In 2006, the effect of soil pH adjustment on control of common scab of potato caused by Streptomyces spp. was evaluated in the field. Soil with an original pH 5.7 was treated with sulfur of 42 kg/10a and calcium hydroxide of 81.6 kg/10a and 184.5 kg/10a to adjust pH 5.0, 6.0 and 6.5, respectively. Additionally, the effect of gypsum (522 kg/10a) on soil pH and disease severity was examined. The soil pH of sulfur treatment plot was continuously declined from 5.13 at tuber initiation time to 5.01 at harvest. The disease severity of sulfur treatment plot was relatively low (22.8%) and the marketable yield of that was high (90.5%). By contrast, the soil pHs treated with calcium hydroxide (81.6 kg/10a and 184.5 kg/10a) were increased (pH 6.06 and 6.49, respectively) at harvest. In addition, calcium hydroxide treatment plots showed higher disease severities (51.0% and 61.1%), and lower marketable yields (55.7% and 37.0%). Meanwhile, the soil pH of gypsum treatment plot was not changed until harvest, and the disease severity and the marketable yield were not significantly different from those of control. The effect of crop rotation was also evaluated with four crops such as welsh onion, soybean, corn and Chinese cabbage. These crops were planted in the infested field from 2005 to 2007, and potatoes were planted in the same field in 2008. The disease severities of potatoes produced after rotations with welsh onion, soybean, corn and Chinese cabbage for 3 years were 13.1%, 16.7%, 28.9% and 30.2%, respectively. However, the disease severity of 73.1% was shown in continuous cropping of potato for 3 years. In addition, similar effects were exhibited in the marketable yield. These results demonstrate that the adjustment of soil pH and crop rotation might be very useful tools to control common scab of potato.
A series of green house experiment was conducted to find but the effect of fertilizer application and inoculation of rhizobium on the changes of amide-N, ureide-N and $NH_4-N$ concentration in stem and root exudates of soybean plant growth. The results obtained were summarized as follows ; 1. Five strains of indigenous Rhizobium japonicum-nitrogen fixing activity($C_2H_2$-reducing activity) was more than 6.4 to 20.1 nmole/hr/tube-were identified from 37 soil samples in 22 areas of farmers field throughout country. 2. These identified 5 strains of rhizobium were obtained high nitrate reductase but low ammonium and nitrite oxidase activities. Among 5 strains of rhizobium the Rhizobium japonicum RjK-134 was applied for this green house experiment. 3. Dry matter yield was increased by the combination of inoculation of Rhizobium japonicum RjK-134 with no fertilizer and without nitrogen fertilizer application. However, dry matter yield was decreased with application of N and NPK with inoculation of rhizobium. 4. The concentrations of amide-N and ureide-N were increased in xylem sap than that of root exudate and higher concentration was obtained ar 30 days after planting than flowering stage (45 days after planting). 5. The combination of NPK application with inoculation of Rhrizobium japonicum RjK-134 enhanced the increase of amide-N and ureide-N concentration in xylem sap and root exudate. 6. High ammonium-N concentration in xylem sap and root exudate were obtained in combination with without-fertilizer under no inoculation of rhizobium and N and NPK application with inoculation of rhizobium.

We have studied physiological responses of mung bean sprout to the treatment of elicitors. Chemicals such as salicylic acid and methyl jasmonic acid are not only the intermediates found in plant defense system but also could affect plant secondary metabolism. We found that mild treatment of salicylic acid and acetyl salicylic acid (aspirin) increase isoflavone production dramatically in mung bean sprout which has very low level of isoflavones compared with soybean sprout. The isoflavone content in salicylic acid treated- and acetyl salicylic acid treated-mung bean sprout was about 2.3 and 2.2 times higher than that of control, respectively. However, the increasing patterns of isoflavone in cotyledon and hypocotyl and root were not identical. The major increase among isoflavone fractions in cotyledon was led by the increase in malonylglycitin and malonyldaidzin level. Whereas, the increase in hypocotyl and root was led by malonyldaidzin. Methyl jasmonic acid did not show statistically significant increase in mung bean sprout. With this result, we were able to propose the non-transgenic method, which can control the isoflavone production in germinating mung bean.
To compare various methods for available soil P test, in terms of correlation between crop yield and amounts of available soil P, a pot experiment was conducted with soybean as test plant using four upland soils differing in phosphorus status. The results are summarized as following. 1. There was highly significant correlation between grain yield and amount of P uptake observed at the floral differentiation stage. Highly significant correlations were also observed between grain yield and available soil P of different methods, and between the amount of P uptake and available soil P. The consistency of correlations between soil P and plant parameters was Olsen${\geq}$Lancaster P>Bray No.1 P${\geq}$Bray No.2 P>Truog P. 2. The magnitute of available soil P values of different methods was in the order of Bray No.2>Lancaster P${\geq}$Bray No.1 P>Olsen P>Truog P. 3. The amounts of available P measured by various methods showed significant correlation with the amounts of Al-P, Fe-P and Ca-P. 4. As conclusion, Olsen-P, Lancaster-P, and Bray-P are reliable information for evaluating soil P status. However, Olesen-P seemed to be better than the others.
Laboratory and pot experiment were carried out to find the symbiotic nitrogen fixing activities and the yield of dry matter on the several selected wild soybean plant. 1. Very high acetylene reducing(nitrogen fixing)activities were observed in isolated strains of Rhinchosis volubilies Lour. (924 nmole), Desmodium oldhami Oliver (844 nmole) and Glycine soja s. et z. (271 nmole/test tube/hr) while Rhizobium japonicum was only 0.6 nmole/test tube/hr. 2. On the serological reaction, eight isolated Rhizobium strains were sufficiently different to distinguish from the Rhizobium japonicum. 3. On the pot experiment with Glycine soja s. et z., A. Number of total nodules were higher in NPK+rice straw+Ca+Mo than in NoPK. However, little differences were observed in mumber of effective nodules. B. Yield of dry matter was increased with increasing of nitrogen content in plants. C. Estimated amount of fixed nitrogen in plant by an individual nodule was higher in NoPK than in NPK+rice straw+Ca+Mo.
Three lepidopteran insect pests of the legume pod borer, Maruca vitrata (Lepidoptera: Pyralidae), the soybean pod worm, Matsumuraeses phaseoli (Lepidoptera: Tortricidae) and an Ostrinia spp. (Lepidoptera: Pyralidae) attacking adzuki bean, Vigna angularis, were confirmed as the major insect pests during the reproductive developmental stage of adzuki bean, and M. vitrata existed dominantly. Almost all plants in the adzuki bean field were injured by at least one of the three species, and $15{\sim}60%$ of reproductive organs were injured. Unbloomed inflorescence occurred continuously through the reproductive stage of adzuki bean. While the flower was many at the early stage and its number soon decreased, the pod was few at the early stage, but its number increased soon and maintained at a constant level. The results suggested that the adzuki bean injured by insect pests compensate the flower loss by developing new inflorescence, but the compensation do not cause the development of new pods. While M. vitrata and M. phaseoli were observed in flowers, pods and stem mainly during the first half of reproductive stage of adzuki bean, Ostrinia spp. was observed only in pods and stem during the second half. In addition, while all instars of larvae of M. vitrata were observed, larvae between the third and fifth instars for M. pahseoli and Ostrinia spp. were observed.
Phenylalanine ammonia-lyase (PAL; EC 4, 3, 1, 5) genomic clones were isolated from tomato(Lycopersicon esculentum L.) genomic DNA libraries using tomato PAL5 cDNA sequences as probes. The nucleotide sequences of tPAL1, tPAL4 and tPAL5 were compared. tPAL5 contains an open reading frame encoding a polypeptide of 722 amino acids, interrupted by a 710 bp intron in the codon for the amino acid 139. tPAL1 encodes a polypeptide of 249 amino acids which is much shorter than tPAL5 gene due to a premature stop codon and does not contain an intron. tPAL4 encodes a polypeptide of 357 amino acids, interrupted by a 305 bp intron in the codon for the amino acid 138. Premature stop codons observed in tPAL1 and tPAL4 gene produce a short polypeptide rather than a normal polypeptide (722 aa). tPALl shows 87.2% homology with tPAL4 and 85.3% homology with tPAL5 gene whereas tPAL4 showes 91.4% homology with tPAL5 at nucleotide level. In general, phylogenetic analysis showed that genes isolated from tomato, potato, and sweet potato were belong to the same group and another dicot plants such as parsley, bean, soybean, pea and alfalfa formed another group. PAL genes isolated from rice and yeast showed very low homology with other PAL genes and formed the other group.

AbstractLiving modified organisms (LMO) are one of the most widespread products of modern biotechnology after DNA discovery. Due to the decline of grain self-sufficiency rate and the increase of reliance on LMO imports in Korea, a series of concerns with regard to safety of living modified(LM) crops has been raised. The aim of this study is to establish the detection methods for unintentional release or growing of LMO plants in environmental conditions. To detect LM crop events, general concepts of specific primer design and PCR conditions were provided by the Joint Research Centre (JRC). The certified reference materials of seven LM events (4 soybean, 2 cotton and 1 corn) were obtained from the Institute for Reference Materials and Measurements (IRMM) and the American Oil Chemists' Society (AOCS). Genomic DNA from seven LM events were purified and PCR amplifications were carried out by using individual event-specific primer sets. LM-specific PCR products of all seven events were efficiently amplified by our methods. The results indicate that the established detection method for LMOs is suitable as a scientific tool to monitor whether the crops found in natural environments are LMOs.

To develop a commercial product using the mycoparasitic fungus Simplicillium lamellicola BCP, the scale-up of conidia production from a 5-l jar to a 5,000-l pilot bioreactor, optimization of the freeze-drying of the fermentation broth, and preparation of a wettable powder-type formulation were performed. Then, its disease control efficacy was evaluated against gray mold diseases of tomato and ginseng plants in field conditions. The final conidial yields of S. lamellicola BCP were $3.3{\times}10^9conidia/ml$ for a 5-l jar, $3.5{\times}10^9conidia/ml$ for a 500-l pilot vessel, and $3.1{\times}10^9conidia/ml$ for a 5,000-l pilot bioreactor. The conidial yield in the 5,000-l pilot bioreactor was comparable to that in the 5-l jar and 500-l pilot vessel. On the other hand, the highest conidial viability of 86% was obtained by the freeze-drying method using an additive combination of lactose, trehalose, soybean meal, and glycerin. Using the freeze-dried sample, a wettable powder-type formulation (active ingredient 10%; BCP-WP10) was prepared. A conidial viability of more than 50% was maintained in BCP-WP10 until 22 weeks for storage at $40^{\circ}C$. BCP-WP10 effectively suppressed the development of gray mold disease on tomato with control efficacies of 64.7% and 82.6% at 500- and 250-fold dilutions, respectively. It also reduced the incidence of gray mold on ginseng by 65.6% and 81.3% at 500- and 250-fold dilutions, respectively. The results indicated that the new microbial fungicide BCP-WP10 can be used widely to control gray mold diseases of various crops including tomato and ginseng.
A foliage contact herbicidal substance was separated from ethyl ether fraction in n-hexane extract of Saussurea lappa roots and identified as dehydrocostus lactone [(3aS,6aR,9aR,9bS)-3,6,9-trimethylidene-3a,4,5,6a,7,8,9a,9b-octahydroazuleno[5,4-d]furan-2-one](DHCL). When DHCL at 4,000 ppm was foliage-applied to two grasses and two broadleaf plants, greater than 85% necrotic injury was obtained from large crabgrass, maize and soybean, whereas only about 40% necrotic injury appeared in black nightshade, indicating that DHCL has no gross morphological selectivity, but shows difference in contact response among the plant species tested. Conductivity in incubation medium of the leaf disks treated with DHCL increased as the incubation time continued. Relatively low contact injury in black nightshade as compared with the other three plant species tested was attributed to decrease in absorption of DHCL due to relatively high amount of cuticle. DHCL did not require light in the herbicidal action and there were no inhibitory effects on seed germination and cell elongation. Acetyl-CoA carboxylase activity was inhibited by 30% and 58% at $100\;{\mu}M$ and $1000\;{\mu}M$ DHCL, respectively. These results suggested that the herbicidal action of DHCL was related with inhibition of fatty acid synthesis which in turn caused to weaken cell membrane integrity.
