K-5 and its mutant Spo-Dl216 were respectively bioassayed against Culex pipiens var. pollens mosquito larvae. The B. sphaeriucs 1593 K-5 showed toxic activity against the larvae. LC$_{50}$ values (cells/$m\ell$) was 2.6$\times$10$^2$. Also the LC$_{50}$ ($\mu\textrm{g}$ Protein/$m\ell$) of the crude extract was 10.26. However, B. sphaericus Spo-Dl216 didn't show toxic activity against the larvae. The soluble cytoplasmic toxin in broken B. sphaeriucs 1593k-5 cells was partially purified by gel permeation chromatography and ion exchange chromatography. Among the fractions of the gel permeation chromatography only a single fraction was found to be toxic. LC$_{50}$ values ($\mu\textrm{g}$ protein/$m\ell$) of the active fraction was 0.182. The active fraction of the gel permeation was subjected to ion exchange chromatography. Only a single fraction showed toxic activity and its LC$_{50}$ values ($\mu\textrm{g}$ protein/$m\ell$) was 0.02..02.

Comparison of Physicochemical and Functional properties of Soymilk with Addition of Onion (양파 첨가 형태를 달리한 두유의 이화학적 및 기능적 특성 비교)

  • Onions contain antioxidant flavonoids and bioactive sulfur compounds. These substances are more abundant in the peel than in onion flesh. For this reason, whole onions including peels were added to soy milk to produce soy milk with whole onions (SWO), whereas peeled onions were added to soy milk to produce soy milk with peeled onions (SPO). The functional and antioxidant properties of these two kinds of soy milk were then analyzed and compared. Compared to control soy milk (CS) without onion powder, treated samples (to which freeze dried onion powder was added at 1.4, 1.6, 1.8, and 2.0%, respectively) showed significantly increased amounts of quercetin, isoflavone, and total phenol (p=0.05). The magnitude of the increase rose as the amount of added onion powder increased and when onion powder contained onion peels. With regards to antioxidant activity (DPPH and ABTS), SWO showed a greater value than SPO. The sensory evaluation scores of SWO and SPO were lower than CS for roughness and swallowing, as adding onion powder increased solid contents and viscosities. However, higher overall acceptability were obtained 1.8SPO and 1.8SWO.

Urease Characteristics and Phylogenetic Status of Bacillus paralicheniformis

  • In 2015, Bacillus paralicheniformis was separated from B. licheniformis on the basis of phylogenomic and phylogenetic studies, and urease activity was reported as a phenotypic property that differentiates between the two species. Subsequently, we have found that the urease activity of B. paralicheniformis is strain-specific, and does not reliably discriminate between species, as strains having the same urease gene cluster were identified in B. licheniformis and B. sonorensis, the closest relatives of B. paralicheniformis. We developed a multilocus sequence typing scheme using eight housekeeping genes, adk, ccpA, glpF, gmk, ilvD, pur, spo0A, and tpi to clearly identify B. paralicheniformis from closely related Bacillus species and to find a molecular marker for the rapid identification of B. paralicheniformis. The scheme differentiated 33 B. paralicheniformis strains from 90 strains formerly identified as B. licheniformis. Among the eight housekeeping genes, spo0A possesses appropriate polymorphic sites for the design of a B. paralichenofomis-specific PCR primer set. The primer set designed in this study perfectly separated B. paralicheniformis from B. licheniformis and B. sonorensis.

The Effect of Action Observation Training on Sit to Walk with Chronic Stroke Patients

  • Purpose: This study was performed to examine the effect of action observation training on sit to-walk times in chronic stroke patients. Methods: Twelve stroke patients were randomly allocated to two groups, as follows: an action observation training (AOT) group and a scenery picture observation (SPO) group. The AOT group was engaged in video observation for 2 minutes 30 seconds with 12 ADL functional activities related to the sit-to-walk task: physical training was carried out in two repetitions lasting 2 minutes 30 seconds. The landscape imagery control group observed landscape picture for 2 minutes 30 seconds and then carried out the same physical training. The intervention involved a total of 12 sessions over 4 weeks, and each session lasted 30 minutes. The pre and post-tests measured the timed up and go test (TUG), Sit to stand test (STS), and Dynamic gait index (DGI). The Wilcoxon signed ranks test was used to compare pre-test and post-test result, and the Mann-Whitney U test was employed for comparison between groups. Results: The TUG time was significantly different between the AOT group and the SPO group. According to the findings, the experimental group's TUG was significantly decreased (p<0.05). In comparison of the two groups, there was no significant in STS or DGI between the AOT group and SPO group. Conclusion: This study showed that the intervention can be used to decrease TUG time. Our research suggests that action observation training has a positive effect on sit-to-walk times in patients with chronic stroke.

Expression of the Promoter for the Maltogenic Amylase Gene in Bacillus subtilis 168

  • An additional amylase, besides the typical $\alpha-amylase,$ was detected for the first time in the cytoplasm of B. subtilis SUH4-2, an isolate from Korean soil. The corresponding gene (bbmA) encoded a malto­genic amylase (MAase) and its sequence was almost identical to the yvdF gene of B. subtilis 168, whose function was unknown. Southern blot analysis using bbmA as the probe indicated that this gene was ubiquitous among various B. subtilis strains. In an effort to understand the physiological function of the bbmA gene in B. subtilis, the expression pattern of the gene was monitored by measuring the $\beta-galactosidase$ activity produced from the bbmA promoter fused to the amino terminus of the lacZ struc­tural gene, which was then integrated into the amyE locus on the B. subtilis 168 chromosome. The pro­moter was induced during the mid-log phase and fully expressed at the early stationary phase in defined media containing $\beta--cyclodextrin\;(\beta-CD),$ maltose, or starch. On the other hand, it was kept repressed in the presence of glucose, fructose, sucrose, or glycerol, suggesting that catabolite repression might be involved in the expression of the gene. Production of the $\beta-CD$ hydrolyzing activity was impaired by the spo0A mutation in B. subtilis 168, indicating the involvement of an additional regu­latory system exerting control on the promoter. Inactivation of yvdF resulted in a significant decrease of the $\beta-CD$ hydrolyzing activity, if not all. This result implied the presence of an additional enzyme(s) that is capable of hydrolyzing $\beta-CD$ in B. subtilis 168. Based on the results, MAase encoded by bbmA is likely to be involved in maltose and $\beta-CD$ utilization when other sugars, which are readily usable as an energy source, are not available during the stationary phase.

DYNAMICAL CHARACTERISTICS OF SUNSPOT CHROMOSPHERES II. ANALYSIS OF CA II H, K AND ${\lambda}8498$ LINES OF A SUNSPOT (SPO 5007) FOR OSCILLATORY MOTIONS

  • We have analyzed the time series of Ca II H,K and ${\lambda}8498$ line profiles taken for a sunspot (SPO 5007) with the Echelle spectrograph attached to Vacuum Tower Telescope at Sacramento Peak Solar Observatory. Each set of spectra was taken simultaneously for 20 minutes at a time interval of 30 seconds. A total of 40 photographic films for each line was scanned by a PDS at Korea Astronomy Observatory. The central peak intensity of Ca II H ($I_{max}$), the intensity measured at ${\Delta}{\lambda}=-0.1{\AA}$ from the line center of ${\lambda}8498(I_{{\lambda}8489})$, the radial velocity ($V_r$) and the Doppler width (${\Delta}{\lambda}_D$) estimated from Ca II H have been measured to study the dynamical behaviors of the sunspot chromosphere. Fourier analysis has been carried out for these measured quantities. Our main results are as follows: (1) We have confirmed the 3-minute oscillation being dominant throughout the umbra. The period of oscillations jumps from 180 sec in the umbra to 500 to 1000 sec in the penumbra. (2) The nonlinear character of the umbral oscillation is noted from the observed sawtooth shaped radial velocity fluctuations with amplitudes reaching up to $5{\sim}6\;km/sec$. (3) The spatial distribution of the maximum powers shows that the power of oscillations is stronger in the umbra than in the penumbra. (4) The spatial distributions of the time averaged < $I_{max}$ > and < $V_r$ > across the spot are found to be nearly axially symmetric, implying that the physical quantities derived from the line profiles of Ca II H and ${\lambda}8498$ are inherently associated with the geometry of the magnetic field distribution of the spot. (5) The central peaks of the CaII H emission core lead the upward motions of the umbral atmosphere by $90^{\circ}$, while no phase delay is found in intensities between $I_{max}$ and $I_{{\lambda}8498}$, suggesting that the umbral oscillation is of standing waves.

Sensitivity of Bordetella bronchise ptica Isolated from Pigs Affected by Infectious Atrophic Rhinitis to Chemotherapeutic Agents (전염성위축성비염돈(傳染性萎縮性鼻炎豚)으로부터 분리(分離)한 Bordetella bronchiseptica의 화학요법제(化學療法劑)에 대(對)한 감수성시험(感受性試驗))

  • A total of 98 strains of Bordetella bronckiseptica isolated from pigs affected the infectious atrophic rhinitis(AR) during 1978 were surveyed for drug sensitivity to 26 chemotherapeutic agents, and minimum inhibitory concentration(MIC), incidence rate of resistant strain and resistant patern from the strains which were obtained from the different pig farm in Jeonnam province were examined. The results obtained are summarized as follows. 1. Most of the strains tested were resistant to Ampicillin (AB, PC), spiramycin(SPO, sulfa drugs (SD) (MIC:$400.0{{\mu}g/ml}$) and streptomycin(SM) (MIC:$200.0{{\mu}g/ml}$). Of the 75.0% of strains were also resistant to penicillin(PC) (MIC:$200.0{{\mu}g/ml}$) and of the 14.3 of strains were inhibited to grow to tetracycline(TC), chlortetracycline(CTC), oxytetracyc-line(OTC), erythromycin(EM), tylosin(TS), leucomycin (LM) and chloramphenicol (CP) (MIC:$6.25{{\mu}g/ml}$). On the other hand, most of the strains tested were inhibited to grow to kanamycin(KM), gentamycin(GM) neomycin(NM) (MIC:$25.0{{\mu}g/ml}$) and to colistin(CL) (MIC:$12.5{{\mu}g/ml}$). 2. Incidence rate of resistant strains to main chemotherapeutic agents was 100.0% of sulfa drugs, 96.4% of streptomycin, 85.7% of penicillin, tetracycline, chloramphenicol and erythromycin, 46.4% of gentamycin, 17.9% of colistin and 0.0% of kanamycin and nalidixic acid.
