국내산 두류 품종에 대한 품질 분석 (Qualities Analysis of Domestic Soybean Cultivars)

  • To analyze the quality characteristics of soybean (Glycine max), the approximate analysis, physical properties, mineral and free sugar content of domestic soybean cultivars were analyzed. The moisture content was about 9.15~11.84%. The fat content of Cheonga, Taekwang, and Jinpung cultivars were 17.93~18.37%, which was significantly higher than that of others. Protein content of Cheonga and Seonyu were 36.15~36.70%, significantly higher than that of others. As for the weight and shape of soybeans, the Daewon was the largest allele, and the seed cover ratio was the highest in the Seonyu. In terms of water absorption rate, the Seonyu showed the highest absorption rate at 246.19%, and the Daewon showed the lowest absorption rate. Soaked soybeans had a hardness of 2.80~4.73 kg/㎠, which did not show low hardness in the sample with high moisture absorption. The grinded soybeans showed higher lightness and yellowness values than the raw soybeans, and the redness was decreased. Soybean minerals contained high K, P, Mg, and Ca content. Overall, The Taekwang contained more minerals than others. Stachyose, sucrose, glucose and fructose were detected as free sugars in soybeans. The total free sugar content of Taekwang was the lowest at 3.47%, and the Cheonga (6.80%) was the highest.

Analysis of genome variants in dwarf soybean lines obtained in F6 derived from cross of normal parents (cultivated and wild soybean)

  • Plant height is an important component of plant architecture and significantly affects crop breeding practices and yield. We studied DNA variations derived from F5 recombinant inbred lines (RILs) with 96.8% homozygous genotypes. Here, we report DNA variations between the normal and dwarf members of four lines harvested from a single seed parent in an F6 RIL population derived from a cross between Glycine max var. Peking and Glycine soja IT182936. Whole genome sequencing was carried out, and the DNA variations in the whole genome were compared between the normal and dwarf samples. We found a large number of DNA variations in both the dwarf and semi-dwarf lines, with one single nucleotide polymorphism (SNP) per at least 3.68 kb in the dwarf lines and 1 SNP per 11.13 kb of the whole genome. This value is 2.18 times higher than the expected DNA variation in the F6 population. A total of 186 SNPs and 241 SNPs were discovered in the coding regions of the dwarf lines 1282 and 1303, respectively, and we discovered 33 homogeneous nonsynonymous SNPs that occurred at the same loci in each set of dwarf and normal soybean. Of them, five SNPs were in the same positions between lines 1282 and 1303. Our results provide important information for improving our understanding of the genetics of soybean plant height and crop breeding. These polymorphisms could be useful genetic resources for plant breeders, geneticists, and biologists for future molecular biology and breeding projects.

콩나물 생장 중 사포닌의 변화 (Changes of Saponin during the Cultivation of Soybean Sprout)

  • 은하콩의 조 사포닌은 4.59mg/g이었으며, 콩나물로 발아 생장하면서 사포닌 함량이 증가하여 재배 $5{\sim}6$일째에 $5.30{\sim}5.33mg/g$으로 가장 높은 함량을 보였으며, 6일째 이후에는 사포닌 함량이 감소하였다. 6일 동안 재배하여 자엽, 줄기 및 뿌리의 조 사포닌 함량은 자엽 4.17mg/g에 비해 줄기 7.46mg/g과 뿌리 7.45mg/g으로 거의 2배정도 높은 함량을 보였으며, 뿌리와 줄기는 비슷한 수준이었다. TLC를 이용한 부위별 사포닌 밴드 패턴으로부터 14종이 확인되었다. 줄기와 뿌리 부위는 콩에서 발견되는 여러 가지 사포닌이 함유되어 있으며, 줄기와 뿌리 부위는 Rf 0.5이하에서 비슷한 패턴으로 뚜렷한 밴드를 나타냈다. 콩의 soyasaponin group B 사포닌함량과 조성은 5일 동안 콩나물로 생장하면서 IV는 8.7배, I는 7배, V는 3.3배, II는 2배 그리고 III는 1.4배로 각각 증가하였으며, 특히 IV와 I이 많이 증가하였다. 콩나물 부위별 사포닌 함량은 자엽 부분에서 비교적 적은 양이 고루 함유하였다. 줄기는 자엽에 비해 I은 10.53배, III는 10.49배, V는 8.14배, IV는 5.72배 그리고 II는 1.45배로 더 많았다. 뿌리는 자엽에 비해 V는 9.37배, I는 5.54배 IV는 4.86배 그리고 III는 2.77배 많았지만, II는 자엽보다 2.96배 더 적었다. 콩나물의 줄기와 뿌리가 자라면서 주로 soyasaponin I, IV 및 V가 증가하였다. 이와 같이 콩나물의 사포닌은 성장함에 따라 또는 부위에 따라서 각각 다르게 생합성이 조절되는 것으로 생각된다.
