Objective: This study aimed to evaluate the protective effects of Socheongryong-tang (SCRT) on elastase-induced lung injury. Materials and Methods: The extract of SCRT was treated to A549 cells and elastase-induced COPD mice model. Then, various parameters such as cell-based cytoprotective activity and histopathological findings were analyzed. Results: SCRT showed a protective effect on elastase-induced cytotoxicity in A549 cells. This effect was correlated with analysis for caspase 3 levels, collagen and elastin contents, and gene expression of TNF-${\alpha}$ and IL-$1{\beta}$ in A549 cells. SCRT treatment also revealed the protective effect on elastase-induced COPD mice model. This effect was evidenced via histopathological findings including immunofluoresence stains against elastin, collagen, and caspase 3, and protein level of Cdc2, cyclin B1, and phospho-Erk1/2 in lung tissue. Conclusion: These data suggest that SCRT has pharmaceutical properties on COPD. This study provides scientific evidence for the efficacy of SCRT for clinical application to patients with COPD.
Objectives : Socheongryong-Tang (小靑龍湯, SCRT) has been widely used to treat respiratory disease. In this study, we investigated the protective effects of SCRT on hydrogen peroxide-induced hepatotoxicity. Methods : In the mouse primary liver cells, SCRT was pretreated for 1 h, and 1 mM $H_2O_2$ was treated to mouse primary liver cells. Cell viability was analyzed by using 3- 4,5-Dimethylthiazol-2-yl-2,5-diphenyltetrazolium bromide (MTT) assay. Also, the activity of AST, ALT and LDH were measured for the evaluation the protective effect of SCRT on $H_2O_2$-induced hepatotoxicity. Intracellular ROS level was analyzed by FACS. Results : SCRT pretreatment decreased $H_2O_2$-induced hepatotoxicity and intracellular ROS production. Pretreatment of SCRT significantly reduced the cytotoxic effect induced by $H_2O_2$, associated with reducing DNA fragmentation and AST, ALT, LDH activities. Conclusions : These results suggest that SCRT has protective effect against $H_2O_2$-induced hepatotoxicity.

A patients with quadriplegia and a history of pancreatectomy was administered Socheongryong-tang to reduce sputum. The patient's sputum decreased, but a sudden increase in blood sugar was observed. Ephedra is known to stimulate the sympathetic nerves to increase the blood pressure and blood sugar, but in healthy people, changes in blood sugar are not significant due to corrections by hormone regulation. However, this case suggests that ephedra can cause significant blood sugar increases in patients who undergo pancreatectomy or who have diabetes mellitus. Even though ephedra is highlighted for its usefulness in sputum reduction and in weight control, it should be carefully prescribed to patients with diabetes.

Socheongryong-tang (SCRT) was one of the major traditional herbal medicines wildly used in the treatment of respiratory disease. SCRT is being commercially produced in the form of mix extracts powder and soft dry extract by different extract methods in the Korean Herbal Pharmacopeia (KHP). In this study, the contents of marker components and biological activities of the SCRT mix extract powder were compared with those of the SCRT decoction. To analyze the marker components of SCRT, nine marker from eight herbal preparations were chosen. And the method using high performance liquid chromatography (HPLC) with diode-array detector method was established for the simultaneous analysis. Method validation was accomplished by linearity, precision test, and recovery test. The contents of nine marker components in this extract was ascertained by ratio. The biological activities were examined the effect of SCRT on anti-oxidation and pro-inflammation mediated by LPS-stimulation. We confirmed that both of SCRT mix extrct powder and decoction have the similar contents on total polyphenol and flavonoid and inhibited the secretion of nitric oxide (NO), $IL-1{\beta}$, IL-6, tumor necrosis factor $(TNF)-{\alpha}$ and the expression of iNOS, COX-2, $IL-1{\beta}$, IL-6, $TNF-{\alpha}$. These results suggest that SCRT mix extract powder and decoction have a significant correlation.

Background : Allergic rhinitis is an inflammation of the nasal mucosa which is characterized by sneezing, coughing itchy nose, mouth and throat, congestion and/or nasal discharge. Object : We have studied effects of the Okbyeongpung-san plus Socheongryong-tang on the change of the amounts of IL-4, II-5, $IFN-{\gamma}$ and total IgE in rats OVA-induced allergic rhinitis. Method : The 15 rats were divided into three groups ; normal group, control group, and sample group. To induce allergic rhinitis in control group and sample group , rats were sensitized intraperitoneally with 0.1% ovalbumin(OVA) solution 3 times at intervals of 1 week. Then intranasal sensitization was performed by diffusing 0.1% ovalbumin(OVA) solution 3 times at intervals of 2 days. After that time, rats in the sample group were oral administration treated by Okbyeongpung-san plus Socheongryong-tang 28 days. We observed the change of the amounts of IL-4, II-5, $IFN-{\gamma}$ and total IgE in rats in each grout. Result : 1. In Total IgE study, the treated group was proved significant inhibitory effect(p<0.05) 2. In Interleukin-4(IL-4) study, the treated group was proved significant inhibitory effect(p<0.001> 3. In IL-5 study, the treated group was proved significant inhibitory effect(p<0.001> 4. In Interferone-${\gamma}(IFN-{\gamma})$ study, the treated group was proved significant inhibitory effect(p<0.005) Conclusion : According to the above results, it is considered that the Okbyeongpung-san flus Socheongyong-tang has inhibitory effects on the allergic rhinitis of rats.

Objectives : The purpose of this study is to assess the effectiveness and safety of Socheongryong-tang(SCRT) for allergic rhinitis(AR). Methods : We searched randomized controlled trials(RCTs) that used SCRT for AR in 8 databases(PubMed, Cochrane Library, CNKI, CiNii Articles, OASIS, NDSL, KISS, KMbase) from their inception until August 2019. The primary outcome was effective rate and scores evaluating the improvement of AR symptoms. The secondary outcome was quality of life scale, adverse events, recurrence rate, and laboratory indicators. Two researchers assessed the risk of bias in the included trials through the Cochrane Risk of Bias tool independently. The study synthesized outcomes using RevMan 5.3. Results : Eighteen RCTs involving 1686 participants were included in this review. The risk of bias was unclear for the majority of the included studies. Meta-analysis of 12 RCTs showed that there was no statistically significant difference between the SCRT group and usual care group in the effective rate(RR 1.18, 95% CI(0.98, 1.41), p=0.09, I2=46%). Meta-analysis of 5 RCTs showed that the combination treatment group of SCRT and usual care was significantly higher than the usual care group in the effective rate(RR 1.24, 95% CI(1.12, 1.38), p<0.0001, I2=0%). The SCRT group was more effective in improving nasal symptoms and quality of life than the placebo group according to one RCT. Mild adverse events such as dry mouth were identified in 5 RCTs, but no serious adverse events were reported. Conclusion : This review showed that in terms of the effective rate for AR, there was no statistically significant difference between SCRT and usual care and the combination treatment of SCRT and usual care was more effective than usual care. There were no serious adverse events. However, it is difficult to make a definite conclusion because of few included studies and heterogeneity between studies, and the quality of included studies was mostly insufficient. Further well-designed randomized controlled trials are needed.

Objective : Allergic rhinitis (AR) is an allergic inflammation of the nasal airways. Socheongryong-Tang traditionally has been used in Korea for treatment of allergy diseases. In the present study, we investigated whether SCRT suppresses the progression of AR in vivo and in vitro. Method : In this study, we investigated the effect of oral administration of SCRT on the Ovalbumin(OVA)-induced allergic rhinitis mouse models. Additionally, to find a possible explanation for the anti-inflammation effects of SCRT, we evaluated the inflammatory cytokine levels in LPS-stimulated mouse inflammation cell (RAW264.7) and mouse splenocyte. Results : The finding of this study demonstrated that SCRT reduced mast cells and inflammation cells infiltration in OVA-induced nasal cavity. Additionally, SCRT inhibited the production of inflammatory cytokines in splenocyte. However SCRT don't inhibited the production of inflammatory cytokines in RAW264.7 cells. Conclusion : Taken together, our results showed that oral administration of SCRT beneficial effects in allergic rhinitis, suggesting that SCRT might be a useful candidate for the treatment of allergic rhinitis.
Objectives : Asthma is a chronic inflammatory disorder in which the airway smooth muscle undergoes exaggerated constriction and is abnormally responsive to external stimuli and clinically manifests dyspnea, cough, and wheezing. Recently in Korea, the number of asthma patients has tended to increase as air pollution increases and new allergens appear. This study aimed to identify the therapeutic effects of Sochongryong-tang, which has been used on asthmatic patients in Oriental Medicine for a long time. Methods : We studied 10 asthmatic patients who had visited the Division of Respiratory System, Department of Internal Medicine of Kyunghee Oriental Medical Center from February 1, 2001 to June 30, 2001, which examined the patients by the pulmonary function test (PFT) and Quality of Life Questionnaire for adult Korean Asthmatics (QLQAKA) before and after taking Sochongryong-tang for two weeks. The data was analyzed using paired t-test. Results : After treatment with Sochongryong-tang for two weeks, FEV 1.0 showed a significant increase of 14.2%, FVC a significant increase of 9.3% and PEFR a significant increase of 16.2% in results of patients examined asthmatic by PFT. In the results of QLQAKA, the mean of scores increased significantly over 0.5 points in total score, symptom domain, emotion domain, and environmental domain. Conclusions : This study shows that Sochongryong-tang has the effect of improvements of pulmonary function and quality of life in asthmatic patients.
In the present study, the author intended to investigate whether three oriental medical prescriptions named Jeongcheonhwadam-tang(JHT), Haengso-tang(HST), Socheungryong-tang(SCRT) significantly affect mucin release from cultured hamster tracheal surface epithelial (HTSE) cells. The results were as follows: (1) JHT significantly inhibited mucin release from cultured HTSE cells, without significant cytotoxicity : (2) HST significantly inhibited mucin release from cultured HTSE cells, without significant cytotoxicity : (3) SCRT significantly inhibited mucin release from cultured HTSE cells, without significant cytotoxicity : (4) JHT, HST chiefly inhibited the 'mucin' release and did not significantly affect the release of the other releasable glycoproteins with less molecular weight than mucin. These results suggest that the three herbal prescriptions specifically inhibit the release of mucin: (5) JHT significantly inhibited the expression levels of MUC SAC mRNA. This result suggests that JHT affects the synthesis of mucin at gene level in cultured HTSE cells. All agents showed no significant cytotoxicity. In view of these results, further investigation of the effects of JHT and HST are likely to be instrumental in yielding novel agents from oriental medical prescriptions which have inhibitory effects or expectorative effects on airway mucus secretion.
Objectives : The objective of present study was to investigate the vasorelaxant effects of 10 traditional Korean Herbal Prescriptions (KHP) on isolated rat thoracic aorta precontracted with potassium chloride (KCl). Methods : An electric extractor was used to extract KHP in distilled water for 3h. Rat aorta rings were isolated and were precontracted using KCl in organ chambers containing 10 ml Krebs Henseleit (KH) buffer. KHP extracts were added in increasing concentrations (10-1000 ㎍/㎖) to investigate vasorelaxant effects. The vasorelaxant responses induced by KHP were expressed as a percentage in response to contraction generated by KCl. Results : Among the 10 KHP, Gamisoyo-san, Galgeun-tang, Gyeji-tang, Gwakhyangjeonggi-san, Daeyoung-jeon, and Socheongryong-tang showed significant vasorelaxant effect at high concentration. In contrast, Gyejibokryeong-hwan constricted more the aorta rings precontracted by KCl. And Gumiganghwal-tang, Guibi-tang, Saengmaek-san showed no significant effect. Also, rat aorta rings treated with Gyejibokryeong-hwan or Gyeji-tang after pre-relaxation by amlodipine did not cause any significant change. Conclusion : Thus, these results provide the experimental evidence as useful herbal prescriptions for the treatment of hypertension and suggest guidelines in conjunction with other western drugs, including amlodipine.
