Effect of Lecithin on Dermal Safety of Nanoemulsion Prepared from Hydrogenated Lecithin and Silicone Oil

  • In this study, a hydrogenated lecithin-containing nanoemulsion was prepared from hydrogenated lecithin and silicone oil. Tween-60 and liquid paraffin, widely known emulsifiers, were used as standard substances, and high shear was produced by utilizing a high shear homogenizer and microfluidizer. The properties of the nanoemulsion prepared with hydrogenated lecithin were evaluated by measuring interfacial tension, dynamic interfacial tension, droplet size, zeta-potential, friction force, skin surface hygrometery, and dermal safety. The interfacial tension of lecinol S10/silicone oil was lower than that of lecinol S10/liquid paraffin. The nanoemulsion prepared from hydrogenated lecithin shows lower zeta-potential, skin surface hygrometery, and friction force compared with a general emulsion. The silicone nanoemulsion prepared from hydrogenated lecithin showed a zero value in the patch test and thus exhibits high dermal safety.

  • Allergy is conditions when a hypersensitivity reaction happens with a certain element, called as an allergen, which is commonly not reactive to ordinary individuals. Allergic diseases involve various organs or systems in the body. The purpose of allergy tests is to make a diagnosis of allergic diseases and to identify the affecting allergens. In vivo tests, more relevant in clinical situation, include skin test, patch test and provocation test. In in vitro tests, there are specific IgE test, histamine releasing assay, and lymphocyte activation test, safer and more objective than in vivo tests. In the view point of clinical practice, skin test, provocation test, total IgE test and specific IgE test were reviewed in depth.

  • Nanoscale observation of attachment systems of animals has revealed various exquisite multiscale architectures for essential functions such as gecko's locomotion, beetles' wing fixation, octopuses' sucking and crawling. In particular, the hierarchical 3-dimensional hexanonal nano-architectures in the tree frog's adhesion is known to have the capability of the enhancement of adhesion forces on the wet or rough surfaces due to the conformal contacts against rough surfaces and water-drainable micro channels. Here, we report that tree frog-inspired patches using unique artificial 3-dimensional hexagonal structures can be exploited to form reversibly enhanced adhesion against various highly curved and rough surfaces in dry and wet condition. To investigate the adhesion effect of micro-channels, we changed the arrangement of microstructure and spacing gaps between micro-channels. In addition, we introduced the 3-dimensional hexagonal hierarchical architectures to artificial patches to enhance to conformal contacts on the various rough surfaces such as skin and organs. Using the robust adhesion properties, we demonstrated the self-drainable and comfortable skin-attachable devices which can measure EKG (electrokardiogramme) for in-vitro diagnostics. As a result, bio-inspired programmable nano-architectures can be applied in versatile devices such as, medical patches, skin-attachable electronics etc., which would shed light on future smart, directional and reversible adhesion systems.

  • In vitro cell culture system has been proposed as a promising alternative model to in vivo skin irritation test. These studies were performed to screen the cytotoxicity effects of surfactants using normal human skin fibroblasts. Cell membrane integrity assessed by the leakage of lactate dehydrogenase (LDH) and mitochondrial integrity by MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromides reduction test were affected in a dose dependent manner. The irritation potential of surfactants to human skin patch test, and the changes of capillary permeability by rabbit intradermal safety test were assessed as in vivo methods. Our results suggest that LDH leakage assay and MTT reduction test using cultured human fibroblasts could be predictive for the irritancy of various surfactants in human, and LDH assay is superior correlated with in vivo test (r=0.886) to MTT test with in vivotest (r=0.757).

A study to effectiveness the usefulness of skin patches containing chlorogenic acid and garcinia (클로로겐산이 함유된 피부 패치의 유용성 평가를 위한 연구)

  • The purpose of study was to evaluate the usefulness a skin patch containing chlorogenic acid to the skin to identify changes in body fat and weight. 16 volunteers, consisting of 8 experimental groups and 8 control groups, participated in the study, and the study period was conducted for 4 weeks from Aug 03, 2020 to Aug 31, 2020. As a result of the inter-group effect test, there was no significant difference in body fat (F=4.38, p<.055), but it can be said that the experimental group had a positive effect because the difference in posterior score was larger than that of the control group. In the change of body weight, it was found that there was a significant difference in weight loss in the experimental group (F=9.43, p<.008). It will be meaningful as a preliminary study applying patches to studies related to obesity, and we hope that the results of the study will be used as basic data for research on patch materials in the future.

Effects of Poly-N-acetyl Glucosamine(pGlcNAc) Patch on Wound Healing in db/db Mouse (Poly-N-acetyl-glucosamine이 당뇨병 쥐에서 창상치료에 미치는 영향)

  • Purpose: Poly-N-acetyl glucosamine(PGlcNAc) nanofiber-based materials, produced by a marine microalga, have been characterized as effective hemostatic and angiogenic agents. The similarity between PGlcNAc patch and the natural extracellular matrix allows it to support new healthy tissue growth in an injured area and to encourage fluid absorption. In this study, we hypothesized that a poly-N-acetyl glucosamine fiber patch(PGlcNAc patch) may enhance wound healing in the db/db mouse. Methods: PGlcNAc patches were applied on one square centimeter, full-thickness, skin wounds in the db/db mouse model. Wounds(n=15 per group) were dressed with a PGlcNAc nanofiber patch for 1 hour(1 h), 24 hours(24 h) or left untreated(NT). After the application time, patches were removed and wounds were allowed to heal spontaneously. The rate of wound closure was evaluated by digital analysis of unclosed wound area in course of time. At day 10, wounds(n=7 per group) were harvested and quantified with immunohistochemical markers of proliferation(Ki-67) and vascularization (platelet endothelial cell adhesion molecule, PECAM-1). Results: Wounds dressed with PGlcNAc patches for 1 hour closed faster than control wounds, reaching 90% closure in 16.6 days, nine days faster than untreated wounds. Granulation tissue showed higher levels of proliferation and vascularization following 1 h treatment than the 24 h and NT groups. In addition to its hemostatic properties, the PGlcNAc material also appears to accelerate wound closure in healing-impaired genetically diabetic mice. Conclusion: This material, with its combination of hemostatic and wound healing properties, has the potential to be effective agent for the treatment of complicated wounds.

  • Both immune reaction and hypersensitivity reaction are occurred by the same mechanism, the antigen and antibody reaction. The favorable result of this reaction towards a host is called clinically an immune reaction and the opposite results is called an hypersensitivity reaction. Type IV hypersensitivity reaction is a delayed type which is related to the cellular immune reaction and a contact hypersensitivity is included in this type. Various dental materials such as metal (mercury, nickel, chrome, cobalt), resin and eugenol are etiologic substances. Patch test kit is composed of test substance with a controlled concentration which respond only to a susceptible patient and an aluminum chamber, and etiologic substances for hypersensitivity can be easily and comfortably found just by applying the kit to the patient's skin. In this case report, the patch test was performed to a patients with oral lichen planus and the allergen, restorative material was found. After removal of the matching restoration from the patient's mouth, the symptom was improved.

  • Carcinous pain is severe and continuous so difficult to alleviate. Western drugs give rise to reaction such as anorexia, constipation, vomiting, general weakness and dyspnea, And they cause tolerence. External treatment is a way to put a patch or rub a liquid on the skin. The patch and liquid are made from herb medicine. In china, they have a good effect of alleviating carcinous pain to make use of external treatment. Usually they use the medicine with effect of reprecussion (消腫), removing obstruction(散結), activating blood flow and removing blood stasis (活血化瘀) such like Borneloum(?片). Bufonis Venenum (蟾?), Aconiti ciliane Tubber(草烏), Arisaematis Rhizoma(南星), Asari herba cum Radice (細辛) etc. We can get the alleviating effect more quickly and prevent reaction through external treatment.

Proliposomal Clenbuterol Patch for Transdermal Delivery (프로리포솜을 이용한 클렌부테롤의 경피흡수 제제화)

  • Proliposomal patch of clenbuterol, ${\beta}_2-agonist$ bronchodilator, was prepared and its feasibility as a novel transdermal drug delivery system was examined. Proliposomal granules containing clenbuterol was prepared by a standard method using sorbitol and lecithin with (Rx 2) or without cholesterol (Rx 1). The porous structure of sorbitol in the proliposomes was maintained allowing tree flowability of the granules. Following contact with water, the granules were converted probably to liposomes almost completely within several minutes. It indicates that proliposomes may be hydrated, when they are applied on the skin under occlusive condition in vivo, by the sweat to form liposomes. Clenbuterol release from Rx 1 and Rx 2 proliposomes to pH 7.4 isotonic phospate buffer (PBS) across cellulose membrane (mol. wt. cut-off of 12000-14000) was retarded significantly compared with that from the mixture of clenbuterol powder and blank proliposomes. Interestingly, proliposomes prepared with lecithin and cholesterol (i.e., Rx 2 proliposomes) showed much more retarded release of clenbuterol than proliposomes prepared only with lecithin (i.e.. Rx 1 proliposomes), indicating that clenbuterol release from proliposomes can be controlled by the addition of cholesterol to the proliposomes. Proliposomal patches were prepared using PVC film as an occlusive backing sheet, two sides adhesive tape (urethane, 1.45 mm thickness) as a reservoir for proliposome granules and Millipore MF-membrane (0.45 mm pore size) as a drug release-controlling membrane. Rx 1 or Rx 2 proliposomes containing 4.6 mg of clenbuterol were loaded into the reservoir of the patch. Clenbuterol release from the patches to pH 7.4 PBS was determined using USP paddle (50 rpm)-over-disc release method. Clenbuterol release from the proliposomal patches was much more retarded even than from a matrix type clenbuterol patch (Boehringer Ingelheim ltd). Being consistent with clenbuterol release from the proliposomal granules, the release from the patches was highly dependent on the presence of cholesterol in the proliposomes : Patches containing Rx 2 proliposomes showed several fold slower drug release than patches containing Rx 1 proliposomes. When the patch containing Rx 1 proliposomes was applied on to the back of a hair-removed rat, clenbuterol concentration in the rat blood was maintained during 6-72 hrs. Transdermal absorption of clenbuterol from the patch was accelerated when the patch was prehydrated with 50 ml of pH 7.4 PBS before topical application. Above results indicate that sustained transdermal delivery of clenbuterol is feasible using proliposomal patches if the cholesterol content and pore size of the release rate-controlling membrane of patches, for example, are appropriately controlled.

Total Phenolic Compound, Total Flavonoid Compound And Anti-Inflammatory Inhibitory Effects of Psidium Guajava Leaf Extract (구아바 잎 추출물의 총 폴리페놀과 플라보노이드 함량 및 항 염증 억제 효과)

  • It was intended to check a possibility of activity of Psidium guajava leaf extract as anti-oxidant and anti-inflammatory material. Total polyphenol and total flavonoid content of Psidium guajava leaf extract was checked. Cream having inhibitory effect on inflammation through toxicity and NO production inhibition in RAW 264.7 cell was manufactured, and skin safety was evaluated. It was confirmen that total polyphenol and flavonoid content of Psidium guajava leaf extract was 126.4 mg/g and 223.17 mg/g respectively, which was high content. According to the results of checking toxicity through cell viability in RAW 264.7 cell, cytotoxicity was not shown. And NO production indicating inflammatory disease was inhibited concentration-dependently. According to the results of carrying out single patch test after manufacturing the cream containing the Psidium guajava leaf extract, skin irritation did not occur for 24 h to put patch on skin or for 24 h after removing the patch. Putting these results together, it was verified that there was possibility of application as raw materials for cosmetics, which would have anti-oxidant activity owing to the high polyphenol and flavonoid content of Psidium guajava leaf extract and anti-inflammatory material through NO production inhibition.
