콩(Glycine max Merrill) 및 녹두(Phaseolus radiatus L.) 종자의 발아일별 ATP 함량변화 (Fluctuation of ATP Content in Soybean and Mungbean Seeds with Germinating Time)

  • ATP contents of soybean(Glycine max Merrill) and mungbean (Phaseolus radiatus L.) seeds being germinated with Hoagland solution at $30^{\circ}C$ for 6 days were determined. In pregerminated seed, ATP contents in soybean and mumgbean were 11.4 and 63.0$\mu$g/g fresh seed respectively. During germination, the highest ATP content of soybean seeds was 550% of initial content on 2nd day and that of mungbean was 480% on 1st day after germination. ATP content in cotyledon of soybean and mungbean were increased up to 4th and 1st day after germination respectively, thereafter both were decreased, but those in the root, including the hypocotyl, of both seeds were continuously increased with germination progress.

  • Recently new soybean saponins with D DMP (2,5-dihydroxy-6-methyl-2,3,- dihydro-4H-pyran-4-one) moiety have been isolated from legumes. The purpose of this study is to characterize ROS scavenging activities of DDMP saponins ($\alpha$g, $\beta$g saponin) isolated from Glycine max (L.) Merrill. The scavenging activity on OH was examined in terms of lipid peroxidation in the rat liver homogenates and the same activity on $O_2$ was also determined in the xanthine-xanthine oxidase system, respectively. Up to 0.25 mg DDMP saponins ($\alpha$g and $\beta$g saponins) did not cause any significant effects on the prevention of lipid peroxidation as compared with the control group. In terms of superoxide scavenging activities, 0.25 and 0.5 mg $\alpha$g saponin inhibits only 2.6% and 5.5% (p<0.05) of the control group, respectively. However, $\alpha$g saponin dose-dependently (p<0.01, r=0.955) inhibits the formation of superoxide radical unto 21.3% of the control group with a maximal dose of 0.5 mg (p<0.01), equivalent to 0.17 units of superoxide dismutase activity.

Plant Regeneration from Embryogenic Suspension Cultures of Soybean (Glycine max L. Merrill)

  • In order to establish efficient plant regeneration from embryogenic suspension cultures of soybean, Glycine max L, we examined the effects of auxin type and concentration, cytokinin type and concentration, and amino acid type and concentration on the growth of embryogenic clumps from induced callus, and the effect of desiccation of mature somatic embryos obtained from these clumps on the frequency of somatic embryo germination. Embryogenic callus was induced from the edge of the cotyledons cultured on MS medium containing 6% sucrose, 40 mg/L 2,4-D, 0.2% gelrite and pH 5.7. The growth of embryogenic clumps was best in early staged, embryogenic callus that was placed in suspension culture of MS medium containing 5 mg/L 2,4-D and 0.5 mg/L asparagine. Single somatic embryos were isolated from the clumps and plated on the same medium for maturation. When the mature single somatic embryos were desiccated for 96 h, somatic embryo germination came up to approximately 90%. The plantlets germinated after embryos desiccation for 2 weeks were transfered to MS medium containing 3% sucrose,0.2% gelrite and pH 5.7.

Comparison of 12 Isoflavone Profiles of Soybean (Glycine max (L.) Merrill) Seed Sprouts from Three Different Countries

  • The levels of 12 isoflavones were measured in soybean (Glycine max (L.) Merrill) sprouts of 68 genetic varieties from three countries (China, Japan, and Korea). The isoflavone profile differences were analyzed using data mining methods. A principal component analysis (PCA) revealed that the CSRV021 variety was separated from the others by the first two principal components. This variety appears to be most suited for functional food production due to its high isoflavone levels. Partial least squares discriminant analysis (PLS-DA) and orthogonal projections to latent structures discriminant analysis (OPLS-DA) showed that there are meaningful isoflavone compositional differences in samples that have different countries of origin. Hierarchical clustering analysis (HCA) of these phytochemicals resulted in clusters derived from closely related biochemical pathways. These results indicate the usefulness of metabolite profiling combined with chemometrics as a tool for assessing the quality of foods and identifying metabolic links in biological systems.

  • 본 연구는 유전자재조합(genetically modified, GM) 콩(Glycine max L. MERRILL)의 검출에 빠르고 감도가 높은 기술을 모색하기 위하여 수행되었다. 기존의 PCR (polymerase chain reaction) 방법과 새로운 방법으로 시도된 DHPLC(denaturing high performance liquid chromatography) 방법으로 유전자재조합 콩을 확인하였다. DHPLC 방법은 기존의 PCR방법에서의 전기영동이 방법 대신 컬럼을 이용하여 분석할 수 있다. 때문에 분석시간도 60분에서 20분으로 단축시킬 수 있다. 검출한계도 PCR방법에서는 15 $ng/{\mu}L$$10^{-4}$ 농도까지 검출할 수 있었고, DHPLC 방법으로는 $15ng/{\mu}L$$10^{-5}$ 농도까지 검출 할 수 있었다. 따라서 유전자재 조합 콩 검출법으로 DHPLC 분석법이 PCR 분석법보다 빠르고 정확한 효과적인 방법임을 확인하였다.

  • A protein isolate was prepared from black soybean (Glycine max L. Merrill) that possessed higher antioxidant activity than ordinary white soy protein isolates. The isolate was partially hydrolyzed by alcalase to reduce the allergenicity of black soybean. Alcalase remarkably reduced the molecular mass of the major soybean allergens that have molecular weights of 53, 38, and 24 kDa. Hydrolytic breakdown occurred more effectively in Gly m Bd 30K than in Gly m Bd 60K or Gly m Bd 28K. Alcalase hydrolysis increased the solubility and hydrophobicity of the black soybean protein isolate. The foaming activity and stability of black soybean proteins were highly increased by the partial hydrolysis.
