화장품에 배합되는 원료 중에 피부 모공을 막아 여드름의 초기단계인 면포를 유발함으로써 염증성 여드름으로 악화시키는 사례가 보고되고 있다. 본 연구는 '여드름 피부에 사용이 적합한' 면포 비유발성 화장품의 객관적인 평가 기준을 마련하고자 하였다. 외국 임상기관에서 실시하고 있는 non-comedogenic test를 조사하고 등 상부 반복 폐쇄 첩포를 통해 면포 유발 평가방법을 확립하고자 연구를 수행하였다. 또한 안면부 사용성 시험을 동일 피험자에게 추가로 진행하였다. 등 상부 반복 폐쇄 첩포를 통해 면포를 채취하여 분석한 결과 시험시료로 사용한 보습제와 자외선 차단제가 면포를 유발하지 않음을 확인할 수 있었고, 동일한 시험제품을 안면부에 사용하여 얻은 면포 유발 결과와 Global acne grading system (GAGS)의 여드름 육안평가 결과는 상관관계가 없었다. 또한 사진 판독을 통해 분석하는 면포와 모낭의 구별을 용이하게 하기 위해 면포 채취 표본에 Oil red O staining을 실시하였다. 염색을 한 경우의 표본이 염색을 하지 않은 표본과 비교하여 결과 간 높은 일치도를 확인하였다. 본 연구에서는 Oil red O staining을 통해 객관성과 신뢰성을 증진시킨 새로운 버전의 화장품 면포 비유발성 평가법을 확립하였다.
본 연구에서는 항산화 물질로 잘 알려진 quercetin과 그 배당체인 rutin을 함유하는 poly(${\varepsilon}$-caprolactone)-b-poly(ethylene glycol) 미셀을 제조하여, 활성물질(quercetin, rutin)의 in vitro 피부 흡수 증진에 관한 연구를 수행하였다. 입자크기는 PCL-b-PEG 고분자의 농도가 증가함에 따라 미셀의 초기 입자 크기가 증가하는 경향을 보였다. 고분자 미셀의 표면 전위(Zeta potential)는 비교적 일정함을 확인하였다. 제조한 고분자 미셀의 피부 흡수력을 평가하기 위하여, 용액 상태의 활성물질을 미셀의 대조군으로 하여 Franz cell을 이용한 투과실험을 진행한 결과 용액 상태보다 미셀에서 더 높게 나타났음을 확인하였다. 또한 화장품 소재로서의 안전성 평가를 위한 인체 피부 일차자극 실험(patch test) 결과 어떠한 피부 자극도 관찰되지 않았다.
Consumers have recently preferred to purchase extensive UV intercepting products, which are waterproof and free from side effects on skin. Testing Cytoroxicity in SR method, cell survivial ratio of UV-B interceptors decreased above 0.08W/V%, and so did that of UV-A interceptors above 0.06W/V%. Also, Patch-test of inorganic UV interceptors resulted in no skin irritation even below 10.0 and 11.25. UV interceptors in the sunlight showed yellowish discoloration in 5 to 14 days. In absorption curves, UV-B was most suitable for Octyl methoxycinnamate and UV-A for Butyl methoxy dibenzoylmethane. Fro this reason, Nylonpoly UVA/UVB the material of OMC and BMDM coated with Nylon & polyethylene, was used as the organic UV interceptor. And zinc oxide and titanium dioxide was used as inorganic UV ibterceptors. The appropriate mixture ratio of ZnO and TiO2 was 6 to 4.6% of ZnO, 4% of TiO2 and 5% of Nylonpoly UVA/UVB were all combined with our sunscreen cream. The SPF value of in-vivo applied to a guinea pig was 34.9 and that of in-vivo was 38.5. Cyclomerhicone and dimerthicone were used in water-in-Silicone system. Ceryl diverhicone and sorbitan sesquioleate were used as emulsifiers and MgSO4, 7H2O, Mg-stearate/Mg-Al-stearate copolymer as emulsification stabilizers. In practical application, each SPF duration of O/W type and W/S type containing sunscreen cream of the same content showed that W/S type of sunscreen cream was 5 times as durable as the other. This product is fit for using in swimming, climbing or skiing. This research is to minimize skin trouble used by UV interceptors and to make one with proper softness, skin safety and UV intercepting efficiency.
Transdermal drug delivery(TDS) offers many important advantages. For instance, it is easy and painless, it protects the active compound from gastric enzymes, and it avoids the hepatic first-pass effect. Also, it is simple to terminate the therapy if any adverse or undesired effect occurs. But skin is a natural barrier, and only a few drugs can penetrate the skin easily and in sufficient quantities to be effective. Therefore, in recent years, numerous studies have been conducted in the area of penetration enhancement. The most commonly used transdermal system is the skin patch using various types of technologies. Compared with other method of dosage, it is possible to use for a long term. It is also possible to stop the drug dosage are stopped if the drug dosage lead to side effect. Polysaccharide, such as xanthan gum and algin were selected as base materials of TDS. Also, these polymers were characterized in terms of enhancers and drug contents. Among these polysaccharide, the permeation rate of Paroxetine such as lipophilic drug was the fastest in xanthan gum matrix in vitro. We used glycerin, PEG400 and PEG800 as enhancers. Since dermis has more water content(hydration) than the stratum corneum, skin permeation rate at steady state was highly influenced when PEG400 was more effective for lipophilic drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate.

두피 유래 유산균 Leuconostoc mesenteroides 추출물을 함유한 화장품을 인체 피부에 첩포하여 자극 유무를 검증하고 기초화장품의 소재로써 피부 안전성을 규명하고자 하였다. L. mesenteroides 추출물 1%와 PEG-60 Hydrogenated Castor Oil 0.6%를 함유한 토너와 L. mesenteroides 추출물 3%, Glyceryl Stearate 0.7%를 함유한 로션을 제조하였다. 제조한 토너는 32명, 로션은 33명의 연구대상자들의 등 부위에 각각 24시간 첩포하여 자극지수를 산출한 결과, 토너는 0.016점, 로션은 0점이 도출되어 모두 무자극으로 나타나 피부에 안전함을 확인하였다. 이와 같은 결과와 기존 선행연구에서 보고된 L. mesenteroides의 항산화, 항알레르기, 항염능 등을 미루어 보아 이는 저농도에서 기초화장품의 소재로 피부에 안전하고 기타 화장품에 적용 시 피부 무자극 소재로 연구 가치가 있을 것으로 사료된다.
인체의 두피 각질에서 유산균 Leuconostoc mesenteroides을 분리하여 배양하고 이를 함유한 가용화 에센스를 피부에 첩포한 후 자극 유무 검증을 통해 화장품의 소재로 활용 가능성을 판단하고자 하였다. 1% 농도로 희석한 L. mesenteroides 배양액과 이를 1% 함유한 에센스를 제조하여 L. mesenteroides 배양액은 31명, 에센스는 32명의 피시험자들의 등 부위에 24시간 첩포하고 이를 제거한 후 30분, 24시간, 48시간 후의 상태를 피부과 전문의가 육안으로 판독하여 자극 지수를 산출하였다. 이 결과 L. mesenteroides 배양액은 0.011점, 에센스는 0점이 도출되어 모두 무자극으로 피부에 안전한 것으로 나타났다. 본 연구에서의 일차자극시험 결과와 선행 연구에서 보고된 항알레르기, 항산화, 항염 활성능 등을 미루어 보아 저 농도의 L. mesenteroides 배양액은 피부에 안전하여 향후 기초화장품에 적용 시 피부자극이 없는 소재로 활발한 연구가 가능할 것으로 기대된다.
화장품은 일반인이 장기간 연용하는 제품으로 인체 피부에 대한 안전성이 매우 중요하다. 화장품에 의한 접촉피부염은 자극성 물질이 피부에 접촉 후 침투하여 유발하는 염증반응으로 활성화된 면역세포에서 염증을 매개하는 다양한 인자를 분비함으로써 시작된다. 본 연구에서는 국산 생약 추출물을 이용하여 RAW264.7 대식 세포에서 염증 관련 인자에 대한 영향을 통해 항염증 활성을 스크리닝 하였다. 51종의 국산 생약 추출물 중 측백, 측백엽(초), 향부자, 형개, 동과자, 산약, 산약(초), 상지, 송절, 택사의 에탄올 추출물 10종이 lipopolysaccharide(LPS)에 의해 유도된 세포독성을 감소시킴과 동시에 염증 관련 인자인 Nitric oxide (NO), interleukin (IL)-1${\beta}$, IL-6, tumor necrosis factor (TNF)-${\alpha}$의 생성을 억제하는 것을 확인하였다. 또한, 이들 생약 추출물은 인체피부에서도 자극을 유발하지 않음을 인체 첩포시험을 통해 검증하였다. 따라서, 항염증 활성이 확인된 10종 국산 생약 추출물은 피부 자극을 예방할 수 있는 화장품 소재로서의 활용이 가능할 것으로 사료된다.
For proper wound healing, dermal contraction and remodeling are critical; during the natural healing process, differentiated fibroblasts called "myofibroblasts" typically undertake these functions. For severe wounds, however, a critical mass of dermal matrix and fibroblasts are lost, making self-regeneration impossible. To overcome this impairment, synthetic wound patches with embedded functional cells can be used to promote healing. In this study, we developed a polydioxanone (PDO)-based cell-embedded sheet on which dermal fibroblasts were cultured and induced for differentiation into myofibroblasts, whereby the following combinatorial physicochemical stimuli were also applied: aligned topology, electric field (EF), and growth factor. The results show that both the aligned topology and EF synergistically enhanced the expression of alpha smooth-muscle actin (${\alpha}$-SMA), a key myofibroblast marker. Our proof-of-concept (POC) experiments demonstrated the potential applicability of a myofibroblast-embedded PDO sheet as a wound patch.
아토피 피부염은 만성염증 피부질환으로서 유전적 요소, 환경적 요소, 면역반응의이상, 피부장벽단백질의 기능 이상에 의하여 발생한다. 본 연구의 목적은 피부장벽단백질의 발현을 측정할 수 있는 ELISA 키트 개발에 있다. AriNo와 D-Squame 패치를 이용하여 비침습적으로 피부에서 단백질을 얻을 수 가 있었고, AriNO가 좀 더 많은 단백질을 획득하였다. 0.1% Triton X-100용액이 다른 용해용액인 0.1 M Tris-HCL, 5 mM KOH, 0.1% Tween-20보다 높은 단백질 수율을 나타냈다. 피부장벽 단백질 측정을 위한 ELISA 키트 개발을 위하여 분자생물학적인 방법을 이용하여 필라그린과 인보루크린의 재조합단백질을 생산하였고, 이에 대한 단일클론항체를 면역학적인 방법을 이용하여 만들었다. 아토피 피부염 환자의 피부에서는 필리그린의 발현이 유의하게 줄어들었고, 인보루크린은 정상인과 아토피 피부염 환자에서 차이를 나타내지 않았다. 본 연구의 결과를 통하여 아토피 피부염에서 피부장벽단백질의 중요성을 규명하였고, 향후 아토피 피부염의 진단키트를 개발하는데, 유용할 것이다.
