Enhancement of Phenanthrene Solubilization and Biodegradation with Biosurfactants Produced by Rhodococcus erythropolis (Rhodococcus erythropolis를 통하여 얻어진 생계면활성제에 의한 phenanthrene의 용해도 및 생분해능력 향상)

  • Effects of the biosurfactant produced by Rhodococcus erythropolis on the solubilization and biodegradation of phenanthrene were investigated. Based on surface tension measurements, the average critical micelle concentration of the biosurfactant was estimated to be about 16mg TOC/L. The apparent solubility of phenanthrene increased linearly with the addition of biosurfactants above the CMC, and the concentration of solubilized phenanthrene was 38.9mg/L in 322mg TOC/L biosurfactant solution. The weight-solubilization ratio of biosurfactants for phenanthrene was approximately 118.8mg/g, this value was over 5 times greater than that of sodium dodecyl sulfate. Using a known phenanthrene degrader, batch phenanthrene biodegradation experiments were conducted with and without biosurfactants in liquid culture. The rate and extent of phenanthrene mineralization by the phenanthrene degrader with biosurfactants were much greater than those without biosurfactants. The greater phenanthrene mineralization observed in the presence of biosurfactants is attributed to the increased phenanthrene concentration in the aqueous culture due to the partitioning of the compound to biosurfactant micelles. The biosurfactant did not exhibit any toxic effect on mineralization of glucose by the phenanthrene-degrader.

  • [ $1m^3$ ] solid hydrate contains up to $200m^3$ of natural gas, depending on pressure and temperature. Such large volume of natural gas hydrate can be utilized to store and transport large quantity of natural gas in a stable condition. So, in the present investigation, experiments carried out for the formation of natural gas hydrate governed by pressure, temperature, and gas compositions, etc.. The results show that the equilibrium pressure of structure II natural gas hydrate (is approximately 65% lower and the solubility is approximately three times higher than structure I methane hydrate). Also, the subcooling conditions of the structure I and II must be above 9K and 11K in order to form hydrate rapidly regardless of gas components, but the pressure increase is more advantageous than the temperature decrease in order to increase the gas consumption. And utilizing nozzles for spraying water in the form of droplets into the natural gas dramatically reduces the hydrate formation time and increases its solubility at the same time.

Enhanced Bioremediation of Phenanthrene Using Biosurfactant (생물계면활성제를 이용한 Phenanthrene의 생물학적 처리)

  • This study was carried out 1) to investigate the pH effect on solubilization of phenanthrene by biosurfactant in aqueous system and 2) to evaluate the pH effect on the biodegradation rate of phenanthrene in the presence and the absence of the biosurfactant by phenanthrene degraders. Tween 80, which is a chemically synthesized surfactant, showed greater solubilizing capacity than rhamnolipid. The solubilization capacity can be expressed as a MSR(molar solubilization ratio=moles of organic compounds solubilized per mole of surfactant). The calculated MSR of Tween 80 and rhamnolipid were 0.1449 and 0.0425 respectively. The kinetic study of phenanthrene solubilization by rhamnolipid showed that solubilization mechanism could reach equilibrium within 24 hours. Addition of 240 ppm rhamnolipid solution, which concentration is 4.3 times of Critical Micelle Concentration(CMC), caused 9 times solubility enhancement compared to water solubility. The highest solubilities were detected around a pH range of 4.5-5.5. Changes in apparent solubility with the changes in pH are possibly related to the fact that the rhamnolipid, an anionic surfactant, can form different structures depending on the pH. Two biodegradation experiments were performed in the absence and the presence of rhamnolipid, with the cell growth investigated using a spread plate method. The specific growth rates at pH 6 and 7 were higher than at the other pH, and the HPLC analysis data, for the total phenanthrene loss, confirmed the trends in the $\mu$(specific growth rate) values. In presence of rhamnolipid, maximum $\mu$ values shifted from around pH 5 which showed maximum enhancement of solubility in the abiotic experiment, compared to the $\mu$ values obtained without the biosurfactant. In this study, the increase in the observed specific grow rate(1.44 times) was not as high as the increase in solubilization(5 times). This was supported by the fact all the solubilized phenanthrene is not bioavailable to microorganisms.

Absorption of Methyl Mercaptan into Aqueous Diethanolamine Solutions (Diethanolamine 수용액에서의 Methyl Mercaptan의 흡수)

  • The kinetics of the reaction of methyl mercaptan into aqueous diethanolamine were studied over a range in temperature ($20^\~60{\circ}C$) and amine concentrations (0-40 $wt\%$) using a wetted-sphere absorber. The physicochemical properties needed to interpret the data are the solubility and diffusivity of methyl mercaptan in the aqueous diethanolamine solution. The density and the viscosity were obtained and correlated in the experimental range. The Wilke Chang equation was applied to estimate the diffusion coefficient. The enhancement factor was found to be high temperature is below than low temperature. It means the absorption rate with chemical reaction is lower than the physical absorption rate.

  • The productivity of natural flavors in alfalfa suspension cultures is low partially due to their low solubility. Thus, by formation inclusion complex of cyclodextrin(CD) with flavor compounds enhancement of the productivity of flavor was attempted in this research. we observed that CD formed inclusion complex with flavor compounds by GC/MS analysis. Also, there was indication that CD played a role as an elicitor in alfalfa suspension cultures.

  • Bulk hetero junction (BHJ) polymer solar cells (PSCs) are one of the most promising fields as alternative energy source. Especially, the development of new p-type conjugated polymer is one of the main issues to get core technology. In this study, a series of varied ratio of 3,6-carbazole in poly[9-(heptadecan-9-yl)-9H-carbazole-2,7-diyl-alt-(5,6-bis-(octyloxy)-4,7-di(thiophen-2-yl)benzo-[1,2,5]-thia-diazole)-5,5-diyl] were designed and synthesized. These polymers have good solubility and film formability than PCDTBT which is well known promising material. Investigation of the photovoltaic properties of these new polymers indicated that polymer with 2% of 3,6-carbazole provided higher PCE (3.8% to 4.9%) with enhanced JSC, FF, VOC. We found origin of this improvement using several methods, one of which is reduced bimolecular recombination in polymer.

Enhancement of the Water-resistance and Physical Properties of Sodium Alginate Film

  • To improve water-resistance and physical properties of sodium alginate film, effects of sodium alginate and plasticizer concentrations, divalent cation types and concentrations, and immersion time of films into divalent cation solutions on sodium alginate films were evaluated, based on elongation strength (ES), elongation rate (E), water vapor permeability (WVP), and water solubility (WS). Film made from 1.5% sodium alginate solution (w/w) had low WVP and WS, which are optimal characteristics for application of film preparation. Addition of plasticizer increased E and WS. Less than 2% $CaCl_2$ addition and 15min immersion time reduced WVP, WS, and E significantly (p<0.05). Sodium alginate films treated with $CuCl_2$, and $ZnCl_2$ solutions had lower WVP and WS, whereas $MgCl_2$ had no influence on improving water resistance of films.

  • In order to synthesize polymer particle containing inorganic material, styrene and nbutylmetacrylate were copolymerized with alumina by dispersion polymerization. The ratio in weight of styrene to n-butyl methacrylate was 3:1. Poly(N-vinyl pyrrolidon) and 2,2'-azobis(isobutyronitrile) were added as stabilizer and as initiator, respectively. The change of particle size was investigated with concentration of initiator, the type of medium, the mixed solubility parameter (${\delta}_{mix}$) of medium, and coupling agent. The enhancement in concentration of initiator resulted in slight increase of particle size. The increase of polarizability in medium also yielded the increase of particle size. In case of changing the ratio of isopropanol to distilled water, we could find relationship of $[{\delta}_{mix}]^{-4.01}\;{\propto}$ particle size and $[{\delta}_{mix}]^{-0.83}\;{\propto}$ particle size distribution(PSD). The type and the concentration of coupling agent showed no effect on the particle size and PSD.

Dietary ε-Polylysine Decreased Serum and Liver Lipid Contents by Enhancing Fecal Lipid Excretion Irrespective of Increased Hepatic Fatty Acid Biosynthesis-Related Enzymes Activities in Rats

  • ${\varepsilon}$-Polylysine (EPL) is used as a natural preservative in food. However, few studies have been conducted to assess the beneficial functions of dietary EPL. The purpose of this study was to elucidate the mechanism underlying the inhibition of neutral and acidic sterol absorption and hepatic enzyme activity-related fatty acid biosynthesis following EPL intake. EPL digest prepared using an in vitro digestion model had lower lipase activity and micellar lipid solubility and higher bile acid binding capacity than casein digest. Male Wistar rats were fed an AIN-93G diet containing 1% (wt/wt) EPL or L-lysine. After 4 weeks of feeding these diets, the marked decrease in serum and liver triacylglycerol contents by the EPL diet was partly attributed to increased fecal fatty acid excretion. The activities of hepatic acetyl-coenzyme A carboxylase and glucose-6-phosphate dehydrogenase, which are key enzymes of fatty acid biosynthesis, were enhanced in rats fed EPL diet. The increased fatty acid biosynthesis activity due to dietary EPL may be prevented by the enhancement of fecal fatty acid excretion. The hypocholesterolemic effect of EPL was mediated by increased fecal neutral and acidic sterol excretions due to the EPL digest suppressing micellar lipid solubility and high bile acid binding capacity. These results show that dietary EPL has beneficial effects that could help prevent lifestyle-related diseases such as hyperlipidemia and atherosclerosis.

Improvement of Solubility of Atorvastatin Calcium Using Self-Microemulsion Drug Delivery System(SMEDDS) (자가미세유화를 통한 아토르바스타틴 칼슘의 난용성 개선)

  • SMEDDS is mixture of oils, surfactants, and cosurfactants, which are emulsified in aqueous media under conditions of gentle agitation and digestive motility that would be encountered in the gastro-intestinal(GI) tract. The main purpose of this work is to prepare self-microemulsifying drug delivery system(SMEDDS) for oral bioavailability enhancement of a poorly water soluble drug, atorvastatin calcium. Solubility of atorvastatin calcium was determined in various vehicles. Pseudo-ternary phase diagrams were constructed to identity the efficient self-emulsification region and particle size distributions of the resultant micro emulsions were determined using a laser diffraction sizer. Optimized formulations for in vitro dissolution and bioavailability assessment were $Capryol^{(R)}$ 90(50%), Tetraglycol(16%), and $Cremophor^{(R)}$ EL(32%). The release rate of atorvastatin from SMEDDS was significantly higher than the conventional tablet ($Lipitor^{(R)}$), 2-fold. Our studies illustrated the potential use of SMEDDS for the delivery of hydrophobic compounds, such as atorvastatin calcium by the oral route.
