Development of a Porous Scaffold-Manufacturing Method by Blending Silk Fibroin and Agarose Polymer Solutions

  • Low-melting-temperature agarose gel solution, as a novel porogen was combined with a silk fibroin solution to generate interconnected porous networks. The porosity of the resultant silk fibroin-agarose scaffolds was greater than that of the scaffolds generated with agarose and deionized water. The porosities of silk fibroin scaffolds containing agarose gel at 0.5%, 1.0%, 1.5%, 2.0% [w/v] were 110.9%, 111.7%, 120.9%, and 123.0%, respectively. Lastly, the internal space generated in scaffolds after dissolution of the agarose gel provides a good environment for cell growth and movement within the scaffold.

  • Receptor for advanced glycation end products (RAGE) is one of the single transmembrane domain containing receptors and causes various inflammatory diseases including diabetes and atherosclerosis. RAGE extracellular domain has three consecutive IgG-like domains (V-C1-C2 domain) which interact with various soluble ligands including heparan sulfate or HMGB1. Studies have shown that each ligand induces different oligomeric forms of RAGE which results in a ligand-specific signal transduction. The structure of mouse RAGE bound to heparan sulfate has been previously determined but the electron density map of heparan sulfate was too ambiguous that the exact position of heparin sulfate could not be defined. Furthermore, the complex structure of human RAGE and heparin sulfate still remains elusive. Therefore, to determine the structure, human RAGE was overexpressed using bacterial expression system and crystallized using the sitting drop method in the condition of 0.1 M sodium acetate trihydrate pH 4.6, 8 % (w/v) polyethylene glycol 4,000 at 290 K. The crystal diffracted to 3.6 Å resolution and the space group is C121 with unit cell parameters a= 206.04 Å, b= 68.64 Å, c= 98.73 Å, α= 90.00°, β= 90.62°, γ= 90.00°.

  • Superior orbital fissure syndrome is characterized by ophthalmoplegia, ptosis of the eye, reflex dilation of the pupil, and anesthesia of the upper eyelid and forehead. This syndrome may be the result of craniofacial fractures as well as neoplasms of the retrobulbar space, hematomas in the orbital muscle cone and retrobulbar space, and hematoma and infection of the cavernous sinus. A case of superior orbital fissure syndrome is described.

Effect of Light on the Vegetative Growth of Viola rossii, V. japonica, and V. dissecta var. chaerophylloides Populations (고깔제비꽃, 왜제비꽃, 남산제비꽃 집단의 광조건에 따른 영양생장적 특징)

  • In this study we investigated the relationship between characteristics of vegetative growth of three species of Viola (V. rossii, V. japonica, and V. dissecta var. chaerophylloides) and light in order to explain how these three species could live together under a closed space in a forest. This studywas carried out at five sites in Mudeung-san (Mt.) on 1994, 1995, and 2000 and additional investigations were performed on 2007 and 2010 at the same sites. In order to find the relationship, three main investigations were conducted: counting number of individuals for each investigated year, measuring dry mass and counting developed leaves per plant, and finding presenting patterns of each species with reference to incidence of light. As a result, we found that three species could share the closed space in a forest because of their different adaptive strategies like V. rossii as Kstrategist, V. japonica as r-strategist, and V. dissecta var. chaerophylloides as an intermediator between two species.

Accordingly, basic purpose which are pleasantness, the safe and the convenience the mobile multi-function as well as networking with controlling of temperature, security, health-test, home-entertainment, home-office and consider environment together.

  • The arginylation branch of the N-end rule pathway is a ubiquitin-mediated proteolytic system in which post-translational conjugation of Arg by ATE1-encoded Arg-tRNA-protein transferase to N-terminal Asp, Glu, or oxidized Cys residues generates essential degradation signals. Here, we characterized the ATE1−/− mice and identified the essential role of N-terminal arginylation in neural tube development. ATE1-null mice showed severe intracerebral hemorrhages and cystic space near the neural tubes. Expression of ATE1 was prominent in the developing brain and spinal cord, and this pattern overlapped with the migration path of neural stem cells. The ATE1−/− brain showed defective G-protein signaling. Finally, we observed reduced mitosis in ATE1−/− neuroepithelium and a significantly higher nitric oxide concentration in the ATE1−/− brain. Our results strongly suggest that the crucial role of ATE1 in neural tube development is directly related to proper turn-over of the RGS4 protein, which participate in the oxygen-sensing mechanism in the cells.

  • The Wnt signaling pathway is well known to play major roles in skeletal development and homeostasis. In certain aspects, fracture repair mimics the process of bone embryonic development. Thus, the importance of Wnt signaling in fracture healing has become more apparent in recent years. Here, we summarize recent research progress in the area, which may be conducive to the development of Wnt-based therapeutic strategies for bone repair.

  • Kombucha mutualistic community (KMC) is composed by acetic acid bacteria and yeasts, producing fermented tea with health benefits. As part of the BIOlogy and Mars EXperiment (BIOMEX) project, the effect of Mars-like conditions on the KMC was analyzed. Here, we analyzed metagenome-assembled genomes (MAGs) of the Komagataeibacter, which is a predominant genus in KMC, to understand their roles in the KMC after exposure to Mars-like conditions (outside the International Space Station) based on functional genetic elements. We constructed three MAGs: K. hansenii, K. rhaeticus, and K. oboediens. Our results showed that (i) K. oboediens MAG functionally more complex than K. hansenii, (ii) K. hansenii is a keystone in KMCs with specific functional features to tolerate extreme stress, and (iii) genes related to the PPDK, betaine biosynthesis, polyamines biosynthesis, sulfate-sulfur assimilation pathway as well as type II toxin-antitoxin (TA) system, quorum sensing (QS) system, and cellulose production could play important roles in the resilience of KMC after exposure to Mars-like stress. Our findings show the potential mechanisms through which Komagataeibacter tolerates the extraterrestrial stress and will help to understand minimal microbial composition of KMC for space travelers.

Purification, crystallization and X-ray crystallographic analysis of nicotinamidase Pnc1 from Kluyveromyces lactis

  • Pnc1 converts nicotinamide to nicotinic acid to generate NAD+ through the Preiss-Handler pathway that is one of the NAD+-salvage pathway. By reducing levels of nicotinamide, an inhibitor of the NAD+-dependent histone deacetylase Sir2, yeast Pnc1 contributes gene silencing. In this study, to understand the structural features and molecular mechanism of nicotinamidase Pnc1, we overexpressed, purified, and crystallized the N-terminally His6-tagged Pnc1 protein from Kluyveromyces lactis and obtained X-ray diffraction data at a resolution of 2.2 Å. The crystals of the K. lactis Pnc1 (KlPnc1) belonged to space group P212121 with unit cell parameters a=38.5, b=77.3, c=83.3, and α=β=γ= 90°. There is one molecule in the asymmetric unit.

  • We tested if the treatment of Beauveria bassiana would influence invertebrate communities in litter and soils by multivariate analysis. The PCA (principal components analysis) was used for the analysis. Using the distances between communities in the ordination space, we carried out statistical tests whether any factors would influence structures of the communities. We did not found any significant effects of the Beauveria treatment on invertebrate communities in both litter and soils.
